1Novel ACTG1 mutation causing autosomal dominant non-syndromic hearing impairment in a Chinese family显示文摘The γ-actin (ACTG1) gene is a cytoplasmic nonmuscle actin gene, which encodes a major cytoskeletal protein in the sensory hair cells of the cochlea. Mutations in ACTG1 were found to cause autosomal dominant, progressive, sensorineural hearing loss linked to the DFNA 20/26 locus on chromosome 17q25.3 in European and American families, respectively. In this study, a novel missense mutation (c.364A>G; p.I122V) co-segregated with the affected individuals in the family and did not exist in the unaffected family members and 150 unrelated normal controls. The alteration of residue Ile122 was predicted to damage its interaction with actin-binding proteins, which may cause disruption of hair cell organization and function. 2Host cell apoptosis induced by infection with duck swollen head hemorrhagic disease virus显示文摘This study aimed to examine the host cell apoptosis in the tissues of Peking ducks infected with duck swollen head hemorrhagic disease virus(DSHDV).The dynamic changes associated with apoptosis occurring in the internal tissues were evaluated at different time points postinoculation(PI) by performing hematoxylin and eosin(HE) staining,followed by light microscopy,terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL) assay,and transmission electron microscopy(TEM).The results showed that DSHDV infection could induce apoptosis in host cells,including those of the bursa of Fabricius(BF),thymus,spleen,liver,intestinal tract,kidney,and esophagus.The apoptotic index(AI) values increased with time from 2 to 72 h PI,and the highest values were recorded at 72 h PI.Further,cell death due to classic necrosis was observed in the dying or deceased ducks after 72 h PI.In conclusion,host cell apoptosis can be induced by DSHDV and may play an important role in the pathogenesis of duck viral swollen head hemorrhagic disease(DVSHD).Chuanfeng Li Anchun Cheng Mingshu Wang Chanjuan Shen Na Zhang Yi Zhou Dekang Zhu Qihui Luo Renyong Jia Xiaoyue Chen 2009Progress in Natural Science:Materials International2009,19,1:3

4Design of All-Fused-Ring Electron Acceptors with High Thermal,Chemical,and Photochemical Stability for Organic Photovoltaics显示文摘High-performance donor-acceptor electron acceptors containing 2-(3-oxo-2,3-dihydro-1H-inden-1-ylidene)malononitrile(INCN)-type terminals are labile toward photooxidation and basic conditions,and new molecular designs toward electron acceptors that can achieve both high power conversion efficiencies and high stability are urgently needed.By replacing the central benzene ring in the classical ladder-type n-type semiconductor,2,2′-(indeno[1,2-b]fluorene-6,12-diylidene)dimalononitrile,with the electron-rich 4,4,9,9-tetrahexyl-4,9-dihydro-s-indaceno[1,2-b:5,6-b′]dithiophene,we report herein the design of 2,2′-(7,7,15,15-tetrahexyl-7,15-dihydro-sindaceno[1,2-b:5,6-b′]diindeno[1,2-d]thiophene-2,10(2H)-diylidene)dimalononitrile(ITYM),a new type of all-fused-ring electron acceptor(AFRA).A threestep reaction including a key Pd-catalyzed double C-H activation/intramolecular cyclization is established for the efficient synthesis of such type of electron acceptors.ITYM is confirmed by singlecrystal X-ray analysis,which shows a planar nonacyclic structure with strongπ-πstacking.Compared with the classical carbon-bridged INCN-type acceptors,ITYM exhibits extraordinary stability with very promising performance.The AFRA concept opens a new avenue toward high-efficiency and-stability organic photovoltaics(OPVs).Xiaozhang Zhu Songjun Liu Qihui Yue Wuyue Liu Shaoming Sun Shengjie Xu 2021CCS Chemistry2021,3,6:1
5Missense mutations in CSX/NKX_(2.5)are associated with atrial septal defects显示文摘Objective: To study the gene mutations of homeobox transcription factor ( CSX /NKX2.5) associated with a Chinese familywith secundum atrial septal defect ( ASD) . Methods: Polymerase chain reaction and DNA sequencing were used to check allthe members in the family with ASD, and single strand conformation polymorphism analysis ( SSCP) was used to check 126normal control people for detecting the mutations of CSX /NKX2.5 gene. Results: Three mutations, G270A( Glu32Lys) , G378A( Glu68Lys) andG390A ( Glu72Lys) were identified in CSX /NKX2.5 gene of ASD patients. However, the other members in thefamily with ASD and the control did not have such gene mutations. 7Evidence that Natural Selection is the Primary Cause of the Guanine-cytosine Content Variation in Rice Genes显示文摘Cereal genes are classified into two distinct classes according to the guanine-cytosine(GC)content at the third codonsites(GC_3).Natural selection and mutation bias have been proposed to affect the GC content.However,there has beencontroversy about the cause of GC variation.Here,we characterized the GC content of 1092 paralogs and other single-copygenes in the duplicated chromosomal regions of the rice genome(ssp.indica)and classified the paralogs into GC_3-richand GC_3-poor groups.By referring to out-group sequences from Arabidopsis and maize,we confirmed that the averagesynonymous substitution rate of the GC_3-rich genes is significantly lower than that of the GC_3-poor genes.Furthermore,we explored the other possible factors corresponding to the GC variation including the length of coding sequences,thenumber of exons in each gene,the number of genes in each family,the location of genes on chromosomes and the proteinfunctions.Consequently,we propose that natural selection rather than mutation bias was the primary cause of the GCvariation.Xiaoli Shi Xiyin Wang Zhe Li Qihui Zhu Ji Yang Song Ge Jingchu Luo 2007Journal of Integrative Plant Biology2007,49,9:0
8Blockchain-Empowered Dynamic Spectrum Management for Space-Air-Ground Integrated Network显示文摘Space-air-ground integrated network is capable of providing seamless and ubiquitous services to cater for the increasing wireless communication demands of emerging applications. However, how to efficiently manage the heterogeneous resources and protect the privacy of connected devices is a very challenging issue, especially under the highly dynamic network topology and multiple trustless network operators. In this paper, we investigate blockchain-empowered dynamic spectrum management by reaping the advantages of blockchain and software defined network(SDN), where operators are incentive to share their resources in a common resourced pool. We first propose a blockchain enabled spectrum management framework for space-air-ground integrated network, with inter-slice spectrum sharing and intra-slice spectrum allocation. Specifically, the inter-slice spectrum sharing is realized through a consortium blockchain formed by the upper-tier SDN controllers, and then a graph coloring based channel assignment algorithm is proposed to manage the intra-slice spectrum assignment. A bilateral confirmation protocol and a consensus mechanism are also proposed for the consortium blockchain. 10Dynamic Spectrum Anti-Jamming with Distributed Learning and Transfer Learning显示文摘Physical-layer security issues in wireless systems have attracted great attention.In this paper,we investigate the spectrum anti-jamming(AJ)problem for data transmissions between devices.Considering fast-changing physical-layer jamming attacks in the time/frequency domain,frequency resources have to be configured for devices in advance with unknown jamming patterns(i.e.the time-frequency distribution of the jamming signals)to avoid jamming signals emitted by malicious devices.This process can be formulated as a Markov decision process and solved by reinforcement learning(RL).Unfortunately,stateof-the-art RL methods may put pressure on the system which has limited computing resources.As a result,we propose a novel RL,by integrating the asynchronous advantage actor-critic(A3C)approach with the kernel method to learn a flexible frequency pre-configuration policy.Moreover,in the presence of time-varying jamming patterns,the traditional AJ strategy can not adapt to the dynamic interference strategy.To handle this issue,we design a kernelbased feature transfer learning method to adjust the structure of the policy function online.Simulation results reveal that our proposed approach can significantly outperform various baselines,in terms of the average normalized throughput and the convergence speed of policy learning.Xinyu Zhu Yang Huang Delong Liu Qihui Wu Xiaohu Ge Yuan Liu 2023China Communications2023,20,12:0
11Expression of estrogen receptor alpha,nerve growth factor,interleukin-2,and androgen receptor in the cerebellum of ovariectomized rats following soybean isoflavone treatment显示文摘BACKGROUND: Studies have shown that estrogen receptor alpha (ERα), nerve growth factor(NGF), interleukin-2 (IL-2), and androgen receptor (AR) expression in the cerebellum decreaseswhen estrogen levels decrease in vivo. Soybean isoflavone, a type of non-steroid estrogen withsimilar molecular structure and function to estradiol, exhibits estrogen-like characteristics.OBJECTIVE: To investigate the effects of various doses of soybean isoflavone on expression ofERα, NGF, IL-2, and AR in the cerebellum of ovariectomized rat, and to determine whether there is adose-dependent effect.DESIGN, TIME AND SETTING: Controlled trial at the cellular and molecular level. The study wasperformed at the Experimental Animal Engineering Center, College of Veterinary Medicine, SichuanAgricultural University from July 2006 to May 2008.MATERIALS: Soybean isoflavone, comprised of daidzin, genistein and isoflavone, was provided byTaiyuan Yuantai Biochemical Industry, China. The ERα, NGF, IL-2, and AR in situ hybridization kit,rabbit anti-rat ERα, NGF, IL-2, and AR monoclonal antibodies, and SABC kit were purchased fromWuhan Boster Biological Technology, China.METHODS: A total of 50 female, Sprague Dawley rats, aged 3 months, were randomly assigned to5 groups, with 10 animals in each group. With the exception of the sham-operation group (abdominalcavity opening alone), all rats underwent bilateral ovariectomy. At 14 days after surgery, rats in thehigh-, middle-, and low-dose soybean isoflavone groups were subcutaneously injected with 1.5, 1.0,and 0.5 mg/kg soybean isoflavone, respectively, every 2 days for 6 consecutive weeks. Rats in thesham-operation and ovariectomized groups were subcutaneously injected with absolute alcohol(0.5 mL/kg).MAIN OUTCOME MEASURES: Expression levels and distribution of ERα, NGF, IL-2, and AR in thecerebellum were detected by immunohistochemistry and in situ hybridization.RESULTS: Compared with the sham-operation group, immunoreactive products and hybridizationsignals of ERα, NGF, IL-2, and AR were significantly decreased in the cerebellar cortex and nuclei ofovariectomized rats (P< 0.05 or P< 0.01), but increased following soybean isoflavone treatment. Inparticular, levels of the high-dose soybean isoflavone group were almost restored to levels of thesham-operation group (P > 0.05). The immunoreactive products were primarily located in thecytoplasm and neurites, and rarely in the cell membrane and nuclei. However, the hybridizationsignals were predominantly located in the nuclei, but rarely in the cytoplasm, cell membrane, orneurites.CONCLUSION: Soybean isofiavone upregulated ERα, NGF, IL-2, and AR protein and geneexpression in a dose-dependent manner, and played an important role in sustaining and protectingstructure and function of cerebellar neurons. 15Mako:A Graph-based Pattern Growth Approach to Detect Complex Structural Variants显示文摘Complex structural variants(CSVs) are genomic alterations that have more than two breakpoints and are considered as the simultaneous occurrence of simple structural variants.However,detecting the compounded mutational signals of CSVs is challenging through a commonly used model-match strategy.As a result,there has been limited progress for CSV discovery compared with simple structural variants.Here,we systematically analyzed the multi-breakpoint connection feature of CSVs,and proposed Mako,utilizing a bottom-up guided model-free strategy,to detect CSVs from paired-end short-read sequencing.Specifically,we implemented a graph-based pattern growth approach,where the graph depicts potential breakpoint connections,and pattern growth enables CSV detection without pre-defined models.Comprehensive evaluations on both simulated and real datasets revealed that Mako outperformed other algorithms.Notably,validation rates of CSVs on real data based on experimental and computational validations as well as manual inspections are around 70%,where the medians of experimental and computational breakpoint shift are 13 bp and 26 bp,respectively.Moreover,the Mako CSV subgraph effectively characterized the breakpoint connections of a CSV event and uncovered a total of 15 CSV types,including two novel types of adjacent segment swap and tandem dispersed duplication.Further analysis of these CSVs also revealed the impact of sequence homology on the formation of CSVs.Mako is publicly available at http://gffzz188fe103f8f1460ascfw06nubko5v6bfp.ffgz.tsg.suse.edu.cn/xjtu-omics/Mako.Jiadong Lin Xiaofei Yang Walter Kosters Tun Xu Yanyan Jia Songbo Wang Qihui Zhu Mallory Ryan Li Guo Chengsheng Zhang The Human Genome Structural Variation Consortium Charles Lee Scott E.Devine Evan E.Eichler Kai Ye 2022Genomics, Proteomics & Bioinformatics2022,20,1:0
16JAX-CNV:A Whole-genome Sequencing-based Algorithm for Copy Number Detection at Clinical Grade Level显示文摘We aimed to develop a whole-genome sequencing(WGS)-based copy number variant(CNV)calling algorithm with the potential of replacing chromosomal microarray assay(CMA)for clinical diagnosis.JAX-CNV is thus developed for CNV detection from WGS data.The performance of this CNV calling algorithm was evaluated in a blinded manner on 31 samples and compared to the 112 CNVs reported by clinically validated CMAs for these 31 samples.The result showed that JAX-CNV recalled 100%of these CNVs.Besides,JAX-CNV identified an average of 30 CNVs per individual,representing an approximately seven-fold increase compared to calls of clinically validated CMAs.Experimental validation of 24 randomly selected CNVs showed one false positive,i.e.,a false discovery rate(FDR)of 4.17%.A robustness test on lowercoverage data revealed a 100%sensitivity for CNVs larger than 300 kb(the current threshold for College of American Pathologists)down to 10×coverage.For CNVs larger than 50 kb,sensitivities were 100%for coverages deeper than 20×,97%for 15×,and 95%for 10×.We developed a WGS-based CNV pipeline,including this newly developed CNV caller JAX-CNV,and found it capable of detecting CMA-reported CNVs at a sensitivity of 100%with about a FDR of 4%.We propose that JAX-CNV could be further examined in a multi-institutional study to justify the transition of first-tier genetic testing from CMAs to WGS.JAX-CNV is available at http://gffzz188fe103f8f1460ascfw06nubko5v6bfp.ffgz.tsg.suse.edu.cn/TheJacksonLaboratory/JAX-CNV.Wan-Ping Lee Qihui Zhu Xiaofei Yang Silvia Liu Eliza Cerveira Mallory Ryan Adam Mil-Homens Lauren Bellfy Kai Ye Charles Lee Chengsheng Zhang 2022Genomics, Proteomics & Bioinformatics2022,20,6:0

18Energy band alignment of 2D/3D MoS_(2)/4H-SiC heterostructure modulated by multiple interfacial interactions显示文摘The interfacial properties of MoS_(2)/4H-SiC heterostructures were studied by combining first-principles calculations and X-ray photoelectron spectroscopy.Experimental(theoretical)valence band offsets(VBOs)increase from 1.49(1.46)to 2.19(2.36)eV with increasing MoS_(2) monolayer(1L)up to 4 layers(4L).A strong interlayer interaction was revealed at 1L MoS_(2)/SiC interface.Fermi level pinning and totally surface passivation were realized for 4H-SiC(0001)surface.About 0.96e per unit cell transferring forms an electric field from SiC to MoS_(2).Then,1L MoS_(2)/SiC interface exhibits type I band alignment with the asymmetric conduction band offset(CBO)and VBO.For 2L and 4L MoS_(2)/SiC,Fermi level was just pinning at the lower MoS_(2)1L.The interaction keeps weak vdW interaction between upper and lower MoS_(2) layers.They exhibit the type II band alignments and the enlarged CBOs and VBOs,which is attributed to weak vdW interaction and strong interlayer orbital coupling in the multilayer MoS_(2).High efficiency of charge separation will emerge due to the asymmetric band alignment and built-in electric field for all the MoS_(2)/SiC interfaces.The multiple interfacial interactions provide a new modulated perspective for the next-generation electronics and optoelectronics based on the 2D/3D semiconductors heterojunctions.Huili Zhu Zifan Hong Changjie Zhou Qihui Wu Tongchang Zheng Lan Yang Shuqiong Lan Weifeng Yang 2023Frontiers of physics2023,18,1:0
