| 2 | High-quality genome assembly of 'Cuiguan' pear (Pyrus pyrifolia) as a reference genome for identifying regulatory genes and epigenetic modifications responsible for bud dormancy显示文摘Dormancy-associated MADS-box(DAM)genes serve as crucial regulators of the endodormancy cycle in rosaceous plants.Although pear DAM genes have been identified previously,the lack of a high-quality reference genome and techniques to study gene function have prevented accurate genome-wide analysis and functional verification of such genes.Additionally,the contribution of other genes to the regulation of endodormancy release remains poorly understood.In this study,a high-quality genome assembly for'Cuiguan'pear(Pyrus pyrifolia),which is a leading cultivar with a low chilling requirement cultivated in China,was constructed using PacBio and Hi-C technologies.Using this genome sequence,we revealed that pear DAM genes were tandemly clustered on Chr8 and Chr15 and were differentially expressed in the buds between'Cuiguan'and the high-chilling-requirement cultivar'Suli'during the dormancy cycle.Using a virus-induced gene silencing system,we determined the repressive effects of DAM genes on bud break.Several novel genes potentially involved in the regulation of endodormancy release were identified by RNA sequencing and H3K4me3 chromatin immunoprecipitation sequencing analyses of‘Suli'buds during artificial chilling using the new reference genome.Our findings enrich the knowledge of the regulatory mechanism underlying endodormancy release and chilling requirements and provide a foundation for the practical regulation of dormancy release in fruit trees as an adaptation to climate change. | Yuhao Gao Qinsong Yang Xinhui Yan Xinyue Wu Feng Yang Jianzhao Li Jia Wei Junbei Ni Mudassar Ahmad Songling Bai Yuanwen Teng | 2021 | Horticulture Research2021,8,1: | 5 |
| 3 | Characterization of a disease susceptibility locus for exploring an efficient way to improve rice resistance against bacterial blight显示文摘Bacterial blight caused by Xanthomonas oryzae pv.oryzae(Xoo) is the most harmful bacterial disease of rice worldwide.Previously,we characterized major disease resistance(MR) gene xa25,which confers race-specific resistance to Xoo strain PXO339.The xa25 is a recessive allele of the SWEET13 locus,but SWEET13's interaction with PXO339 and how efficiently using this locus for rice breeding still need to be defined.Here we show that the SWEET 13 allele from rice Zhenshan 97 is a susceptibility gene to PXO339.Using this allele's promoter to regulate xa25 resulted in disease,suggesting that the promoter is a key determinant in SWEET13 caused disease in Zhanshan 97 after PXO339 infection.PXO339 transcriptionally induces SWEET13 to cause disease.Partial suppressing SWEET13 expression leads to a high level of resistance to PXO339.Thus,the transcriptionally suppressed SWEET13 functions as xa25 in resistance to PXO339.Hybrid rice is widely grown in many countries.However,recessive MR genes have not been efficiently used for disease resistance breeding in hybrid rice production for both parents of the hybrid have to carry the same recessive gene.However,the suppressed SWEET 13 functions dominantly,which will have advantage to improve the resistance of hybrid rice to xa25-incomptible Xoo. | Qi Cheng Weihua Mao Wenya Xie Qinsong Liu Jianbo Cao Meng Yuan Qinglu Zhang Xianghua Li Shiping Wang | 2017 | Science China(Life Sciences)2017,60,3: | 4 |

| 17 | Electroactive and antibacterial wound dressings based on Ti_(3)C_(2)T_(x)MXene/poly(ε-caprolactone)/gelatin coaxial electrospun nanofibrous membranes显示文摘Endogenous electric fields(EFs)are capable of regulating the behaviors of skin cells in wound healing.However,majority of current dressings are primarily engaged in the passive repair of defective tissue,as they lack the ability to actively respond to physiological electrical signals.In this work,a series of nanofibrous membranes(NFMs)were fabricated by coaxial electrospinning,combining the good mechanical properties of poly(ε-caprolactone)(PCL),the bioactivity of gelatin and the electroactivity of Ti_(3)C_(2)T_(x)MXene,as electroactive and antibacterial dressings for cutaneous wound healing.The obtained NFMs exhibited suitable mechanical properties and hydrophilicity,excellent electroactivity,antibacterial activity,and biocompatibility.Especially,Ti_(3)C_(2)T_(x)MXene/PCL/gelatin-6(MPG-6,6 wt.%of Ti_(3)C_(2)T_(x)MXene in sheath spinning liquids)showed the optimal conductivity and antibacterial activity.Excitingly,this scaffold significantly promoted the adhesion,proliferation,and migration of NIH 3T3 cells under the electrical stimulation(ES).The in vivo evaluation in a full-thickness wounds defect model demonstrated that the MPG-6 films significantly accelerated wound closure,increased granulation tissue formation,increased collagen deposition,and promoted wound vascularization.In summary,the versatile scaffold is expected to be an ideal candidate as wound dressings due to its ability to promote the transmission of physiological electrical signals and thus improved the therapeutic outcomes of wound regeneration. | Shixin Xu Chen Du Miaomiao Zhang Ruoying Wang Wei Feng Chengwei Wang Qinsong Liu Wen Zhao | 2023 | Nano Research2023,16,7: | 0 |
