2Identification of a Newly Isolated Getah Virus in the China-Laos Border,China显示文摘In this study, we isolated a virus strain(YN12031) from specimens of Armigeres subalbatus collected in the China-Laos border. BHK-21 cells infected with YN12031 exhibited an evident cytopathic effect(CPE) 32 h post-infection. The virus particles were spherical, 70 nm in diameter, and enveloped; they also featured surface fibers. Molecular genetic analysis revealed that YN12031 was closely related to alpha viruses such as Chikungunya virus and Sindbis virus, and located in the same clade as MM2021, the prototype of Getahvirus(GETV) isolated in Malaysia in 1955. Phylogenetic analysis of the E2 and capsid genes further revealed that YN12031 was located in the same clade as the Russian isolate LEIV/16275/Mag. Analysis of the homology of nucleotides and amino acids in the coding area and E2 gene demonstrated that the YN12031 isolated from the China-Laos border(tropical region) was related closest to the LEIV/16275/Mag isolate obtained in Russia(North frigid zone area) among other isolates studied. These results suggest that GETV can adapt to different geographical environments to propagate and evolve. 3Long noncoding RNAs in neurodevelopment and Parkinson’s disease显示文摘Long noncoding RNAs(lnc RNAs) are RNA molecules comprising more than 200 nucleotides, which are not translated into proteins. Many studies have shown that lnc RNAs are involved in regulating a variety of biological processes, including immune, cancer, stress, development and differentiation at the transcriptional, epigenetic or post-transcriptional levels. Here, we review the role of lnc RNAs in the process of neurodevelopment, neural differentiation, synaptic function, and pathogenesis of Parkinson’s disease(PD). These pathomechanisms include protein misfolding and aggregation, disordered protein degradation, mitochondrial dysfunction, oxidative stress, autophagy, apoptosis, and neuroinflammation. 6Zika virus infection induces RNAi-mediated antiviral immunity in human neural progenitors and brain organoids显示文摘The re-emergence of Zika virus(ZIKV)in the Western Hemisphere has resulted in global public health crisis since 2015.ZIKV preferentially infects and targets human neural progenitor cells(hNPCs)and causes fetal microcephaly upon maternal infection.hNPCs not only play critical roles during fetal brain development,but also persist in adult brain throughout life.Yet the mechanism of innate antiviral immunity in hNPCs remains largely unknown.Here,we show that ZIKV infection triggers the abundant production of virus-derived small interfering RNAs in hNPCs,but not in the more differentiated progenies or somatic cells.Ablation of key RNAi machinery components significantly enhances ZIKV replication in hNPCs.Furthermore,enoxacin,a broad-spectrum antibiotic that is known as an RNAi enhancer,exerts potent anti-ZIKV activity in hNPCs and other RNAi-competent cells.Strikingly,enoxacin treatment completely prevents ZIKV infection and circumvents ZIKV-induced microcephalic phenotypes in brain organoid models that recapitulate human fetal brain development.Our findings highlight the physiological importance of RNAi-mediated antiviral immunity during the early stage of human brain development,uncovering a novel strategy to combat human congenital viral infections through enhancing RNAi.Yan-Peng Xu Yang Qiu Boya Zhang Guilai Chen Qi Chen Miao Wang Fan Mo Jiuyue Xu Jin Wu Rong-Rong Zhang Meng-Li Cheng Na-Na Zhang Bao Lyu Wen-Liang Zhu Meng-Hua Wu Qing Ye Da Zhang Jiang-Hong Man Xiao-Feng Li Jie Cui Zhiheng Xu Baoyang Hu Xi Zhou Cheng-Feng Qin 2019Cell Research2019,29,4:8

8Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8

12GmCRY1s modulate gibberellin metabolism to regulate soybean shade avoidance in response to reduced blue light显示文摘Soybean is an important legume crop that displays the classic shade avoidance syndrome(SAS),including exaggerated stem elongation,which leads to lodging and yield reduction under density farming conditions.Here,we compared the effects of two shade signals,low red light to far-red light ratio(R:FR)and low blue light(LBL),on soybean status and revealed that LBL predominantly induces excessive stem elongation.We used CRISPR-Cas9-engineered Gmcry mutants to investigate the functions of seven cryptochromes(GmCRYs)in soybean and found that the four GmCRY1s overlap in mediating LBL-induced SAS.Lightactivated GmCRY1s increase the abundance of the bZlP transcription factors STF1 and STF2,which directly upregulate the expression of genes encoding GA2 oxidases to deactivate GA1 and repress stem elongation.Notably,GmCRY1b overexpression lines displayed multiple agronomic advantages over the wild-type control under both dense planting and intercropping conditions.Our study demonstrates the integration of GmCRY1-mediated signals with the GA metabolic pathway in the regulation of LBL-induced SAS in soybean.It also provides a promising option for breeding lodging-resistant,high-yield soybean cultivars in the future.Xiangguang Lyu Qican Cheng Chao Qin Yinghui Li Xinying Xu Ronghuan Ji Ruolan Mu Hongyu Li Tao Zhao Jun Liu Yonggang Zhou Haiyan Li Guodong Yang Qingshan Chen Bin Liu 2021Molecular Plant2021,14,2:4

14Epidemiological Characteristics of Plague in the Meriones unguiculatus Plague Focus--Inner Mongolia Autonomous Region,China,1950−2019显示文摘From 1950 to 2019,a total of 267 plague cases in humans were reported in the Inner Mongolia Autonomous Region with 133 deaths and 10,710 Yersinia pestis isolates;21 of these cases were reported from the Meriones unguiculatus(M.unguiculatus)plague focus with 6 deaths and 6,771 isolates.According to the plague situation in the Inner Mongolia Autonomous Region and the implementation of local preventive measures,the prevalence of plague in humans in the M.unguiculatus plague focus of the Inner Mongolia Autonomous Region could be divided into four stages.The first stage was the initial stage of plague prevention and control(1950–1959),with 0.80 cases annually and a case fatality rate of 37.50%.The second stage was the plague eradication stage(1960–1979),with 0.15 cases annually and a case fatality rate of 33.33%.The third stage was the plague surveillance stage(1980-1999),with 0.25 cases annually and a case fatality rate of 20.00%.The fourth stage is the comprehensive prevention and control stage under the emergency system(2000-2019),with 0.25 cases annually and a case fatality rate of 20.00%.The surveillance of rodent density(1981-2019)and studies on plague-related factors among M.unguiculatus have shown that the higher the M.unguiculatus density,the higher the nocturnal rodent capture rate(r=0.670,p<0.05)and the higher the indirect hemagglutination assay(IHA)positive rate of M.unguiculatus(r=0.344,p<0.05);the higher the percentage of hosts infected,the lower the M.unguiculatus density(r=-0.361,p<0.05)and the lower the IHA positive rate of M.unguiculatus(r=-0.337,p<0.05);the higher the percentage of nests infected with fleas,the lower the IHA positive rate of M.unguiculatus(r=-0.348,p<0.05).Together,these results suggest that it is necessary to simultaneously monitor the pathogens,serology,and vector index of M.unguiculatus to accurately reflect the plague prevalence among local animals.Although bubonic plague is the main plague type of the M.unguiculatus plague focus,severe pneumonic plague or septic plague may be secondary when the bubonic plague is misdiagnosed or not treated in time.In addition,the plague in animals is relatively virulent in the M.unguiculatus plague focus,and the risk of spreading to humans is higher.For plague in the Inner Mongolia Autonomous Region,comprehensive control efforts should be aimed at the M.unguiculatus focus,covering host animals,vector insects,and humans.Boxi Liu Dayu Zhang Yuhuang Chen Zhaokai He Jun Liu Dongyue Lyu Weiwei Wu Ran Duan Shuai Qin Junrong Liang Huaiqi Jing Xin Wang 2020China CDC weekly2020,2,49:3
15Risk prediction models for lung cancer:Perspectives and dissemination显示文摘Objective: The objective was to systematically assess lung cancer risk prediction models by critical evaluation of methodology, transparency and validation in order to provide a direction for future model development.Methods: Electronic searches(including PubMed, EMbase, the Cochrane Library, Web of Science, the China National Knowledge Infrastructure, Wanfang, the Chinese BioMedical Literature Database, and other official cancer websites) were completed with English and Chinese databases until April 30 th, 2018. Main reported sources were input data, assumptions and sensitivity analysis. Model validation was based on statements in the publications regarding internal validation, external validation and/or cross-validation.Results: Twenty-two studies(containing 11 multiple-use and 11 single-use models) were included. Original models were developed between 2003 and 2016. Most of these were from the United States. Multivariate logistic regression was widely used to identify a model. The minimum area under the curve for each model was 0.57 and the largest was 0.87. The smallest C statistic was 0.59 and the largest 0.85. Six studies were validated by external validation and three were cross-validated. In total, 2 models had a high risk of bias, 6 models reported the most used variables were age and smoking duration, and 5 models included family history of lung cancer.Conclusions: The prediction accuracy of the models was high overall, indicating that it is feasible to use models for high-risk population prediction. However, the process of model development and reporting is not optimal with a high risk of bias. This risk affects prediction accuracy, influencing the promotion and further development of the model. 16Ascorbate peroxidase 1 confers resistance to southern corn leaf blight in maize显示文摘Southern corn leaf blight (SCLB), caused by Bipolarismaydis, is one of the most devastatingdiseases affecting maize production. However,only one SLCB resistance gene, conferring partialresistance, is currently known, underscoring theimportance of isolating new SCLB resistancerelatedgenes. Here, we performed a comparativeproteomic analysis and identified 258 proteinsshowing differential abundance during the maizeresponse to B. maydis. These proteins included anascorbate peroxidase (Zea mays ascorbate peroxidase1 (ZmAPX1)) encoded by a gene locatedwithin the mapping interval of a previously identifiedquantitative trait locus associated with SCLBresistance. ZmAPX1 overexpression resulted inlower H_(2)O_(2) accumulation and enhanced resistanceagainst B. maydis. Jasmonic acid (JA)contents and transcript levels for JA biosynthesisand responsive genes increased in ZmAPX1-overexpressing plants infected with B. maydis,whereas Zmapx1 mutants showed the oppositeeffects. We further determined that low levels of H_(2)O_(2) are accompanied by an accumulation of JAthat enhances SCLB resistance. These resultsdemonstrate that ZmAPX1 positively regulatesSCLB resistance by decreasing H_(2)O_(2) accumulationand activating the JA-mediated defensesignaling pathway. This study identified ZmAPX1as a potentially useful gene for increasing SCLBresistance. 18Novel non-synonymous mutations of PAX8 in a cohort of Chinese with congenital hypothyroidism显示文摘Background:The transcription factor paired box 8 (PAX8) was associated with type 2 congenital non-goitrous hypothyroidism (CHNG2), a clinical phenotype of congenital hypothyroidism (CH). Though studied in a few regions with different ethnicities, the incidence of PAX8 mutations varied, even among Chinese cohorts in different regions. This study aimed to identify and characterize PAX8 mutations and explore the prevalence of its mutations in another cohort of CH. Methods: The 105 unrelated Chinese patients with CH were collected from four major hospitals. Exomes of the 105 samples were sequenced by Hiseq 2000 platform to identify mutations of PAX8 on genomic DNAs extracted from peripheral blood samples. Luciferase reporter assays were used to assess the effects of mutations on the transcription of thyroid peroxidase (TPO). Results: Three PAX8 mutations in four subjects were identified in 105 samples. One variant, rsl 89229644, was detected in two subjects, and categorized as uncertain significance. The other two missense mutations (275T>C/Ile92Thr and 398G>A/Argl33Gln) were not detected in three large-scale genotyping projects, namely 1000 Genome Project, Exome Aggregation Consortium and GO Exome Sequencing Project. Functional studies for the two mutations revealed that they could impair the transcription ability of PAX8 on one of its target genes, TPO. Therefore, the two mutations were causative for the pathogenesis of CHNG2. After combining the studies of PAX8 mutations, an average frequency of 1.74%(21/1209) could be obtained in Chinese patients with CH. 19Effect of glucose on the soil bacterial diversity and function in the rhizosphere of Cerasus sachalinensis显示文摘Most cherry orchards in China have low organic carbon content,though carbon is very important for plant growth.The changes in soil carbon and bacterial diversity were determined after different amounts of 12C-glucose were added to the rhizosphere of Cerasus sachalinensis.Soil bacteria diversity was measured using high throughput sequencing,and bacteria containing 13C-glucose were identified using DNA-SIP methods.The results demonstrated that soil microbial biomass carbon(MBC)content and the soil respiratory rate were increased at 3 and 7 days after adding glucose.The soil organic carbon(SOC)content was decreased on the 7th day in the treatment where the added glucose-C was equivalent to the MBC content.SOC content was decreased on the 15th day after adding glucose-C equivalent to five times that of the soil MBC.Compared to the controls,the relative abundance of taxa at the phylum level displayed no significant change in the treatments with glucose-C added as 10%and equal amount of soil MBC 3–30 days after treatment.However,the relative abundance of Proteobacteria increased significantly in the treatment with the addition of glucose-C equivalent to five times of soil MBC.The main changes were observed in the bacteria in several genera including A4,Flavisolibacter,Aquicella,and Candidatus Solibacter.DNA-SIP results indicated that the relative abundance of the Proteobacteria and Pseudomonas was the highest;these were the primary bacteria phylum and genus,respectively,from day 3 to day 15.In conclusion,the changing pattern demonstrated that with the addition of more glucose,the range of the bacterial communities changed more.Proteobacteria and Pseudomonas may be the bacteria promoting priming effect.Wenjie Zhou Xu Qin Deguo Lyu Sijun Qin 2021Horticultural Plant Journal2021,7,4:2
20Modulating effects of Astragalus polysaccharide on immune disorders via gut microbiota and the TLR4/NF-κB pathway in rats with syndrome of dampness stagnancy due to spleen deficiency显示文摘The syndrome of dampness stagnancy due to spleen deficiency(DSSD)is relatively common globally.Although the pathogenesis of DSSD remains unclear,evidence has suggested that the gut microbiota might play a significant role.Radix Astragali,used as both medicine and food,exerts the effects of tonifying spleen and qi.Astragalus polysaccharide(APS)comprises a macromolecule substance extracted from the dried root of Radix Astragali,which has many pharmacological functions.However,whether APS mitigates the immune disorders underlying the DSSD syndrome via regulating gut microbiota and the relevant mechanism remains unknown.Here,we used DSSD rats induced by high-fat and low-protein(HFLP)diet plus exhaustive swimming,and found that APS of moderate molecular weight increased the body weight gain and immune organ indexes,decreased the levels of interleukin-1β(IL-1β),IL-6,and endotoxin,and suppressed the Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)pathway.Moreover,a total of 27 critical genera were significantly enriched according to the linear discriminant analysis effect size(LEfSe).APS increased the diversity of the gut microbiota and changed its composition,such as reducing the relative abundance of Pseudoflavonifractor and Paraprevotella,and increasing that of Parasutterella,Parabacteroides,Clostridium XIVb,Oscillibacter,Butyricicoccus,and Dorea.APS also elevated the contents of short-chain fatty acids(SCFAs).Furthermore,the correlation analysis indicated that 12 critical bacteria were related to the body weight gain and immune organ indexes.In general,our study demonstrated that APS ameliorated the immune disorders in DSSD rats via modulating their gut microbiota,especially for some bacteria involving immune and inflammatory response and SCFA production,as well as the TLR4/NF-κB pathway.This study provides an insight into the function of APS as a unique potential prebiotic through exerting systemic activities in treating DSSD.Wenxiao ZHAO Chenchen DUAN Yanli LIU Guangying LU Qin LYU Xiumei LIU Jun ZHENG Xuelian ZHAO Shijun WANG Haijun ZHAO 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7:1
