1肝内和肝外胆管癌肿瘤抑制基因启动子甲基化分析显示文摘目的 探讨胆管癌的表遗传学改变。方法 用甲基化特异PCR(MSP)法 ,检测了 12个候选肿瘤抑制基因 (APGE cad herin/CDH1,MGMT ,RASSF1A ,GSTP ,RAR β ,p14 ARF,p15 INK4b,p16INK4a,p73 ,hMLH1,DAPK)启动子在 72例胆管癌中的甲基化情况 ,其中肝内和肝外胆管癌各 3 6例 ,10例良性胆管上皮作为对照。结果 85 %的胆管癌至少有一个肿瘤抑制基因的甲基化 ,在胆管癌中 ,肿瘤抑制基因的甲基化顺序是 :RASSF1A(65 %) ,p15 INK4b(5 0 %) ,p16INK4a(5 0 %) ,APC(4 6%) ,E cadherin/CDH1(4 3 %) ,p14 ARF(3 8%) ,p73 (3 6%) ,MGMT(3 3 %) ,hMHL1(2 5 %) ,GSTP(14 %) ,RAR β(14 %)和DAPK(3 %)。虽然单个肿瘤抑制基因的甲基化可见于良性胆管上皮 ,但是多个肿瘤抑制基因的甲基化只见于胆管癌。约 70 %(5 0 /72 )的胆管癌有 3个或 3个以上的肿瘤抑制基因的甲基化 ,5 2 %(3 8/72 )有 4个或 4个以上肿瘤抑制基因的甲基化。多个肿瘤抑制基因的协同甲基化 ,和RASSF1A ,p15 INK4b,p16INK4a和 /或hMHL1密切相关。RASSF1A的甲基化在肝外胆管癌 (83 %)较肝内胆管癌更常见 (4 7%) (P =0 . 0 0 3 ) ,而GSTP更多见于肝内胆管癌(肝内 3 1%,肝外 6%,P =0 . 0 12 ) ,本研究提示肿瘤抑制基因启动子CpG岛的甲基化在胆管癌中是一?YANG Bin Michael G.House GUO Mingzhou James G.Herman Douglas P.Clark 2005胃肠病学和肝病学杂志2005,14,1:46

3Dynamic Prediction Method of Production Logistics Bottleneck Based on Bottleneck Index显示文摘In modern manufacturing pattern,there are many uncertain factors in the modern manufacturing process,such as changes of product attribute,changes of manufacturing resources' state,and so on,which cause production logistics bottleneck frequently shift,and make decisions of production planning and control based on formed bottleneck deviated from practical production process.Considering these factors,present researches mainly apply afterwards control to optimize production process to passively adapt to bottleneck changes.If the direction of bottleneck shifting can be accurately forecasted,the transition from afterwards control of chasing bottleneck to beforehand control can be realized.Therefore,aiming at the phenomenon of production logistics bottleneck shifting under uncertain manufacturing circumstances,this paper starts off with dynamic property of capability and requirement and then builds the concepts of bottleneck degree and bottleneck index to describe dynamic bottleneck characteristic of production unit;taken production capability,production load and quality assurance capability into consideration,mathematical model of bottleneck index is established to measure bottleneck degree accurately,consequently,quantitative research on mechanism of production logistics shifting is achieved.Based on bottleneck index,the prediction model of production logistics bottleneck is founded to predict dynamic change of bottleneck accurately.Finally,an example of forecasting and monitoring the production logistics bottleneck in one manufacturing shop is given to testify the validation and practicability of the prediction method.LIU Mingzhou TANG Juan GE Maogen JIANG Zengqiang HU Jing LING Lin 2009Chinese Journal of Mechanical Engineering2009,22,5:22
4糖尿病视网膜病变患病率的流行病学调查:在中国6省13473名糖尿病患者中的横断面研究显示文摘目的本研究旨在探究中国糖尿病患者中糖尿病视网膜病变(DR)和视力威胁性糖尿病视网膜病变(STDR)的患病率和严重程度。研究设计、设置与参与者本研究为横断面流行病学调查,研究对象为中国内地地区的糖尿病患者(N=13473)。研究对象为被内科医师诊断为糖尿病,并转诊到筛查门诊的糖尿病患者(其中住院患者占1/3,城市居民占1/3,农村居民占1/3)。研究对象的每只眼睛应用彩色、免散瞳和非立体眼底照相机拍摄2张眼底照片,并根据英国DR分级指南进行分级。主要结果和测量指标DR与STDR的患病率和严重程度。结果在参与本研究的13473例糖尿病患者中,共发现4591例DR和1769例STDR患者,DR的总体患病率为34.08%[95%可信区间(凹)33.28%~34.88%],其中STDR的总体患病率为13.13%(95%C112.56%~13.70%)。在所有研究对象中,12780例参与了眼底照片分级(94.86%)。年龄大于65岁的研究对象患DR或STDR的可能性较低(P〈0.001)。性别对是否患病没有显著影响(P〉0.05)。STDR患者与非STDR患者相比,视力损伤更为严重(P〈0.001)。中国北方DR和STDR的患病率高于南方(P〈0.001)。DR的患病率与糖尿病患病时间f比值比(鲫)2.63;95%C12.42~2.86;P〈0.001]以及糖尿病发病年龄(OR 0.38;95%CI为0.35~0.41;P〈0.001)密切相关。结论DR和STDR在中国内地地区的患病率偏高,并且不同地区患病率存在差异。在DR中,非增殖性DR更常见,但STDR更需要及时的诊断和治疗,特别是在经济欠发达地区。本研究强调了中国内地地区DR筛查和治疗的重要性。Liu, Yan Song, Yifan Tao, Liyuan Qiu, Weiqiang Lv, Huibin Jiang, Xiaodan Zhang, Mingzhou Li, Xuemin 2017英国医学杂志中文版2017,20,11:23

7DNA methylation-mediated repression of miR-181a/135a/302c expression promotes the microsatellite-unstable colorectal cancer development and 5-FU resistance via targeting PLAG1显示文摘Micro sate Hite instability(MSI) defines a subtype of colorectal cancer(CRC) with typical clinicopathologic characteristics. CRCs with MSI(MSI CRCs) frequently acquire accelerated carcinogenesis and 5-FU resistance, and the exact underlying mechanism remains incompletely understood. Our previous study has identified the microRNA(miRNA) expression profile in MSI CRCs. In this study, three miRNAs(miR-181 a, miR-135 a and miR-302 c) were validated by qRT-PCR to be dramatically decreased in 67 CRC samples. Proliferation and apoptosis assays demonstrated that miR-181 a/135 a/302 c function as tumor suppressors via repressing PLAG1/IGF2 signaling. Moreover, we presented compelling evidence that restoration of miR-181 a/135 a/302 c expression promoted sensitivity of MSI CRC cells to 5-FU treatment. miR-181 a/135 a/302 c exerted their effect on chemoresistance through attenuating PLAG1 expression. Notably, the hypermethylation status of MSI CRC accounts for the decrements of miR-181 a/135 a/302 c. 8Building a sequence map of the pig pan-genome from multiple de novo assemblies and Hi-C data显示文摘Pigs were domesticated independently in the Near East and China,indicating that a single reference genome from one individual is unable to represent the full spectrum of divergent sequences in pigs worldwide.Therefore,12 de novo pig assemblies from Eurasia were compared in this study to identify the missing sequences from the reference genome.As a result,72.5 Mb of nonredundant sequences(~3% of the genome)were found to be absent from the reference genome(Sscrofa11.1)and were defined as pan-sequences.Of the pan-sequences,9.0 Mb were dominant in Chinese pigs,in contrast with their low frequency in European pigs.One sequence dominant in Chinese pigs contained the complete genic region of the tazarotene-induced gene 3(TIG3)gene which is involved in fatty acid metabolism.Using flanking sequences and Hi-C based methods,27.7% of the sequences could be anchored to the reference genome.The supplementation of these sequences could contribute to the accurate interpretation of the 3D chromatin structure.A web-based pan-genome database was further provided to serve as a primary resource for exploration of genetic diversity and promote pig breeding and biomedical research.Xiaomeng Tian Ran Li Weiwei Fu Yan Li Xihong Wang Ming Li Duo Du Qianzi Tang Yudong Cai Yiming Long Yue Zhao Mingzhou Li Yu Jiang 2020Science China(Life Sciences)2020,63,5:7

10Characterization particulate matter from several Chinese cooking dishes and implications in health effects显示文摘Cooking fume produced by oil and food at a high temperature releases large amount of fine particulate matter(PM) which have a potential hazard to human health. This chamber study investigated particle emission characteristics originated from using four types of oil(soybean oil, olive oil, peanut oil and lard) and different kinds of food materials(meat and vegetable). The corresponding emission factors(EFs) of number, mass, surface area and volume for particles were discussed. Temporal variation of size-fractionated particle concentration showed that olive oil produced the highest number PM concentration for the entire cooking process. Multiple path particle dosimetry(MPPD) model was performed to predict deposition in the human respiratory tract. Results showed that the pulmonary airway deposition fraction was the largest. It was also found that particles produced from olive oil led to the highest deposition. 11Amplification of the miR-181c/d cluster is inversely correlated with PDCD4 expression in gastric cancer显示文摘miR-181c/d is dysregulated in gastric cancer(GC).We investigated the amplification and expression of miR-181c/d and its predicted target genes in GC.Amplification of miR-181c/d was quantified by genomic real-time PCR in GC and adjacent normal tissues,as well as the levels of mature miR-181c/d was performed by real-time PCR in the same tissues.The potential target genes of miR-181c/d were predicted using bioinformatics software.Expression of one potential target gene,PDCD4,was measured by semiquantitative RT-PCR,real-time PCR,and immunohistochemistry.Next,the relationship between miR181c/d expression and PDCD4 expression was analyzed.Results indicated that the amplification and expression of miR-181c/d were significantly higher in GC than in adjacent normal tissues(primary miR-181c/d,P\0.001;miR-181c,P=0.0344;miR-181d,P=0.0153),and there was a strong correlation between mature miR-181c/d and primary miR-181c/d.Thirty-two target genes were predicted,including PDCD4 which is a known tumor suppressor gene.Expression of PDCD4 was significantly down-regulated in GC as compared to adjacent normal tissues and was inversely correlated with miR-181c/d expression in GC(miR-181c and PDCD4:R=-0.496,P=0.008;miR-181d and PDCD4:R=-0.454,P=0.003).Therefore,miR-181c/d may play a pivotal role in the pathogenesis of GC by downregulating PDCD4 expression.Yuanming Pan Rui Xing Juan An Jiantao Cui Wenmei Li Mingzhou Guo Youyong Lu 2014Chinese Science Bulletin2014,59,19:6
12Microbial bioavailability of dissolved organic nitrogen(DON) in the sediments of Lake Shankou,Northeastern China显示文摘Dissolved organic nitrogen(DON)extracted from Lake Shankou sediments using KCl was isolated into hydrophobic and hydrophilic fractions.The bioavailabilities of the hydrophobic and hydrophilic fractions to three types of bacterial communities collected from sediments,activated sludge and compost products were examined.The DON recoveries obtained by DAX-8 and cation exchange resins treatment were 96.17% ± 1.58% and 98.14% ±0% for the samples obtained from N4 and N14 stations,respectively.After 25 days of incubation at 25°C,most DON(59% to 96%)was degraded.Hydrophilic DON exhibited a higher reduction rate than hydrophobic DON during the growth phase.Untreated wastewater from Changshuihe town was the main degradable DON source to station N4,and 93% of hydrophilic DON and 80% of hydrophobic DON were degraded.Station N14 received a large amount of refractory DON from forest soils and exhibited DON degradation rates of 82% and 71% for the hydrophilic and hydrophobic fractions,respectively.Amino acid contents and fluorescence intensities were also analyzed.Approximately 27% to 74% of amino acids were taken up by day 5,and their concentration gradually increased in the following days due to the decomposition of dissolved proteins.Parallel factor analysis resulted in identification of tryptophan-like proteins,tyrosine-like proteins and FA-like substances.During the growth phase,40%–51% of the tryptophan-like proteins were taken up by bacteria,and the accumulation of tyrosine-like proteins was attributed to the release of biotic substances.The concentration of the FA-like substances decreased due to microbial decomposition.Mingzhou Su Jingtian Zhang Shouliang Huo Beidou Xi Fei Hua Fengyu Zan Guangren Qian Jianyong Liu 2016Journal of Environmental Sciences2016,28,4:5
13Differential expression analysis and regulatory network reconstruction for genes associated with muscle growth and adipose deposition in obese and lean pigs显示文摘During the growth and development of skeletal muscle cells and adipose cells, the regulatory mechanism of micro-effect polygenes determines porcine meat quality, carcass characteristics and other relative quantitative traits. Obese and lean type pig breeds show obvi- ous differences in muscle growth and adipose deposition; however, the molecular mechanism underlying this phenotypic variation remains unknown. We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with muscle growth and adipose deposition in longissimus dorsi muscle at six growth stages (birth, 1, 2, 3, 4 and 5 months) of Landrace (a leaner, Western breed) and Taihu pigs (a fatty, indigenous, Chinese breed). Variance analysis (ANOVA) revealed that differences in the expression of 18 genes in Landrace pigs and three genes in Taihu pigs were very significant (FDR-adjusted permutation, P < 0.01) and di?erences for 22 genes in Landrace pigs and seven genes in Taihu pigs were significant (FDR-adjusted permutation, P < 0.05) among six growth stages. Clustering analysis revealed a high level of signi?cance (FDR-adjusted, P < 0.01) for four gene expression patterns, in which genes that strongly up-regulated were mainly associated with the positive regulation of myofiber formation and fatty acid biogenesis and genes that strongly down-regulated were mainly associated with the inhibition of cell proliferation and positive regulation of fatty acid b-oxidation. Based on a dynamic Bayesian network (DBN) model, gene regulatory networks (GRNs) were reconstructed from time-series data for each pig breed. These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of muscle growth and adipose deposition between the two pig breeds; from these results, some potential key genes could be identified. Quantitative real-time RT-PCR (QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both two pig breeds at different growth stages (r = 0.876 ± 0.095). 15Phase Ⅱ study of apatinib in combination with oral vinorelbine in heavily pretreated HER2-negative metastatic breast cancer and clinical implications of monitoring ctDNA显示文摘Objective:Apatinib is an oral TKI targeting VEGFR-2.Single-agent apatinib treatment has been shown to produce an objective response in patients with pretreated m BC.Oral vinorelbine also holds promise as a treatment of choice in patients with m BC.This study aimed to investigate the efficacy and safety of the oral vinorelbine-apatinib combination in patients with pretreated m BC.In addition,we detected gene variants in ct DNA to explore the therapeutic implications.Methods:This study enrolled patients with HER2-negative m BC who were pretreated with anthracycline/taxanes.Patients were treated with apatinib at 500 mg/425 mg daily plus oral vinorelbine 60 mg/m2 on days 1,8,and 15 of every cycle(3 weeks).The primary endpoint was PFS.The secondary endpoints were ORR,CBR,OS,and safety.Patients eligible for ct DNA detection were evaluated before and during treatment.Results:Forty patients were enrolled.The median PFS was 5.2 months(95%CI,3.4–7.0 months),and the median OS was 17.4 months(95%CI,8.0–27.0 months).The ORR was 17.1%(6/35),and the CBR was 45.7%(16/35).The most common AEs included gastrointestinal reaction,myelosuppression,and hypertension.In 20 patients,ct DNA was detected at baseline and during treatment.A significant difference was found in PFS for undetected vs.detected baseline ct DNA(13.9 months vs.3.6 months,P=0.018).Conclusions:All-oral therapy with apatinib plus vinorelbine displayed objective efficacy in patients with heavily pretreated HER2-negative m BC,with acceptable and manageable toxicity profiles.Patients with no gene variant detected and lower variant allele frequencies in ct DNA at baseline showed longer PFS.Anjie Zhu Peng Yuan Nanlin Hu Mingzhou Li Wenmiao Wang Xue Wang Jian Yue Jiayu Wang Yang Luo Fei Ma Pin Zhang Qing Li Binghe Xu Shanbo Cao Giuseppe Lippi Yoichi Naito Mohammed A.Osman Gustavo N.Marta Gianluca Franceschini Armando Orlandi 2021Cancer Biology & Medicine2021,18,3:4
16An analytical solution for the population balance equation using a moment method显示文摘Brownian coagulation is the most important inter-particle mechanism affecting the size distribution of aerosols.Analytical solutions to the governing population balance equation(PBE) remain a challenging issue.In this work,we develop an analytical model to solve the PBE under Brownian coagulation based on the Taylor-expansion method of moments.The proposed model has a clear advantage over conventional asymptotic models in both precision and efficiency.We first analyze the geometric standard deviation(GSD) of aerosol size distribution.The new model is then implemented to determine two analytic solutions,one with a varying GSD and the other with a constant GSD.The varying solution traces the evolution of the size distribution,whereas the constant case admits a decoupled solution for the zero and second moments.Both solutions are confirmed to have the same precision as the highly reliable numerical model,implemented by the fourth-order Runge-Kutta algorithm,and the analytic model requires significantly less computational time than the numerical approach.Our results suggest that the proposed model has great potential to replace the existing numerical model,and is thus recommended for the study of physical aerosol characteristics,especially for rapid predictions of haze formation and evolution.Mingzhou Yu Jianzhong Lin Junji Cao Martin Seipenbusch 2015Particuology2015,13,1:4
17Alterations in cecal microbiota of Jinhua piglets fostered by a Yorkshire sow显示文摘Gut microbiota play important roles in host health and diseases.Several factors,in particular antibiotics,affect the gut microbiota of pigs.Cross-fostering has been applied as a regular practice to equalize litter size,reduce pre-weaning mortality and increase body weight.However,the effect of cross-fostering on cecal microbiota is unclear.In this study,we fostered three Jinhua pigs to a Yorkshire sow.The fostered Jinhua piglets grew significantly faster than their biological siblings.To explore whether the cecal microbiota of piglets will alter during fostering,we characterized the cecal microbiota of allpiglets by examining the V3 hypervariable region of 16S rDNA.We observed altered cecal microbiota in these piglets using the Illumina HiSeq 2000 platform,and this was accompanied with an increase in growth rate after fostering.The relative abundance of Bacteroidetes in the fostered Jinhua piglets was decreased compared with their biological siblings,although still higher in comparison with their new littermates.Beta-diversity analysis also showed that the cecal microbiota of the adopted Jinhua piglets differed from their biological siblings with a shift toward their step-siblings.Our data show that cecal microbiota of piglets were altered after cross-fostering while the growth rate increased.Lingjin Xian Ying Li Zhi Jiang Jideng Ma Long Jin Lei Chen Chaowei Zhou Jie Zhang Yingkai Liu Li Zhu An-an Jiang Guoqing Tang Jian Xiao Jinyong Wang Bo Zeng Mingzhou Li Xuewei Li 2014Chinese Science Bulletin2014,59,32:4

19Algal uptake of dissolved organic nitrogen in wastewater treatment plants显示文摘The algal uptake of dissolved organic nitrogen(DON) in the anaerobic–anoxic–oxic(A_2O)process was investigated in this study. Anaerobic, aerobic and effluent DON samples from two wastewater treatment plants(WWTPs) were separated into hydrophilic and hydrophobic fractions using a DAX-8 resin coupled with an anion exchange resin and a nanofiltration(NF)pretreatment. Hydrophilic DON accounted for 66.66%–88.74% of the entire DON for the two plants evaluated. After a 15-day incubation, 16.95%–91.75% DON was bioavailable for algal growth, and untreated samples exhibited higher DON bioavailability, with 52.83% DON average uptake rates, compared with the hydrophilic and hydrophobic fractions(45.53% and 44.40%, respectively) because the pretreatment caused the inorganic salt to be resistant to algae. Anaerobic untreated samples, hydrophilic fractions and hydrophobic fractions showed higher DON reduction rates and higher biomass accumulation compared with the other DON fractions due to the decomposition of resistant organics by anaerobic and anoxic bacteria.DON in aerobic and effluent samples of plant A was more bioavailable than that of plant B with usages of 27.49%–55.26% DON. DON bioavailability in the anaerobic–anoxic–oxic process decreased in the following order: anaerobic > effluent > aerobic. The DON contents were reduced after anaerobic treatment in the two plants. 20High-sensitivity infrared vibrational nanospectroscopy in water显示文摘Mid-infrared vibrational spectroscopy is a universal label-free tool for identifying molecular compounds in chemical and biological samples on the basis of their‘fingerprint’vibrational absorption lines.Vibrational spectroscopy with nanometer spatial resolution can reveal the chemical composition of samples at the nanoscale,and several scanning-probe techniques have been developed to address this need1–22.It is also highly desirable to study biological and chemical samples in their native aqueous environments rather than in air.In aqueous environments,however,the sensitivity of the current vibrational nanospectroscopy techniques deteriorates dramatically7,23.Here,we report the first mid-infrared nanospectroscopy technique that retains nanoscale sensitivity and spatial resolution when the sample and the scanning probe are completely immersed in water.This method overcomes challenges including water absorption and scattering and the mechanical damping of cantilever vibrations.We further demonstrate spectroscopy and imaging of 20-to 50-nmthick polymer samples with a 25-nm spatial resolution in the biologically relevant Amide I and II spectral regions.Mingzhou Jin Feng Lu Mikhail A Belkin 2017Light(Science & Applications)2017,6,1:3
