1Molecular network-based intervention brings us closer to ending the HIV pandemic显示文摘Precise identification of HIV transmission among populations is a key step in public health responses.However,the HIV transmission network is usually difficult to determine.HIV molecular networks can be determined by phylogenetic approach,genetic distance-based approach,and a combination of both approaches.These approaches are increasingly used to identify transmission networks among populations,reconstruct the history of HIV spread,monitor the dynamics of HIV transmission,guide targeted intervention on key subpopulations,and assess the effects of interventions.Simulation and retrospective studies have demonstrated that these molecular network-based interventions are more cost-effective than random or traditional interventions.However,we still need to address several challenges to improve the practice of molecular network-guided targeting interventions to finally end the HIV epidemic.The data remain limited or difficult to obtain,and more automatic real-time tools are required.In addition,molecular and social networks must be combined,and technical parameters and ethnic issues warrant further studies.Xiaoxu Han Bin Zhao Minghui An Ping Zhong Hong Shang 2020Frontiers of Medicine2020,14,2:7

3Comprehensive Humoral and Cellular Immune Responses to SARS-CoV-2 Variants in Diverse Chinese Population显示文摘The SARS-CoV-2 variants have been emerging and have made great challenges to current vaccine and pandemic control strategies.It is urgent to understand the current immune status of various Chinese populations given that the preexisting immunity has been established by national vaccination or ex posure to past variants.Using sera from 85 individuals(including 21 convalescents of natural infection,15 cases which suffered a breakthrough infection after being fully vaccinated,and 49 healthy vaccinees),we showed signifcantly enhanced neutralizing activities against SRAS-CoV-2 variants in convalescent sera,especially those who had been fully vaccinated.The neut ralizing antibodies against Omicron were detectable in 75%of convalescents and 4.9%of healthy vaccinees(p=0.006),with a GMT of 289.5,180.9-463.3,and 42.6,31.3-59,respetively.However,the neutralizing activities were weaker in young convalescents(aged<18y),with a detectable rate of 50%and a GMT of 46.4 against Omicron.We also examined and found no pan.sarbecovirus neutralizing activities in vaccinated SARS-CoV-I survivors.A booster dose could further increase the breadth and magnitude of neutralization against WT and variants of concern(VOCs)to different degrees.In addition,we showed that COVID-19-inactivated vaccines can elicit Omicron-specific T-cell responses.The positive rates of ELISpot reactions were 26.7%(4/15)and 43.8%(7/16)in the full vaccination group and the booster vaccination group,respectively,although without statistically significant difference.The neutralizing antibody titers declined while T-cell responses remain consistent over 6 months.These findings will inform the optimization of public health vaccination and intervention strategies to protect diverse populations against SARS-CoV-2 variants.Advances.Breakthrough infection significantly boosted neutralizing activities against SARS-CoV-2 variants a compared to booster immunization with inaclivated vaccine.Vaccine-induced vir us-specific T-cell immunity,on the other hand,may compensate for the shortall.Furthermore,the public health system should target the most vulnerable group due to a poorer protective serological response in both infected and vaccinated adolescents.Jiwei Li Jing Wu Qiuyue Long Yan'an Wu Xiaoyi Hu Yukun He Mingzheng Jiang Jia Li Lili Zhao Shuoqi Yang Xiaoyong Chen Minghui Wang Jianshi Zheng Fangfang Wu Ruiliang Wu Lihong Ren Liang Bu Houzhao Wang Ke Li Lijuan Fu Guojun Zhang Yali Zheng Zhancheng Gao 2022Research2022,,2:1
4Application of single-cell RNA sequencing in head and neck squamous cell carcinoma显示文摘Single-cell RNA sequencing has been broadly applied to head and neck squamous cell carcinoma(HNSCC) for characterizing the heterogeneity and genomic mutations of HNSCC benefiting from the advantage of single-cell resolution. We summarized most of the current studies and aimed to explore their research methods and ideas, as well as how to transform them into clinical applications. Through single-cell RNA sequencing, we found the differences in tumor cells’ expression programs and differentiation tracks. The studies of immune microenvironment allowed us to distinguish immune cell subpopulations, the extensive expression of immune checkpoints, and the complex crosstalk network between immune cells and non-immune cells. For cancerassociated fibroblasts(CAFs), single-cell RNA sequencing had made an irreplaceable contribution to the exploration of their differentiation status, specific CAFs markers, and the interaction with tumor cells and immune cells. 5Effects of hexanal fumigation on fungal spoilage and grain quality of stored wheat显示文摘The prevention of fungal spoilage is an essential consideration in wheat storage.Recent studies have revealed that volatile organic compounds(VOCs),possibly with natural fungicidal properties,could be produced from stored wheat grains.In this study,the antifungal effect of hexanal,a main component of VOCs from stored wheat,against spoilage fungi on agar plate and in high-moisture wheat grains were investigated via the gas fumigation method.And the impact of hexanal fumigation on grain quality was evaluated through analysis of the malondialdehyde content,fatty acid values,germination percentages and vigor of 16%and 18%moisture wheat grains fumigated with 1.66,2.49,and 3.31 mmol/L hexanal vapor.The results of in vitro antifungal experiments on agar plates revealed that the minimum inhibitory concentration and minimum fungicidal(fatal)concentration of hexanal against the five fungi were 4–14 folds and 4–7 folds lower than those of propionic acid,respectively.The fungal spoilage of high-moisture wheat grains inoculated with pure fungal spores and with naturally occurring fungi could be completely inhibited by 1.66 mmol/L hexanal vapor.During 5-week storage of high-moisture wheat grains fumigated with 1.66,2.49,and 3.31 mmol/L hexanal vapor,the malondialdehyde content in high-moisture wheat grains did not change significantly in all samples,and fatty acid values were slightly higher in 18%moisture wheat than in 16%moisture wheat.The germination percentages and vigor of wheat samples decreased with increased hexanal vapor concentrations and moisture content.These results indicated that hexanal fumigation could be used as an alternative chemical control method to prevent the fungal spoilage of postharvest wheat.Shuaibing Zhang Minghui Zheng Huanchen Zhai Ping'an Ma Yangyong Lyu Yuansen Hu Jingping Cai 2021Grain & Oil Science and Technology2021,4,1:0
6Perioperative risk factors for delayed gastric emptying after pancreaticoduodenectomy:a meta-analysis of the complications显示文摘Objective:To evaluate the correlation between the incidence of delayed gastric emptying(DGE)in pancreaticoduodenectomy(PD)and the perioperative possible risk factors.Methods:We searched PubMed,Embase,Cochrane Library,and Medline for studies describing complications related to PD from 2018 to 2022.Statistical analysis was performed using Stata/SE16.0 software.Results:In 6 retrospective studies,1 systematic review,and 1 randomized controlled trial(RCT)experiment,12,419 patients(2174 and 10,245 patients,respectively,in DGE and no delayed gastric emptying[NDGE]groups).The preoperative American Society of Anesthesiologists(ASA)score between DGE and NDGE groups(log odds ratio[OR]=0.23,95%credible interval[CI]:0.11-0.35,P<.01),operation time(mean diff.=21.87,95%CI:15.96-27.78,P<.01),estimated intraoperative blood loss(EIBL;mean diff.=70.67,95%CI:17.75-123.58,P<.05),postoperative pancreatic fistula(POPF)(log OR=0.85,95%CI:0.24-1.46,P<.05)were statistically significant.No significant differences in preoperative body mass index(BMI),preoperative biliary drain-age,and pancreatic texture were observed(P>.05).Conclusion:The incidence of DGE is significantly correlated with the operation time,preoperative ASA score,POPF,and EIBL,which suggests that the surgeon should thoroughly evaluate the patient through adequate preoperative examination before the operation.Finally,standardized practice and perfect technology are undoubtedly necessary to reduce complications.Xiaowei Deng Ting Niu Zhaolong Han Xing Liang Zhiping Fu Liang Tang Guang Yang Judong Li An’an Liu Junfeng Peng Minghui Zheng Chenghao Shao 2023Journal of Pancreatology2023,6,2:0
