| 1 | Inhibitory Effect of 16 Fungicides on Persimmon Anthracnose Pathogen Colletotrichum horii显示文摘In this research,the inhibitory effect of 16 fungicides on Colletotrichum horri causing persimmon anthracnose was investigated using mycelial growth method and spore germination method. The results showed that among the 16 tested fungicides,10% of Difenoconazole WG,33. 5% of Copper quinolate SC,25%of Bromothalonil EC,70% of Mancozeb WP,430 g/L of Tebuconazole SC,50% of Prochloraz-manganese chloride and 400 g/L of Flusilazole EC achieved the best inhibitory effect on mycelial growth of C. horri,with the inhibition rate of 100%; 70% of Polyram WG,33. 5% of Copper quinolate SC,25% of Bromothalonil EC,70% of Mancozeb WP,50% of Chlorobromoisocyanuric acid AF,50% of Triram WP and 400 g/L of Flusilazole achieved the best inhibitory effect on spore germination of C. horri,with the germination rate of 0. In conclusion,33. 5% of Copper quinolate SC,25% of Bromothalonil EC,70% of Mancozeb WP and 400 g/L of Flusilazole EC achieved the best inhibitory effect both in mycelial growth and spore germination,which could be used as the preference fungicides for the control of persimmon anthracnose,and 70% of Polyram WG and 50% of Triram WP achieved the secondly best inhibitory effect,which could be used as alternative fungicides. | 2 | Application of single-cell RNA sequencing in head and neck squamous cell carcinoma显示文摘Single-cell RNA sequencing has been broadly applied to head and neck squamous cell carcinoma(HNSCC) for characterizing the heterogeneity and genomic mutations of HNSCC benefiting from the advantage of single-cell resolution. We summarized most of the current studies and aimed to explore their research methods and ideas, as well as how to transform them into clinical applications. Through single-cell RNA sequencing, we found the differences in tumor cells’ expression programs and differentiation tracks. The studies of immune microenvironment allowed us to distinguish immune cell subpopulations, the extensive expression of immune checkpoints, and the complex crosstalk network between immune cells and non-immune cells. For cancerassociated fibroblasts(CAFs), single-cell RNA sequencing had made an irreplaceable contribution to the exploration of their differentiation status, specific CAFs markers, and the interaction with tumor cells and immune cells. | 3 | Biological Characteristics of Persimmon Anthracnose Pathogen Colletotrichum horii and Screening of Inhibitory Fungicides显示文摘This experiment studied the biological characteristics of Colletotrichum horii causing persimmon anthracnose using the crossing method and blood cell counting plate method,and screened inhibitory fungicides via assessing the effects of 16 common fungicides on the mycelial growth and spore germination. The results showed that the most suitable temperature for mycelial growth of C. horri is 25℃,the most suitable temperature for spore germination is 28℃; the suitable p H for mycelial growth of C. horri is 4. 0-6. 0,the most suitable p H for spore germination is 4. 0; the optimal carbon source is glucose and maltose,and the optimal source of nitrogen is beef extract. Among the 16 common fungicides,33. 5% copper quinolate SC,25% bromothalonil EC and 70% Mancozeb WP have the optimal inhibitory effects on the mycelial growth and spore germination of C. horri,and can be used as preferred agent for prevention and control of persimmon anthracnose,followed by70% Polyram WG,400 g/L Flusilazole EC and 50% Thiram WP,which can be used as alternative agents. | 4 | Incubation strategies of the Black-necked Crane(Grus nigricollis) in relation to ambient temperature and time of day显示文摘Background: The behavior of cranes reflects many of their survival strategies, but little has been known of the incubation strategies of cranes, in which both parents share incubation duties, in response to cold temperatures in alpine environments. The lack of information may restrict the effective conservation of the threatened Black-necked Crane(Grus nigricollis), a biparental bird nesting in high elevation wetlands.Methods: We directly observed and used infrared video cameras from 2014 to 2015 to study the incubation behavior and quantitatively measured the frequency and details of egg turning behavior in the Black-necked Crane at the Yanchiwan National Nature Reserve in western Gansu Province, China.Results: At lower ambient temperatures in the morning, crane parents spent more time on the nest with less recess frequency and prolonged on-bout duration, while at higher temperatures around noon, the parents had more frequent recesses from incubation and shorter periods between nest exchanges. They adjusted the amount of time incubating by varying the recess frequency and the length of on-bout duration. Mean nest attendance and egg turning frequency of females were significantly higher than those of the males. The nest attendance and on-bout duration of females showed a significantly negative relationship with those of males. The two parents responded differently to the change of temperature. Females spent more time on the nest at lower morning temperatures, while males increased their time on the nest at higher temperatures after noon. Higher incubation recess frequency and egg turning frequency were observed at noon, probably because parents spent more time foraging, taking advantage of the lower egg cooling rate.Conclusion: Both Black-necked Crane parents in the alpine environment adjusted their behavior in response to the thermal requirements of eggs and the weather conditions experienced.
