| 1 | GOLM1 exacerbates CD8^(+)T cell suppression in hepatocellular carcinoma by promoting exosomal PD-L1 transport into tumor-associated macrophages显示文摘The immunosuppressive microenvironment plays an important role in tumor progression and immunotherapy responses.Golgi membrane protein 1(GOLM1)is correlated to hepatocellular carcinoma(HCC)progression and metastasis.However,little is known about the role of GOLM1 in regulating the immunosuppressive environment and its impact on immunotherapeutic efficacy in HCC.In this study,GOLM1 was positively correlated with infiltrating tumor-associated macrophages(TAMs)expressed high levels of programmed death-ligand 1(PD-L1)and CD8^(+)T cell suppression in HCC tissues.Both gain-and loss-of-function studies determined a close correlation between GOLM1 and immunosuppression.In the mechanism,GOLM1 promoted COP9 signalosome 5-mediated PD-L1 deubiquitination in HCC cells and increased the transport of PD-L1 into exosomes via suppression of Rab27b expression.Furthermore,co-culture with exosomes derived from HCC cells upregulated the expression of PD-L1 on macrophages.Zoledronic acid in combination with anti-PD-L1 therapy reduced PD-L1^(+)TAMs infiltration and alleviated CD8^(+)T cell suppression,resulting in tumor growth inhibition in the mouse HCC model.Together,our study unveils a mechanism by which GOLM1 induces CD8^(+)T cells suppression through promoting PD-L1 stabilization and transporting PD-L1 into TAMs with exosome dependent.Targeting PD-L1^(+)TAM could be a novel strategy to enhance the efficacy of anti-PD-L1 therapy in HCC. | Jinhong Chen Zhifei Lin Lu Liu Rui Zhang Yan Geng Minghao Fan Wenwei Zhu Ming Lu Lu Lu Huliang Jia Jubo Zhang Lun-Xiu Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 5 |

| 5 | Primary assessment of the diversity of Omicron sublineages and the epidemiologic features of autumn/winter 2022 COVID-19 wave in Chinese mainland显示文摘With the recent ongoing autumn/winter 2022 COVID-19 wave and the adjustment of public health control measures,there have been widespread SARS-CoV-2 infections in Chinese mainland.Here we have analyzed 369 viral genomes from recently diagnosed COVID-19 patients in Shanghai,identifying a large number of sublineages of the SARS-CoV-2 Omicron family.Phylogenetic analysis,coupled with contact history tracing,revealed simultaneous community transmission of two Omicron sublineages dominating the infections in some areas of China(BA.5.2 mainly in Guangzhou and Shanghai,and BF.7 mainly in Beijing)and two highly infectious sublineages recently imported from abroad(XBB and BQ.1).Publicly available data from August 31 to November 29,2022 indicated an overall severe/critical case rate of 0.035%nationwide,while analysis of 5706 symptomatic patients treated at the Shanghai Public Health Center between September 1 and December 26,2022 showed that 20 cases(0.35%)without comorbidities progressed into severe/critical conditions and 153 cases(2.68%)with COVID-19-exacerbated comorbidities progressed into severe/critical conditions.These observations shall alert healthcare providers to place more resources for the treatment of severe/critical cases.Furthermore,mathematical modeling predicts this autumn/winter wave might pass through major cities in China by the end of the year,whereas some middle and western provinces and rural areas would be hit by the upcoming infection wave in mid-to-late January 2023,and the duration and magnitude of upcoming outbreak could be dramatically enhanced by the extensive travels during the Spring Festival(January 21,2023).Altogether,these preliminary data highlight the needs to allocate resources to early diagnosis and effective treatment of severe cases and the protection of vulnerable population,especially in the rural areas,to ensure the country’s smooth exit from the ongoing pandemic and accelerate socio-economic recovery. | Gang Lu Yun Ling Minghao Jiang Yun Tan Dong Wei Lu Jiang Shuting Yu Fangying Jiang Shuai Wang Yao Dai Jinzeng Wang Geng Wu Xinxin Zhang Guoyu Meng Shengyue Wang Feng Liu Xiaohong Fan Saijuan Chen | 2023 | Frontiers of Medicine2023,17,4: | 1 |

| 11 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 14 | Solving multi-objective constrained minimum weighted bipartite assignment problem:a case study on energy-aware radio broadcast scheduling显示文摘This paper proposes a multi-objective constrained minimum weighted bipartite assignment problem(MCMWBAP),which is considered an extension of the classical bipartite matching problem(BMP).We first provide the formulation of the MCMWBAP and prove that it is an NP-hard combinatorial optimization problem.Based on this formulation,multi-objective energy-aware shortwave radio broadcast resource allocation problem(MSRBRAP)application is studied.The goal of this problem is to allocate radio programs to transmission devices to broadcast all radio programs felicitously with a maximized objective of total qualified monitoring sites and a minimized objective of energy consumption.Then,a novel multi-objective hybrid evolutionary algorithm(MOHEA),which is integrated with push and pull initialization,the dynamic resource allocation strategy,and the aggregate local search procedure,is developed to solve the problem.The proposed method is evaluated using two categories of benchmarks for MCMWBAP together with a real scenario case study for MSRBRAP.Furthermore,the key components of MOHEA are analyzed,and the experimental results demonstrate that MOHEA outperforms two classical multi-objective evolutionary algorithms(NSGA-II and MOEA/D),improving working efficiency. | Yupeng ZHOU Mingjie FAN Feifei MA Minghao YIN | 2022 | Science China(Information Sciences)2022,65,8: | 0 |
| 15 | SREBP2 inhibitor betulin sensitizes hepatocellular carcinoma to lenvatinib by inhibiting the mTOR/IL-1βpathway显示文摘Lenvatinib has become the first-line therapy in advanced hepatocellular carcinoma(HCC),but its efficacy is still limited because of the inevitable development of resistance.It has been reported that cellular cholesterol levels are associated with tyrosine kinase inhibitor(TKI)efficacy.Here,we show that betulin,a sterol regulatory elementbinding protein 2(SREBP2)inhibitor,markedly enhances the anti-tumor effect of lenvatinib in HCC both in vitro and in vivo.Our results also show that the combination treatment of lenvatinib and betulin synergistically inhibits the proliferation and clonogenicity of HCC cells.The mRNA and protein expressions of IL-1βare markedly decreased in HCC cells treated with betulin,while the sensitivity of HCC cells to lenvatinib is enhanced.Moreover,we find that the knockdown of IL-1βalso enhances the efficacy of lenvatinib,and recombinant IL-1βprotein rescues cell viability,which is reduced by lenvatinib in HCC cells.Further mechanistic studies indicate that betulin decreases the level of IL-1βin HCC cells by inhibiting the mTOR signaling pathway.Finally,the growth of the tumors in xenograft mouse models subjected to combination treatment is significantly suppressed.In summary,our study reveals that the SREBP2 inhibitor betulin sensitizes hepatocellular carcinoma to lenvatinib by inhibiting the mTOR/IL-1βpathway,which may be a promising therapeutic strategy for patients with HCC. | Minghao Fan Zhenmei Chen Weiqing Shao Yiran Chen Zhifei Lin Chenhe Yi Yitong Li Lu Lu Yu Zhou Jing Lin | 2023 | Acta Biochimica et Biophysica Sinica2023,55,9: | 0 |
