4Stable knockdown of heparanase expression in gastric cancer cells in vitro显示文摘AIM:To develop short hairpin RNA(shRNA)against heparanase,and to determine its effects on heparanase expression and the malignant characteristics of gastric cancer cells. METHODS:Heparanase-specific shRNA was constructed and transferred into cultured the gastric cancer cell line SGC-7901.Stable subclonal cells were screened by G418 selection.Heparanase expression was measured by reverse transcriptase-polymerase chain reaction(RT-PCR),real-time quantitative PCR and Western blotting.Cell proliferation was detected by 2-(4,5-dimethyltriazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT)colorimetry and colony formation assay. The in vitro invasiveness and metastasis of cancer cells were measured by cell adhesion assay,wound healingassay and matrigel invasion assay.The angiogenesis capabilities of cancer cells were measured by tube formation of endothelial cells. RESULTS:Stable transfection of heparanase-specific shRNA,but not of scrambled shRNA and mock vector,resulted in reduced mRNA and protein levels of heparanase.The shRNA-mediated knockdown of heparanase did not affect the cellular proliferation of SGC-7901 cells.However,the in vitro invasiveness and metastasis of cancer cells were decreased after knockdown of heparanase.Moreover,transfection of heparanase-specific shRNA decreased the in vitro angiogenesis capabilities of SGC-7901 cells. 5The 5.5 cal ka BP climate event, population growth, circumscription and the emergence of the earliest complex societies in China显示文摘The emergence of complex society is a milestone in the history of human society evolution. China is one of the few regions in the world where the earliest complex society appeared; however, its driving mechanisms remain unresolved. On the base of available evidence from both archaeology and Holocene climate, in combination with agency theory, this study attempts to address the driving mechanisms for the simultaneous emergence of complex societies in multiple areas of China around 5.5 cal ka BP. It is hypothesized that three factors, including climate change, population growth, and circumscription, jointly act and cause regional population-resource imbalance and trigger inter-group conflicts and wars. Such competitions provide the opportunity for some power-pursuing agents to break the restriction of social leveling mechanism and to become the centralized decision-making leaders, which further lead to the emergence of incipient large-scale complex societies. Increase in extreme climate events during 6.0–5.0 cal ka BP cooling period causes frequent occurrence of resource stress and increase in the frequency of inter-group competitions, which creates conditions for the legitimation, institutionalization, and persistence of centralized leadership, and finally leads to the formation of persistent institutionalized inequity. 6Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 7Geochemical Characters of Water Coproduced with Coalbed Gas and Shallow Groundwater in Liulin Coalfield of China显示文摘To reveal the geochemical characters of water coproduced with coalbed gas and shallow groundwater,water samples were collected from 12 wells of coalbed methane and 7 wells of shallow groundwater.The pH,CODMn,fCO2,total dissolved solids(TDS),total hardness,and concentrations of metasilicic acid,sodium and kalium,calcium ion,magnesium ion,ammonium iron,bicarbonate ion,carbonate,chloride,sulfate ion,nitrate ion,fluoride,lithium,zinc,nickel,manganese,iron,boron,barium,etc.of the samples were measured.Research results showed the following:(1)Concentrations of TDS,chloride,fluoride,sodium and kalium,ammonium,iron,and barium in the water coproduced with coalbed gas exceeded the national standards of China;however,physical,chemical,and biological properties of shallow groundwater could meet the national standard.(2)The water produced from coalbed contained mainly Na–Cl·HCO3,with average TDS of 4588.5 ppm,whereas shallow groundwater contained a mixture of chemicals including Na·Mg·Ca-HCO3·SO4and Na·MgHCO3·SO4,with average TDS of 663.8 ppm.(3)In general,it was observed that bicarbonate and sodium accumulated in a reducing environment and deeper system,while depletion of hydrogen ions and dissolution of sulfate,calcium,and magnesium occurred in a redox environment and shallow system.(4)Sodium and kalium,ammonium,chloride,and bicarbonate ions were the main ions found in the study area.YANG Mei JU Yiwen LIU Guijian TONG Li KANG Yu HOU Quanlin 2013Acta Geologica Sinica(English Edition)2013,87,6:6

11Environmental and Psycho-social Factors Related to Prostate Cancer Risk in the Chinese Population:a Case-control Study显示文摘Objective To study the risk environmental and psycho-social factors associated to prostate cancer(PCa) in Chinese population. Methods 250 PCa patients and 500 controls were enrolled in this case-control study. Information was collected and logistic regression analysis was used to estimate the odds ratios(OR) and 95% confidence intervals(95% CI) for relationship between lifestyle, eating habits and psycho-social factors with PCa risk. Results Green vegetables and green tea were associated with a decreased risk of PCa(OR=0.39, 95% CI: 0.28-0.53; OR=0.59, 95% CI: 0.40-0.87, respectively). Family history of PCa(OR=7.16, 95% CI: 2.01-25.49), history of prostate diseases(OR=2.28, 95% CI: 1.53-3.41), alcohol consumption(OR=1.97, 95% CI: 1.33-2.90), red meat consumption(OR=1.74, 95% CI: 1.20-2.52), barbecued(OR=2.29, 95% CI: 1.11-4.73) or fried(OR=2.35, 95% CI: 1.24-4.43) foods were related with increased PCa risk. Negative psycho-social factors including occupational setbacks(OR=1.61, 95% CI: 1.00-2.59), marital separation(OR=1.94, 95% CI: 1.29-2.91), self-contained suffering(OR=2.37, 95% CI: 1.58-3.55), and high sensitivity to the personal comments(OR=1.73, 95% CI: 1.18-2.54) were related to PCa. Conclusion Regular consumption of green vegetables and green tea may suggest protective effects on PCa. Alcohol consumption, red meat consumption and barbecued or fried foods were associated with PCa. 12Increased N-terminal pro-brain natriuretic peptide level predicts atrial fibrillation after surgery for esophageal carcinoma显示文摘AIM: To evaluate the value of plasma N-terminal pro- brain natriuretic peptide (NT-proBNP) level for predicting postoperative atrial fibrillation (AF) in patients undergoing surgery for esophageal carcinoma. METHODS: NT-proBNP levels were measured in 142 patients 24 h before and 1 h after surgery for esophageal carcinoma. All patients having a preoperative cardiac diagnosis by electrocardiogram (ECG), remained under continuous monitoring for at least 48 h after surgery, and then underwent clinical cardiac evaluation until discharge. RESULTS: Postoperative AF occurred in 11 patients (7.7%). AF patients were significantly older (69.6 ± 12.2 years vs 63.4 ± 13.3 years, P = 0.031) than non-AF patients. There were no significant differences in history of diabetes mellitus, sex distribution, surgical approach, anastomosis site, intraoperative hypotension and postoperative fever. The preoperative plasma NT-proBNP level was significantly higher in patients who developed postoperative AF (121.3 ± 18.3 pg/mL vs 396.1 ± 42.6 pg/mL, P = 0.016). After adjustment for age, gender, chronic obstructive pulmonary disease (COPD), history of cardiac diseases, hypertension, postoperative hypoxia and thoracic-gastric dilation, NT-proBNP levels were found to be associated with the highest risk factor for postoperative AF (odds ratio = 4.711, 95% CI = 1.212 to 7.644, P = 0.008).CONCLUSION: An elevated perioperative plasma BNP level is a strong and independent predictor of postoperative AF in patients undergoing surgery for esophageal carcinoma. 13Lamellar morphology of directional solidified Ti–45Al–6Nb–xW alloys显示文摘Directional solidification of Ti-45Al-6Nb-xW(x=0,0.4,0.8;at%) alloys was performed by Bridgeman method with the stable growth rate of 5,10,and 20 μm·s^(-1).The differential scanning calorimeter(DSC) results indicate that both the eutectic temperature and the transition temperature of α(Ti_3Al) to γ(TiAl) increase with W content increasing from 0 at%to 0.8 at%.For the stable growth rate of 10 μm·s^(-1),the orientations of α_2(Ti_3Al)/γ(TiAl)lamellae change from 45°(0 at%W) to 0° and near 0°(0.4at%and 0.8 at%W) to the crystal growth direction,and the spacing of α_2/γ lamellae decreases with W content increasing from 0 at%to 0.4 at%,while it increases when W content is 0.8 at%.With the increase in growth rate from5 to 10 and 20 μm·s^(-1),the lamellar spacing of α_2/γbecomes smaller,and the lamellar thickness becomes more uniform.Zhi-Yuan Hou Yong-Sheng Li Hao-Jie Mei Kai Hu Guang Chen 2016Rare Metals2016,35,1:4

17Gap-free genome assembly and comparative analysis reveal the evolution and anthocyanin accumulation mechanism of Rhodomyrtus tomentosa显示文摘Rhodomyrtus tomentosa is an important f leshy-fruited tree and a well-known medicinal plant of the Myrtaceae family that is widely cultivated in tropical and subtropical areas of the world.However,studies on the evolution and genomic breeding of R.tomentosa were hindered by the lack of a reference genome.Here,we presented a chromosome-level gap-free T2T genome assembly of R.tomentosa using PacBio and ONT long read sequencing.We assembled the genome with size of 470.35 Mb and contig N50 of∼43.80 Mb with 11 pseudochromosomes.A total of 33382 genes and 239.31 Mb of repetitive sequences were annotated in this genome.Phylogenetic analysis elucidated the independent evolution of R.tomentosa starting from 14.37MYA and shared a recent WGD event with other Myrtaceae species.We identified four major compounds of anthocyanins and their synthetic pathways in R.tomentosa.Comparative genomic and gene expression analysis suggested the coloring and high anthocyanin accumulation in R.tomentosa tends to be determined by the activation of anthocyanin synthesis pathway.The positive selection and up-regulation of MYB transcription factors were the implicit factors in this process.The copy number increase of downstream anthocyanin transport-related OMT and GST gene were also detected in R.tomentosa.Expression analysis and pathway identification enriched the importance of starch degradation,response to stimuli,effect of hormones,and cell wall metabolism during the f leshy fruit development in Myrtaceae.Our genome assembly provided a foundation for investigating the origins and differentiation of Myrtaceae species and accelerated the genetic improvement of R.tomentosa.angping Li Shiqiang Xu Zitong Xiao Jingming Wang Yu Mei Haifei Hu Jingyu Li Jieying Liu Zhuangwei Hou Junliang Zhao Shaohai Yang Jihua Wang 2023Horticulture Research2023,10,3:3
18ADSORBENTS USED IN THE CLEARANCE OF ENDOTOXIN显示文摘A series of modified poly (methyl methacrylate, PMMA) resins were prepared and compared their adsorption abilities to endotoxin. The results showed that adsorbents, which were grafted with tertiary amine and long spacing arms, had the best adsorption capacities and good blood compatibility. It is hopeful to be used as adsorbent in hemoperfusion for clinical clearance of endotoxin. 19Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research.
