| 3 | Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. | 5 | The fabrication of biomimetic biphasic CAN-PAC hydrogel with a seamless interfacial layer applied in osteochondral defect repair显示文摘Cartilage tissue engineering based on biomimetic scaffolds has become a rapidly developing strategy for repairing cartilage defects. In this study, a biphasic CAN-PAC hydrogel for osteochondral defect(OCD)regeneration was fabricated based on the density difference between the two layers via a thermally reactive,rapid cross-linking method. The upper hydrogel was cross-linked by CSMA and NIPAm, and the lower hydrogel was composed of PECDA, AAm and PEGDA. The interface between the two layers was first grafted by the physical cross-linking of calcium gluconate and alginate, followed by the chemical cross-linking of the carbon-carbon double bonds in the other components. The pore sizes of the upper and lower hydrogels were ~ 187.4 and ~ 112.6 μm, respectively. The moduli of the upper and lower hydrogels were ~ 0.065 and~ 0.261 MPa. This prepared bilayer hydrogel exhibited the characteristics of mimetic composition, mimetic structure and mimetic stiffness, which provided a microenvironment for sustaining cell attachment and viability. Meanwhile, the biodegradability and biocompatibility of the CAN-PAC hydrogel were examined in vivo. Furthermore, an osteochondral defect model was developed in rabbits, and the bilayer hydrogels were implanted into the defect. | 6 | Co-occurrence Patterns of Above-ground and Below-ground Mite Communities in Farmland of Sanjiang Plain, Northeast China显示文摘One of the fundamental questions in community ecology is whether communities are random or formed by deterministic mechanisms. Although many efforts have been made to verify non-randomness in community structure, little is known with regard to co-occurrence patterns in above-ground and below-ground communities. In this paper, we used a null model to test non-randomness in the structure of the above-ground and below-ground mite communities in farmland of the Sanjiang Plain, Northeast China. Then, we used four tests for non-randomness to recognize species pairs that would be demonstrated as significantly aggregated or segregated co-occurrences of the above-ground and below-ground mite communities. The co-occurrence pattern of the above-ground mite community was significantly non-random in October, suggesting species segregation and hence interspecific competition. Additionally, species co-occurrence patterns did not differ from randomness in the above-ground mite community in August or in below-ground mite communities in August and October. Only one significant species pair was detected in the above-ground mite community in August, while no significant species pairs were recognized in the above-ground mite community in October or in the below-ground mite communities in August and October. | 7 | Ability to detect weak effective seismic signals by utilizing chaotic vibrator system显示文摘Taking the advantage of CVS (Chaotic Vibrator System) sensitivity of large-scale periodic phase-state response to quasi-periodic or periodic signals,a series of numerical experiments were made to understand the ability of CVS to detect weak effective seismic signals in the common-shot seismic record distorted by strong stochastic noise. The re-sults demonstrate that the large-scale periodic phase-states of CVS are correlated with the signal composition of the quasi-periodic wavelet sequence constructing from horizontal moveout of seismic events,noise strength and the noise distortion de-gree to signal. For the same kind of events,the higher the noise distortion degree is,the lower the detectable SNR can be reached by CVS. For seismic data with the same noise distortion degree,the closer the scanning seismic velocity (the trial moveout ve-locity) approaches to the accurate velocity,the higher the detectable SNR can be reached by CVS. | 8 | Synthesis of TiO_(2) nanotubes film and its light scattering property显示文摘TiO2 nanotubes with diameters of 10 nm and lengths up to 600 nm were fabricated by directly using com-mercial TiO2 powders P25 as the precursors via sonica-tion-hydrothermal combination approach. TiO2 nanotubes were characterized by means of X-ray powder diffractometer (XRD), scanning electron microscope (SEM), selected area electron diffraction pattern (SAED) and transmission elec-tron microscope (TEM). The light scattering property of film electrodes modified with TiO2 nanotubes was studied and revealed that TiO2 nanotubes can be used as the light scat-tering centers to increase the light absorption in dye- sensi-tized solar cells. | 11 | Nucleic acids and analogs for bone regeneration显示文摘With the incidence of different bone diseases increasing, effective therapies are needed that coordinate a combination of various technologies and biological materials. Bone tissue engineering has also been considered as a promising strategy to repair various bone defects. Therefore, different biological materials that can promote stem cell proliferation, migration, and osteoblastic differentiation to accelerate bone tissue regeneration and repair have also become the focus of research in multiple fields. Stem cell therapy, biomaterial scaffolds, and biological growth factors have shown potential for bone tissue engineering; however, off-target effects and cytotoxicity have limited their clinical use. The application of nucleic acids(deoxyribonucleic acid or ribonucleic acid)and nucleic acid analogs(peptide nucleic acids or locked nucleic acids), which are designed based on foreign genes or with special structures, can be taken up by target cells to exert different effects such as modulating protein expression, replacing a missing gene, or targeting specific gens or proteins. Due to some drawbacks, nucleic acids and nucleic acid analogs are combined with various delivery systems to exert enhanced effects, but current studies of these molecules have not yet satisfied clinical requirements. In-depth studies of nucleic acid or nucleic acid analog delivery systems have been performed, with a particular focus on bone tissue regeneration and repair. In this review, we mainly introduce delivery systems for nucleic acids and nucleic acid analogs and their applications in bone repair and regeneration. | 13 | Mild hypothermia effects on matrix metalloproteinase-9 expression in the perihematomal region of rats following experimental intracerebral hemorrhage显示文摘BACKGROUND: Matrix metalloproteinase-9 (MMP-9) expression increases with intracerebral hemorrhage, and participates in the pathophysiological processes of secondary brain injury after intracerebral hemorrhage. OBJECTIVE: To investigate the effects of mild hypothermia on MMP-9 expression and brain edema in the perihematomal region of experimental intracerebral hemorrhage rats. DESIGN, TIME AND SETTING: The randomized, controlled experiment was performed at the Central Laboratory of Shandong Provincial Hospital between May and September 2007. MATERIALS: Seventy-two, Wistar, male rats, 12-weeks old, were used for this study. Rabbit anti-MMP-9 primary antibody was purchased from Boster, China. METHODS: Wistar rats were equally and randomly divided into normothermia and mild hypothermia groups. The two groups each comprised control, 6-hour intracerebral hemorrhage, 24-hour intracerebral hemorrhage, 48-hour intracerebral hemorrhage, 72-hour intracerebral hemorrhage, and 1-week intracerebral hemorrhage subgroups, with six rats in each subgroup. Rat models of intracerebral hemorrhage were established by injecting 100 μL of autologous blood into the rat caudate nucleus. Rats in the mild hypothermia group received four hours of local mild hypothermia immediately following the injection. Intracerebral temperature was maintained at (33 ± 0.5) ℃. Subsequently, intracerebral temperature was spontaneously recovered at 25 ℃. Rats in the control subgroup were not injected with autologous blood and received only with intracerebral hemorrhage. MAIN OUTCOME MEASURES: Brain water content and MMP-9 expression surrounding the hematoma region. RESULTS: MMP-9 expression increased at 6 hours, and brain edema reached a peak at 48 hours after intracerebral hemorrhage. MMP-9 expression was significantly decreased in the mild hypothermia group compared with the normothermia group at each time point (P < 0.05). | 20 | Interception, degradation and contributions of terrestrial organic carbon obtained from lignin analysis in Wujiang River, southwest China显示文摘Understanding the fate of terrestrial organic carbon(OC)in a cascade impoundment system is critical for recognizing the role of carbon sink for reservoirs.Surface sediments collected from eight cascade reservoirs across the Wujiang River,southwestern China,were analyzed for elemental and stable carbon isotopic(δ^13C)composition,and lignin phenols(∑8 andΛ8)to investigate the spatial distribution,contribution,origin and degradation of sedimentary terrestrial OC.The values of total organic carbon(TOC)and∑8 exhibited a remarkable reduction along the upstream-downstream transect suggesting the trapping effect of cascade-damming.A relatively broad range ofδ^13C(-26.61 to-25.54%,95%CI)and C/N(6.80-18.20)indicated mixed allochthonous/autochthonous OC sources in surface sediments.The quantitative simulation indicates that the OC of the sediments mainly was derived from terrestrial organic matter.Soilderived OC rather than C_(3)vascular plant-derived OC makes a major contribution to sedimentary terrestrial OC in reservoirs on karst terrain.As evidenced by lignin compositions andδ^13C,the predominant vascular plant origins of terrestrial OC along the Wujiang River are non-woody angiosperm C_(3)plants.The aged reservoirs showed a trend of increasing contribution of autochthonous OC,which potentially weaken the role of carbon sink for reservoirs.The relationship between runoff inputs,watershed area/water surface area ratios,and water residence time and Λ8 were explored,indicating the natural and anthropogenic influences on terrestrial OC remains very complex in a cascade-damming river. | Xin Lin Yujie Wang Jinhua Zhang Ming Yang Xueping Chen Fushun Wang Jing Ma | 2021 | Acta Geochimica2021,40,6: | 1 |
