9Knockdown of insulin-like growth factor 2 gene disrupts mitochondrial functions in the liver显示文摘Even though insulin-like growth factor 2(IGF2)has been reported to be overexpressed in nonalcoholic fatty liver disease(NAFLD),its role in the progression of NAFLD and the potential mechanism remain largely unclear.Using in vitro models,we found that IGF2 was the key overexpressed gene in steatosis,suggesting a possible association between IGF2 and NAFLD.Interestingly,loss-of-function experiments revealed that inhibition of IGF2 protein impaired mitochondrial biogenesis and respiration.It additionally disrupted the expression changes of mitochondrial fusion and fission-related proteins necessary in maintaining mitochondrial homeostasis.Consistently,IGF2 knockdown reduced the mitochondrial membrane potential and increased the production of reactive oxygen species.Mechanistically,IGF2 regulates mitochondrial functions by modulating the expression of SIRT1 and its downstream gene PGCla.This research opens a new frontier on the role of IGF2 in energy metabolism,which potentially participates in the development of NAFLD.As such,IGF2 is a potential therapeutic target against NAFLD.Weiwei Gui Yiyi Zhu Shuiya Sun Weifen Zhu Bowen Tan Hanxin Zhao Chengxin Shang Fenping Zheng Xihua Lin Hong Li 2021Journal of Molecular Cell Biology2021,13,8:1
10Femtosecond laser micro-nano processing for boosting bubble releasing of gas evolution reactions显示文摘Coupling effect of chemical composition and physical structure is a key factor to construct superaerophobic electrodes.Almost all reports about superaerophobic electrodes were aimed at precisely controlling morphology of loaded materials(constructing specific structure)and ignored the due role of substrate.Nevertheless,in this work,by using high precision and controllable femtosecond laser,hierarchical micro-nano structures with superaerophobic properties were constructed on the surface of silicon substrate(fs-Si),and such special super-wettability could be successfully inherited to subsequent self-supporting electrodes through chemical synthesis.Femtosecond laser processing endowed electrodes with high electrochemical surface area,strong physical structure,and remarkable superaerophobic efficacy.As an unconventional processing method,the reconstructed morphology of substrate surface bears the responsibility of superaerophobicity,thus liberating the structural constraints on loaded materials.Since this key of coupling effect is transferred from the loaded materials to substrate,we provided a new general scheme for synthesizing superaerophobic electrodes.The successful introduction of femtosecond laser will open a new idea to synthesize self-supporting electrodes for gas-involving reactions.Shuai Zhang Lishuang Xu Jie Wu Ying Yang Chengxin Zhang Haiyan Tao Jingquan Lin Licheng Huang Wencheng Fang Keying Shi Xiangting Dong 2022Nano Research2022,15,2:0

12Berberine enhances the anti-hepatocellular carcinoma effect of NK92-MI cells through inhibiting IFN-gamma-mediated PD-L1 expression显示文摘Background and aims:Berberine is one of the most promising clinically tested natural alkaloids,and immunotherapy using natural killer(NK)cells is a potentially effective treatment for hepatocellular carcinoma(HCC).This study aims to elucidate the effect of berberine on the anti-HCC effect of NK92-MI cells.Materials and methods:Human HCC SMMC-7721 and Hep3B cells were co-incubated with NK92-MI cells,berberine(60 or 80 mmol/L),or their combination for 36 h.To evaluate the killing effect of NK92-MI cells on HCC cells,the release of lactate dehydrogenase(LDH)in HCC cells was measured.The anti-tumor effects of berberine,NK92-MI cells,and their combinations were evaluated by MTS,EdU,Tunel,and Western blot assays.A male BALB/c nude mouse subcutaneous tumor model was used to investigate the anti-HCC effect of berberine and NK92-MI cells in vivo.Results:The LDH assay showed that berberine enhanced the cytotoxicity of NK92-MI cells on HCC cells.The combination of berberine and NK92-MI cells demonstrated more obvious anti-HCC effect than did the berberine or NK92-MI single treatment in inhibiting cell proliferation and inducing apoptosis both in vitro and in vivo.Mechanistically,the expression of programmed cell death-ligand 1(PD-L1)in HCC cells was upregulated after co-culture with NK-92MI cells.PD-L1 expression was knocked down,thereby inhibiting the proliferation and promoting apoptosis of HCC cells,and inhibited by berberine that blocked the secretion of interferon gamma(IFN-g),thereby enhancing the anti-tumor effect of NK92-MI cells.Conclusions:Current data show that the immunomodulatory role of berberine is to enhance the cytotoxic effect of NK92-MI cells and inhibit tumor immune escape by reducing the expression of PD-L1.Our study provides a rationale for the clinical application of berberine in combination with NK cells for the treatment of HCC.Kunyuan Wang Chengxin Gu Ganxiang Yu Jiaen Lin Zhilei Wang Qianting Lu Yangzhi Xu Dan Zhao Xiaofeng Jiang Weijian Mai Shiming Liu Hui Yang 2022Liver Research2022,6,3:0
13Application of Brain-Computer-Interface in Awareness Detection Using Machine Learning Methods显示文摘The awareness detection in patients with disorders of consciousness currently relies on behavioral observations and CRS-R tests,however,the misdiagnosis rates have been relatively high.In this study,we applied brain-computer interface(BCI)to awareness detection with a passive auditory stimulation paradigm.12 subjects with normal hearing were invited to collect electroencephalogram(EEG)based on a BCI communication system,in which EEG signals are transmitted wirelessly.After necessary preprocessing,RBF-SVM and EEGNet were used for algorithm realization and analysis.For a single subject,RBF-SVM can distinguish his(her)name stimuli awareness with classification accuracies ranging from 60-95%.EEGNet was used to learn all subjects'data and improved accuracy to 78.04%for characteristics finding and model generalization.Moreover,we completed the supplementary analysis work from the time domain and time-frequency domain.This study applied BCI communication to human awareness detection,proposed a passive auditory paradigm,and proved the effectiveness,which could be an inspiration for brain,mental or physical diseases diagnosis and detection.Kaiqiang Feng Weilong Lin Feng Wu Chengxin Pang Liang Song Yijia Wu Rong Cao Huiliang Shang Xinhua Zeng 2022China Communications2022,19,6:0

15Plant and fungal species interactions differ between aboveground and belowground habitats in mountain forests of eastern China显示文摘Plant and fungal species interactions drive many essential ecosystem properties and processes;however,how these interactions differ between aboveground and belowground habitats remains unclear at large spatial scales.Here,we surveyed 494 pairwise fungal communities in leaves and soils by Illumina sequencing,which were associated with 55 woody plant species across more than 2,000-km span of mountain forests in eastern China.The relative contributions of plant,climate,soil and space to the variation of fungal communities were assessed,and the plant-fungus network topologies were inferred.Plant phylogeny was the strongest predictor for fungal community composition in leaves,accounting for 19.1%of the variation.In soils,plant phylogeny,climatic factors and soil properties explained 9.2%,9.0%and 8.7%of the variation in soil fungal community,respectively.The plant-fungus networks in leaves exhibited significantly higher specialization,modularity and robustness(resistance to node loss),but less complicated topology(e.g.,significantly lower linkage density and mean number of links)than those in soils.In addition,host/fungus preference combinations and key species,such as hubs and connectors,in bipartite networks differed strikingly between aboveground and belowground samples.The findings provide novel insights into cross-kingdom(plant-fungus)species co-occurrence at large spatial scales.The data further suggest that community shifts of trees due to climate change or human activities will impair aboveground and belowground forest fungal diversity in different ways.Teng Yang Leho Tedersoo Pamela S.Soltis Douglas E.Soltis Miao Sun Yuying Ma Yingying Ni Xu Liu Xiao Fu Yu Shi Han-Yang Lin Yun-Peng Zhao Chengxin Fu Chuan-Chao Dai Jack A.Gilbert Haiyan Chu 2023Science China(Life Sciences)2023,66,5:0
