| 1 | Wogonoside inhibits tumor growth and metastasis in endometrial cancer via ER stress-Hippo signaling axis显示文摘Wogonoside,a bioactive flavonoid component derived from Scutellaria baicalensis Georgi,has been reported to inhibit tumor growth in mice bearing various types of cancer cells such as breast cancer,lung cancer,and leukemia cells.However,whether wogonoside could inhibit tumor growth of endometrial cancer has not been elucidated.In this study,we explored the function of wogonoside on tumor growth and the underlying mechanism on endometrial cancer.Firstly,we investigated the effect of wogonoside on endometrial cancer cells and found that wogonoside could significantly decrease cell proliferation and metastasis.Mechanistically,wogonoside could aggravate the extent of ER stress and upregulate the phosphorylation level of Mammalian Ste20-like kinase 1,leading to the activation of the Hippo signaling pathway.Taken together,in vitro and in vivo data demonstrated that wogonoside could be a potent inducer of ER stress and could be further developed into a promising therapy for endometrial cancer. | Shaorong Chen Zhuna Wu Yumin Ke Pingping Shu Caihong Chen Ruying Lin Qirong Shi | 2019 | Acta Biochimica et Biophysica Sinica2019,51,11: | 8 |
| 2 | Preparation, characterization, antimicrobial and cytotoxicity studies of copper/zincloaded montmorillonite显示文摘Background: A series of modified montmorillonites(Mt) including zinc-loaded Mt(Zn-Mt), copper-loaded Mt(Cu-Mt), copper/zinc-loaded Mt with different Cu/Zn ratio(Cu/Zn-Mt-1, Cu/Zn-Mt-2, Cu/Zn-Mt-3) were prepared by an ion-exchange reaction, and characterized using X-ray diffraction(XRD), fourier transformed infrared spectroscopy(FTIR) and transmission electron microscopy(TEM). The specific surface areas, antimicrobial activity and cytotoxicity of the modified Mt were investigated.Results: In the modified Mt, hydrated Cu ions and Zn ions were exchanged in the interlayer space of Mt and the particles were irregular shapes. The results showed that Cu/Zn-Mt enhanced antibacterial and antifungal activity compared with Zn-Mt and Cu-Mt possibly due to the synergistic effect between Cu and Zn. Among the Cu/Zn-Mt with different Cu/Zn raitos, Cu/Zn-Mt with a Cu/Zn ratio of 0.98 or 0.51 showed higher antimicrobial activity against gram-negative bacteria(Escherichia coli), gram-positive bacteria(Staphylococcus aureus), fungi(Candida albicans).Moreover, the antimicrobial activity of Cu/Zn-Mt was correlated with its specific surface area. | 3 | Similar Ability of FbaA with M Protein to Elicit Protective Immunity Against Group A Streptococcus Challenge in Mice显示文摘Group A streptococcus(GAS),an important human pathogen,can cause various kinds of infections including superficial infections and potentially lethal infections,and the search for an effective vaccine to prevent GAS infections has been ongoing for many years. This paper compares the immunogenicity and immunoprotection of FbaA(an Fn-binding protein expressed on the surface of GAS) with that of M protein,the best immunogen of GAS. Assay for immune response showed that FbaA,similar to M protein,could induce protein-specific high IgG titer in BALB/c mice. Furthermore,following GAS challenge,the mice immunized with FbaA showed the same protective rate as those with M protein. | 4 | Dihydromyricetin promotes apoptosis, suppresses proliferation and tumor necrosis factor-α-mediated nuclear factor kappa-B activation in nasopharyngeal carcinoma CNE-2 cells显示文摘OBJECTIVE: To investigate the efficacy of dihydromyricetin(DMY) on nasopharyngeal carcinoma(NPC) cell proliferation, apoptosis and to reveal the underlying mechanism in vitro experiments.METHODS: The CNE-2 cell line was treated with different concentrations of DMY and the effects of DMY on cell viability and proliferation were evaluated using cell counting kit-8(CCK-8) assay and plate colony formation assay. Cellular apoptosis was detected by flow cytometry following Annexin V fluorescein isothiocyanate/propidine iodide staining.Nuclei morphology was observed under a fluorescence microscope following Hoechst 333258 staining. The expression of phosphorylated inhibitor of nuclear factor kappa-B kinase subunit beta(p-IKKβ), phosphorylated inhibitor of nuclear factor kappa-B kinase subunit alpha(p-IKKα), inhibitor of nuclear factor kappa-B alpha(IκB-α), nuclear factor kappa-B(NF-κB)/p65 was examined by Western blot analysis and the nuclear translocation of NF-κB/p65 was observed using a confocal laser scanning microscopy.RESULTS: DMY inhibited the proliferative capability and colony formation of NPC CNE-2 cells. Meanwhile, DMY induced apoptosis of CNE-2 cells in a dose and time-dependent manner via upregulating B-cell lymphoma-2 associated X, but downregulating B-cell lymphoma-2 and pro-caspase-3. Importantly, we found that DMY suppressed tumor necrosis factor alpha(TNF-α)-mediated NF-κB activation via inhibiting p-IKKβ, p-IKKα and blocking NF-κB subunit p65.CONCLUSION: Our experiments demonstrated that DMY had significant antiproliferative and apoptosisinducing effects on CNE-2 cells. Additionally, DMY promoted inactivation of p-IKKβ, p-IKKα,and blocked the nuclear translocation of NF-κB subunit p65. | 7 | A rare variant in COL11A1 is strongly associated with adult height in Chinese Han population显示文摘Human height is a highly heritable trait in which multiple genes are involved. Recent genome-wide association studies(GWASs) have identified that COL11A1 is an important susceptibility gene for human height. To determine whether the variants of COL11A1 are associated with adult and children height,we analyzed splicing and coding single-nucleotide variants across COL11A1 through exome-targeted sequencing and two validation stages with a total 20,426 Chinese Han samples. A total of 105 variants were identified by exome-targeted sequencing, of which 30 SNPs were located in coding region. The strongest association signal was chr1_103380393 with P value of 4.8 * 10^(-7). Chr1_103380393 also showed nominal significance in the validation stage(P = 1.21 * 10^(-6)). Combined analysis of 16,738 samples strengthened the original association of chr1_103380393 with adult height(P_(combined)= 3.1 * 10^(-8)), with an increased height of 0.292sd(standard deviation) per G allele(95% CI:0.19-0.40). There was no evidence(P = 0.843) showing that chr1_103380393 altered child height in 3688 child samples. | 10 | Experimental Characterization and Model Verification of Thermal Conductivity from Mesoporous to Macroporous SiOC Ceramics显示文摘Based on the chemical cross-linking method,this paper uses polydimethylsiloxane with various viscosities of 10 cSt,20 cSt,50 cSt,and 100 cSt to synthesize mesoporous and macroporous SiOC ceramics.Their thermal conductivities are measured by using 3ωmethod with high accuracy.Three typical models for their thermal conductivities,i.e.,series model(SM),maxwell-Eucken 1 model(ME1),and effective medium theory(EMT)model,are utilized to derive the empirical formula through the multi-parameter linear optimization algorithm,which agrees well with the experimental results.The effects of pore size and specific surface area on the overall thermal conductivity of the porous structure are explored.Interestingly,it is found that the thermal conductivities of both gas phase and solid phase inside the porous structure increase with the increasing pore size at the nanometer scale,but the overall thermal conductivity of the porous structure decreases with the increasing pore size.Scanning electron microscopy graphs corroborate that the extension of the heat transfer route and the barrier of more pores between the solid phases together cause the reduction of the gas-solid coupling thermal conductivity of SiOC ceramics with larger pore size.On the contrary,the miniaturization of individual particles through modulating the synthesis parameters can increase the number of small pores in the sample itself to meet the pseudo-lattice vibration conditions,which results in the increment of the gas-solid coupling thermal conductivity and the overall thermal conductivity of the porous structure.These findings would provide meaningful guidance for designing SiOC porous ceramic super-insulation materials with extremely low thermal conductivity. | QIU Lin DU Yanbo BAI Yangyang FENG Yanhui ZHANG Xinxin WU Jin WANG Xiaotian XU Caihong | 2021 | Journal of Thermal Science2021,30,2: | 0 |
