1Mechanisms of resistance to EGFR tyrosine kinase inhibitors显示文摘Since the discovery that non-small cell lung cancer(NSCLC) is driven by epidermal growth factor receptor(EGFR) mutations, the EGFR tyrosine kinase inhibitors(EGFR-TKIs, e.g., ge fi tinib and elrotinib) have been effectively used for clinical treatment. However, patients eventually develop drug resistance. Resistance to EGFR-TKIs is inevitable due to various mechanisms, such as the secondary mutation(T790M), activation of alternative pathways(c-Met, HGF, AXL), aberrance of the downstream pathways(K-RAS mutations, loss of PTEN), impairment of the EGFR-TKIs-mediated apoptosis pathway(BCL2-like 11/BIM deletion polymorphism), histologic transformation, ATP binding cassette(ABC) transporter effusion, etc. Here we review and summarize the known resistant mechanisms to EGFR-TKIs and provide potential targets for development of new therapeutic strategies.Lihua Huang Liwu Fu 2015Acta Pharmaceutica Sinica B2015,5,5:42

3MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells.There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting.After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide,secondary neurosphere assay,soft agar colony assay and MTT assay were performed.Compared with U87 cells,the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar,and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony,and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells.Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu 2010The Journal of Biomedical Research2010,24,6:14
4Ecological-hydrological processes in arid environment: Past, present and future显示文摘Ecohydrology, aiming to study the interactions between terrestrial ecological systems and hydrological cycles as well as their impacts on water management, has been an emerging interdisciplinary research field since the 20 th century. It hosts both natural and human regulated processes that are potentially coupled in complex ways. Understanding the ecological-hydrological processes, the fundamental mechanisms and the connections between them is critical since these processes are not isolated but integrated to impact basin-scale hydrological and biogeochemical functioning of a larger river system, especially in arid environment where water resources are considered to be the source of life. Thus, research on ecological-hydrological processes in arid environment is not only a scientific focus area but also important to sustainable development. Research projects and initiatives involved in observation, measurement, modeling and data assimilation have been well-developed for those purposes over the past 20 years. This review summarizes the historical development of ecohydrology science in China and the state-of-the-art tools available in the research framework. Five grand scientific challenges are listed as prospects and exciting opportunities for the scientific community. 8Hierarchical Honeycomb-Structured Electret/Triboelectric Nanogenerator for Biomechanical and Morphing Wing Energy Harvesting显示文摘Flexible,compact,lightweight and sustainable power sources are indispensable for modern wearable and personal electronics and small-unmanned aerial vehicles(UAVs).Hierarchical honeycomb has the unique merits of compact mesostructures,excellent energy absorption properties and considerable weight to strength ratios.Herein,a honeycomb-inspired triboelectric nanogenerator(h-TENG)is proposed for biomechanical and UAV morphing wing energy harvesting based on contact triboelectrification wavy surface of cellular honeycomb structure.The wavy surface comprises a multilayered thin film structure(combining polyethylene terephthalate,silver nanowires and fluorinated ethylene propylene)fabricated through high-temperature thermoplastic molding and wafer-level bonding process.With superior synchronization of large amounts of energy generation units with honeycomb cells,the manufactured h-TENG prototype produces the maximum instantaneous open-circuit voltage,short-circuit current and output power of 1207 V,68.5μA and 12.4 mW,respectively,corresponding to a remarkable peak power density of 0.275 mW cm^(−3)(or 2.48 mW g^(−1))under hand pressing excitations.Attributed to the excellent elastic property of self-rebounding honeycomb structure,the flexible and transparent h-TENG can be easily pressed,bent and integrated into shoes for real-time insole plantar pressure mapping.The lightweight and compact h-TENG is further installed into a morphing wing of small UAVs for efficiently converting the flapping energy of ailerons into electricity for the first time.This research demonstrates this new conceptualizing single h-TENG device’s versatility and viability for broad-range real-world application scenarios.Kai Tao Zhensheng Chen Haiping Yi Ruirong Zhang Qiang Shen Jin Wu Lihua Tang Kangqi Fan Yongqing Fu Jianmin Miao Weizheng Yuan 2021Nano-Micro Letters2021,13,8:7
9Mycobacterium tuberculosis Mce2E suppresses the macrophage innate immune response and promotes epithelial cell proliferation显示文摘The intracellular pathogen Mycobacterium tuberculosis(Mtb)can survive in the host and cause disease by interfering with a variety of cellular functions.The mammalian cell entry 2(mce2)operon of Mtb has been shown to contribute to tuberculosis pathogenicity.However,little is known about the regulatory roles of Mtb Mce2 family proteins towards host cellular functions.Here we show that the Mce2 family protein Mce2E suppressed the macrophage innate immune response and promoted epithelial cell proliferation.Mce2E inhibited activation of the extracellular signal-regulated kinase(ERK)and Jun N-terminal kinase(JNK)mitogen-activated protein kinase(MAPK)signaling pathways in a non-canonical D motif(a MAPK-docking motif)-dependent manner,leading to reduced expression of TNF and IL-6 in macrophages.Furthermore,Mce2E promoted proliferation of human lung epithelium-derived lung adenoma A549 cells by inhibiting K48-linked polyubiquitination of eEF1A1 in aβstrand region-dependent manner.In summary,Mce2E is a novel multifunctional Mtb virulence factor that regulates host cellular functions in a niche-dependent manner.Our data suggest a potential novel target for TB therapy.Lihua Qiang Jing Wang Yong Zhang Pupu Ge Qiyao Chai Bingxi Li Yi Shi Lingqiang Zhang George Fu Gao Cui Hua Liu 2019Cellular & Molecular Immunology2019,16,4:7

11Dependence of creep age formability on initial temper of an Al-Zn-Mg-Cu alloy显示文摘The initial temper of the material may directly affect the whole creep age forming(CAF)process. In terms of creep deformation and stress relaxation, using the constant-stress creep aging and constant-strain stress relaxation aging tests, the relationship between initial temper and CAF formability is investigated for an Al-Zn-Mg-Cu alloy at 165 °C for 18 h. Three tempers are selected as the initial tempers in CAF, viz., solution, retrogression and re-solution. The CAF formability of this alloy with initial temper of retrogression is the best, and the creep strain of the retrogression tempered specimen after creep aging of 18 h is about 1.21 and 1.34 times than that of the solution and the re-solution tempered specimens, respectively. The calculated stress exponents of this alloy with three initial tempers range from 7.3 to 9.5, indicating that the CAF of this alloy is mainly controlled by the dislocation creep. The various formability for three initial tempers are attributed to different inhibitions of the transgranular precipitates on the dislocation movement. For the retrogression temper, the initial fine and uniformly distributed precipitates are seriously coarsened after6 h of CAF, which minimally inhibit the dislocation movement. 13Study of magneto-optical effects in γ-Fe_2O_3/ZnFe_2O_4 nanoparticle ferrofluids, using circularly polarized light显示文摘When circularly polarized light travels through a ferrofluid film in the presence of an applied magnetic field, the transmitted light will behave as elliptically polarized light, because of magnetic birefringence and dichroism. The angular distribution o the relative intensity of the transmitted light can be obtained using a polarizer, so that the ratio of both the long and short ax es-as well as the orientation angle of the ellipse, corresponding to the direction of the applied magnetic field-can be deter mined, and whether the ferrofluids samples are stable during the measurement can be directly judged from the shape of the distribution curves. Thus, the ratio of the amplitudes Ax/Ay and the added phase difference can be resolved in the elliptically polarized light, and information on both the magnetic birefringence n and the dichroism k can be deduced for the ferrofluid sample. From the orientation angles of both right-handed and left-handed elliptically polarized transmitted light, the direction of the applied magnetic field can be accurately determined. Using circularly polarized light, the magnetic birefringence and dichroism of pure γ-Fe2O3 ferrofluids and γ-Fe2O3/ZnFe2O4 binary ferrofluids were studied. 14The deubiquitinase OTUD1 inhibits colonic inflammation by suppressing RIPK1-mediated NF-κB signaling显示文摘The E3 ubiquitin ligase(E3)-mediated ubiquitination and deubiquitinase(DUB)-mediated deubiquitination processes are closely associated with the occurrence and development of colonic inflammation.Ovarian tumor deubiquitinase 1(OTUD1)is involved in immunoregulatory functions linked to infectious diseases.However,the effect of OTUD1 on intestinal immune responses during colonic inflammatory disorders such as inflammatory bowel disease(IBD)remains unclear.Here,we show that loss of OTUD1 in mice contributes to the pathogenesis of dextran sulfate sodium(DSS)-induced colitis via excessive release of proinflammatory cytokines.In addition,bone marrow transplantation experiments revealed that OTUD1 in hematopoietic cells plays a dominant role in protection against colitis.Mechanistically,OTUD1 physically interacts with receptor-interacting serine/threonine-protein kinase 1(RIPK1)and selectively cleaves K63-linked polyubiquitin chains from RIPK1 to inhibit the recruitment of NF-κB essential modulator(NEMO).Moreover,the expression of OTUD1 in mucosa samples from ulcerative colitis(UC)patients was lower than that in mucosa samples from healthy controls.Furthermore,we demonstrate that the UC-associated OTUD1 G430V mutation abolishes the ability of OTUD1 to inhibit RIPK1-mediated NF-κB activation and intestinal inflammation.Taken together,our study unveils a previously unexplored role of OTUD1 in moderating intestinal inflammation by inhibiting RIPK1-mediated NF-κB activation,suggesting that the OTUD1-RIPK1 axis could be a potential target for the treatment of IBD.Bo Wu Lihua Qiang Yong Zhang Yesheng Fu Mengyuan Zhao Zehui Lei Zhe Lu Yan-Ge Wei Hongmiao Dai Yingwei Ge Mingqiu Liu Xuemei Zhou Zhiqiang Peng Hongchang Li Chun-Ping Cui Jing Wang Hui Zheng Cui Hua Liu Lingqiang Zhang 2022Cellular & Molecular Immunology2022,19,2:3
15Simulation of Evapotranspiration Based on BEPS-TerrainLab V2.0 from 1990 to 2018 in the Dajiuhu Basin显示文摘Accurate estimations of evapotranspiration(ET)are essential for understanding land-atmosphere coupling and atmosphere-underlying surface energy and water vapor exchanges.Based on input data processing,this paper simulates the temporal and spatial variation of ET in the Dajiuhu Basin from 1990 to 2018 using the BEPS-Terrain Lab V2.0 model.Compared with the ET measured by an eddy covariance(EC)tower,the model explained 80.1%of the ET variation.From 1990 to 2018,the average annual ET in the Dajiuhu Basin was 1262.7 mm/yr indicating a downward trend(–27.12 mm/yr).In 2005,a sudden change point was observed based on the Mann-Kendall(MK)test and 3-year moving t-test.Around 2005,the downward trend in ET slowed and the proportional trend of ET to precipitation changed from upward trend to downward trend.Regarding spatial distribution,the ET in the basin’s central part was smaller than that in the basin’s surrounding area,the ET of the southern slope was higher than that of the northern slope,and the decrease in the ET rate on the sunny side was lower than that on the shady side.ET decreased as the elevation increased,with the fastest decrease observed between 2184 and 2384 m.For different landcover types,the average ET exhibited the following order:deciduous forest>mixed forest>wetland>grass>agriculture land.Decreased solar radiation is the main reason for the decreased ET in the Dajiuhu Basin,followed by increased wind speed and relative humidity,which together contribute 83.9%to the ET trend.This paper provides a theoretical basis for the study of ET changes and the mechanism of ET and provides a decision-making reference for water resource management in the Dajiuhu Basin and even the South-to-North Water Transfer Project.WU Zongfan ZHANG Lihua LIU Dandan ZHANG Kang ZHU Zhiru FU Yasheng MA Yongming 2020Chinese Geographical Science2020,30,6:2
16Coupled carbon and sulfur isotope behaviors and other geochemical perspectives into marine methane seepage显示文摘Methane seepage is the signal of the deep hydrocarbon reservoir. The determination of seepage is significant to the exploration of petroleum, gas and gas hydrate. The seepage habits microbial and macrofaunal life which is fueled by the hydrocarbons, the metabolic byproducts facilitate the precipitation of authigenic minerals. The study of methane seepage is also important to understand the oceanographic condition and local ecosystem. The seepage could be active or quiescent at different times. The geophysical surveys and the geochemical determinations reveal the existence of seepage. Among these methods, only geochemical determination could expose message of the dormant seepages. The active seepage demonstrates high porewater methane concentration with rapid SO_4^(2–) depleted, low H_2S and dissolved inorganic carbon(DIC), higher rates of sulfate reduction(SR) and anaerobic oxidation of methane(AOM). The quiescent seepage typically develops authigenic carbonates with specific biomarkers, with extremely depleted ~13C in gas, DIC and carbonates and with enriched ~34S sulfate and depleted ~34S pyrite. The origin of methane, minerals precipitation, the scenario of seepage and the possible method of immigration could be determined by the integration of solutes concentration, mineral composition and isotopic fractionation of carbon, sulfur. Numerical models with the integrated results provide useful insight into the nature and intensity of methane seepage occurring in the sediment and paleooceanographic conditions. Unfortunately, the intensive investigation of a specific area with dormant seep is still limit. Most seepage and modeling studies are site-specific and little attempt has been made to extrapolate the results to larger scales. 17E3 ligase FBXW7 aggravates TMPD-induced systemic lupus erythematosus by promoting cell apoptosis显示文摘Systemic lupus erythematosus(SLE)is a systemic autoimmune disease,and the pathogenesis of SLE has not been fully elucidated.The E3 ubiquitin ligase FBXW7 has been well characterized in cancer as a tumor suppressor that can promote the ubiquitination and subsequent degradation of various oncoproteins;however,the potential role of FBXW7 in autoimmune diseases is unclear.In the present study,we identified that FBXW7 is a crucial exacerbating factor for SLE development and progression in a mouse model induced by 2,6,10,14-tetramethylpentadecane(TMPD).Myeloid cell-specific FBXW7-deficient(Lysm+FBXW7f/f)C57BL/6 mice showed decreased immune complex accumulation,glomerulonephritis,glomerular mesangial cell proliferation,and basemembrane thickness in the kidney.Lysm+FBXW7f/f mice produced fewer anti-Sm/RNP and anti-ANA autoantibodies and showed a decreased MHC II expression in B cells.In Lysm+FBXW7f/f mice,we observed that cell apoptosis was reduced and that fewer CD11b+Ly6Chi inflammatory monocytes were recruited to the peritoneal cavity.Consistently,diffuse pulmonary hemorrhage(DPH)was also decreased in Lysm+FBXW7f/f mice.Mechanistically,we clarified that FBXW7 promoted TMPD-induced cell apoptosis by catalyzing MCL1 degradation through K48-linked ubiquitination.Our work revealed that FBXW7 expression in myeloid cells played a crucial role in TMPD-induced SLE progression in mice,which may provide novel ideas and theoretical support for understanding the pathogenesis of SLE.Zhenlu Chong Chunjing Bao Jia He Tianxiao Chen Lijia Zhong Gaopeng Li Huanle Li Lutong Fang Yinjing Song Guoxiang Fu Xuyan Yang Lihua Lai Yang Liu Qingqing Wang 2018Cellular & Molecular Immunology2018,15,12:2
18In situ atomic-scale observation of dislocation behaviors in twin-structured Pt nanocrystals显示文摘The deformation mechanisms of twin-structured metallic materials have attracted great interest.Though previous theoretical predictions have suggested that the repulsive force of the twin boundary(TB)can significantly affect the deformation of twinstructured metals,it remains unclear whether this prediction applies to experimental conditions.In this paper,the atomic-scaled deformation process of twin-structured Pt nanocrystals was in situ observed using our home-made device in a high-resolution transmission electron microscope.We have shown that the plastic deformation of the twin-structured Pt nanocrystals was governed by full dislocation generation as well as Lomer dislocation(LD)lock formation and destruction.After LD locks were destructed,these full dislocations tended to move towards the surface of the nanocrystals.The findings revealed that due to the ultra-high repulsive force of TB on dislocation,there was no dislocation-TB reaction during the deformation.These findings can enrich our understanding of the dislocation behaviors of twin-structured nanocrystals.GUO YiZhong SUN Tao FU LiBo HAO LongHu HUANG Ming WEI RuJian LUO JunFeng HE Xin WANG XingQuan XIONG XiaoDong WANG LiHua HAN XiaoDong 2021Science China(Technological Sciences)2021,64,3:2

20Mesencephalic astrocyte-derived neurotrophic factor reduces cell apoptosis via upregulating HSP70 in SHSY-5Y cells显示文摘Background:Mesencephalic astrocyte-derived neurotrophic factor(MANF)is a new candidate growth factor for dopaminergic neurons against endoplasmic reticulum stress(ER stress).HSP70 family,a chaperon like heat shock protein family,was proved to be involved in the MANF induced survival pathway in 6-OHDA treated SHSY-5Y cells.However,the ER stress relative transcriptome,in MANF signaling cascades is still investigated.The involvement of HSP70,a 70kd member of HSP70 family,need further to be verified.Methods:The cell apoptosis was assayed by MTT,TUNEL staining and western blot of cleaved Caspase-3.The differentially expressed genes in SHSY-5Y cells under different conditions(control,6-OHDA,6-OHDA+MANF)were investigated by RNA-seq.Expression of HSP70 was further confirmed by real-time PCR.RNAi knockdown for HSP70 was performed to investigate the role of HSP70 in the MANF signaling pathway.Results:MANF inhibits 6-OHDA-induced apoptosis in SHSY-5Y cells.Six ER stress relative genes(HSP70,GRP78,xbp-1,ATF-4,ATF-6,MAPK)were found enriched in 6-OHDA+MANF treatment group.HSP70 was the most significantly up-regulated gene under 6-OHDA+MANF treatment in SHSY-5Y cells.RNAi knockdown for HSP70 inhibits the protective effects of MANF against 6-OHDA toxicity in SHSY-5Y cells.Conclusion:MANF exerts a protective role against 6-OHDA induced apoptosis in SHSY-5Y cells via up-regulating some ER stress genes,including HSP70 family members.The HSP70 expression level plays a key role in MANFmediated survival pathway.Hui Sun Ming Jiang Xing Fu Qiong Cai Jingxing Zhang Yanxin Yin Jia Guo Lihua Yu Yun Jiang Yigang Liu Liang Feng Zhiyu Nie Jianmin Fang Lingjing Jin 2017Translational Neurodegeneration2017,6,1:2
