11HLA-B-associated transcript 3 (Bat3) stabilizes and activates p53 in a HAUSP-dependent manner显示文摘The p53 pathway is a highly complex signaling network including several key regulators.HAUSP is a critical component of the p53 pathway acting as a deubiquitinase for both p53 and its key repressor Mdm2.Here,we identified a novel HAUSP-interacting protein,HLA-B-associated transcript 3(Bat3)and found it to be capable of inducing p53 stabilization and activation via a HAUSP-dependent mechanism,resulting in cell growth inhibition.Surprisingly,the deubiquitylating enzymatic activity of HAUSP was not required for this phenomenon.Co-immunoprecipitation showed that p53 coexisted in a complex with Bat3 and HAUSP in vivo,and HAUSP may serve as a binding mediator to enhance the interaction between p53 and Bat3.Further studies revealed that formation of this three-protein complex interfered with the binding of p53 to its proteasome receptor S5a and promoted the accumulation of p53 in nucleus.Notably,Mdm2 protein abundance is also regulated by Bat3 in the presence of HAUSP.Overexpression of Bat3 and HAUSP increases Mdm2 protein levels without influencing the p53–Mdm2 interaction and Mdm2-mediated p53 ubiquitination,indicating that Bat3–HAUSP-mediated protein stabilization is not specific to p53 and different mechanisms may be involved in Bat3-mediated regulation of p53–Mdm2 pathway.Together,our study unravels a novel mechanism by which p53 is stabilized and activated by HAUSP-mediated interaction with Bat3 and implies that Bat3 might function as a tumor suppressor through the stabilization of p53.Rui Zhang Di Cui TengXue Yue Lang Yunfan Zhang Lianjie Li Haili Sun Yu Kuang Gebin Li Jun Tang 2020Journal of Molecular Cell Biology2020,12,2:0
12Safety and Immunogenicity of SARS-CoV-2 Vaccines in People Living With HIV:A Systematic Review and Meta-analysis of Real-World Studies显示文摘The safety and immunogenicity of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccines in people living with HIV(PLWH)in real-world studies remain controversial.Thus,we conducted a comprehensive systematic review and meta-analysis to address this issue.Data search were conducted from PubMed,Web of Science and EMBASE.Adverse events following vaccination,the risk ratio(RR)of SARS-CoV-2–specific IgG seroconversion and the level of anti–SARS-CoV-2 neutralizing antibodies were compared between the PLWH group and a healthy control group.A total of 10,582 PLWH from 22 studies were included.In our analysis,the incidence of local or systemic adverse events after the first SARS-CoV-2 vaccine dose was not statistically different between PLWH and healthy controls.However,there was a statistical difference after the second dose(RR,0.83;95%CI,0.71–0.98).The seroconversion rate of SARS-CoV-2 IgGantibodies in PLWH was significantly lower than that in the healthy control group(RR,0.94;95%CI,0.89–0.98;I2=80%,P<0.01).The anti–SARS-CoV-2 neutralizing antibody titers in PLWH after full immunization were also significantly lower than those in the healthy control group(RR,0.91;95%CI,0.85–0.98;I2=81%,P<0.01).The safety and tolerance of COVID-19 vaccines in PLWHare acceptable.However,their immunogenicitymay be impaired to a certain extent,characterized by a lower IgGseroconversion rate and neutralizing antibody titers compared with healthy individuals.These findings should provide guidance for optimizing future COVID-19 vaccination strategies among PLWH.Xueying Fan Yangguo Zhao Fan Wu Yue Yuan Bing Lang Di Yin Zhongliang Xu Shiqiang Jiang Huachun Zou Jianhui Yuan Caijun Sun 2023Infectious Microbes & Diseases2023,5,4:0
13The Anatomic Pathology Evaluation of Liver with Diethylinitrosamine Treated via Intraperitoneal Injection Singly and Peros for 90 Days Carcinogenicity Study in F344 Rats显示文摘Objective:To establish the integrity experiment method of short(medium)-term carcinogenicity test pursuant to GLP,make into relative SOP and improve the safeguard in the center.Methods:Diethylinitrosamine(DEN) is known as carcinogenic agent,whose target organ is liver. Using the two-stage carcinogenesis test method,DEN was treated to F344 rats via intraperitoneal injection singly(200 mg/kg),and peros administrated for 90 days(10 ppm). The liver in any group rat will be examined by light microscopy.Results:In pathologic examination,no liver cell tumor was shown in the livers of the rats that were singly treated with a carcinogenic chemical-DEN.Foci of cellular alteration were observed in the livers of these rats. The proliferation lesions of liver from slight to seveity(foci of cellular alteration-hepatocelluar adenoma-hepatocellular carcinoma)were observed in the livers of the rats which exposed peros to a low dose of DEN for 90 days after initiation by a single intraperitoneal injection. The incidence of hepatocelluar tumor was 35% in male animal,which was not shown in the liver of female rat.Conclusion:For current results,it may be possible that low-dose DEN acts as a promotor of hepatocelluar tumor if it was exposed in a population for a long time. It is considered that male hormone has a synergistic effect on hepatocelluar tumor development of DEN. 14TNFR2 is a potent prognostic biomarker for post-transplant lung metastasis in patients with hepatocellular carcinoma显示文摘Objective:Lung metastasis is a common and fatal complication of liver transplantation for hepatocellular carcinoma(HCC).The precise prediction of post-transplant lung metastasis in the early phase is of great value.Methods:The mRNA profiles of primary and paired lung metastatic lesions were analyzed to determine key signaling pathways.We enrolled 241 HCC patients who underwent liver transplantation from three centers.Tissue microarrays were used to evaluate the prognostic capacity of tumor necrosis factor(TNF),tumor necrosis factor receptor 1(TNFR1),and TNFR2,particularly for post-transplant lung metastasis.Results:Comparison of primary and lung metastatic lesions revealed that the TNF-dependent signaling pathway was related to lung metastasis of HCC.The expression of TNF was degraded in comparison to that in para-tumor tissues(P<0.001).The expression of key receptors in the TNF-dependent signaling pathway,TNFR1 and TNFR2,was higher in HCC tissues than in para-tumor tissues(P<0.001).TNF and TNFR1 showed no relationship with patients’outcomes,whereas elevated TNFR2 in tumor tissue was significantly associated with worse overall survival(OS)and increased recurrence risk(5-year OS rate:31.9%vs.62.5%,P<0.001).Notably,elevated TNFR2 levels were also associated with an increased risk of post-transplant lung metastasis(hazard ratio:1.146;P<0.001).Cox regression analysis revealed that TNFR2,Hangzhou criteria,age,and hepatitis B surface antigen were independent risk factors for post-transplant lung metastasis,and a novel nomogram was established accordingly.The nomogram achieved excellent prognostic efficiency(area under time-dependent receiver operating characteristic=0.755,concordance-index=0.779)and was superior to conventional models,such as the Milan criteria.Conclusions:TNFR2 is a potent prognostic biomarker for predicting post-transplant lung metastasis in patients with HCC.A nomogram incorporating TNFR2 deserves to be a helpful prognostic tool in liver transplantation for HCC.Huigang Li Zuyuan Lin Jianyong Zhuo Modan Yang Wei Shen Zhihang Hu Yichen Ding Hao Chen Chiyu He Xinyu Yang Siyi Dong Xuyong Wei Beicheng Sun Shusen Zheng Ren Lang Di Lu Xiao Xu 2023Chinese Journal of Cancer Research2023,35,1:0
15Mechanical properties,corrosion behavior and microstructure evolution of zinc and scandium co-strengthened 5xxx alloy显示文摘Nowadays,it is a challenge to improve the strength of 5xxx aluminium alloy while maintaining its distinctive properties.To address this,the Zn and Sc modified high strength 5xxx aluminium alloys with good intergranular corrosion resistance were prepared through chill casting,and the effects of minor Sc on mechanical properties,corrosion behavior and microstructure evolution of Al-5.0Mg-3.0Zn alloys were systematically investigated.The results show that the Sc containing alloy has high number density and small size of intragranular precipitates,combined with the solid solution strengthening of Sc and the Al_(3)(Sc_(1-x),Zr_(x))second phase strengthening,the strength of Al-5.0Mg-3.0Zn-0.1Sc alloy is obviously enhanced.Meanwhile,the intergranular corrosion resistance of the Sc containing alloy is also improved due to the formation of a large number of low angle grain boundaries and the weakening of the continuity of precipitated phases along the grain boundaries.Yanlin Pan Yue Xu Di Zhang Jishan Zhang Guxin Zhou Yujing Lang 2023Journal of Rare Earths2023,41,11:0
16Sintering kinetics involving grain growth and densification of Mg-PSZ nanopowders显示文摘Sintering kinetics have been found to be effective in judging the evolution of ceramics.By using magnesium oxide-partially stabilized zirconia(Mg-PSZ)powder prepared by co-precipitation as raw materials,the evolution of densification and grain growth for Mg-PSZ ceramics were investigated.The results indicated that the densification of samples was mainly controlled by grain boundary diffusion in intermediate sintering stage.During the sintering process,the grain growth mechanisms included normal grain growth,abnormal grain growth and solid solution drag-controlled grain growth.Interestingly,the apparent activation energy for grain growth of Mg-PSZ ceramics is lower than that of ZrO_(2)–Y_(2)O_(3)ceramics in the solid solution drag-controlled grain grow process,which will cause grain to grow easily.The sintering kinetics and microstructure of Mg-PSZ ceramics were studied,and the kinetic equation of grain growth at different temperatures was established.The results show that the strength difference between Mg-PSZ and yttrium oxide-stabilized zirconia is closely related to the easy grain growth of Mg-PSZ ceramics.Yi-ming She Jie-fu Lang Di An Chao-wei Si Xu-dong Luo Zhi-peng Xie Jie-gang You 2022Journal of Iron and Steel Research(International)2022,29,7:0

The contents of clinical application of women’s contraceptive methods are very wide, involving multiple areas. This consensus deeply discusses the specific contraceptive needs at different statuses, combined with gynecological diseases, postabortion contraception, and postpartum family planning, ensuring the correct use of contraceptive methods under the corresponding status. The top priority of the consensus is the specific contraception consensus section for women combined with gynecological diseases because medical treatment effect as well as side effects should be weighed carefully. 18CPAL, as a New Mediator of Cardiomyocyte Metabolic Alterations and Pyroptosis, Regulates Myocardial Infarction Injury in Mice显示文摘Myocardial infarction (MI), the most serious of the ischemic heart diseases, is accompanied by myocardial metabolic disorders and the loss of cardiomyocytes. Increasing evidence has shown that long noncoding RNAs (lncRNAs) are involved in various pathological conditions such as cancer and cardiovascular diseases (CVDs), and are emerging as a novel biomarker for these disorders. This study aims to investigate the regulatory role and mechanisms of lncRNAs in myocardial remodeling in the setting of MI. We find that post-infarcted hearts exhibit a reduction of adenosine triphosphate (ATP) and an alteration of the glucose and lipid metabolism genes cluster of differentiation 36 (CD36), hexokinase 1 (HK1), and clucose transporter 4 (GLUT4), accompanied by cardiomyocyte pyroptosis. We then identify a previously unknown conserved lncRNA, AK009126 (cardiomyocyte pyroptosis-associated lncRNA, CPAL), which is remarkably upregulated in the myocardial border zone of MI mice. Importantly, the adeno-associated virus 9 (AAV9)-mediated silencing of endogenous CPAL by its short hairpin RNA (shRNA) partially abrogates myocardial metabolic alterations and cardiomyocyte pyroptosis during MI in mice. Mechanistically, CPAL is shown to bind directly to nuclear factor kappa B (NFκB) and to act as an activator of NFκB to induce NFκB phosphorylation in cardiomyocytes. We also find that CPAL upregulates caspase-1 expression at the transcriptional level and consequently promotes the release of interleukin (IL)-18 and IL-1β from cardiomyocytes. 19Improving the resilience of maritime supply chains: The integration of ports and inland transporters in duopoly markets显示文摘The adverse impact of the outbreak of COVID-19 has reduced ports’ operational efficiency. In addition, ports and inland logistics providers are generally independent of each other and difficult to work together, which leads to time loss. Thus, as the core player, ports can integrate with inland logistics providers to improve the efficiency and resilience of maritime supply chains. This study examines the strategic options of two competing maritime supply chains consisting of ports and inland logistics providers. We investigate the impact of cooperation between ports and inland logistics providers and government regulation on the maritime supply chain by comparing members’ optimal pricing and overall social welfare under centralized, decentralized, and hybrid scenarios. Results indicate that the hybrid scenario is an equilibrium strategy for maritime supply chain, although this strategy is not optimal for governments seeking to improve supply chain resilience and maximize social welfare.
