1Salidroside reduces tau hyperphosphorylation via up-regulating GSK-3β phosphorylation in a tau transgenic Drosophila model of Alzheimer’s disease显示文摘Background:Alzheimer’s disease(AD)is an age-related and progressive neurodegenerative disease that causes substantial public health care burdens.Intensive efforts have been made to find effective and safe treatment against AD.Salidroside(Sal)is the main effective component of Rhodiola rosea L.,which has several pharmacological activities.The objective of this study was to investigate the efficacy of Sal in the treatment of AD transgenic Drosophila and the associated mechanisms.Methods:We used tau transgenic Drosophila line(TAU)in which tau protein is expressed in the central nervous system and eyes by the Gal4/UAS system.After feeding flies with Sal,the lifespan and locomotor activity were recorded.We further examined the appearance of vacuoles in the mushroom body using immunohistochemistry,and detected the levels of total glycogen synthase kinase 3β(t-GSK-3β),phosphorylated GSK-3β(p-GSK-3β),t-tau and p-tau in the brain by western blot analysis.Results:Our results showed that the longevity was improved in salidroside-fed Drosophila groups as well as the locomotor activity.We also observed less vacuoles in the mushroom body,upregulated level of p-GSK-3βand downregulated p-tau following Sal treatment.Conclusion:Our data presented the evidence that Sal was capable of reducing the neurodegeneration in tau transgenic Drosophila and inhibiting neuronal loss.The neuroprotective effects of Sal were associated with its up-regulation of the p-GSK-3βand down-regulation of the p-tau.Bei Zhang Qiongqiong Li Xingkun Chu Suya Sun Shengdi Chen 2016Translational Neurodegeneration2016,5,1:14

5Intelligent diagnosis of the solder bumps defects using fuzzy C-means algorithm with the weighted coefficients显示文摘Solder bump technology has been widely used in electronic packaging. With the development of solder bumps towards higher density and finer pitch, it is more difficult to inspect the defects of solder bumps as they are hidden in the package. A nondestructive method using the transient active thermography has been proposed to inspect the defects of a solder bump, and we aim at developing an intelligent diagnosis system to eliminate the influence of emissivity unevenness and non-uniform heating on defects recognition in active infrared testing. An improved fuzzy c-means(FCM) algorithm based on the entropy weights is investigated in this paper. The captured thermograms are preprocessed to enhance the thermal contrast between the defective and good bumps. Hot spots corresponding to 16 solder bumps are segmented from the thermal images. The statistical features are calculated and selected appropriately to characterize the status of solder bumps in FCM clustering. The missing bump is identified in the FCM result, which is also validated by the principle component analysis. 7Transmission Properties of a New Glass Ceramic and Doped with Co^(2+) as Saturable Absorber for 1.54 μm Er Glass Short Pulse Laser显示文摘The preparation and characteristics of a new transparent glass ceramic were described. Crystal phase particles with nanometer size were successfully precipitated in glass matrix, which was confirmed to be one of indium aluminum zinc oxide compounds (InxAlyZnzO). The presence of aluminum (Al) and indium (In) impurities in the zinc oxides (ZnO) crystal lattice leads to some changes of the carrier concentration in the material and then promote the sharply changes of transmission spectra in IR range wavelength. And subsequently, the IR cut-off edge blue shifted from 5.5 μm in base glass to 3 μm in transparent glass ceramic sample. 8Particle shape effect on hydrodynamics and heat transfer in spouted bed:A CFD-DEM study显示文摘Spouted bed has drawn much attention due to its good heat and mass transfer efficiency in many chemical units.Investigating the flow patterns and heat and mass transfer inside a spouted bed can help optimize the spouting process.Therefore,in this study,the effects of particle shape on the hydrodynamics and heat transfer in a spouted bed are investigated.This is done by using a validated computational fluid dynamics-discrete element method(CFD-DEM)model,considering volume-equivalent spheres and oblate and prolate spheroids.The results are analysed in detail in terms of the flow pattern,microstruc-ture,and heat transfer characteristics.The numerical results show that the prolate spheroids(Ar=2.4)form the largest bubble from the beginning of the spouting process and rise the highest because the fluid drag forces can overcome the interlocking and particle-particle frictional forces.Compared with spherical particles,ellipsoidal spheroids have better mobility because of the stronger rotational kinetic energy resulting from the rough surfaces and nonuniform torques.In addition,the oblate spheroid system exhibits better heat transfer performance benefiting from the larger surface area,while prolate spheroids have poor heat transfer efficiency because of their orientation distribution.These findings can serve as a reference for optimizing the design and operation of complex spouted beds.Dianyu E Peng Zhou Suya Guo Jia Zeng Jiaxin Cui Youyuan Jiang Yuanxiang Lu Zeyi Jiang Zhengquan Li Shibo Kuang 2022Particuology2022,20,10:0
9Clinicopathologic characterization and abnormal autophagy of CSF1R-related leukoencephalopathy显示文摘Background:CSF1R-related leukoencephalopathy,also known as hereditary diffuse leukoencephalopathy with spheroids(HDLS),is a rare white-matter encephalopathy characterized by motor and neuropsychiatric symptoms due to colony-stimulating factor 1 receptor(CSF1R)gene mutation.Few of CSF1R mutations have been functionally testified and the pathogenesis remains unknown.Methods:In order to investigate clinical and pathological characteristics of patients with CSF1R-related leukoencephalopathy and explore the potential impact of CSF1R mutations,we analyzed clinical manifestations of 15 patients from 10 unrelated families and performed brain biopsy in 2 cases.Next generation sequencing was conducted for 10 probands to confirm the diagnosis.Sanger sequencing,segregation analysis and phenotypic reevaluation were utilized to substantiate findings.Functional examination of identified mutations was further explored.Results:Clinical and neuroimaging characteristics were summarized.The average age at onset was 35.9±6.4 years(range 24–46 years old).Younger age of onset was observed in female than male(34.2 vs.39.2 years).The most common initial symptoms were speech dysfunction,cognitive decline and parkinsonian symptoms.One patient also had marked peripheral neuropathy.Brain biopsy of two cases showed typical pathological changes,including myelin loss,axonal spheroids,phosphorylated neurofilament and activated macrophages.Electron microscopy disclosed increased mitochondrial vacuolation and disorganized neurofilaments in ballooned axons.A total of 7 pathogenic variants(4 novel,3 documented)were identified with autophosphorylation deficiency,among which c.2342C>T remained partial function of autophosphorylation.Western blotting disclosed the significantly lower level of c.2026C>T(p.R676*)than wild type.The level of microtubule associated protein 1 light chain 3-II(LC3-II),a classical marker of autophagy,was significantly lower in mutants expressed cells than wild type group by western blotting and immunofluorescence staining.Conclusions:Our findings support the loss-of-function and haploinsufficiency hypothesis in pathogenesis.Autophagy abnormality may play a role in the disease.Repairing or promoting the phosphorylation level of mutant CSF1R may shed light on therapeutic targets in the future.However,whether peripheral polyneuropathy potentially belongs to CSF1R-related spectrum deserves further study with longer follow-up and more patients enrolled.Wo-Tu Tian Fei-Xia Zhan Qing Liu Xing-Hua Luan Chao Zhang Liang Shang Ben-Yan Zhang Si-Jian Pan Fei Miao Jiong Hu Ping Zhong Shi-Hua Liu Ze-Yu Zhu Hai-Yan Zhou Suya Sun Xiao-Li Liu Xiao-Jun Huang Jing-Wen Jiang Jian-Fang Ma Ying Wang Shu-Fen Chen Hui-Dong Tang Sheng-Di Chen Li Cao 2019Translational Neurodegeneration2019,8,1:0
10Targeting neuroplasticity in neurodegenerative diseases stimulation techniques显示文摘Deficits in synaptic transmission and plasticity are thought to contribute to the pathophysiology of Alzheimer's disease(AD)and Parkinson's disease(PD).Several brain stimulation techniques are currently available to assess or modulate human neuroplasticity,which could offer clinically useful interventions as well as quantitative diagnostic and prognostic biomarkers.In this review,we discuss several brain stimulation techniques,with a special emphasis on transcranial magnetic stimulation and deep brain stimulation(DBS),and review the results of clinical studies that applied these techniques to examine or modulate impaired neuroplasticity at the local and network levels in patients with AD or PD.The impaired neuroplasticity can be detected in patients at the earlier and later stages of both neurodegenerative diseases.However,current brain stimulation techniques,with a notable exception of DBS for PD treatment,cannot serve as adequate clinical tools to assist in the diagnosis,treatment,or prognosis of individual patients with AD or PD.Targeting the impaired neuroplasticity with improved brain stimulation techniques could offer a powerful novel approach for the treatment of AD and PD.Ti-Fei Yuan Wei-Guang Li Chencheng Zhang Hongjiang Wei Suya Sun Nan-Jie Xu Jun Liu Tian-Le Xu 2020Translational Neurodegeneration2020,9,4:0
11HPDA/Zn as a CREB Inhibitor for Ultrasound imaging and Stabilization of Atherosclerosis Plaque+显示文摘Thrombosis,secondary to rupture of unstable plaque,is a fatal risk factor for myocardial infarction and ischemic stroke.At present,more novel methods are needed for the diagnosis and treatment of vulnerable plaque.Here,we report a hollow polydopamine/Zn(HPDA/Zn)ultrasound contrast agent.Through western-blot,Elisa,and other experiments,we found that in addition to having a good contrast-enhancement capability in ultrasound imaging in vitro and in vivo,HPDA/Zn also has the effect of reducing the expres-sion of CREB.CREB protein and its downstream-regulated proteins and factors are closely related to the stability of plaque.HPDA/Zn has the effect of reducing the expression of CREB protein,which leads to the decrease of expression of MMP-9,the regulatory pro-tein downstream of the CREB protein.In addition,it also reduces the secretion of inflammatory factors hs-CRP and IL-17A.Thus,HPDA/Zn can stabilize plaque by inhibiting CREB and reducing plaque vulnerable markers and inflammatory factors.In a word,HPDA/Zn is a kind of ultrasound contrast agent,which can stabilize plaques by inhibiting CREB protein.Linzi Chen Zhenqi Jiang Lifei Yang Ye Fang Shuwei Lu Ozioma U.Akakuru Shuaishuai Huang Juan Li Suya Ma Aiguo Wu 2023Chinese Journal of Chemistry2023,41,2:0

13Mechanical Properties and Oxidation Behaviors of Self-Healing SiCf/SiC-SiBCN Composites Exposed to H_(2)O/O_(2)/Na_(2)SO_(4)Environments显示文摘The oxidation behaviors and their influence on the mechanical properties of self-healing SiCf/SiC-SiBCN composites were investigated in H_(2)O/O_(2)and H_(2)O/O_(2)/Na_(2)SO_(4)environments at 1200‒1350℃for 100 h.As the temperatures increase from 1200 to 1350℃,the oxidation rate constants increase from 0.45×10^(–7)to 1.58×10^(–7)mg^(2)/(mm^(4) h)in H_(2)O/O_(2),and from 1.02×10^(–7)to 3.42×10^(–7)mg^(2)/(mm^(4) h)in H_(2)O/O_(2)/Na_(2)SO_(4).The involvement of Na_(2)SO_(4)leads to the formation of a loose lamellar oxide layer,the breakage of the SiBCN/CVI-SiC interface and the decrease in the oxide viscosity,thus accelerating the oxidation of the composites.The composites show the maximum retention rate of strength(102%,535.71 MPa)after oxidation in H_(2)O/O_(2)at 1200℃due to the good self-healing capacity of the produced glass,while the minimum(82%,430.56 MPa)in H_(2)O/O_(2)/Na_(2)SO_(4)at 1350℃caused by the severe microstructural corrosion derived from Na_(2)SO_(4).Suya Ji Bin Liang Chenglong Hu Shengyang Pang Rida Zhao Jian Li Sufang Tang 2023Acta Metallurgica Sinica(English Letters)2023,36,11:0
