1Microtubule-associated deacetylase HDAC6 promotes angiogenesis by regulating cell migration in an EB1-dependent manner显示文摘Angiogenesis,a process by which the preexisting blood vasculature gives rise to new capillary vessels,is associated with a variety of physiologic and pathologic conditions.However,the molecular mechanism underlying this important process remains poorly understood.Here we show that histone deacetylase 6(HDAC6),a microtubule-associated enzyme critical for cell motility,contributes to angiogenesis by regulating the polarization and migration of vascular endothelial cells.Inhibition of HDAC6 activity impairs the formation of new blood vessels in chick embryos and in angioreactors implanted in mice.The requirement for HDAC6 in angiogenesis is corroborated in vitro by analysis of endothelial tube formation and capillary sprouting.Our data further show that HDAC6 stimulates membrane ruffling at the leading edge to promote cell polarization.In addition,microtubule end binding protein 1(EB1)is important for HDAC6 to exert its activity towards the migration of endothelial cells and generation of capillary-like structures.These results thus identify HDAC6 as a novel player in the angiogenic process and offer novel insights into the molecular mechanism governing endothelial cell migration and angiogenesis.Dengwen Li Songbo Xie Yuan Ren Lihong Huo Jinmin Gao Dandan Cui Min Liu Jun Zhou 2011Protein & Cell2011,2,2:13

3Medicinals with properties of warming Yang and tonifying Qi in terms of Traditional Chinese Medicine: their effects on left ventricular ejection fraction and aldosterone in rats with induced failure heart显示文摘OBJECTIVE: To investigate on Left ventricular ejection fraction value and aldosterone of two medicinals of Traditional Chinese Medicine(TCM) with the properties of warming Yang and tonifying Qi in terms of TCM theory.METHODS: An animal model of coronary ligation of heart failure after myocardial infarction was employed to study the influence of these two kinds of drugs on three batches of rats. On the basis of the average score of left ventricle ejection fraction during the investigation, there were some different groups, including Wen Yang(the warming Yang)group, Yi Qi(tonifying Qi) group, Wen Yang and Yi Qi group, captopril group, digoxin group. In additional, an artificial operation group was set for compari-son. The systemic intervention using these medicinal and drugs was taken effects on the 2nd day after the operation of myocardial infraction(MI) with once a day. At week one, two, and four after the MI treatment, evaluated were EF values, and ferritin,angiotensin-II and aldosterone in the rats' plasma.RESULTS: At week one and week two, the medicinal of Wen Yang, Yi Qi, Wen Yang pluse Yi Qi, and digoxin could improve left ventricular ejection fraction in rats with heart failure; Compared to the model group, captopril Left ventricular EF value increased, but there was not significant. At week four,heart failure and left ventricular EF values was improved in the intervention group and the other four captopril drug intervention. At week one in the rats with drug intervention, the medicinals of Yi Qi, Wen Yang plus Yi Qi, and captopril could inhibit activation in vivo hormone aldosterone in heart failure rats; aldosterone in Wen Yang group and digoxin group were not different from that in the model group at week two and four.CONCLUSION: Chinese medicinals with properties of Wen Yang, Yi Qi have significant effect on improving left ventricular EF in rats with heart failure; compared to Yi Qi medicinals, Wen Yang medicinals that inhibit the effectiveness of the time required for the activation of the role of aldosterone. 4Effect of diacylglycerol acyltransferase 2overexpression in 3T3-L1 is associated to an increase in mono-unsaturated fatty acid accumulation显示文摘Background:Fatty acid(FA) composition is the most important parameter affecting the flavor and nutritional value of the meat.The final and the only committed step in the biosynthesis of triglycerides is catalyzed by diacylglycerol acyltransferase 2(DGAT2).The role of DGAT2 in lipid accumulation has been demonstrated in adipocytes,However,little is known about the effect of DGAT2 on the FA composition of these cells.Methods:To investigate the role of DGAT2 in regulating lipid accumulation,FA composition and the expression of adipogenic genes,we cloned the open reading frame of the porcine DGAT2 gene and established 3T3-L1 cells that overexpressed DGAT2.Cells were then cultured in differentiation medium(DM) without FA,with a mixture of FAs(FA-DM),or containing a ^(13)C stable isotope-labeled FA mixture(IFA-DM).The FA composition of adipocytes was analyzed by gas chromatography-mass spectrometry and gas chromatography-isotope ratio mass spectrometry.Quantitative PCR and western blotting were employed to detect expression of adipogenic genes in 3T3-L1 adipocytes cultured with FA-DM for 12 d.Results:The triacylglyceride(TAG) content was significantly higher in 3T3-L1 adipocytes overexpressing DGAT2 than in control cells.When cultured in DM or FA-DM for 12 d,cells overexpressing DGAT2 showed a higher proportion of unsaturated FAs(C16:1 and C18:1).However,when cells overexpressing DGAT2 were cultured with FA-DM for30 min,the FA composition was almost identical to that of controls.Further,the proportion of stable isotope-labeled FAs were similar in 3T3-L1 adipocytes overexpressing DGAT2 and control cells cultured in IFA-DM for 12 d.These results collectively indicate that the higher proportion of mono-unsaturated FAs,C16:1 and C18:1,may originate from de novo FA synthesis but not from the uptake of specific FAs from the medium.This hypothesis is further supported by evidence that both mRNA and protein expression of genes involved in FA synthesis(ACACA,FASN,SCD1,and A-FABP)were significantly higher in cells overexpressing DGAT2 than in control cells.Conclusions:In conclusion,our study revealed that TAG accumulation,the proportion of MUFAs,and the expression of adipogenic genes were higher in 3T3-L1 cells overexpressing DGAT2 than in control cells.Zhiqi Zhang Gang Shu Xiaotong Zhu Junming Guo Han Cai Songbo Wang Lina Wang Ping Gao Qianyun Xi Yongliang Zhang Li Yuan Qingyan Jiang 2014Journal of Animal Science and Biotechnology2014,5,4:3
5The luminescence properties of Sr_3Gd(BO_3)_3:Tb^(3+) phosphors under vacuum ultraviolet excitation显示文摘Tb3+-activated Sr3Gd(BO3)3 green phosphors were prepared by conventional solid-state reaction. The vacuum ultraviolet (VUV) excitation, photoluminescence (PL) and decay properties of the phosphors in the visible range were investigated. The excitation spectrum showed a strong broad band from 160 to 200 nm with a maximum at 183 nm which was adjacent to the VUV excitation light source of 172 nm. Under excitation at 172 nm, the optimum co-doping concentration of Tb3+ was 10 mol%, and the emission intensity of Sr3Gd0.9(BO3)3:0.1Tb3+ was comparable to that of commercial Zn2SiO4:Mn2+. The strongest emission peak of Sr3Gd0.9(BO3)3:0.1Tb3+ was at 543 nm with chromaticity coordinates of (0.2626, 0.4922) and a lifetime of 2.32 ms. 10Study on crystallization behavior of Tibet butter显示文摘Tibetan butter(TB),generally called butter,is a solid oil product extracted from yak milk in the QinghaiTibet plateau area.However,due to the limitations of raw material sources and production technology,there is a shortage of TB,so it is important to find substitutes of TB.This paper studied the crystallization behavior of six kinds of commercial TB products in Tibet to provide the theoretical basis for the development of TB substitutes for the production of TB lamps and flowers.This study assessed the crystallization behaviors of the TB samples,including isothermal crystallization process and non-isothermal crystallization process.The microstructure,isothermal crystallization of TB were evaluated by polarized light microscopy and low-pulse NMR spectrometry,respectively.The non-isothermal crystallization process of TB under temperature scanning were investigated using Rheometer,and the crystallization behavior under different cooling rates were determined by diferential scanning calorimetry(DSC)procedures.The results showed that the TB was crystallized at a higher supercooling(-10,0 and 10℃).Dimensional growth is dominant;at 20℃,spiral growth dominates.The TB has complex crystallization nucleation behavior.The crystal types of TB are mainlyβ’-crystal form andβ’-like crystal form.The lower the cooling rate,the shorter the crystallization induction time,and the more the number of crystal nuclei.Therefore,during producing TB substitutes in the factory,the cooling rate can be controlled at10℃/min to reduce energy consumption and production costs.Chuanguo Ma Songbo Wang Tianlei Si Jieyu Li Xiaowei Chen 2021Grain & Oil Science and Technology2021,4,2:1

13Oxidation of benzene to phenol with N_(2)O over a hierarchical Fe/ZSM-5 catalyst显示文摘Catalytic oxidation of benzene with N_(2)O to phenol over the hierarchical and microporous Fe/ZSM-5-based catalysts in a continuous fixedbed reactor was investigated.The spent catalyst was in-situ regenerated by an oxidative treatment using N_(2)O and in total 10 reaction-regeneration cycles were performed.A 100% N_(2)O conversion,93.3% phenol selectivity,and high initial phenol formation rate of 16.49±0.06mmol_(phenol gcatalyst)^(-1)h^(-1)at time on stream(TOS) of 5 min,and a good phenol productivity of 147.06 mmol_(phenol gcatalyst)^(-1)during catalyst lifetime of 1800 min were obtained on a fresh hierarchical Fe/ZSM-5-Hi2.8 catalyst.With the reaction-regeneration cycle,N_(2)O conversion is fully recovered within TOS of 3 h,moreover,the phenol productivity was decreased ca.2.2±0.8% after each cycle,leading to a total phenol productivity of ca.0.44 ton_(pheol kg_(catalyst)^(-1)estimated for 300 cycles.Catalyst characterizations imply that the coke is rapidly deposited on catalyst surface in the initial TOS of 3 h(0.28 mgc_(gcatalyst)^(-1)min^(-1)) and gradually becomes graphitic during the TOS of 30 h with a slow formation rate of 0.06 mgc g_(catalyst)^(-1)min^(-1).Among others(e.g.,the decrease of textural property and acidity),the nearly complete coverage of the active Fe-O-Al sites by coke accounts for the main catalyst deactivation.Besides these reversible deactivation characteristics related to coking,the irreversible catalyst deactivation is also observed with the reaction-regeneration cycle.The latter is reflected by a further decreased amount of the active Fe-O-Al sites,which agglomerate on catalyst surface with the cycle,likely associated with the hard coke residue that is not completely removed by the regeneration.Cui Ouyang Jianwei Li Yaqi Qu Song Hong Songbo He 2023Green Energy & Environment2023,8,4:0
14Redox-dependent regulation of end-binding protein 1 activity by glutathionylation显示文摘Cytoskeletal proteins are susceptible to glutathionylation under oxidizing conditions,and oxidative damage has been implicated in several neurodegenerative diseases.End-binding protein 1(EB1)is a master regulator of microtubule plus-end tracking proteins(+TIPs)and is critically involved in the control of microtubule dynamics and cellular processes.However,the impact of glutathionylation on EB1 functions remains unknown.Here we reveal that glutathionylation is important for controlling EB1 activity and protecting EB1 from irreversible oxidation.In vitro biochemical and cellular assays reveal that EB1 is glutathionylated.Diamide,a mild oxidizing reagent,reduces EB1 comet number and length in cells,indicating the impairment of microtubule dynamics.Three cysteine residues of EB1 are glutathionylated,with mutations of these three cysteines to serines attenuating microtubule dynamics but buffering diamide-induced decrease in microtubule dynamics.In addition,glutaredoxin 1(Grx1)deglutathionylates EB1,and Grx1 depletion suppresses microtubule dynamics and leads to defects in cell division orientation and cell migration,suggesting a critical role of Grx1-mediated deglutathionylation in maintaining EB1 activity.Collectively,these data reveal that EB1 glutathionylation is an important protective mechanism for the regulation of microtubule dynamics and microtubule-based cellular activities.Miao Chen Jian Wang Yang Yang Tao Zhong Peng Zhou Huixian Ma Jingrui Li Dengwen Li Jun Zhou Songbo Xie Min Liu 2021Science China(Life Sciences)2021,64,4:0

16Mako:A Graph-based Pattern Growth Approach to Detect Complex Structural Variants显示文摘Complex structural variants(CSVs) are genomic alterations that have more than two breakpoints and are considered as the simultaneous occurrence of simple structural variants.However,detecting the compounded mutational signals of CSVs is challenging through a commonly used model-match strategy.As a result,there has been limited progress for CSV discovery compared with simple structural variants.Here,we systematically analyzed the multi-breakpoint connection feature of CSVs,and proposed Mako,utilizing a bottom-up guided model-free strategy,to detect CSVs from paired-end short-read sequencing.Specifically,we implemented a graph-based pattern growth approach,where the graph depicts potential breakpoint connections,and pattern growth enables CSV detection without pre-defined models.Comprehensive evaluations on both simulated and real datasets revealed that Mako outperformed other algorithms.Notably,validation rates of CSVs on real data based on experimental and computational validations as well as manual inspections are around 70%,where the medians of experimental and computational breakpoint shift are 13 bp and 26 bp,respectively.Moreover,the Mako CSV subgraph effectively characterized the breakpoint connections of a CSV event and uncovered a total of 15 CSV types,including two novel types of adjacent segment swap and tandem dispersed duplication.Further analysis of these CSVs also revealed the impact of sequence homology on the formation of CSVs.Mako is publicly available at http://gffzz188fe103f8f1460asoocc5f9ubbcf6xwq.ffgz.tsg.suse.edu.cn/xjtu-omics/Mako.Jiadong Lin Xiaofei Yang Walter Kosters Tun Xu Yanyan Jia Songbo Wang Qihui Zhu Mallory Ryan Li Guo Chengsheng Zhang The Human Genome Structural Variation Consortium Charles Lee Scott E.Devine Evan E.Eichler Kai Ye 2022Genomics, Proteomics & Bioinformatics2022,20,1:0
17Lithocholic acid promotes skeletal muscle regeneration through the TGR5 receptor显示文摘Lithocholic acid(LCA)is a classical secondary bile acid formed by the metabolism of gut microbiota.The TGR5 receptor(also known as G protein-coupled receptor 1,GPBAR1)is an important bile acid membrane receptor that mediates a variety of metabolic processes in vivo.In recent years,most studies have focused on the role of bile acid receptors in the intestine and liver.However,there are few reports on its effect on skeletal muscle regeneration,and the specific mechanism remains unclear.Therefore,it is necessary to investigate the mechanism of the TGR5 receptor in the regulation of skeletal muscle regeneration.The results demonstrate that muscle injection with LCA significantly reduces the necrosis rate of injured muscle and improves muscle injury.Moreover,treatment of C2C12 cells with LCA significantly increases AKT/mTOR/FoxO3 phosphorylation through the TGR5 receptor,enhances MyoG transcription and reduces FBXO32 transcription.These findings indicate that LCA can activate the TGR5/AKT signaling pathway,inhibit protein degradation and promote protein synthesis to enhance the myogenic process and promote skeletal muscle regeneration.Lijuan Sun Fan Li Weihao Tan Weijie Zhao Yongxiang Li Xiaotong Zhu Ping Gao Gang Shu Songbo Wang Qingyan Jiang Lina Wang 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
