3Effects of Salvia miltiorrhiza on intestinal microflora in rats with ischemia/reperfusion liver injury显示文摘BACKGROUND: Hepatic ischemia/reperfusion injury may induce intestinal microflora imbalance. Salvia miltiorrhiza is effective in promoting blood circulation and counteracting peroxidation in tissues. The aim of the present study was to determine the effects of Salvia miltiorrhiza on intestinal mi- croflora, endotoxemia, and bacterial translocation in rats with hepatic I/R injury. METHODS: Sprague-Dawley rats in specific pathogen free grade were divided into 3 groups: group I(n =6) for sham operation: groups ( n = 7) for liver ische- mia for 20 minutes and reperfusion for 22 hours. Group was also pretreated with 4 ml/day of Salvia miltiorrhiza solu- tion (250 mg/kg) by daily gavage for 7 days. The levels of serum alanine aminotransferase (ALT), aspartate amino- transferase (AST), malondialdehyde ( MDA) and supero- xide dismutase ( SOD ) in liver tissues, serum endotoxin, intestinal bacterial counts, intestinal mucosal histology and bacterial translocation were studied. RESULTS: The levels of ALT, AST, plasma endotoxin and MDA in liver tissues were decreased more markedly in group (57.57 ± 18.08 U/L, 147.57 ±40.84 U/L, 0.42 ± 0.144 EU/ml and 0. 52 ±0.19 nmol/mg-prot respectively) in group 295.9±216.92 U/L, 0.80± 0.262 EU/ml and 0.72±0.12 nmol/mg-prot; P <0.05-0.01 respectively). Liver SOD activity was increased more sig- nificantly in group (318.47±64.62 U/mg-prot) than in group U/mg-prot, P<0.05). The counts of Bifidobacteria and Bacteroides increased more significantly in group than in group but were similar to those in group I. Bacterial translocation to the kidney in group was 50% (5/10), whereas no bacterial translocation to the kidney occurred in the other two groups (P <0. 01). 5A multiplex PCR for differential detection of Echinococcus granulosus sensu stricto,Echinococcus multilocularis and Echinococcus canadensis in China显示文摘Background:Echinococcosis caused byEchinococcus is one of the most major infectious diseases in north-west highland of China.E.granulosus sensu strict,E.multilocularis,andE.canadensis are known to be the only three species related to human health transmitting in the areas.To achieve targeted treatment and control of echinococcosis,the accurate identification and discrimination of the species are important.However,currently the available diagnostic approaches do not present ideal results either in accuracy or efficiency.Methods:In the study,a set of primers were designed to aim at the three human-pathogenicEchinococcus species in China.The one-step multiplex PCR assay was developed and evaluated for the specificity and sensitivity.A total of 73parasitic lesions and 41 fecal materials obtained from human and various animals collected in the clinic and the field were tested to assess the applicability of this method.Results:The multiplex PCR effectively detected the individual DNA from the targeted species and their random mixtures generating with distinguishable expected size of products.The detection limit of the assay for each of the three species was 5 pg/μl when they were tested separately.When DNA mixtures of the targeted species containing the same concentration were used as templates,the lowest amount of DNA which can be detected was 50 pg/μl,10 pg/μl and 5 pg/μl forE.granulosus s.s.,E.multilocularis,andE.canadensis respectively.No cross-reactivity was observed when DNA from eight genetically close species was used as control templates.The multiplex PCR identifications of all samples were in line with the original sequencing results except for those infected withE.shiquicus,which showed negative signals in the developed assay.Of all the tested stool materials,16 were previously found positive forEchinococcus by visual and microscopic examination.Among these 16 samples,13 were confirmed by the multiplex PCR,and the other three tested negative.Additionally,the multiplex PCR identified another 14 positive feces from the remained 25 stool samples which absence of worms.Conclusions:The developed multiplex PCR shows advantages in fast diagnosis and large-scale epidemiological investigation,which proven to be a promising tool utilized in clinic and surveillance system.Jing-Ye Shang Guang-Jia Zhang Sha Liao Yan Huang Wen-Jie Yu Wei He Guang-You Yang Tiao-Ying Li Xing-Wang Chen Bo Zhong Qian Wang Qi Wang Rui-Rui Li Hao Wang 2019Infectious Diseases of Poverty2019,8,4:11

7Advanced oxidation of bromide-containing drinking water: A balance between bromate and trihalomethane formation control显示文摘Addition of H2O2 has been employed to repress bromate formation during ozonation of bromide-containing source water.However,the addition of H2O2 will change the oxidation pathways of organic compounds due to the generation of abundant hydroxyl radicals,which could afect the removal efcacy of trihalomethane precursors via the combination of ozone and biological activated carbon(O3-BAC).In this study,we evaluated the efects of H2O2 addition on bromate formation and trihalomethane formation potential(THMFP)reduction during treatment of bromide-containing(97.6–129.1μg/L)source water by the O3-BAC process.At an ozone dose of 4.2 mg/L,an H2O2/O3(g/g)ratio of over 1.0 was required to maintain the bromate concentration below 10.0μg/L,while a much lower H2O2/O3ratio was sufcient for a lower ozone dose.An H2O2/O3(g/g)ratio below 0.3 should be avoided since the bromate concentration will increase with increasing H2O2 dose below this ratio.However,the addition of H2O2 at an ozone dose of 3.2 mg/L and an H2O2/O3 ratio of 1.0 resulted in a 43% decrease in THMFP removal when compared with the O3-BAC process.The optimum H2O2/O3(g/g) ratio for balancing bromate and trihalomethane control was about 0.7–1.0.Fractionation of organic materials showed that the addition of H2O2 decreased the removal efcacy of the hydrophilic matter fraction of DOC by ozonation and increased the reactivity of the hydrophobic fractions during formation of trihalomethane,which may be the two main reasons responsible for the decrease in THMFP reduction efcacy.Overall,this study clearly demonstrated that it is necessary to balance bromate reduction and THMFP control when adopting an H2O2 addition strategy.Yongjing Wang Jianwei Yu Po Han Jing Sha Tao Li Wei An Juan Liu Min Yang 2013Journal of Environmental Sciences2013,25,11:9

10Urodynamic assessment of bladder storage function after radical hysterectomy for cervical cancer显示文摘Background:After radical hysterectomy for cervical cancer,the most common complication is lower urinary tract symptoms.Post-operatively,bladder capacity can alter bladder function for a prolonged period.This study aimed to identify factors affecting bladder storage function.Methods:A multicenter,retrospective cohort study was conducted.Information of patients with stages IA2 to IIB cervical cancer with urodynamic study results were retrospectively collected from nine hospitals between June 2013 and June 2018 according to the inclusion criteria.Demographic,surgical,and oncological data were collected.The univariate and multivariate logistic regression was used to identify clinical factors associated with bladder storage function.Results:Two hundred and three patients with cervical cancer had urodynamic testing post-operatively.Ninety-five(46.8%)patients were diagnosed with stress urinary incontinence(SUI).The incidence of low bladder compliance(LBC)was 23.2%.Twenty-seven(13.3%)patients showed detrusor overactivity(DO).Fifty-seven patients(28.1%)presented with a decreased maximum cystometric capacity(DMCC).The probability of composite bladder storage dysfunction was 68.0%.Multivariate analysis confirmed that laparoscopy represents a protective factor for SUI with an odds ratio of 0.498(P=0.034).Patients who underwent a nerve-sparing procedure were less odds to experience SUI(P=0.014).A significant positive correlation between LBC and DO was observed(P<0.001).A greater length of the resected vagina and chemoradiotherapy were common risk factors for LBC and DO,while radiotherapy exerted a stronger effect than chemotherapy.Additionally,patients who received chemoradiotherapy frequently developed a DMCC.The follow-up time was not correlated with bladder storage function.Conclusion:A nerve-sparing procedure without longer resected vagina is recommended for protecting the bladder storage function.Ting-Ting Cao Hong-Wu Wen Yu-Nong Gao Qiu-Bo Lyu Hui-Xin Liu Sha Wang Shi-Yan Wang Hua-Xin Sun Na Yu Hai-Bo Wang Yi Li Zhi-Qi Wang Olivia H.Chang Xiu-Li Sun Jian-Liu Wang 2020Chinese Medical Journal2020,,19:7
11The Relationship between Tectonic Subsidence and BSR of Upper Neogene in the Deep-Water Area of the Northern Continental Slope, South China Sea显示文摘BSR (Bottom Simulating Reflector) occurs widely in the strata since the late Miocene in the deep-water area of the northern continental slope of South China Sea (SCS). It is an important seismic reference mark which identifies the gas hydrate and its distribution influenced by the tectonic movements. Single-point basin modeling was conducted using 473 points in the study area. To discuss the relationships between the tectonic subsidence and BSR, the volume and rate of tectonic subsidence in each geological time have been simulated. The results show that there are three tectonic accelerate subsidence processes in the study area since the late Miocene, especially since 1.8Ma the tectonic subsidence accelerates more apparently. Since the Late Miocene to Pleistocene, the rate of tectonic subsidence in deep-water underwent a transformation from weak to strong. The ratio of tectonic subsidence to the total subsidence was relatively high (65-70%). 13Biofilm-inspired adhesive and antibacterial hydrogel with tough tissue integration performance for sealing hemostasis and wound healing显示文摘Uncontrolled bleeding and infection can cause significant increases in mortalities.Hydrogel sealants have attracted extensive attention for their ability to control bleeding.However,because interfacial water is a formidable barrier to strong surface bonding,a challenge remains in finding a product that offers robust tissue adhesion combined with anti-infection properties.Inspired by the strong adhesive mechanism of biofilm and mussels,we report a novel dual bionic adhesive hydrogel(DBAH)based on chitosan grafted with methacrylate(CS-MA),dopamine(DA),and N-hydroxymethyl acrylamide(NMA)via a facile radical polymerization process.CS-MA and DA were simultaneously included in the adhesive polymer for imitating the two key adhesive components:polysaccharide intercellular adhesin(PIA)of staphylococci biofilm and 3,4-dihydroxy-L-phenylalanine(Dopa)of mussel foot protein,respectively.DBAH presented strong adhesion at 34 kPa even upon three cycles of full immersion in water and was able to withstand up to 168 mm Hg blood pressure,which is significantly higher than the 60–160 mm Hg measured in most clinical settings.Most importantly,these hydrogels presented outstanding hemostatic capability under wet and dynamic in vivo movements while displaying excellent antibacterial properties and biocompatibility.Therefore,DBAH represents a promising class of biomaterials for high-efficiency hemostasis and wound healing.Wei Han Bo Zhou Kai Yang Xin Xiong Shifang Luan Yu Wang Zheng Xu Peng Lei Zhengshan Luo Jian Gao Yijing Zhan Guopu Chen Lei Liang Rui Wang Sha Li Hong Xu 2020Bioactive Materials2020,5,4:6

15Dmrt1 regulates the immune response by repressing the TLR4 signaling pathway in goat male germline stem cells显示文摘Double sex and mab-3-related transcription factor 1(Dmrt1),which is expressed in goat male germline stem cells(mGSCs)and Sertoli cells,is one of the most conserved transcription factors involved in sex determination.In this study,we highlighted the role of Dmrt1 in balancing the innate immune response in goat mGSCs.Dmrt1 recruited promyelocytic leukemia zinc finger(Plzf),also known as zinc finger and BTB domain-containing protein 16(Zbtb16),to repress the Toll-like receptor 4(TLR4)-dependent inflammatory signaling pathway and nuclear factor(NF)-κB.Knockdown of Dmrt1 in seminiferous tubules resulted in widespread degeneration of germ and somatic cells,while the expression of proinflammatory factors were significantly enhanced.We also demonstrated that Dmrt1 stimulated proliferation of mGSCs,but repressed apoptosis caused by the immune response.Thus,Dmrt1 is sufficient to reduce inflammation in the testes,thereby establishing the stability of spermatogenesis and the testicular microenvironment.Yu-Dong Wei Xiao-Min Du Dong-Hui Yang Fang-Lin Ma Xiu-Wei Yu Meng-Fei Zhang Na Li Sha Peng Ming-Zhi Liao Guang-Peng Li Chun-Ling Bai Wei-Shuai Liu Jin-Lian Hua 2021Zoological Research2021,42,1:5
16LIN28A inhibits DUSP family phosphatases and activates MAPK signaling pathway to maintain pluripotency in porcine induced pluripotent stem cells显示文摘LIN28A,an RNA-binding protein,plays an important role in porcine induced pluripotent stem cells(piPSCs).However,the molecular mechanism underlying the function of LIN28A in the maintenance of pluripotency in piPSCs remains unclear.Here,we explored the function of LIN28A in piPSCs based on its overexpression and knockdown.We performed total RNA sequencing(RNA-seq)of piPSCs and detected the expression levels of relevant genes by quantitative real-time polymerase chain reaction(qRT-PCR),western blot analysis,and immunofluorescence staining.Results indicated that piPSC proliferation ability decreased following LIN28A knockdown.Furthermore,when LIN28A expression in the shLIN28A2 group was lower(by 20%)than that in the negative control knockdown group(shNC),the pluripotency of piPSCs disappeared and they differentiated into neuroectoderm cells.Results also showed that LIN28A overexpression inhibited the expression of DUSP(dual-specificity phosphatases)family phosphatases and activated the mitogen-activated protein kinase(MAPK)signaling pathway.Thus,LIN28A appears to activate the MAPK signaling pathway to maintain the pluripotency and proliferation ability of piPSCs.Our study provides a new resource for exploring the functions of LIN28A in piPSCs.Xiao-Long Wu Zhen-Shuo Zhu Xia Xiao Zhe Zhou Shuai Yu Qiao-Yan Shen Ju-Qing Zhang Wei Yue Rui Zhang Xin He Sha Peng Shi-Qiang Zhang Na Li Ming-Zhi Liao Jin-Lian Hua 2021Zoological Research2021,42,3:5
17Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. 20Highway constructions on the Qinghai-Tibet Plateau:Challenge,research and practice显示文摘Highway constructions on the Qinghai-Tibet Plateau(QTP)face great challenges induced by the unique local environmental,geological,and engineering conditions.The large area of permafrost,great temperature variability,strong UV rays,and complex geological conditions are the major factors that adversely influence the longterm performance of pavement systems.Since 1960s,Chinese engineers and researchers have started conducting research on the QTP to enhance the performance and durability of pavement systems.The present paper provide a comprehensive review of challenge,research and practice of highway constructions on the QTP including the special environmental and geological conditions,history of highway constructions on the QTP,major challenges and the state-of-the-art technology of subgrade constructions on permafrost,developments of the pavement structures and materials,performance prediction and maintenance methods of pavement surfaces,and applications of the research achievements on the first expressway on the QTP(i.e.,Gongyu Expressway).Based on the comprehensive literature review,it can be found that(1)frost heave and thaw weakening induced subgrade disease and longitudinal cracks on the pavement surface are complex coupled water-thermal-load problems.Engineering solutions are focusing on active cooling and thermal insulation methods,which can help to reduce temperature variations in the subgrade and thus improving its stability,(2)the harsh environmental and construction conditions may reduce the early strength and induce premature damage of cement-treated base materials.Some field validations showed that geocell-reinforced or asphalt-treated flexible base materials can provide better long-term performance,(3)the large temperature variability and strong UV rays can significantly accelerate aging of asphalt binders and greatly reduce the service life of asphalt mixtures.Various binder modification methods were developed for improving their viscoelasticity and enhance the low-temperature cracking resistance of pavement surface materials but are still lack of field validation data and comparisons of their life cycle costs.Therefore,it is recommended that a demonstration research project build test sections to examine a range of pavement structures and materials,and compare their long-term performance and life cycle costs,which can serve as important reference for future highway constructions on the QTP.Aimin Sha Biao Ma Hainian Wang Liqun Hu Xuesong Mao Xilan Zhi Huaxin Chen Yu Liu Feng Ma Zhuangzhuang Liu Rui He Wei Si Xuhao Wang Cheng Li 2022Journal of Road Engineering2022,2,1:4
