1On the tuning of electrical and thermal transport in thermoelectrics: an integrated theory–experiment perspective显示文摘During the last two decades,we have witnessed great progress in research on thermoelectrics.There are two primary focuses.One is the fundamental understanding of electrical and thermal transport,enabled by the interplay of theory and experiment;the other is the substantial enhancement of the performance of various thermoelectric materials,through synergistic optimisation of those intercorrelated transport parameters.Here we review some of the successful strategies for tuning electrical and thermal transport.For electrical transport,we start from the classical but still very active strategy of tuning band degeneracy(or band convergence),then discuss the engineering of carrier scattering,and finally address the concept of conduction channels and conductive networks that emerge in complex thermoelectric materials.For thermal transport,we summarise the approaches for studying thermal transport based on phonon–phonon interactions valid for conventional solids,as well as some quantitative efforts for nanostructures.We also discuss the thermal transport in complex materials with chemical-bond hierarchy,in which a portion of the atoms(or subunits)are weakly bonded to the rest of the structure,leading to an intrinsic manifestation of part-crystalline part-liquid state at elevated temperatures.In this review,we provide a summary of achievements made in recent studies of thermoelectric transport properties,and demonstrate how they have led to improvements in thermoelectric performance by the integration of modern theory and experiment,and point out some challenges and possible directions.Jiong Yang Lili Xi Wujie Qiu Lihua Wu Xun Shi Lidong Chen Jihui Yang Wenqing Zhang Ctirad Uher David J Singh 2016npj Computational Materials2016,,1:24
2A rare Waxy allele coordinately improves rice eating and cooking quality and grain transparency显示文摘In rice(Oryza sativa), amylose content(AC) is the major factor that determines eating and cooking quality(ECQ). The diversity in AC is largely attributed to natural allelic variation at the Waxy(Wx)locus. Here we identified a rare Wx allele, Wx^(mw) ,which combines a favorable AC, improved ECQ and grain transparency. Based on a phylogenetic analysis of Wx genomic sequences from 370 rice accessions, we speculated that Wx^(mw) may have derived from recombination between two important natural Wx alleles, Wx^(in) and Wx^(b). We validated the effects of Wx^(mw) on rice grain quality using both transgenic lines and near-isogenic lines(NILs). When introgressed into the japonica Nipponbare(NIP) background, Wx^(mw) resulted in a moderate AC that was intermediate between that of NILs carrying the Wx^(b)allele and NILs with the Wx^(mp) allele. Notably, mature grains of NILs fixed for Wx^(mw) had an improved transparent endosperm relative to soft rice. Further, we introduced Wx^(mw) into a high-yielding japonica cultivar via molecular marker-assisted selection: the introgressed lines exhibited clear improvements in ECQ and endosperm transparency. 4PD-L1 is a direct target of cancer-FOXP3 in pancreatic ductal adenocarcinoma(PDAC),and combined immunotherapy with antibodies against PD-L1 and CCL5 is effective in the treatment of PDAC显示文摘High expression of PD-L1 marks the poor prognosis of pancreatic ductal adenocarcinomas(PDAC).However,the regulatory mechanism of PD-L1 remains elusive.We recently reported that cancer Forkhead box protein 3(Cancer-FOXP3 or C-FOXP3)promoted immune evasion of PDAC by recruiting Treg cells into PDAC via upregulation of CCL5.In this study,we confirmed that PD-L1 was overexpressed in PDAC samples from two independent cohorts of patients with radical resection.Moreover,C-FOXP3 was colocalized and correlated with the expression of PD-L1 in tumor cells at the mRNA and protein levels,and this finding was confirmed by the The Cancer Genome Atlas(TCGA)database.Chromatin immunoprecipitation(ChIP)revealed that C-FOXP3 directly bound to the promoter region of PD-L1 in pancreatic cancer cells.Furthermore,overexpression of C-FOXP3 activated the luciferase reporter gene under the control of the PD-L1 promoter.However,mutation of the binding motif-a completely reversed the luciferase activity.In addition,C-FOXP3-induced upregulation of PD-L1 effectively inhibited the activity of CD8+T cells.Based on our recent finding that the CCL-5 antibody achieved a better response to PDAC models with high C-FOXP3 levels,we further demonstrated that the PD-L1 antibody strengthened the antitumor effect of CCL-5 blockade in xenograft and orthotopic mouse models with high C-FOXP3 levels.In conclusion,C-FOXP3 directly activates PD-L1 and represents a core transcription factor that mediates the immune escape of PDAC.Combined blockade of PD-L1 and CCL-5 may provide an effective therapy for patients with PDAC that have high C-FOXP3 levels.Xiuchao Wang Xin Li Xunbin Wei Haiping Jiang Chungen Lan Shengyu Yang Han Wang Yanhui Yang Caijuan Tian Zanmei Xu Jiangyan Zhang Jihui Hao He Ren 2020Signal Transduction and Targeted Therapy2020,5,1:11
5Long-distance ranging with high precision using a soliton microcomb显示文摘Laser-based light detection and ranging(lidar)plays a significant role in both scientific and industrial areas.However,it is difficult for existing lidars to achieve high speed,high precision,and long distance simultaneously.Here,we demonstrate a high-performance lidar based on a chip-scaled soliton microcomb(SMC)that can realize all three specialties simultaneously.Aided by the excellent properties of ultrahigh repetition rate and the smooth envelope of the SMC,traditional optical frequency comb(OFC)-based dispersive interferometry is heavily improved and the measuring dead zone induced by the mismatch between the repetition rate of the OFC and resolution of the optical spectrum analyzer is totally eliminated.Combined with an auxiliary dual-frequency phase-modulated laser range finder,the none-dead-zone measurable range ambiguity is extended up to 1500 m.The proposed SMC lidar is experimentally implemented in both indoor and outdoor environment.In the outdoor baseline field,real-time,high-speed(up to 35 k Hz)measurement of a long distance of^1179 m is achieved with a minimum Allan deviation of 5.6μm at an average time of 0.2 ms(27 nm at an average time of 1.8 s after high-pass filtering).The present SMC lidar approaches a compact,fast,high-precision,and none-dead zone long-distance ranging system,aimed at emerging applications of frontier basic scientific research and advances in industrial manufacturing.JINDONG WANG ZHIZHOU LU WEIQIANG WANG FUMIN ZHANG JIAWEI CHEN YANG WANG JIHUI ZHENG SAI T.CHU WEI ZHAO BRENT E.LITTLE XINGHUA QU WENFU ZHANG 2020Photonics Research2020,8,12:8
6Quantitative nanoscale mapping of three-phase thermal conductivities in filled skutterudites via scanning thermal microscopy显示文摘In the last two decades, a nano structuring paradigm has been successfully applied in a wide range of thermoelectric materials, resulting in significant reduction in thermal conductivity and superior thermoelectric performance. These advances, however, have been accomplished without directly investigating the local thermoelectric properties, even though local electric current can be mapped with high spatial resolution. In fact, there still lacks an effective method that links the macroscopic thermoelectric performance to the local micro structures and properties. Here, we show that local thermal conductivity can be mapped quantitatively with good accuracy, nanometer resolution and one-to-one correspondence to the micro structure using a three-phase skutterudite as a model system. Scanning thermal microscopy combined with finite element simulations demonstrate close correlation between sample conductivity and probe resistance,enabling us to distinguish thermal conductivities spanning orders of magnitude, yet resolving thermal variation across a phase interface with small contrast. 8Prognostic significance of combining high mobility group Box-1 and OV-6 expression in hepatocellular carcinoma显示文摘The inflammatory environment and existence of cancer stem cells are critical for progression and intrahepatic recurrence of hepatocellular carcinoma(HCC) after curative resections. Here, we investigated the prognostic significance of combining high mobility group box 1(HMGB1) expression and hepatic progenitor marker OV6 in hepatocellular carcinoma. Expression of HMGB1 and OV6 was evaluated using immunohistochemistry profiling in tissue microarrays containing samples from 208 HCC patients. Invasive clinical or pathological factors were found in patients with high expression of HMGB1 or OV6. Higher HMGB1 was associated with poorer clinical outcomes, and independently related to elevated 5-year recurrence incidence(85.5% vs. 62.4%, P<0.001). We also found that more OV6 positive staining was correlated with poor prognosis of HCC patients(P<0.001). Notably, expression of HMGB1 was positively correlated with OV6 in density(R2=0.032, P<0.001) and reversely related to HCC outcomes. Abnormal expression of HMGB1 in combination with positive staining of OV6 displayed poorer prognostic performance than single biomarker alone(area under curve(AUC) survival=0.696). 9Efficacy of moxibustion by stimulating acupoints of Danzhong(CV 17) and Ganshu(BL 18) on hyperplasia of mammary gland in rats显示文摘OBJECTIVE: To evaluate the efficacy of moxibustion, through stimulating acupoints of Danzhong(CV 17) and Ganshu(BL 18) in rats with hyperplasia of mammary gland(HMG) which induced by estrogen and progestogen.METHODS: Thirty female Sprague-Dawley rats were randomly divided into saline control group,HMG model group, and HMG moxibustion group with 10 in each group. Saline control was the group injected by saline. HMG model were created by injection of estrogen and progestogen. Moxibustion group was also injected of estrogen and progesto-gen with moxibustion at the same time. The Changes of nipple diameter and height were measured.The rats' skin temperature was recorded by an infrared thermal camera at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). Pathological changes of mammary gland in rats were also observed under light microscope.RESULTS: The diameter and height of the nipples in model group were prominently bigger and higher than that in control group(P < 0.01). The diameter and height in moxibustion group were prominently smaller and lower than that in model group(P < 0.01), and there was no significant difference between moxibustion group and control group.Compared with control group, skin temperature of the nipples, mammary area, and acupoints Danzhong(CV 17) and Ganshu(BL 18) decreased prominently in model group(P < 0.01-0.05). Compared with model group, skin temperature of that in moxibustion group increased prominently(P <0.05).CONCLUSION: Treatment with moxibustion can effectively decrease the HMG rats' nipple diameter and height, and increase the skin temperature in HMG model rats at the nipples, mammary areas,Danzhong(CV 17) and Ganshu(BL 18). 10Possibility of women treated with fertility-sparing surgery fornon-epithelial ovarian tumors to safely and successfully becomepregnant--a Chinese retrospective cohort study among 148cases显示文摘This study was performed to evaluate the oncological and reproductive outcomes of childbearing-agewomen treated with fertility-sparing surgery (FSS) for non-epithelial ovarian tumors in China. One hundred andforty eight non-epithelial ovarian tumor women treated with FSS between January I, 2000 and August 31, 2015from two medical centers in China were identified. Progression-free survival (PFS) was 88.5%, whereas overallsurvival (OS) was 93.9%. Univariate analysis suggested that delivery after treatment is related to PFS (P = 0.023),whereas histology significantly influenced OS. Cox regression analysis suggested that only histology was associatedwith PFS and OS (P 〈 0.05). Among the 129 women who completed adjuvant chemotherapy (ACT), nonedeveloped amenorrhea. Among the 44 women who desired pregnancy, 35 (79.5%) successfully had 51 gestationsincluding 35 live births without birth defects. Non-epithelial ovarian tumors can achieve fulfilling prognosis afterFSS and chemotherapy. Histology might be the only independent prognostic factor for PFS and OS. FSS followedby ACT appeared to have little or no effect on fertility. Meanwhile, postoperative pregnancy did not increase thePFS or OS. 11Hypoxic microenvironment induced spatial transcriptome changes in pancreatic cancer显示文摘Objective: Hypoxia is a significant feature of solid tumors, including pancreatic ductal adenocarcinoma(PDAC). It is associated with tumor invasion, metastasis, and drug resistance. However, the spatial distribution of hypoxia-related heterogeneity in PDAC remains unclear.Methods: Spatial transcriptomics(STs), a new technique, was used to investigate the ST features of engrafted human PDAC in the ischemic hind limbs of nude mice. Transcriptomes from ST spots in the hypoxic tumor and the control were clustered using differentially-expressed genes. These data were compared to determine the spatial organization of hypoxia-induced heterogeneity in PDAC. Clinical relevance was validated using the Tumor Cancer Genome Atlas and KM-plotter databases. The CMAP website was used to identify molecules that may serve as therapeutic targets for PDAC.Results: ST showed that the tumor cell subgroups decreased to 7 subgroups in the hypoxia group, compared to 9 subgroups in the control group. Different subgroups showed positional characteristics and different gene signatures. Subgroup 6 located at the invasive front showed a higher proliferative ability under hypoxia. Subgroup 6 had active functions including cell proliferation, invasion, and response to stress. Expressions of hypoxia-related genes, LDHA, TPI1, and ENO1, induced changes. CMAP analysis indicated that ADZ-6482, a PI3 K inhibitor, was targeted by the invasive subgroup in hypoxic tumors.Conclusions: This study is the first to describe hypoxic microenvironment-induced spatial transcriptome changes in PDAC, and to identify potential treatment targets for PDAC. 17Identification of a dominant murine T-cell epitope in recombinant protein P29 from Echinococcus granulosus显示文摘Echinococcus granulosus causes echinococcosis,an important zoonotic disease worldwide and a major public health issue.Vaccination is an economical and practical approach for controlling E.granulosus.We have previously revealed that a recombinant protein P29(rEg.P29)is a good vaccine candidate against E.granulosus.However,T cell immunogenic epitopes have not been identified.In the present study,we use rEg.P29-immunized mice as models to screen immunogenic epitopes for the construction of a novel multi-epitope vaccine.We search for immunodominant epitopes from an overlapping peptide library to screen the peptides of rEg.P29.Our results confirm that rEg.P29 immunization in mice elicits the activation of T cells and induces cellular immune responses.Further analyses show that a T cell epitope within amino acids 86–100 of rEg.P29 elicits significant antigen-specific IFN-γproduction in CD4^(+) and CD8^(+) T cells and promotes specific T-cell activation and proliferation.Collectively,these results provide a reference for the construction of a novel vaccine against broad E.granulosus genotypes based on epitopes of rEg.P29.Yongxue Lv Yazhou Zhu Liangliang Chang Jihui Yang Yinqi Zhao Jiaqing Zhao Yana Wang Mingxing Zhu Changyou Wu Wei Zhao 2022Acta Biochimica et Biophysica Sinica2022,54,4:1
