1Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 8Advances in the study of helminth mitochondrial genomes and their associated applications显示文摘Helminths, including flatworms and roundworms, are abundant organisms that have a variety of life histories. Of these, the genera Schistosoma, Echinococcus, Trichinella are notable parasites of veterinary and medical importance, and cause substantial socio- economic losses throughout China and the rest of the world. Genetic markers in the mitochondrial (mt) genome have proven use- ful for systematic, ecological, evolutionary and population studies, and the growth of mt genomic research has increased in the last two decades. Technological improvements, such as the long-polymerase chain reaction method and high-throughput se- quencing have allowed minute amounts of DNA from single worms, biopsy samples or microscopic organisms to be used for whole mt genome characterization. To facilitate the retrieval, annotation and analyses of mitochondrial features, multiple data- bases and specific software have also been designed and established. This review focuses on current progress, applications and perspectives regarding helminth mt genomics. To date, the complete mt genomes for 93 species of helminths have been sequenced and analyzed. Analyses of the mt genes, including gene content, arrangement, composition and variation have revealed unique features among the helminths when compared with other metazoans. This provides important data concerning their functional and comparative mitochondrial genomics, molecular taxonomy and characterization, population genetics and systematics, and evolu- tionary history. Moreover, mt genome data for parasitic helminths are important for diagnosis, epidemiology and ecology of in- fections. 10Characteristics of TP53 germline variants and their correlations with Li-Fraumeni syndrome or Li-Fraumeni-like syndrome in Chinese tumor patients显示文摘Li-Fraumeni syndrome(LFS),a rare autosomal-dominant inheritance condition,is associated with a family cancer history as well as pathogenic/likely-pathogenic TP53 germline variants(P/LP TP53 GV).The current clinical methods for detecting LFS are limited.Here,we retrospectively investigate P/LP TP53 GV among Chinese cancer patients by next-generation sequencing and evaluate its relationship with a family cancer history.A total of 270 out of 19,226 cancer patients have TP53 GV,including 53 patients with P/LP TP53 GV.Patients with P/LP TP53 GV are mainly found in male with glioma,lung cancer or sarcoma.The median age of diagnosis for P/LP TP53 GV patients is significantly lower than that of non-P/LP TP53 GV patients(31-years vs.53-years;P<0.01).One LFS patient and 3 Li-Fraumeni-like syndrome(LFL)patients are among the 26 followed-up P/LP TP53 GV patients.Among 25 types of P/LP TP53 GV,the highest variant frequencies occurred at codon 175 and 248.p.M237 I,p.R158 H,p.C238 Y and p.C275 R,are firstly identified among the Chinese LFS/LFL patients.This study reports the(P/LP)TP53 GV characteristics of Chinese pan-cancer patients.These findings suggest analyzing the P/LP TP53 GV in cancer patients is an effective strategy for identifying cancer predisposition syndrome.Panwen Tian Xiaoyan Zhang Sheng Yang Yu Fang Hongling Yuan Wei Li Honglin Zhu Fangping Zhao Jinlei Ding Yunshu Zhu Sizhen Wang Guochen Sun Hongbin Ni Tonghui Ma Ting Lei 2022Journal of Genetics and Genomics2022,49,7:0
11Design of B/N Co-doped micro/meso porous carbon electrodes from CNF/BNNS/ZIF-8 nanocomposites for advanced supercapacitors显示文摘Boron(B)and nitrogen(N)co-doped 3D hierarchical micro/meso porous carbon(BNPC)were successfully fabricated from cellulose nanofiber(CNF)/boron nitride nanosheets(BNNS)/zinc-methylimidazolate framework-8(ZIF-8)nanocomposites prepared by 2D BNNS,ZIF-8 nanoparticles,and wheat straw based CNFs.Herein,CNF/ZIF-8 acts as versatile skeleton and imparts partial N dopant into porous carbon structure,while the introduced BNNS can help strengthen the hierarchical porous superstructure and endow abundant B/N co-dopants within BNPC matrix.The obtained BNPC electrode possesses a high specific surface area of 505.4 m2/g,high B/N co-doping content,and desirable hydrophilicity.Supercapacitors assembled with BNPC-2(B/N co-doped porous carbon with a CNF/BNNS mass ratio of 1꞉2)electrodes exhibited exceptional electrochemical performance,demonstrating high capacitance stability even after 5000 charge-discharge cycles.The devices exhibited outstanding energy density and power density,as well as the highest specific capacitance of 433.4 F/g at 1.0 A/g,when compared with other similar reports.This study proposes a facile and sustainable strategy for efficiently fabrication of rich B/N co-doped hierarchical micro/meso porous carbon electrodes from agricultural waste biomass for advanced supercapacitor performance.Zhen Shang Xingye An Shuangxi Nie Na Li Haibing Cao Zhengbai Cheng Hongbin Liu Yonghao Ni Liqin Liu 2023Journal of Bioresources and Bioproducts2023,8,3:0
12Data-driven based variable geometry design显示文摘Conventionally,the method to make up for the missing data of middle-shallow layer in the obstacle area is by variable geometry,for example,deviating physical points and adding sources and receivers.And the missing data of middle-shallow layer is evaluated according to the effective coverage of the target layer.Since the traditional method doesn't consider the actual seismic data,it is impossible to actually predict the gap of section and the imaging effect.The paper proposes the evaluation method of data-driven based variable geometry:Firstly,the obstacle avoidance design is realized according to the coordinate range and safe distance of the obstacle area;Secondly,the local similarity of each common image gather(CIG)is calculated,and the contribution of the sources and receivers to the target area is also calculated;Thirdly,according to the variable geometry design,choose the required trace to perform sorting and stacking according to the contribution of the sources and receivers in the CIG,the stack data volume of the whole work area is generated;finally,evaluate the missing data in the obstacle area by the extracted seismic stacked sections in different direction and guide the designer in the infilling plan.Meanwhile,for area with very low signal to noise ratio(SNR),the new method can be used to evaluate the imaging potential and guide the survey design.The new method has achieved very good effect in the production,and the analysis result is very consistent with the processed result of the actual seismic data.Yinpo Xu Cheng Yin Xuefeng Zou Yingjie Pan Yudong Ni Hongbin Yi 2021Earthquake Research Advances2021,1,S01:0

14Detection of FOXO1 break-apart status by fluorescence in situ hybridization in atypical alveolar rhabdomyosarcoma显示文摘The morphologies of alveolar rhabdomyosarcoma(ARMS) are various. Some cases entirely lack an alveolar pattern and instead display a histological pattern that overlaps with embryonal rhabdomyosarcoma(ERMS). The method of pathological diagnosis of ARMS and ERMS has been updated in the 4th edition of the World Health Organization's guidelines for classification of skeletal muscle tumors. Under the new guidelines, there is still no molecular test to distinguish between these two subtypes of rhabdomyosarcoma(RMS). In the present study, we applied fluorescent in situ hybridization(FISH) and found that the Forkhead box O1(FOXO1) gene broke apart, amplified, and displayed an aneuploid signal that was related to the RMS pathological subtype.Aside from the fact that FOXO1 break-apart and its amplification were correlated with atypical ARMS, aneuploidies were usually found in atypical ERMS. 15N-doped carbon nanocube with zinc oxide sodiophilic sites enables a superior sodium metal anode显示文摘The metallic Na has been regarded as the most promising anode for next-generation sodium metal batteries(SMBs)owing to its high theoretical specific capacity,low redox potential,and low cost.The practical applications of Na metal,however,have still been severely hindered by the uncontrolled sodium dendrites growth during Na deposition and stripping processes,which leads to low Coulombic efficiency and poor cycling stability.In this study,sub-nano zinc oxide(ZnO)uniformly dispersed in threedimensional(3D)porous nitrogen-doped(N-doped)carbon nanocube(ZnO@NC)was acquired as a stable host for dendrite-free Na metal anode.Benefiting from the in-situ electrochemically formed sodiophilic nucleation site(NaZn13 alloy)and the enriched pore structure,rapid and uniform sodium deposition behavior can be performed.As expected,the ZnO@NC electrode delivers impressive electrochemical performance,an ultra-high areal capacity of 20 mAh·cm−2 in the half-cell can be maintained for 2,000 h.In the symmetrical-cell,it can also exhibit up to 3,000 h at 3 mA·cm−2 and 3 mAh·cm−2 with low polarization potential.Furthermore,in the full-cell that matches with Prussian blue(PB)cathode,the Na@ZnO@NC anode performs the outstanding long-cycling and rate performance.Therefore,this work provides an effective strategy to inhibit the growth of Na dendrites for the development of high-safety and long-cycling SMBs.Yijuan Li Pan Xu Hongbin Ni Jirong Mou Tangchao Xie Hong Xiao Hengji Zhu Quanfeng Dong Shaoming Huang 2023Nano Research2023,16,1:0
