1Anomaly Detection of Industrial Control Systems Based on Transfer Learning显示文摘Industrial Control Systems(ICSs)are the lifeline of a country.Therefore,the anomaly detection of ICS traffic is an important endeavor.This paper proposes a model based on a deep residual Convolution Neural Network(CNN)to prevent gradient explosion or gradient disappearance and guarantee accuracy.The developed methodology addresses two limitations:most traditional machine learning methods can only detect known network attacks and deep learning algorithms require a long time to train.The utilization of transfer learning under the modification of the existing residual CNN structure guarantees the detection of unknown attacks.One-dimensional ICS flow data are converted into two-dimensional grayscale images to take full advantage of the features of CNN.Results show that the proposed method achieves a high score and solves the time problem associated with deep learning model training.The model can give reliable predictions for unknown or differently distributed abnormal data through short-term training.Thus,the proposed model ensures the safety of ICSs and verifies the feasibility of transfer learning for ICS anomaly detection.Weiping Wang Zhaorong Wang Zhanfan Zhou Haixia Deng Weiliang Zhao Chunyang Wang Yongzhen Guo 2021Tsinghua Science and Technology2021,26,6:11
2Stem cell transplantation for treatment of cerebral ischemia in rats Effects of human umbilical cord blood stem cells and human neural stem cells显示文摘BACKGROUND: Exogenous neural stem cell transplantation promotes neural regeneration. However, various types of stem cells transplantation outcomes remain controversial. OBJECTIVE: To explore distribution, proliferation and differentiation of human neural stem cells (hNSCs) and human umbilical cord blood stem cells (hUCBSCs) following transplantation in ischemic brain tissue of rats, and to compare therapeutic outcomes between hNSCs and hUCBSCs. DESIGN, TIME AND SETTING: Randomized controlled animal studies were performed at the Experimental Animal Center of Nanjing Medical University and Central Laboratory of Second Affiliated Hospital of Nanjing Medical University of China from September 2008 to April 2009. MATERIALS: hNSCs were harvested from brain tissue of 10-13 week old fetuses following spontaneous abortion, and hUCBSCs were collected from umbilical cord blood of full-term newborns at the Second Affiliated Hospital of Nanjing Medical University of China. hNSCs and hUCBSCs were labeled by 5-bromodeoxyuridine (BrdU) prior to transplantation. METHODS: Rat models of cerebral ischemia were established by the suture method. A total of 60 healthy male Sprague Dawley rats aged 7-9 weeks were randomly assigned to hNSC transplantation, hUCBSC transplantation and control groups. The rat models in the hNSC transplantation, hUCBSC transplantation and control groups were infused with hNSC suspension, hUCBSC suspension and saline via the caudal vein, respectively. MAIN OUTCOME MEASURES: The distribution, proliferation and differentiation of hNSCs and hUCBSCs in ischemic brain tissue were observed using immunohistochemical methods. Neurological function in rats was assessed using the neurological severity score. RESULTS: The number of BrdU-positive cells was significantly greater in the hNSC transplantation group compared with hUCBSC transplantation group at 14 days following transplantation (P < 0.05). The number of BrdU-positive cells reached a peak at 28 days following transplantation. Nestin-positive, glial fibrillary acidic protein-positive, cyclic nucleotide 3' phosphohydrolase-positive and neuron specific enolase-positive cells were visible following transplantation. No significant difference was determined in the constituent ratio of various cells between hNSC and hUCBSC transplantation groups (P > 0.05). The neurological severity score was significantly decreased in rats at 21 days following transplantation (P < 0.05). No significant difference was detected in neurological severity score between hNSC and hUCBSC transplantation groups at various time points (P > 0.05). 6A drug-loaded composite coating to improve osteogenic and antibacterial properties of Zn-1Mg porous scaffolds as biodegradable bone implants显示文摘Zinc(Zn)alloy porous scaffolds produced by additive manufacturing own customizable structures and biodegradable functions,having a great application potential for repairing bone defect.In this work,a hydroxyapatite(HA)/polydopamine(PDA)composite coating was constructed on the surface of Zn-1Mg porous scaffolds fabricated by laser powder bed fusion,and was loaded with a bioactive factor BMP2 and an antibacterial drug vancomycin.The microstructure,degradation behavior,biocompatibility,antibacterial performance and osteogenic activities were systematically investigated.Compared with as-built Zn-1Mg scaffolds,the rapid increase of Zn2+,which resulted to the deteriorated cell viability and osteogenic differentiation,was inhibited due to the physical barrier of the composite coating.In vitro cellular and bacterial assay indicated that the loaded BMP2 and vancomycin considerably enhanced the cytocompatibility and antibacterial performance.Significantly improved osteogenic and antibacterial functions were also observed according to in vivo implantation in the lateral femoral condyle of rats.The design,influence and mechanism of the composite coating were discussed accordingly.It was concluded that the additively manufactured Zn-1Mg porous scaffolds together with the composite coating could modulate biodegradable performance and contribute to effective promotion of bone recovery and antibacterial function.Zhenbao Zhang Aobo Liu Jiadong Fan Menglin Wang Jiabao Dai Xiang Jin Huanze Deng Xuan Wang Yijie Liang Haixia Li Yantao Zhao Peng Wen Yanfeng Li 2023Bioactive Materials2023,,9:2

8Overexpression of amplified in breast cancer 1 (AIB1) gene promotes lung adenocarcinoma aggressiveness in vitro and in vivo by upregulating C-X-C motif chemokine receptor 4显示文摘Background:We previously found that overexpression of the gene known as amplified in breast cancer 1(AIB1)was associated with lymph node metastasis and poor prognosis in patients with lung adenocarcinoma.However,the role of AIB1 in that malignancy remains unknown.The present study aimed to investigate the function of AIB1 in the process of lung adenocarcinoma cell metastasis.Methods:A series of in vivo and in vitro assays were performed to elucidate the function of AIB1,while real-time PCR and Western blotting were utilized to identify the potential downstream targets of AIB1 in the process of lung adenocarcinoma metastasis.Rescue experiments and in vitro assays were performed to investigate whether the invasive-ness of AIB1-induced lung adenocarcinoma was mediated by C-X-C motif chemokine receptor 4(CXCR4).Results:The ectopic overexpression of AIB1 in lung adenocarcinoma cells substantially enhanced cell migration and invasive abilities in vitro and tumor metastasis in vivo,whereas the depletion of AIB1 expression substantially inhibited lung adenocarcinoma cell migration and invasion.CXCR4 was identified as a potential downstream target of AIB1 in lung adenocarcinoma.The knockdown of AIB1 greatly reduced CXCR4 gene expression at both the transcription and protein levels,whereas the knockdown of CXCR4 in cells with AIB1 ectopic overexpression diminished AIB1-induced migration and invasion in vitro and tumor metastasis in vivo.Furthermore,we found a significant positive association between the expression of AIB1 and CXCR4 in lung adenocarcinoma patients(183 cases),and the co-overexpression of AIB1 and CXCR4 predicted the poorest prognosis.Conclusions:These findings suggest that AIB1 promotes the aggressiveness of lung adenocarcinoma in vitro and in vivo by upregulating CXCR4 and that it might be usable as a novel prognostic marker and/or therapeutic target for this disease.Liru He Haixia Deng Shiliang Liu Jiewei Chen Binkui Li Chenyuan Wang Xin Wang Yiguo Jiang Ningfang Ma Mengzhong Liu Dan Xie 2018Cancer Communications2018,38,1:2

11Variant rs8400 enhances ALKBH5 expression through disrupting miR-186 binding and promotes neuroblastoma progression显示文摘Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3p caused by this genetic variation in ALKBH5 promotes the occurrence and development of neuroblastoma through the ALKBH5-SPP1 axis.Qian Guan Huiran Lin Wenfeng Hua Lei Lin Jiabin Liu Linqing Deng Jiao Zhang Jiwen Cheng Zhonghua Yang Yong Li Jun Bian Haixia Zhou Suhong Li Li Li Lei Miao Huimin Xia Jing He Zhenjian Zhuo 2023Chinese Journal of Cancer Research2023,35,2:0

14Dietary phytochemicals: As a potential natural source for treatment of Alzheimer's Disease显示文摘Alzheimer's disease(AD)is a common neurodegenerative disease,which seriously impairs human health and life.At present,scientists have proposed more than a dozen hypotheses about the pathogenesis of AD,including the tau propagation hypothesis.However,the exact ultimate pathogenic factor of AD remains unknown.Based on the current hypotheses,some anti-AD drugs(e.g.,donepezil and Ketamine)have been developed and used in clinical treatment,which fall into two main categories,acetylcholinesterase inhibitors(AChEIs)and N-methyl-D-aspartate(NMDA)receptor antagonists,the former representative drug is donepezil,and the latter representative drug is memantine.Since these drugs have undesirable side effects,it is necessary to find safer alternatives for AD treatment.Interestingly,dietary phytochemicals have the advantages of wide source,safety,and high biological activity,which is the natural route for screening anti-AD drugs.In this study,several representatives’dietary phytochemicals with anti-AD effect,including resveratrol,lycopene,gallic acid,berberine,ginsenoside Rg1,pseudoginsenoside-F11,ginsenoside Rh2,artemisinin,and torularhodin were selected from the published data over the last 10 years and their potential molecular mechanisms and clinical applications reviewed in the treatment of AD.Zhuo Ren Haixia Yang Chenhui Zhu Daidi Fan Jianjun Deng 2023Food Innovation and Advances2023,2,1:0
15Rumen microbiota-host transcriptome interaction mediates the protective effects of trans-10,cis-12 CLA on facilitating weaning transition of lambs显示文摘Developing alternatives to antibiotics for prevention of gastrointestinal dysbiosis in early-weaning farmed animals is urgently needed.This study was to explore the potential effects of trans-10,cis-12 conjugated linoleic acid(CLA)on maintaining ruminal homeostasis of young ruminants during the weaning transition period.Thirty neonatal lambs were selected(6 lambs per group)and euthanized for rumen microbial and epithelial analysis.The lambs were weaned at 28 d and experienced the following 5 treatments:euthanized on d 28 as the pre-weaning control(CON0),fed starter feed for 5(CON5)or 21(CON21)d,fed starter feed with 1%of CLA supplemented for 5(CLA5)or 21(CLA21)d.Results showed that the average daily weight gain and dry matter intake were significantly higher in CLA5 than CON5 group.As compared with the CON5 and CON21 group,the relative abundances of volatile fatty acid(VFA)producing bacteria including Bacteroides,Treponema,Parabacteroides and Anaerovibrio,as well as the concentrations of acetate,butyrate and total VFA were significantly increased in CLA5 and CLA21 group,respectively.Integrating microbial profiling and epithelial transcriptome results showed that 7 downregulated inflammatory signaling-related host genes IL2RA,CXCL9,CD4,CCR4,LTB,SPP1,and BCL2A1 with CLA supplementation were significantly negatively correlated with both VFA concentration and VFA producing bacteria,while 3(GPX2,SLC27A2 and ALDH3A1)and 2(GSTM3 and GSTA1)upregulated metabolism-related genes,significantly positively correlated with either VFA concentration or VFA producing bacteria,respectively.To confirm the effects of CLA on epithelial signal transduction,in vitro experiment was further conducted by treating rumen epithelial cells without or with IL-17A+TNF-αfor 12 h after pretreatment of 100μM CLA or not(6 replicates per treatment).The results demonstrated the anti-inflammatory effect of CLA via suppressing the protein expression of NF-кB p-p65/p65 with the activation of peroxisome proliferator-activated receptor gamma(PPARγ).In conclusion,CLA supplementation enhanced the ruminal microbiota-driven transcriptional regulation in healthy rumen epithelial development via rumen VFA production,and CLA may therefore serve as an alternative way to alleviate early-weaning stress and improve physiological and metabolic conditions of young ruminants.Chunlei Yang Xiangfei Deng Peter Lund Haixia Liu Xingwang Ding Zhengwei Fu Naifeng Zhang Jinjun Li Lifeng Dong 2023Animal Nutrition2023,,1:0
