| 2 | Isolation of Lymphocytes and Their Innate Immune Characterizations from Liver,Intestine,Lung and Uterus显示文摘In steady-state conditions, the number and distribution of lymphocyte populations are under homeostatic control.New lymphocytes are continuously produced in primary and secondary lymphoid organs and then achieve immune-competence within different tissues, and they must challenge with resident cells for survival. The first step in the study of tissue lymphoid cells is their isolation in intact and viable form appropriate for establishment of in vitro culture systems. For reasons of simplicity, cell purity, cell yields and various purposes, lymphocytes obtained from different tissues in different labs were subjected to diverse protocols. | 6 | Multiple-Sized Amphiphilic Janus Gold Nanoparticles by Ligand Exchange at Toluene/Water Interface显示文摘Amphiphilic Janus gold nanoparticles(GNPs) are prospected to encapsulate drug molecules in cancer therapy and to serve as heterogeneous catalysts at oil/water interfaces, where Janus GNPs with different sizes are required. In this work, multiple-sized precursor GNPs were synthesized by seeded growth method protected with tris(hydroxymethyl)phosphine oxide(THPO) ligand molecule, and a ligand exchange reaction with triphenylphosphine(PPh3) at the toluene/water interface was employed to prepare amphiphilic Janus GNPs. UV-vis and transmission electron microscopy(TEM) analyses indicate that the as-prepared GNPs are nanocrystals with average diameters of 2.3 nm, 9.5 nm, 16.1 nm and 18.8 nm, respectively. | 7 | Ecological Benefits of Compound Ridge Culture in Hillside Orchard in the Three Gorge Reservoir Area显示文摘Based on contour tillage of wide ridge and furrow covering and agro-forestry, this paper sets up a compound ridge culture mode in hillside orchards and introduces a long-term observation to the mode's effects on ecological benefits of water and soil conservation. A five-year study shows that compound ridge culture in hillside orchards is effective in water and soil conservation, especially in reducing soil erosion. Compared with the traditional management modes of orchards, compound ridge culture has reduced runoff amount by 41.96%-57.96%, soil erosion amount by 55.47%-67.75%. | 9 | Chlorine-passivated superatom Al37clusters for nonlinear optics显示文摘Utilizing a facile top-down synthetic procedure, here we report the finding of a chlorine-passivated Al_(37) superatom cluster. It is demonstrated that the presence of electrophilic groups, severing as protecting ligands, alters the valence electron count of the metallic core and stabilize the as-prepared aluminum clusters especially when even-numbered chlorine atoms are located at equilibrium positions. Following the discussion regarding their reasonable stabilities, we illustrate the feasible reaction pathways in forming such chlorine-passivated Al_(37) superatom clusters which bear delocalized superatomic orbitals with five valence 3P^5 electrons shifting to the chlorine ligands indicative of a closed electron shell 2F^(14) of the metal core. The successful synthesis of such chlorine-protected aluminum clusters evidences the compatibility of general theory of cluster chemistry in both gas phase and wet chemistry. | 13 | An Upper Crustal Ophiolite Remnant within the Feather River Ultramafic Belt, California:Tectonomagmatic Origins and Implications for Its Evolution显示文摘The 150-km-long Feather River ultramafic belt(FRB) of the northern Sierra Nevada,California,consists of serpentinized ultramafic rocks,gabbroic rocks,with lesser amounts of amphibolitic rocks. The gabbroic rocks contain metagabbro and dikes within it. Based on the electron microprobe analyses,the metagabbro consists of typical igneous composition for clinopyroxene and early hornblende,indicating that they bear low grade metamorphism. The gabbroic rocks show Nb and Ta depletion in primitive-mantle normalized plots that is similar to typical island arc rocks. We present that the metagabbro within the ultramafic rocks is forming in the suprasubduction zone setting. Tectonic models are presented for the metagabbro and amphibolitic rocks in FRB. The low P burial metamorphism of the upper plate metagabbro and the high grade amphibolitic rocks are typical of relationships of upper plate ophiolites to metamorphic soles. The amphibolitic rocks record as metamorphic sole forming during subduction initiation beneath the ultramafic rocks and metagabbro. | 19 | Targeting autophagy overcomes cancer-intrinsic resistance to CAR-T immunotherapy in B-cell malignancies显示文摘Background:Chimeric antigen receptor T(CAR-T)therapy has substantially revolutionized the clinical outcomes of patients with hematologic malignan-cies,but the cancer-intrinsic mechanisms underlying resistance to CAR-T cells remain yet to be fully understood.This study aims to explore the molecular deter-minants of cancer cell sensitivity to CAR-T cell-mediated killing and to provide a better understanding of the underlying mechanisms and potential modulation to improve clinical efficacy.Methods:The human whole-genome CRISPR/Cas9-based knockout screening was conducted to identify key genes that enable cancer cells to evade CD19 CAR-T-cell-mediated killing.The in vitro cytotoxicity assays and evaluation of tumor tissue and bone marrow specimens were further conducted to confirm the role of the key genes in cancer cell susceptibility to CAR-T cells.In addition,the specific mechanisms influencing CAR-T cell-mediated cancer clearance were elucidated in mouse and cellular models.Results:The CRISPR/Cas9-based knockout screening showed that the enrich-ment of autophagy-related genes(ATG3,BECN1,and RB1CC1)provided protec-tion of cancer cells from CD19 CAR-T cell-mediated cytotoxicity.These findings were further validated by in vitro cytotoxicity assays in cells with genetic and pharmacological inhibition of autophagy.Notably,higher expression of the three autophagy-related proteins in tumor samples was correlated with poorer respon-siveness and worse survival in patients with relapsed/refractory B-cell lymphoma after CD19 CAR-T therapy.Bulk RNA sequencing analysis of bone marrow samples from B-cell leukemia patients also suggested the clinical relevance of autophagy to the therapeutic response and relapse after CD19 CAR-T cell ther-apy.Pharmacological inhibition of autophagy and knockout of RB1CC1 could dramatically sensitize tumor cells to CD19 CAR-T cell-mediated killing in mouse models of both B-cell leukemia and lymphoma.Moreover,our study revealed that cancer-intrinsic autophagy mediates evasion of CAR-T cells via the TNF-α-TNFR1 axis-mediated apoptosis and STAT1/IRF1-induced chemokine signaling activation.Conclusions:These findings confirm that autophagy signaling in B-cell malig-nancies is essential for the effective cytotoxic function of CAR-T cells and thereby pave the way for the development of autophagy-targeting strategies to improve the clinical efficacy of CAR-T cell immunotherapy. | Lu Tang Huan Zhang Fen Zhou Qiuzhe Wei Mengyi Du Jianghua Wu Chenggong Li Wenjing Luo Jie Zhou Xindi Wang Zhaozhao Chen Yinqiang Zhang Zhongpei Huang Zhuolin Wu Yuxi Wen Huiwen Jiang Danying Liao Haiming Kou Wei Xiong Heng Mei Yu Hu | 2024 | Cancer Communications2024,44,3: | 0 |
