1The Roles of Innate Immune Cells in Liver Injury and Regeneration显示文摘For predominant abundance with liver-specific Kupffer cells, natural killer (NK) cells, and natural killer T (NKT) cells and their rapid responses to several stimuli, the liver is considered as an organ with innate immune features. In contrast to their roles in the defense of many infectious agents like hepatitis viruses and parasites, hepatic innate immune cells are also involved in the immunopathogenesis of human clinical liver diseases and several murine hepatitis models such as concanavalin A (Con A), lipopolysaccharide (LPS), or polyinosinic-polycytidylic acid (Poly I:C)-induced liver injury. In this review, the destructive roles of NK cells, NKT cells and Kupffer cells in the processes of immune-mediated liver injury and regeneration will be discussed, and some putative mechanisms involving the impairment of liver regeneration caused by activated hepatic innate immune cells are also proposed.Zhongjun Dong 2007Cellular & Molecular Immunology2007,4,4:24
2The Involvement of Heat Shock Proteins in Murine Liver Regeneration显示文摘Partial hepatectomy (PHx) in mammals is a very common experimental model to investigate the process of liver regeneration. The surgery itself could give birth to a series of stresses, such as the temporary raise of body temperature and the ischaemia-reperfusion injury. Heat shock proteins (HSPs) were a family of stress-inducible proteins involved in maintaining cell homeostasis and regulating the immune system. In our study, we intended to investigate the expression and role of HSPs in liver regeneration. Using RT-PCR and Western blotting, we determined the expression in regenerating liver of HSP27, HSP60, HSP70 and HSP90 in mRNA level and protein level, respectively, with mice treated with sham operation as controls. We also used quercertin as an inhibitior of HSPs to explore their effects on liver regeneration. We found that hepatic expression of HSPs increased at the early phase of liver regeneration and declined to the constitutively low level later. Moreover, quercetin pretreatment delayed the progress of liver regeneration in mice via inhibition of HSPs. 3BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD.Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du 2018Cellular & Molecular Immunology2018,15,12:9
4Isolation of Lymphocytes and Their Innate Immune Characterizations from Liver,Intestine,Lung and Uterus显示文摘In steady-state conditions, the number and distribution of lymphocyte populations are under homeostatic control.New lymphocytes are continuously produced in primary and secondary lymphoid organs and then achieve immune-competence within different tissues, and they must challenge with resident cells for survival. The first step in the study of tissue lymphoid cells is their isolation in intact and viable form appropriate for establishment of in vitro culture systems. For reasons of simplicity, cell purity, cell yields and various purposes, lymphocytes obtained from different tissues in different labs were subjected to diverse protocols. 5Combined deficiency of SLAMF8 and SLAMF9 prevents endotoxin-induced liver inflammation by downregulating TLR4 expression on macrophages显示文摘Classical signaling lymphocyte activating molecule(SLAM)family receptors are abundant within many types of immune cells,whereas the nonclassical SLAM family receptors SLAMF8 and SLAMF9,which uniquely lack cytoplasmic signaling motifs,are highly expressed by myeloid cells.Due to the potential redundancy,whether these two receptors regulate macrophage function remains largely unknown.Here,we show that SLAMF8 and SLAMF9 co-regulate macrophage-mediated liver inflammation.To overcome the redundancy,we generated mice that simultaneously lacked SLAMF8 and SLAMF9 using CRISPR-Cas9 technology.Although macrophage differentiation was not altered by the combined deficiency of SLAMF8 and SLAMF9,the loss of these two receptors significantly protected against lipopolysaccharide(LPS)-induced liver injury.SLAMF8 and SLAMF9 double-deficient mice had a prolonged survival rate and less infiltration of inflammatory cells.The depletion of macrophages using clodronate liposomes abolished the effects of SLAMF8 and SLAMF9 deficiencies on LPS-induced liver injury,which demonstrates that these receptors are required for macrophage activation following LPS challenge.Moreover,the deficiency of SLAMF8 and SLAMF9 suppressed the secretion of inflammatory cytokines by downregulating the expression of Toll-like receptor-4(TLR4),a receptor that specifically binds LPS,which led to decreased mitogen-activated protein kinases(MAPK)signaling activation.Notably,combined injections of truncated extracellular SLAMF8 and SLAMF9 proteins significantly alleviated LPS-induced liver injury.Thus,our findings provide insights into the role of SLAMF8 and SLAMF9 in endotoxin-induced liver injury and suggest that SLAMF8 and SLAMF9 are potential therapeutic targets for acute hepatic injury.Xiaokang Zeng Guangao Liu Wanwen Peng Junming He Chenxu Cai Wei Xiong Shasha Chen Meixiang Yang Zhongjun Dong 2020Cellular & Molecular Immunology2020,17,2:5
6Osteoporosis affects both post-yield microdamage accumulation and plasticity degradation in vertebra of ovariectomized rats显示文摘Estrogen withdrawal in postmenopausal women increases bone loss and bone fragility in the vertebra. Bone loss with osteoporosis not only reduces bone mineral density(BMD), but actually alters bone quality, which can be comprehensively represented by bone post-yield behaviors. This study aimed to provide some information as to how osteoporosis induced by estrogen depletion could influence the evolution of post-yield microdamage accumulation and plastic deformation in vertebral bodies. This study also tried to reveal the part of the mechanisms of how estrogen deficiency-induced osteoporosis would increase the bone fracture risk. A rat bilateral ovariectomy(OVX)model was used to induce osteoporosis. Progressive cyclic compression loading was developed for vertebra testing to elucidate the post-yield behaviors. BMD, bone volume fraction, stiffness degradation, and plastic deformation evolution were compared among rats raised for 5 weeks(ovx5w and sham5 w groups) and 35 weeks(ovx35w and sham35 w groups) after sham surgery and OVX. The results showedthat a higher bone loss in vertebral bodies corresponded to lower stiffness and higher plastic deformation. 7Cryo-electron microscopy reconstructions of two types of wild rabbit hemorrhagic disease viruses characterized the structural features of Lagovirus显示文摘Rabbit hemorrhagic disease was described in China in 1984 and can cause hemorrhagic necrosis of the liver within two or three days after infection.The etiological agent,rabbit hemorrhagic disease virus(RHDV),belongs to the Lagovirus genus in the Caliciviridae family.Compared to other calicivirus,such as rNV and SMSV,the structure of Lagovirus members is not well characterized.In this report,structures of two types of wild RHDV particles,the intact virion and the core-like particle(CLP),were reconstructed by cryo-electron microscopy at 11Åand 17Å,respectively.This is the first time the 3D structure of wild caliciviruses CLP has been provided,and the 3D structure of intact RHDV virion is the highest resolution structure in Lagovirus.Comparison of the intact virion and CLP structures clearly indicated that CLP was produced from the intact virion with the protrusion dissociated.In contrast with the crystal structures of recombinant Norovirus and San Miguel sea lion virus,the capsomers of RHDV virion exhibited unique structural features and assembly modes.Both P1 and P2 subdomains have interactions inside the AB capsomer,while only P2 subdomains have interaction inside CC capsomer.The pseudo atomic models of RHDV capsomers were constructed by homology modeling and density map fitting,and the rotation of RHDV VP60 P domain with respect to its S domain,compared with SMSV,was observed.Collectively,our cryo-electron microscopic studies of RHDV provide close insight into the structure of Lagovirus,which is important for functional analysis and better vaccine development in the future.Zhongjun Hu Xiaojuan Tian Yujia Zhai Wei Xu Dong Zheng Fei Sun 2010Protein & Cell2010,1,1:3

10Long-wavelength(red to near-infrared) emissive carbon dots: Key factors for synthesis, fluorescence mechanism, and applications in biosensing and cancer theranostics显示文摘Carbon dots(CDs), as a new member of carbon nanostructures, have been widely applied in extensive fields due to their exceptional physicochemical properties. While, the emissions of most reported CDs are located in the blue to green range under the excitation of ultraviolet or blue light, which severely limits their practical applications, especially in photovoltaic and biological fields. Studies that focused on synthesizing CDs with long-wavelength(red to near-infrared) emission/excitation features(simply named L-w CDs) and exploring their potential applications have been frequently reported in recent years. In this review, we analyzed the key influence factors for the synthesis of CDs with long wavelength and multicolor(containing long wavelength) emissive properties, discussed possible fluorescence mechanism,and summarized their applications in sensing and cancer theranostics. 12Experimental study on mechanical properties of granite under different cooling methods after thermal treatment显示文摘Static uniaxial compression tests were conducted on 16 granite specimens after thermal treatment using a heating device and an electro-hydraulic servo pressure-testing machine. The effects of air cooling and water cooling on the physical and mechanical properties of the high-temperature granite specimens were studied. Test results showed that the longitudinal wave velocities of the high-temperature specimens gradually decreased after they were cooled by water and air. The peak stress and elastic modulus of the samples decreased gradually with an increase in temperature, whereas their peak strain increased gradually. The effects of peak stress and peak strain were considerably more evident when cooling by water than by air. 13Eupalinolide B inhibits hepatic carcinoma by inducing ferroptosis and ROS-ER-JNK pathway显示文摘Primary hepatic carcinoma is a common malignant tumor.The classic molecular targeted drug sorafenib is costly and is only effective for some patients.Therefore,it is of great clinical significance to search for new molecular targeted drugs.Eupalinolide B(EB)from Eupatorium lindleyanum DC.is used to treat chronic tracheitis in clinical practice.However,the role of EB in hepatic carcinoma is unknown.In this study,we first measure the effect of EB on tumor growth in a xenograft model and PDX model.The cell proliferation and migration are also detected in human hepatocarcinoma cell lines(SMMC-7721 and HCCLM3).Then,we investigate cell cycle,cell apoptosis,cell necrosis,cell autophagy,and ferroptosis by flow cytometry,western blot analysis and electron microscopy.The results demonstrate that EB exerts anti-proliferative activity in hepatic carcinoma by blocking cell cycle arrest at S phase and inducing ferroptosis mediated by endoplasmic reticulum(ER)stress,as well as HO-1 activation.When HO-1 is inhibited,EB-induced cell death and ER protein expression are rescued.The migration-related mechanism consists of activation of the ROS-ER-JNK signaling pathway and is not connected to ferroptosis.In summary,we first discover that EB inhibits cell proliferation and migration in hepatic carcinoma,and thus EB is a promising anti-tumor compound that can be used for hepatic carcinoma.Yonghui Zhang Haoyang Zhang Jinage Mu Meiyue Han Zhihao Cao Feng Dong Tingting Wang Lian Pan Wujing Luo Jiaxin Li Huan Liu Lishan Jin Wenxuan Ding Yong Wei Xuesong Deng Dan Liu Xiuzhen He Yi Pang Xiao Mu Zhongjun Wu Dilong Chen 2022Acta Biochimica et Biophysica Sinica2022,54,7:1

15Chronic Nitrogen Fertilization Modulates Competitive Interactions Among Microbial Ammonia Oxidizers in a Loess Soil显示文摘Nitrogen(N) application may lead to niche segregation of soil ammonia-oxidizing archaea(AOA) and bacteria(AOB), thereby reducing the competitive interactions between AOA and AOB due to higher ammonium substrate availability. However, the adaptive mechanisms of AOA and AOB under N enrichment remain poorly understood. Stable isotope probing(SIP) microcosm incubation was employed to reveal community changes of active AOA and AOB in a loess soil from a field experiment growing potatoes that received no N(control, CK), low N(LN, 75 kg N ha^(-1)), and high N(HN, 375 kg N ha^(-1)). The results showed that the soil potential nitrification rate(PNR) was measured by culturing of the soil samples from the field experiment. Soil PNR was significantly increased in HN by87.5% and 67.5% compared with CK and LN, respectively. Compared with CK, the^(13)C-amoA genes of soil AOA and AOB in HN had 2.58 × 10~4 and 1.55 × 10~6 copies, representing 1.6-and 16.2-fold increase respectively. It was indicated that AOB dominated soil ammonia oxidation. A phylogenetic analysis of the^(13)C-amoA gene showed that N application significantly increased the proportion of54 d9-like AOA up to 90% in HN, while the Nitrososphaera gargensis-like and Nitrososphaera viennensis-like AOA were inhibited and completely disappeared. Nitrogen application also resulted in the community shift of active AOB-dominant group from Nitrosospira briensis-like to Nitrosospira sp. TCH711-like. 16Determination of carbon-fixing potential of Bathyarchaeota in marine sediment by DNA stable isotope probing analysis显示文摘Bathyarchaeota is believed to play a crucial role in the global carbon cycle due to its vast biomass,broad distribution,and diverse habitat.However,its physiological and metabolic features are hard to determine without pure culture.While metagenomic analyses have shown that Bathyarchaeota has a complete inorganic carbon fixation(Wood-Ljungdahl,WL)pathway,no direct functional confirmation has been reported.To explore the inorganic carbon fixation ability of Bathyarchaeota,we used lignin and sodium bicarbonate-^(13)C(NaH^(13)CO_(3))in the long-term incubation of marine sediment samples.We found that Bathyarchaeota grew continuously in the cultivation system with lignin,and its abundance increased up to 15.3 times after10 months,increasing its fraction of all archaea from 30%to 80%.We monitored theδ^(13)C of total organic carbon to identify microbial carbon fixation in the cultivation systems,finding that it increased in the first month while NaH^(13)CO_(3)was present but only increased continuously afterward when lignin was also present.Furthermore,ultracentrifugation was performed on DNA extracted from samples at different cultivation stages to separate DNA of different buoyant densities,and bathyarchaeotal and bacterial 16S ribosomal RNA(r RNA)gene abundance were quantified using qPCR.Compared to bacteria,bathyarchaeotal 16S rRNA tended to be concentrated in heavy layers after 4 months of incubation with lignin and NaH^(13)CO_(3),indicating that Bathyarchaeota DNA contained^(13)C through proliferation based on lignin utilization and NaH^(13)CO_(3)assimilation,proving the carbon fixation capacity of Bathyarchaeota.Wenyue LIANG Tiantian YU Liang DONG Zhongjun JIA Fengping WANG 2023Science China Earth Sciences2023,66,4:0
17Computational fluid dynamics analysis and experimental hemolytic performance of three clinical centrifugal blood pumps: Revolution, Rotaflow and CentriMag显示文摘Centrifugal blood pumps have become popular for adult extracorporeal membrane oxygenation(ECMO)due to their superior blood handling and reduced thrombosis risk featured by their secondary flow paths that avoid stagnant areas.However,the high rotational speed within a centrifugal blood pump can introduce high shear stress,causing a significant shear-induced hemolysis rate.The Revolution pump,the Rotaflow pump,and the CentriMag pump are three of the leading centrifugal blood pumps on the market.Although many experimental and computational studies have focused on evaluating the hydraulic and hemolytic performances of the Rotaflow and CentriMag pumps,there are few on the Revolution pump.Furthermore,a thorough direct comparison of these three pumps'flow characteristics and hemolysis is not available.In this study,we conducted a computational and experimental analysis to compare the hemolytic performances of the Revolution,Rotaflow,and CentriMag pumps operating under a clinically relevant condition,i.e.,the blood flow rate of 5 L/min and pump pressure head of 350 mmHg,for adult ECMO support.In silico simulations were used to characterize the shear stress distributions and predict the hemolysis index,while in vitro blood loop studies experimentally determined hemolysis performance.Comparative simulation results and experimental data demonstrated that the CentriMag pump caused the lowest hemolysis while the Revolution pump generated the highest hemolysis.Dong Han Joshua L.Leibowitz Lu Han Shigang Wang Ge He Bartley P.Griffith Zhongjun J.Wu 2022Medicine in Novel Technology and Devices2022,,3:0
18Effects of two strategies on afterglow behavior of Lu_(2)O_(3):Eu single crystal scintillator:Co-doping with Pr^(3+)and solid solution with Sc_(2)O_(3)显示文摘In this paper,effect of two strategies on afterglow behavior of Lu_(2)O_(3):Eu single crystal scintillato r,Pr^(3+)codoping and solid solution with Sc_(2)O_(3),were studied systematically.Two groups of Lu_(2)O_(3):5 at%Eu,x at%Pr(x=0,0.2,0.5,1,2 and 5)and(Lu1-yScy)_(2)O_(3):5 at%Eu(y=0,20 at%,50 at%and 70 at%)single crystals were grown by floating zone(FZ)method in air atmosphere.The structures of as-grown crystals were determined by X-ray diffraction(XRD).The scintillation,photoluminescence properties and carrier trap states were investigated through afterglow,X-ray excitation luminescence(XEL),transmittance,photoluminescence excitation(PLE)and photoluminescence(PL),PL decay and thermal stimulated luminescence(TSL)curves.It is found that with the increase of Pr^(3+)concentration,the afterglow level of the system decreases at the expense of scintillation luminescence efficiency.Meanwhile,although Sc_(2)O_(3):Eu presents much lower afterglow intensity than Lu_(2)O_(3):Eu,the addition of Sc_(2)O_(3)will just increase the afterglow level of the(Lu1-yScy)_(2)O_(3):5 at%Eu single crystal system.Possible mechanisms for above phenomena are discussed based on experimental results.Zhongjun Xue Dongzhou Ding Yating Sima Zuyao Zhou Hanrui Dong Shuwen Zhao He Feng 2023Journal of Rare Earths2023,41,5:0

20A Histological Study on Subacute Toxicity of 'Baikening' in Hy-Line Brown Roosters显示文摘[Objective]A histological study was conducted to investigate the toxicity of'Baikening'and the toxic effect of'Baikening'on roosters after longerterm administration at different dose levels.[Method]Thirty 21-day-old Hy-Line Brown roosters were administered with 0.5 and 1.0 ml of'Baikening'aqueous extract through drinking water once a day for continuous 14 d.Roosters in control group and experimental groups after drug administration for 14 d and drug withdrawal for 7 d were euthanized;livers,kidneys,thymuses,spleens and bursas of Fabricius were collected and weighed for comparative analysis.Various tissues were prepared into paraffin-embedded slices and stained by H.E.,to observe histological changes of various organs under an optical microscope.[Result]After drug administration for 14 d,all the roosters in experimental groups survived;there was no visible pathological change in various organs of Hy-Line Brown roosters in each group;organ weights of Hy-Line Brown roosters in various groups showed no significant difference;observation results of tissue slices indicated that there was no pathological change in fine structures,nuclei and cytoplasm of various organs of Hy-Line Brown roosters in different groups;various blood vessels in different organs also presented no change in blood circulation or other pathological changes.[Conclusion]'Baikening'aqueous extract(oral administration)has no toxic and side effect on roosters in the subacute toxicity experiment.Qiumei SHI Ping SHEN Shuzhen DONG Zhongjun HU 2013Agricultural Biotechnology2013,2,6:0
