3Matrine attenuates oxidative stress and cardiomyocyte apoptosis in doxorubicin-induced cardiotoxicity via maintaining AMPKα/UCP2 pathway显示文摘Oxidative stress and cardiomyocyte apoptosis are involved in the pathogenesis of doxorubicin(DOX)-induced cardiotoxicity. Matrine is well-known for its powerful anti-oxidant and anti-apoptotic capacities. Our present study aimed to investigate the effect of matrine on DOX-induced cardiotoxicity and try to unearth the underlying mechanisms. Mice were exposed with DOX to generate DOX-induced cardiotoxicity or normal saline as control. H9 C2 cells were used to verify the effect of matrine in vitro.DOX injection triggered increased generation of reactive oxygen species(ROS) and excessive cardiomyocyte apoptosis, which were significantly mitigated by matrine. Mechanistically, we found that matrine ameliorated DOX-induced uncoupling protein 2(UCP2) downregulation, and UCP2 inhibition bygenipin could blunt the protective effect of matrine on DOX-induced oxidative stress and cardiomyocyte apoptosis. Besides, 50-AMP-activated protein kinase α2(Ampkα2) deficiency inhibited matrine-mediated UCP2 preservation and abolished the beneficial effect of matrine in mice. Besides, we observed that matrine incubation alleviated DOX-induced H9 C2 cells apoptosis and oxidative stress level via activating AMPKα/UCP2, which were blunted by either AMPKα or UCP2 inhibition with genetic or pharmacological methods. 4Radio Propagation and Wireless Coverage of LSAA-Based 5G Millimeter-Wave Mobile Communication Systems显示文摘Millimeter-wave(mm Wave) communications will be used in fifth-generation(5G) mobile communication systems, but they experience severe path loss and have high sensitivity to physical objects, leading to smaller cell radii and complicated network architectures. A coverage extension scheme using large-scale antenna arrays(LSAAs) has been suggested and theoretically proven to be cost-efficient in combination with ultradense small cell networks. To analyze and optimize the LSAA-based network deployments, a comprehensive survey of recent advances in statistical mmWave channel modeling is first presented in terms of channel parameter estimation, large-scale path loss models, and small-scale cluster models. Next, the measurement and modeling results at two 5G candidate mmWave bands(e.g., 28 GHz and 39 GHz) are reviewed and compared in several outdoor scenarios of interest, where the propagation characteristics make crucial contributions to wireless network designs. 5Erucic acid from Isatis indigotica Fort. suppresses influenza A virus replication and inflammation in vitro and in vivo through modulation of NF-kB and p38 MAPK pathway显示文摘Isatis indigotica Fort.(Ban-Lan-Gen)is an herbal medicine prescribed for influenza treatment.However,its active components and mode of action remain mostly unknown.In the present study,erucic acid was isolated from Isatis indigotica Fort.,and subsequently its underlying mechanism against influenza A virus(IAV)infection was investigated in vitro and in vivo.Our results demonstrated that erucic acid exhibited broad-spectrum antiviral activity against IAV resulting from reduction of viral polymerase transcription activity.Erucic acid was found to exert inhibitory effects on IAV or viral(v)RNA-induced pro-inflam-matory mediators as well as interferons(IFNs).The molecular mechanism by which erucic acid with antiviral and anti-inflammatory properties was attributed to inactivation of NF-kB and p38 MAPK signaling.Furthermore,the NF-kB and p38 MAPK inhibitory effect of erucic acid led to diminishing the transcriptional activity of interferon-stimulated gene factor 3(ISGF-3),and thereby reducing IAV-triggered pro-inflammatory response amplification in IFN-β-sensitized cells.Additionally,IAV-or vRNA-triggered apoptosis of alveolar epithelial A549 cells was prevented by erucic acid.In vivo,erucic acid administration consistently displayed decreased lung viral load and viral antigens expression.Meanwhile,erucic acid markedly reduced CD8+cytotoxic T lymphocyte(CTL)recruitment,pro-apoptotic signaling,hyperactivity of multiple signaling pathways,and exacerbated immune inflammation in the lung,which resulted in decreased lung injury and mortality in mice with a mouse-adapted A/FM/1/47-MA(H1N1)strain infection.Our findings provided a mechanistic basis for the action of erucic acid against IAV-mediated inflammation and injury,suggesting that erucic acid may have a therapeutic potential in the treatment of influenza.Xiaoli Liang Yuan Huang Xiping Pan Yanbing Hao Xiaowei Chen Haiming Jiang Jing Li Beixian Zhou Zifeng Yang 2020Journal of Pharmaceutical Analysis2020,10,2:10
6Dynamic contrast-enhanced MRI versus ^(18)F-FDG PET/CT: Which is better in differentiation between malignant and benign solitary pulmonary nodules?显示文摘Objective: To prospectively compare the discriminative capacity of dynamic contrast enhanced-magnetic resonance imaging(DCE-MRI) with that of ^(18)F-fluorodeoxyglucose(^(18)F-FDG) positron emission tomography/computed tomography(PET/CT) in the differentiation of malignant and benign solitary pulmonary nodules(SPNs).Methods: Forty-nine patients with SPNs were included in this prospective study. Thirty-two of the patients had malignant SPNs, while the other 17 had benign SPNs. All these patients underwent DCE-MRI and ^(18)F-FDG PET/CT examinations. The quantitative MRI pharmacokinetic parameters, including the trans-endothelial transfer constant(K^(trans)), redistribution rate constant(K_(ep)), and fractional volume(Ve), were calculated using the Extended-Tofts Linear two-compartment model. The ^(18)F-FDG PET/CT parameter, maximum standardized uptake value(SUV_(max)), was also measured. Spearman's correlations were calculated between the MRI pharmacokinetic parameters and the SUV_(max) of each SPN. These parameters were statistically compared between the malignant and benign nodules. Receiver operating characteristic(ROC) analyses were used to compare the diagnostic capability between the DCE-MRI and ^(18)F-FDG PET/CT indexes.Results: Positive correlations were found between K^(trans) and SUV_(max), and between K_(ep) and SUV_(max)(P<0.05).There were significant differences between the malignant and benign nodules in terms of the K^(trans), K_(ep) and SUV_(max) values(P<0.05). The areas under the ROC curve(AUC) of K^(trans), K_(ep) and SUV_(max) between the malignant and benign nodules were 0.909, 0.838 and 0.759, respectively. The sensitivity and specificity in differentiating malignant from benign SPNs were 90.6% and 82.4% for K^(trans); 87.5% and 76.5% for K_(ep); and 75.0% and 70.6%for SUV_(max), respectively. 7Assessment of the Ecological Security in the Three Gorges Reservoir Area by Using the Ecological Footprint Method显示文摘The ecological footprint concept and its calculation models are useful for the measurement of the sustainable level of social and economic development.The ecological security situation of the Three Gorges Reservoir Area(TGRA) was evaluated using this concept in this study.The construction of the Three Gorges Reservoir has led to the change in the ecology and immigration status of TGRA.The ecological footprint method is an important means to study the regional ecological security.Our results suggested that,by excluding the areas for biodiversity conservation(12% of the total land),the ecological footprint per capita was 0.57895 ha,which exceeded the ecological carrying capacity in TGRA.The total ecological deficit was found to be 11,522,193.34 ha,accounting for 95.02% of the ecological carrying capacity.These findings suggested that the ecological security of TGRA was not good.In order to compensate for the ecological deficit,it was essential to introduce natural resources from other regions.WANG Haiming WANG Hai'e SUN Hongyang WANG Xiaolan LIAO Xiaoyong CHEN Zhijian LI Xianwei 2012Journal of Mountain Science2012,9,6:9
8Prehistoric agriculture development in the Yunnan-Guizhou Plateau, southwest China: Archaeobotanical evidence显示文摘The origin, development and expansion of prehistoric agriculture in East Asia have been widely investigated over the past two decades using archaeobotanical analysis from excavated Neolithic and Bronze Age sites. Research on prehistoric agriculture has predominantly focused in the valleys of the Yellow River and the Yangtze River. Agricultural development during the Neolithic and Bronze Age periods in the Yunnan-Guizhou Plateau of southwest China, an important passageway for human migration into Southeast Asia, still remains unclear. In this paper, based on macrofossil and microfossil analysis and radiocarbon dating at the Shilinggang site, we investigate plant subsistence strategies in the Nujiang River valley during the Bronze Age period. Combined with previous archaeobotanical studies in the Yunnan-Guizhou Plateau, we explore agricultural development processes in this area during the Neolithic and Bronze Age. Our results indicate that rice and foxtail millet were cultivated in Shilinggang around 2500 cal a BP. Three phases of prehistoric agricultural development in the Yunnan-Guizhou Plateau can be identified: rice cultivation from 4800–3900 cal a BP, mixed rice and millet crop(foxtail millet and broomcorn millet) cultivation from 3900–3400 cal a BP, and mixed rice, millet crop and wheat cultivation from 3400–2300 cal a BP. 9Defective Kernel 39 encodes a PPR protein required for seed development in maize显示文摘RNA editing is a posttranscriptional process that is important in mitochondria and plastids of higher plants. All RNA editing-specific trans-factors reported so far belong to PLS-class of pentatricopeptide repeat(PPR)proteins. Here, we report the map-based cloning and molecular characterization of a defective kernel mutant dek39 in maize. Loss of Dek39 function leads to delayed embryogenesis and endosperm development, reduced kernel size, and seedling lethality. Dek39 encodes an E subclass PPR protein that targets to both mitochondria and chloroplasts, and is involved in RNA editing in mitochondrial NADH dehydrogenase3(nad3) at nad3-247 and nad3-275. C-to-U editing of nad3-275 is not conserved and even lost in Arabidopsis, consistent with the idea that no close DEK39 homologs are present in Arabidopsis. However, the amino acids generated by editing nad3-247 and nad3-275 are highly conserved in many other plant species, and the reductions of editing at these two sites decrease the activity of mitochondria NADH dehydrogenase complex I,indicating that the alteration of amino acid sequence is necessary for Nad3 function. 10Geochemistry of Pyrite from the Gangcha Gold Deposit, West Qinling Orogen, China: Implications for Ore Genesis显示文摘The Gangcha gold deposit was discovered in 2011 in the Xiahe-Hezuo region, West Qinling Orogen, China. Five types of pyrite have been identified in the ore according to the detailed mineral paragenetic studies. Geochemical data are presented for type I pyrite(py1) rim-core zonation and for the different types of pyrite based on in-situ laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS) and electron probe microanalysis(EPMA). The results show that pyrites are characterized with heterogeneous Au concentrations, which indicate that Au occurs mainly as micro-or nano-particle native inclusions. Time-resolved depth profiles demonstrate that As, Co and Ni occur as solid solutions in pyrite, whereas Cu, Pb, Zn and Ag occur mainly as inclusions. Trace element concentrations differ between rims and cores in py1, and the differences also exist within different pyrite types. 11Refined chronology of prehistoric cultures and its implication for re-evaluating human-environment relations in the Hexi Corridor,northwest China显示文摘The reconstruction of high-resolution chronologies for prehistoric cultures is a prerequisite for understanding the history of human evolution and its relationship with environmental change, and is valuable for exploring the trajectory of transcontinental cultural exchanges in prehistoric time. The Hexi Corridor of northwest China was one of the earliest centers for long-distance culture exchange in the prehistoric world. The timing and sequence of cultural changes in this area remains poorly understood resulting from the lack of radiometric age control. This paper presents a refined radiocarbon(14 C) chronology to resolve the timing of human occupation and cultural evolution in the Hexi Corridor. Radiocarbon dating of crop remains, which have an annual life cycle, has the advantage of eliminating problems such as the carbon stored in ‘old wood’. As a result,14C dates from crop remains are used to test the validity of the14C dates derived from charcoal, bone and plant remains. The resultant database of ages is modelled using the Bayesian approach. The updated chronology presented in this paper shows generally good agreement with the original dates used to constrain cultural evolution in the Hexi Corridor. However, the occupation of the corridor by the Shajing and Shanma Late Bronze Age-Early Iron Age cultures appears to have taken place ~300 years later than previously thought. 12Polybrominated diphenyl ether (PBDE) in blood from children (age 9–12) in Taizhou, China显示文摘Polybrominated diphenyl ethers (PBDEs) as ubiquitous persistent organic pollutants have attracted much attention in recent years. Exposure to PBDEs could induce a high health risk for children. The aim of this study was to investigate the PBDEs exposure of children (9–12 years) from Taizhou, China. Fifty-eight blood samples were collected in one school in a mountainous area in Taizhou. The concentrations of ∑9PBDEs (sum of BDE-28, -47, -99, -100, -153, -154, -183, -197 and -209) ranged from 2.66 to 33.9 ng/g lipid wet (lw) with a median of 7.22 ng/g lw. These concentrations were lower than those of children in USA, but close to European and Asian general population levels. The results showed that children in Taizhou countryside were at a low level of PBDEs exposure. The predominant congener was BDE-209, followed by BDE-28, -47, -197 and -153. 13Effects of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze-Yellow rivers, Tibetan Plateau of China显示文摘Improving our knowledge of the effects of environmental factors(e.g.soil conditions,precipitation and temperature) on belowground biomass in an alpine grassland is essential for understanding the consequences of carbon storage in this biome.The object of this study is to investigate the relative importance of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze and Yellow rivers,Tibetan Plateau.Soil organic carbon(SOC),total nitrogen(TN) and total phosphorous(TP) contents and belowground biomass were measured at 22 sampling sites across an alpine meadow on the Tibetan Plateau.We analyzed the data by using the redundancy analysis to determine the main environmental factors affecting the belowground biomass and the contribution of each factor.The results showed that SOC,TN and TP were the main factors that influenced belowground biomass,and the contribution of SOC,TN and TP on biomass was in the range of 47.87%–72.06% at soil depths of 0–30 cm.Moreover,the combined contribution of annual mean temperature(AMT) and mean annual precipitation(MAP) on belowground biomass ranged from 0.92% to 4.10%.A potential mechanism for the differences in belowground biomass was caused by the variations in soil nitrogen and phosphorous,which were coupled with SOC.A significant correlation was observed between MAP and soil nutrients(SOC,TN and TP) at the soil depth of 0–10 cm(P<0.05).We concluded that precipitation is an important driving force in regulating ecosystem functioning as reflected in variations of soil nutrients(SOC,TN and TP) and dynamics of belowground biomass in alpine grassland ecosystems.WANG Haiming SUN Jian LI Weipeng WU Jianbo CHEN Youjun LIU Wenhui 2016Journal of Arid Land2016,8,6:5

15Enhanced ozonation degradation of atrazine in the presence of nano-ZnO: Performance, kinetics and effects显示文摘Enhanced ozonation degradation of atrazine(ATZ) with nano-ZnO(nZnO) as catalyst and the influences of the operational parameters have been investigated through semi-continuous experiments in this study. The results demonstrated that the combination of ozone(O_3) and nZnO showed an obvious synergetic effect and the ATZ degradation conformed to pseudo-first-order kinetics. An improvement of ATZ degradation efficiency by 41.8% and pseudo-first-order rate constant by more than a factor of four was obtained in the O_3/nZnO process after 5 min of reaction compared to O_3 alone. Meanwhile, the degradation efficiency of ATZ was gradually enhanced with increasing nZnO dosage and initial pH in the range from 3.0 to 8.0, and a higher amount of ATZ was degraded when the initial concentration of ATZ rose from 0.5 to 5 mg/L. Additionally, sulfate ion, chloride ion, nitrate ion and low concentrations of humic acid substances led to enhancement of the ATZ degradation. The notable decrease of ATZ removal efficiency observed in the presence of radical scavengers and the results of free radical tests indicated thatUOH is the dominant active radical species. 16Spatial distribution of IL4 controls iNKT cell-DC crosstalk in tumors显示文摘The spatiotemporal distribution of cytokines orchestrates immune responses in vivo,yet the underlying mechanisms remain to be explored.We showed here that the spatial distribution of interleukin-4(IL4)in invariant natural killer T(iNKT)cells regulated crosstalk between iNKT cells and dendritic cells(DCs)and controlled iNKT cell-mediated T-helper type 1(Th1)responses.The persistent polarization of IL4 induced by strong lipid antigens,that is,α-galactosylceramide(αGC),caused IL4 accumulation at the immunological synapse(IS),which promoted the activation of the IL4R-STAT6(signal transducer and activator of transcription 6)pathway and production of IL12 in DCs,which enhanced interferon-γ(IFNγ)production in iNKT cells.Conversely,the nonpolarized secretion of IL4 induced by Th2 lipid antigens with a short or unsaturated chain was incapable of enhancing this iNKT cell-DC crosstalk and thus shifted the immune response to a Th2-type response.The nonpolarized secretion of IL4 in response to Th2 lipid antigens was caused by the degradation of Cdc42 in iNKT cells.Moreover,reduced Cdc42 expression was observed in tumorinfiltrating iNKT cells,which impaired IL4 polarization and disturbed iNKT cell-DC crosstalk in tumors.Lu Wang Zhilan Liu Lili Wang Qielan Wu Xiang Li Di Xie Huimin Zhang Yongdeng Zhang Lusheng Gu Yanhong Xue Ting Yue Gang Liu Wei Ji Haiming Wei Tao Xu Li Bai 2020Cellular & Molecular Immunology2020,17,5:4
17Application of Optical Tweezers in the Research of Molecular Interaction between Lymphocyte Function Associated Antigen-1 and Its Monoclonal Antibody显示文摘Lymphocyte function associated antigen-1 (CD11a/CD18, LFA-1) plays an important role in the structure of the immunological synapse and is required for efficient lysis of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. To study the activation mode of LFA-1 on the NK cell surface, optical tweezers were used in the work. As an emerging technology, optical tweezers are widely used to manipulate microscopic objects and measure the forces of molecular interactions in the field of biological research. In our study, a new platform was constructed to study the single molecular behavior of receptor on cell surface using optical tweezers. Based on the platform, the interaction between an NK cell and a polystyrene microsphere coated with anti-LFA-1 antibody was observed. The result confirmed that the adhesion forces between an NK cell and a polystyrene bead were time-dependent. 18Magnetoresponsive nanozyme:magnetic stimulation on the nanozyme activity of iron oxide nanoparticles显示文摘The iron oxide nanoparticles(IONPs)that combine the nanozyme activity and magnetothermal properties have attracted significant interest for various biomedical applications.However,the effect of magnetic stimulation in fine-tuning the nanozyme activities remains unclear.Here,we have constructed a series of IONPs with different magneto-thermal conversion abilities,and systematically study the effect of magnetic field stimulation on the peroxidase(POD)activity of IONPs.The results show that POD activity is effectively amplified via an in situ alternating magnetic field(AMF)stimulation with no solution temperature rise,and the degree of activity enhancement is closely related to the magnetic heating ability of the IONPs,confirming the origin of activity enhancement arises from the local magnetothermal effect.As the first report to prove magnetothermal regulation on nanozyme activity and to shed lights on the underlying correlation between activity enhancement and the intrinsic specific absorption rate(SAR),this work is expected to provide important support for future design of new magnetoresponsive nanozymes in various practical applications.Yuan He Xiaoyong Chen Ye Zhang Yanyun Wang Mengyao Cui Galong Li Xiaoli Liu Haiming Fan 2022Science China(Life Sciences)2022,65,1:3

The maize opaque endosperm and small germ 1 (osl) mutant has opaque endosperm and a small embryo,Here, we cloned OSi1 and show that it encodes a putative transcription factor containing an RWP-RK domain-Transcriptional analysis indicated that OSi1 expression is elevated in early endosperm development, especially in the basal endosperm transfer layer (BETL), conducting zone (CZ), and central starch endospcrm (CSE) cells. RNA sequencing (RNA-Seq) analysis of the osi mutant revealed sharp downregulation of certain genes in specific cell types, including ZmMRP-1 and Megl in BETL cells and a majority of zein-and starch-related genes in CSE cells. Using a haploid induction system, we show that wild-type endosperm could rescue the smaller size of osi embryo, which suggests that nutrients are allocated by tho wild type endosperm. Therefore, our dsta imply that the network regulated by 051 accomplishes a key step in nutrient allocation between endosperm and embryo within maize seeds. 20Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients.We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment.We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients.The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone.The cure rate,changes of oxygen saturation and interference,and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group,and 32 patients to the control group.The cure rate in the tocilizumab group was higher than that in the control group,but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions,the hypoxia ameliorated earlier after tocilizumab treatment,and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative.For patients with bilateral pulmonary lesions and elevated IL-6 levels,tocilizumab could be recommended to improve outcome.Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu 2021Frontiers of Medicine2021,15,3:2
