7Domesticated cynomolgus monkey embryonic stem cells allow the generation of neonatal interspecies chimeric pigs显示文摘Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.Rui Fu Dawei Yu Jilong Ren Chongyang Li Jing Wang Guihai Feng Xuepeng Wang Haifeng Wan Tianda Li Libin Wang Ying Zhang Tang Hai Wei Li Qi Zhou 2020Protein & Cell2020,11,2:4
8Transfer of β-hydroxy-β-methylbutyrate from sows to their offspring and its impact on muscle fiber type transformation and performance in pigs显示文摘Background: Previous studies suggested that supplementation of lactating sows with β-hydroxy-β-methylbutyrate(HMB) could improve the performance of weaning pigs, but there were little information in the muscle fiber type transformation of the offspring and the subsequent performance in pigs from weaning through finishing in response to maternal HMB consumption. The purpose of this study was to determine the effect of supplementing lactating sows with HMB on skeletal muscle fiber type transformation and growth of the offspring during d 28 and180 after birth. A total of 20 sows according to their body weight were divided into the control(CON, n = 10) or HMB groups(HMB, n = 10). Sows in the HMB group were supplemented with β-hydroxy-β-methylbutyrate calcium(HMB-Ca) 2 g/kg feed during d 1 to 27 of lactation. After weaning, 48 mixed sex piglets were blocked by sow treatment and fed standard diets for post-weaning, growing, finishing periods. Growth performance was recorded during d 28 to 180 after birth. Pigs were slaughtered on d 28(n = 6/treatment) and 180(n = 6/treatment) postnatal and the longissimus dorsi(LD) was collected, respectively.Results: The HMB-fed sows during lactation showed increased HMB concentration(P < 0.05) in milk and LD of weaning piglets(P < 0.05). In addition, offsprings in HMB group had a higher finishing BW and lean percentage than did pigs in CON group(P < 0.05), meanwhile, compared with pigs from sows fed the CON diet, pigs from sows fed HMB diet showed higher type Ⅱ muscle fiber cross-sectional area(CSA), elevated myosin heavy chain(MyHC) Ⅱb and Sox6 mRNA, and fast-MyHC protein levels in LD(P < 0.05).Conclusions: HMB supplemented to sow diets throughout lactation increases the levels of HMB in maternal milk and skeletal muscle of pigs during d 28 after birth and promotes subsequent performance of pigs between d 28 and 180 of age by enhancing glycolytic muscle fiber transformation.Haifeng Wan Jiatao Zhu Caimei Wu Pan Zhou Yong Shen Yan Lin Shengyu Xu Lianqiang Che Bin Feng Jian Li Zhengfeng Fang De Wu 2017Journal of Animal Science and Biotechnology2017,8,2:2

10Synthesis and ordered aggregation in water of a blue light-emitting PEO-PHP-PEO triblock oligomer显示文摘A novel blue light-emitting coil-rod-coil triblock oligomer, PEO-PHP-PEO, which consists of para-hexaphenyl (PHP) as the rigid rod and poly(ethylene oxide) (PEO, M w = 750) as the flexible coil, has been synthesized. It is the first soluble poly(para-phenylene) molecule containing no lateral substituent, which can dissolve not only in organic solvents such as chloroform and THF, but also in water. The critical micelle concentration (CMC) of PEO-PHP-PEO in water is 5.25×10-4 mol·L-1 and the surface tension at CMC, 鉉MC, is 61.29 mN·m-1. The absorption peak of the molecularly dissolved PEO-PHP-PEO in water is centered at 323 nm and the fluorescence spectrum has a maximum at 432 nm and shows the vibronic fine structure typical of molecular dissolution. The photoluminescence spectrum of the aggregate solution is similar to that of the PHP polycrystal film, indicating that the ordered packing of PHP block has taken place due to a combination of hydrophobic interaction and p-p stacking. 11Self-organization of unimolecular micelles in beam stream for functional mesoporous metal oxide nanofibers显示文摘The use of linear amphiphilic block copolymers as templates is an important method for the preparation of mesoporous materials.However,the obtained assemblies are usually sensitive to synthetic conditions,which impedes the preparation of such mesoporous materials in certain environments.Herein,we report a universal strategy applying an amphiphilic multiarm triblock copolymer in the preparation of mesoporous metal oxide nanofibers(NFs)using one metal oxide(TiO_(2),ZrO_(2),WO_(3),CeO_(2)),or two(TiO_(2)/WO 3,TiO_(2)/ZrO_(2),TiO_(2)/CeO_(2))and three(TiO_(2)/WO_(3)/CuO)metal oxides as composites.The template consists of modified β-cyclodextrin as the center of the macromolecule which is attached sequentially to a block of polystyrene,poly(acrylic acid),and poly(ethylene oxide).Under electrospinning conditions,stable unimolecular micelles are formed and effectively co-assemble with metal ions to form fibrous nanostructures.As indicated by various characterization methods,the synthesized TiO_(2) and its derived composite NFs maintain a straight and continuous fibrous structure after calcination,and TiO_(2) NFs exhibit uniform mesopores of 10.8 nm in diameter and a large Brunauer-Emmett-Teller surface area of 143.3 m^(2)g^(−1).Benefiting from the characteristic structure,still present after modification,Pt-decorated mesoporous TiO_(2) NFs display excellent ability in the visible-light photocatalytic degradation of tetracycline,which is superior to the commercial P25 catalyst.This study reveals a promising strategy for the preparation of fibrous mesoporous metal oxides.Chongfei Gu Xiaoshan Fan Guihua Zhu Yuchi Fan Haifeng Wang Tao Zhao Qi Xiao Yuan Fang Xiaopeng Li Wan Jiang Lianjun Wang Pengpeng Qiu Wei Luo 2022Fundamental Research2022,2,5:1

14SYNTHESIS AND CHARACTERIZATION OF A NOVEL BLUE LIGHT EMITTING ROD-COIL DIBLOCK OLIGOMER显示文摘Four new rod-coil diblock molecules, which had well-defined para-phenylene oligomers (PPP, from biphenyl to quinquephenyl) as rigid segments and poly(ethylene oxide) (PEO, Mw ca. 750) as flexible segments, were synthesized. The chemical structure and the number of repeating unit in PEO coil and PPP rod were characterized by 1H-NMR and MALDITOF mass spectrometry. The diblock oligomers showed good solubility in common organic solvents and aggregated in water as characterized by a blue shift in UV-vis absorption spectra, a red shift in emission spectra, quenching of the fluorescence and formation of the microfibrilla and fractal-like structures. 15Generation of tetraploid complementation mice from embryonic stem cells cultured with chemical defined medium显示文摘Mouse embryonic stem cells(mESCs)derived from inner cell mass(ICM)of pre-implantation embryos,can maintain undifferentiated state when cultured in N2B27 medium supplemented with GSK3inhibitor CHIR99021 and MEK inhibitor PD0325901(‘‘2i’’)and leukemia inhibitor factor(LIF).Compare to conventional culture medium,all components of this medium are defined.With the N2B27 medium,‘‘2i’’and LIF,mESCs can contribute to the germline of the chimeric embryos,however,whether the‘‘all-ES cells’’mice can been generated by tetraploid complementation is unclear yet,while the tetraploid complementation serve as a golden standard to assess the pluripotency of ES cells.Here,our study showed that mESCs derived and cultured with the N2B27 complete medium could generate fertile mice by tetraploid complementation.In addition,the survival rate of tetraploid complementation mice produced by inbred mES cell lines is higher than the conventional culture condition,and increased the percentage of Oct4 positive cells contrast to conventional medium either.Therefore,the N2B27 medium supplemented with‘‘2i’’and LIF is an alternative choice forthe derivation and long-term culture of mouse embryonic stem cells.Chunjing Feng Haifeng Wan Xiao-Yang Zhao Liu Wang Qi Zhou 2014Chinese Science Bulletin2014,59,22:1
