| 1 | Duck egg drop syndrome virus:an emerging Tembusu-related flavivirus in China显示文摘Duck egg drop syndrome virus(DEDSV) is a newly emerging pathogenic flavivirus isolated from ducks in China.DEDSV infection mainly results in severe egg drop syndrome in domestic poultry,which leads to huge economic losses.Thus,the discovery of ways and means to combat DEDSV is urgent.Since 2010,a remarkable amount of progress concerning DEDSV research has been achieved.Here,we review current knowledge on the epidemiology,symptomatology,and pathology of DEDSV.A detailed dissection of the viral genome and polyprotein sequences,comparative analysis of viral antigenicity and the corresponding potential immunity against the virus are also summarized.Current findings indicate that DEDSV should be a distinct species from Tembusu virus.Moreover,the adaption of DEDSV in wildlife and its high homology to pathogenic flaviviruses(e.g.,West Nile virus,Japanese encephalitis virus,and dengue virus),illustrate its reemergence and potential to become a zoonotic pathogen that should not be overlooked.Detailed insight into the antigenicity and corresponding immunity against the virus is of clear significance for the development of vaccines and antiviral drugs specific for DEDSV. | LIU PeiPei LU Hao LI Shuang WU Ying George Fu GAO SU JingLiang | 2013 | Science China(Life Sciences)2013,56,8: | 28 |

This increased the output of influenza virus sequence data as well as the research publication and other reports. In order to fight against H5N1 avian flu in a comprehensive way, we designed and started to set up the Website for Avian Flu Information (http://gffzz859ce0d435cb4b67hx06cvpqopcxx6nox.ffgz.tsg.suse.edu.cn) from 2004. Other than the influenza virus database available, the website is aiming to integrate diversified information for both researchers and the public. From 2004 to 2009, we collected information from all aspects, i.e. reports of outbreaks, scientific publications and editorials, policies for prevention, medicines and vaccines, clinic and diagnosis. Except for publications, all information is in Chinese. Till April 15, 2009, the cumulative news entries had been over 2000 and research papers were approaching 5000. By using the curated data from Influenza Virus Resource, we have set up an influenza virus sequence database and a bioinformatic platform, providing the basic functions for the sequence analysis of influenza virus. | 14 | Crystal structure of histidine-containing phosphocarrier protein from Thermoanaerobacter tengcongensis MB4 and the implications for thermostability显示文摘Protein thermostability is an inherent characteristic of proteins from thermophilic microorganisms,and therefore enables these organisms to survive at extreme temperatures.Although it is well-known that thermostable proteins are critical for the growth of thermophilic organisms,the structural basis of protein thermostability is not yet fully understood.The histidine-containing phosphocarrier (HPr) protein,a phosphate shuttle protein in the phosphoenolpyruvate-dependent sugar transport system (PTS) of bacterial species,is an ideal model for investigating protein thermostability with respect to its small size and deficiency in disulphide bonds or cofactors.In this study,the HPr protein from Thermoanaerobacter tengcongensis (TtHPr) is cloned and purified.Crystal structure with good quality has been determined at 2.3 resolution,which provides a firm foundation for exploring the thermostable mechanism.However,it shows that the crystal structure is conserved and no clue can be obtained from this single structure.Furthermore,detailed comparison of sequence and structure with the homologs from mesoor thermophilic bacteria shows no obvious rule for thermostability,but the extra salt-bridge existing only in thermophilic bacteria might be a better explanation for thermostability of HPr.Thus,mutations are performed to interrupt the salt-bridge in HPrs in thermophilic bacteria.Using site-directed mutations and the circular dichroism method,thermostability is evaluated,and the mutational variations are shown to have a faster denaturing rate than for wild-type viruses,indicating that mutations cause instability in the HPrs.Understanding the higher-temperature resistance of thermophilic and hyperthermophilic proteins is essential to studies on protein folding and stability,and is critical in engineering efficient enzymes that can work at a high temperature. | FENG ChunYan GAO Feng LIU YiWei WANG GuiHua PENG Hao MA YanHe YAN JingHua GAO George Fu | 2011 | Science China(Life Sciences)2011,54,6: | 2 |
