Disease profile and plasma neutralizing activity of postvaccination Omicron BA.1 infection in Tianjin,China:a retrospective study显示文摘Dear Editor,SARS-CoV-2 Omicron variant BA.1 first emerged on the Chinese mainland on January 8,2022,in Tianjin and caused a large wave of infections.1 Four rounds of mass testing of its~14 million residents were launched on January 9,12,15,and 20,respectively.As of February 7,2022,a total of 430 individuals were tested positive for Omicron BA.1,with no new infections detected for the following 16 days.In the Chinese mainland, the most delivered vaccines are inactivated vaccines. As of January 8,2022,up to 93.2%of Tianjin’s residents had been vaccinated with at least one dose.2 We investigated the protection conferred by vaccination against Omicron BA.1 by examining the breakthrough infections among vaccinees as compared with unvaccinated cases. Our study population comprised all 430 cases during the outbreak of Omicron BA.1 in Tianjin. | Hong Zheng Yunlong Cao Xiaosu Chen Fengmei Wang Ye Hu Weiliang Song Yangyang Chai Qingqing Gu Yansong Shi Yingmei Feng Shuxun Liu Yan Xie Xiaoliang Sunney Xie Wentao Jiang Zhongyang Shen | 2022 | Cell Research2022,32,8: | 12
In vivo self-assembled small RNAs as a new generation of RNAi therapeutics显示文摘RNAi therapy has undergone two stages of development, direct injection of synthetic siRNAs and delivery with artificial vehicles or conjugated ligands;both have not solved the problem of efficient in vivo siRNA delivery. Here,we present a proof-of-principle strategy that reprogrammes host liver with genetic circuits to direct the synthesis and self-assembly of siRNAs into secretory exosomes and facilitate the in vivo delivery of siRNAs through circulating exosomes.By combination of different genetic circuit modules,in vivo assembled siRNAs are systematically distributed to multiple tissues or targeted to specific tissues(e.g.,brain),inducing potent target gene silencing in these tissues. The therapeutic value of our strategy is demonstrated by programmed silencing of critical targets associated with various diseases, including EGFR/KRAS in lung cancer,EGFR/TNC in glioblastoma and PTP1B in obesity.Overall, our strategy represents a next generation RNAi therapeutics, which makes RNAi therapy feasible. | Zheng Fu Xiang Zhang Xinyan Zhou Uzair Ur-Rehman Mengchao Yu Hongwei Liang Hongyuan Guo Xu Guo Yan Kong Yuanyuan Su Yangyang Ye Xiuting Hu Wei Cheng Jinrong Wu Yanbo Wang Yayun Gu Sheng-feng Lu Dianqing Wu Ke Zen Jing Li Chao Yan Chen-Yu Zhang Xi Chen | 2021 | Cell Research2021,31,6: | 10
Trade-offs and Synergies of Ecosystem Services in the Taihu Lake Ba- sin of China显示文摘Understanding the spatial interactions among multiple ecosystem services is crucial for ecosystem services management. Ecosystem services, including crop production, freshwater supply, aquatic production, net primary production, soil conservation, water conservation, flood regulation, forest recreation, were measured at 1-km grid scale covering the Taihu Lake Basin(TLB) of China. Our objective is to get a comprehensive understanding of the spatial distributions, trade-offs, synergies of multiple ecosystem services across the TLB. Our results found that: 1) majority of ecosystem services were clustered in space and had a similar spatial distribution pattern with the geographical resource endowment. Most of the landscape contributed a high supply of no services, one or two, and a low supply of three to seven services. 2) There were high correlation between forest recreation and freshwater supply and regulating services. Aquatic production had low correlation with other services. 3) The changes of provisioning services led to trade-offs between regulating services and cultural services in the TLB, while synergies mainly occurred among the provisioning service. 4) The spatial relationships of multiple services are consistent at 1-km spatial scale, counties and provinces. This research could help integrate multiple ecosystem services across scales and serve as a reference for decision making. | QIAO Xuning GU Yangyang ZOU Changxin WANG Lei LUO Juhua HUANG Xianfeng | 2018 | Chinese Geographical Science2018,28,1: | 5
Mineral phase and structure changes during roasting of fine-grained carbonaceous gold ores and their effects on gold leaching efficiency显示文摘While roasting has been widely applied to reduce the negative effect of carbonaceous matters on gold extraction from fine-grained carbonaceous gold ores, the phase and structure changes of minerals during roasting and their in fluences on the leachi ng rate of gold have not been fully understood. This limits the extraction of carbonaceous gold deposits. The current work examines the oxidation process of a fine-grained carbonaceous gold ore during roasting using a range of techniques including X-ray diffraction (XRD), seanning electron microscopy (SEM), Energy Dispersive Spectrometer (EDS) analysis and pore structure analysis together with gold leaching tests. The results show that during the process of oxidative roasting, the carbonaceous matters (organic carbon and graphitic carbon) and pyrite were completely decomposed at 600 ℃ with the carbonaceous components burned and pyrite oxidized into hematite. At 650 ℃, while dolomite was decomposed into calcia, magnesia, calcium sulfate etc., the calcine structure became loose and porous, leading to a high gold leaching rate from the roasted product. Above 750 ℃, the porous calcite structure started to collapse along with the agglomeration, leading to the secondary encapsulation of gold particles, which contributed to the sharp drop in the gold leaching rate of the roasted product. This study suggests optimum phase and structure changes of minerals during roasting to achieve maximum gold extraction from fine-grained carbonaceous gold deposits. | Jianping Jin Yuexin Han Hui Li Yangyang Huai Yongjun Peng Xiaotian Gu Wei Yang | 2019 | Chinese Journal of Chemical Engineering2019,27,5: | 4

A large-scale proteogenomic atlas of pear显示文摘Pear is an important fruit tree that is widely distributed around the world. The first pear genome map was reported from our laboratory approximately 10 years ago.To further study global protein expression patterns in pear,we generated pear proteome data based on 24 major tissues. The tissue-resolved profiles provided evidence of the expression of 17953 proteins. We identified 4294 new coding events and improved the pear genome annotation via the proteogenomic strategy based on 18090 peptide spectra with peptide spectrum matches>1.Among the eight randomly selected new short coding open reading frames that were expressed in the style, four promoted and one inhibited the growth of pear pollen tubes.Based on gene coexpression module analysis,we explored the key genes associated with important agronomic traits, such as stone cell formation in fruits. The network regulating the synthesis of lignin,a major component of stone cells, was reconstructed, and receptor-like kinases were implicated as core factors in this regulatory network. Moreover,we constructed the online database PearEXP(http://gffzz20aea264798e4804hfcwfu9qcffpo60wb.ffgz.tsg.suse.edu.cn)to enable access to the pear proteogenomic resources. This study provides a paradigm for in-depth proteogenomic studies of woody plants. | Peng Wang Xiao Wu Zebin Shi Shutian Tao Zhe Liu Kaijie Qi Zhihua Xie Xin Qiao Chao Gu Hao Yin Mengyu Cheng Xiaoyu Gu Xueying Liu Chao Tang Peng Cao Shaohang Xu Baojin Zhou Tingting Gui Yangyang Bian Juyou Wu Shaoling Zhang | 2023 | Molecular Plant2023,16,3: | 0
Spatial variability between glacier mass balance and environmental factors in the High Mountain Asia显示文摘High Mountain Asia(HMA) region contains the world’s highest peaks and the largest concentration of glaciers except for the polar regions, making it sensitive to global climate change. In the context of global warming, most glaciers in the HMA show various degrees of negative mass balance, while some show positive or near-neutral balance. Many studies have reported that spatial heterogeneity in glacier mass balance is strongly related to a combination of climate parameters. However, this spatial heterogeneity may vary according to the dynamic patterns of climate change at regional or continental scale. The reasons for this may be related to non-climatic factors. To understand the mechanisms by which spatial heterogeneity forms, it is necessary to establish the relationships between glacier mass balance and environmental factors related to topography and morphology. In this study, climate, topography, morphology, and other environmental factors are investigated. Geodetector and linear regression analysis were used to explore the driving factors of spatial variability of glacier mass balance in the HMA by using elevation change data during 2000–2016. The results show that the coverage of supraglacial debris is an essential factor affecting the spatial heterogeneity of glacier mass balance, followed by climatic factors and topographic factors, especially the median elevation and slope in the HMA. There are some differences among mountain regions and the explanatory power of climatic factors on the spatial differentiation of glacier mass balance in each mountain region is weak, indicating that climatic background of each mountain region is similar. Therefore, under similar climatic backgrounds, the median elevation and slope are most correlated with glacier mass balance. The interaction of various factors is enhanced, but no unified interaction factor plays a primary role. Topographic and morphological factors also control the spatial heterogeneity of glacier mass balance by influencing its sensitivity to climate change. In conclusion, geodetector method provides an objective framework for revealing the factors controlling glacier mass balance. | ZHANG Zhen GU Zhengnan Hu Kehong XU Yangyang ZHAO Jinbiao | 2022 | Journal of Arid Land2022,14,4: | 0
Roles of light transmission aggregometry and CYP2C19 genotype in predicting ischaemic complications during interventional therapy for intracranial aneurysms显示文摘Background and purpose Light transmission aggregometry(LTA)and CYP2C19 genotype analysis are commonly used to evaluate the antiplatelet effects of clopidogrel during the interventional treatment of intracranial aneurysms. The aim of this study was to determine which test can predict ischaemic events during these treatments. Methods Patient demographic information, imaging data, laboratory data and ischaemic complications were recorded.LTA and CYP2C19 genotype results were compared, and multiple linear regression was performed to examine factors related to platelet reactivity.Multivariate regression analysis was performed to determine whether LTA and CYP2C19 could predict ischaemic complications and to identify other clinical risk factors.Receiver operating characteristic curve analysis was conducted to calculate the cut-off value for predicting ischaemic complications.A subgroup analysis was also performed for different CYP2C19 genotype metabolisers,as well as for patients with flow diverters and traditional stents. Results A total of 379 patients were included,of which 22 developed ischaemic events.Maximum platelet aggregation induced by ADP(ADP-MPA)could predict ischaemic events(p<0.001;area under the curve,0.752(95%CI 0.663 to 0.842)),and its cut-off value was 41.5%.ADP-MPA(p=0.001)and hypertension duration>10 years(p=0.022)were independent risk factors for ischaemic events, while the CYP2C19 genotype was not associated with ischaemic events. In the subgroup analysis,ADP-MPA could predict ischaemic events in fast metabolisers(p=0.004)and intermediate metabolisers(p=0.003).The cut-off value for ischaemic events was lower in patients with flow diverters(ADP-MPA=36.4%)than in patients with traditional stents(ADP-MPA=42.9%).Conclusions ADP-MPA can predict ischaemic complications during endovascular treatment of intracranial aneurysms.Patients with flow diverters require stronger antiplatelet medication than patients with traditional stents. | Yangyang Zhou Wenqiang Li Chao Wang Ruhang Xie Yongnan Zhu Qichen Peng Limin Zhang Hongqi Zhang Yuxiang Gu Shiqing Mu Jian Liu Xinjian Yang | 2023 | Stroke & Vascular Neurology2023,8,4: | 0
