| 1 | Diffusion-tensor imaging as an adjunct to dynamic contrastenhanced MRI for improved accuracy of differential diagnosis between breast ductal carcinoma in situ and invasive breast carcinoma显示文摘Objective:To determine the value of diffusion-tensor imaging(DTI) as an adjunct to dynamic contrastenhanced magnetic resonance imaging(DCE-MRI) for improved accuracy of differential diagnosis between breast ductal carcinoma in situ(DCIS) and invasive breast carcinoma(IBC).Methods:The MRI data of 63 patients pathologically confirmed as breast cancer were analyzed.The conventional MRI analysis metrics included enhancement style,initial enhancement characteristic,maximum slope of increase,time to peak,time signal intensity curve(TIC) pattern,and signal intensity on FST2 WI.The values of apparent diffusion coefficient(ADC),directionally-averaged mean diffusivity(D_(avg)),exponential attenuation(EA),fractional anisotropy(FA),volume ratio(VR) and relative anisotropy(RA)were calculated and compared between DCIS and IBC.Multivariate logistic regression was used to identify independent factors for distinguishing IBC and DCIS.The diagnostic performance of the diagnosis equation was evaluated using the receiver operating characteristic(ROC) curve.The diagnostic efficacies of DCEMRI,DWI and DTI were compared independendy or combined.Results:EA value,lesion enhancement style and TIC pattern were identified as independent factor for differential diagnosis of IBC and DCIS.The combination diagnosis showed higher diagnostic efficacy than a single use of DCE-MRI(P=0.02),and the area of the curve was improved from 0.84(95%CI,0.67-0.99) to0.94(95%CI,0.85-1.00).Conclusions:Quantitative DTI measurement as an adjunct to DCE-MRI could improve the diagnostic performance of differential diagnosis between DCIS and IBC compared to a single use of DCE-MRI. | Yuan Wang Xiaopeng Zhang Kun Cao Yanling Li Xiaoting Li Liping Qi Lei Tang Zhilong Wang Shunyu Gao | 2015 | Chinese Journal of Cancer Research2015,27,2: | 12 |
| 2 | Coagulation efficiency and flocs characteristics of recycling sludge during treatment of low temperature and micro-polluted water显示文摘Drinking water treatment sludge,characterized as accumulated suspended solids and organic and inorganic matter,is produced in large quantities during the coagulation process.The proper disposal,regeneration or reuse of sludge is,therefore,a significant environmental issue.Reused sludge at low temperatures is an alternative method to enhance traditional coagulation efficiency.In the present study,the recycling mass of mixed sludge and properties of raw water (such as pH and turbidity) were systematically investigated to optimize coagulation efficiency.We determined that the appropriate dosage of mixed sludge was 60 mL/L,effective initial turbidity ranges were below 45.0 NTU,and optimal pH for DOMs and turbidity removal was 6.5-7.0 and 8.0,respectively.Furthermore,by comparing the flocs characteristics with and without recycling sludge,we found that floc structures with sludge were more irregular with average size growth to 64.7 μm from 48.1 μm.Recycling sludge was a feasible and successful method for enhancing pollutants removal,and the more irregular flocs structure after recycling might be caused by breakage of reused flocs and incorporation of powdered activated carbon into larger flocs structure.Applied during the coagulation process,recycling sludge could be significant for the treatment of low temperature and micro-polluted source water. | Zhiwei Zhou Yanling Yang Xing Li Wei Gao Heng Liang Guibai Li | 2012 | Journal of Environmental Sciences2012,24,6: | 12 |
| 3 | SHP2 promotes proliferation of breast cancer cells through regulating Cyclin D1 stabilityviathe PI3K/AKT/GSK3β signaling pathway显示文摘Objective:The tyrosine phosphatase SHP2 has a dual role in cancer initiation and progression in a tissue type-dependent manner.Several studies have linked SHP2 to the aggressive behavior of breast cancer cells and poorer outcomes in people with cancer.Nevertheless,the mechanistic details of how SHP2 promotes breast cancer progression remain largely undefined.Methods:The relationship between SHP2 expression and the prognosis of patients with breast cancer was investigated by using the TCGA and GEO databases.The expression of SHP2 in breast cancer tissues was analyzed by immunohistochemistry.CRISPR/Cas9 technology was used to generate SHP2-knockout breast cancer cells.Cell-counting kit-8,colony formation,cell cycle,and EdU incorporation assays,as well as a tumor xenograft model were used to examine the function of SHP2 in breast cancer proliferation.Quantitative RT-PCR,western blotting,immunofluorescence staining,and ubiquitination assays were used to explore the molecular mechanism through which SHP2 regulates breast cancer proliferation.Results:High SHP2 expression is correlated with poor prognosis in patients with breast cancer.SHP2 is required for the proliferation of breast cancer cellsin vitro and tumor growthin vivo through regulation of Cyclin D1 abundance,thereby accelerating cell cycle progression.Notably,SHP2 modulates the ubiquitin–proteasome-dependent degradation of Cyclin D1viathe PI3K/AKT/GSK3βsignaling pathway.SHP2 knockout attenuates the activation of PI3K/AKT signaling and causes the dephosphorylation and resultant activation of GSK3β.GSK3βthen mediates phosphorylation of Cyclin D1 at threonine 286,thereby promoting the translocation of Cyclin D1 from the nucleus to the cytoplasm and facilitating Cyclin D1 degradation through the ubiquitin–proteasome system.Conclusions:Our study uncovered the mechanism through which SHP2 regulates breast cancer proliferation.SHP2 may therefore potentially serve as a therapeutic target for breast cancer. | Yue Yuan Yanling Fan Zicong Gao Xuan Sun He Zhang Zhiyong Wang Yanfen Cui Weijie Song Zhaosong Wang Fei Zhang Ruifang Niu | 2020 | Cancer Biology & Medicine2020,17,3: | 11 |

| 5 | Quantifying public health benefits of environmental strategy of PM2.5 air quality management in Beijing–Tianjin–Hebei region,China显示文摘In 2013,China issued 'Air Pollution Prevention and Control Action Plan(Action Plan)' to improve air quality.To assess the benefits of this program in Beijing-Tianjin-Hebei(BTH)region,where the density of population and emissions vary greatly,we simulated the air quality benefit based on Ben MAP to satisfy the Action Plan.In this study,we estimate PM_(2.5) concentration using Voronoi spatial interpolation method on a grid with a spatial resolution of 1 × 1 km^2.Combined with the exposure-response function between PM_(2.5) concentration and health endpoints,health effects of PM_(2.5) exposure are analyzed.The economic loss is assessed by using the willingness to pay(WTP) method and human capital(HC) method.When the PM_(2.5) concentration falls by 25% in BTH and reached 60 μg/m^3 in Beijing,the avoiding deaths will be in the range of 3175 to 14051 based on different functions each year.Of the estimated mortality attributable to all causes,3117 annual deaths were due to lung cancer,1924 – 6318 annual deaths were due to cardiovascular,and343 – 1697 annual deaths were due to respiratory.Based on WTP,the estimated monetary values for the avoided cases of all cause mortality,cardiovascular mortality,respiratory mortality and lung cancer ranged from 1110 to 29632,673 to 13325,120 to 3579,1091 to 6574 million yuan,respectively.Based on HC,the corresponding values for the avoided cases of these four mortalities were 267 to 1178,161 to 529,29 to 143 and 261 million yuan,respectively. | Li Chen Mengshuang Shi Suhuan Li Shuang Gao Hui Zhang Yanling Sun Jian Mao Zhipeng Bai Zhongliang Wang Jiang Zhou | 2017 | Journal of Environmental Sciences2017,29,7: | 6 |

| 7 | Melanopsin-expressing retinal ganglion cell loss and behavioral analysis in the Thy1-CFP-DBA/2J mouse model of glaucoma显示文摘In this study,the role of melanopsin-expressing retinal ganglion cells(mRGCs) in the glaucoma-induced depressive behavioral response pattern was investigated.The CFP-D2 transgenic glaucoma animal model from five age groups was used in this study.Immunohistochemical labeling,quantitative analysis of mRGC morphology,open field test(OFT),and statistical analysis were used.In comparison with C57 BL/6 mice,the age-matched CFP-D2 mice had significantly elevated intraocular pressure(IOP).We observed parallel morphological changes in the retina,including a reduction in the density of cyan fluorescent protein(CFP) expressing cells(cells mm 2 at 2 months of age,1309±26;14 months,878±30,P<0.001),mRGCs(2 months,48±3;14 months,19±4,P<0.001),Brn3b-expressing RGCs(2 months,1283±80;14 months,950±31,P<0.001),Brn-3b expressing mRGCs(5 months,50.17%±5.5%;14 months,12.61%±3.8%,P<0.001),and reduction in the dendritic field size of mRGCs(mm2 at 2 months,0.077±0.015;14 months,0.065±0.015,P<0.05).CFP-D2 mice had hyperactive locomotor activity patterns based on OFT findings of the total distance traveled,number of entries into the center,and time spent in the center of the testing apparatus.The glaucoma induced hyperactive response pattern could be associated with dysfunctional mRGCs,most likely Brn-3b-positive mRGCs in CFP-D2 mice. | ZHANG Qi VUONG Helen HUANG Xin WANG YanLing BRECHA Nicholas C. | 8 | Utility of CT in differentiating liver metastases of well-differentiated gastroenteropancreatic neuroendocrine neoplasms from poorly-differentiated neuroendocrine neoplasms显示文摘Objective: To determine the capability of dynamic enhanced computed tomography(CT) to differentiate liver metastases(LMs) of well-differentiated from poorly-differentiated gastroenteropancreatic neuroendocrine neoplasms(GEP-NENs).Methods: Patients with LMs of GEP-NENs who underwent dynamic enhanced CT examination in Peking University Cancer Hospital from January 2009 to October 2015 were included and data were retrospectively analyzed. We assessed the qualitative and quantitative CT features to identify the significant differentiating CT features of LMs of poorly-differentiated GEP-NENs from those of well-differentiated GEP-NENs using univariate analysis and a multivariate logistic regression model.Results: The study included 22 patients with LMs of well-differentiated GEP-NENs and 32 patients with LMs of poorly-differentiated GEP-NENs. Univariate analysis revealed statistically significant differences between the LMs of well-and poorly-differentiated GEP-NENs in terms of feeding arteries(36.4% vs. 75.0%, χ2=8.061,P=0.005), intratumoral neovascularity(18.2% vs. 59.4%, χ2=9.047, P=0.003), lymphadenopathy(27.3% vs. 81.2%,χ2=15.733, P<0.001), tumor-to-aortic ratio in the hepatic arterial and portal venous phase(T-A/AP: 0.297±0.080 vs.0.251±0.059, t=2.437, P=0.018; T-A/PVP: 0.639±0.138 vs. 0.529±0.117, t=3.163, P=0.003) and tumor-to-liver ratio in the hepatic arterial phase(T-L/AP: 1.108±0.267 vs. 0.907±0.240, t=2.882, P=0.006). The LMs of poorlydifferentiated GEP-NENs showed more feeding arteries, more intratumoral neovascularity, more lymphadenopathy and a lower tumor-to-aortic ratio. | 9 | Fast mapping of a chlorophyll b synthesis-deficiency gene in barley(Hordeum vulgare L.) via bulked-segregant analysis with reduced-representation sequencing显示文摘Bulked-segregant analysis coupled with next-generation sequencing(BSA-seq) has emerged as an efficient tool for genetic mapping of single genes or major quantitative trait loci controlling(agronomic) traits of interest. However, such a mapping-by-sequencing approach usually relies on deep sequencing and advanced statistical methods. Application of BSA-Seq based on construction of reduced-representation libraries and allele frequency analysis permitted anchoring the barley pale-green(pg) gene on chromosome 3 HL. With further marker-assisted validation, pg was mapped to a 3.9 Mb physical-map interval. In the pg mutant a complete deletion of chlorophyllide a oxygenase(HvCAO) gene was identified.Because the product of this gene converts Chl a to Chl b, the pg mutant is deficient in Chl b.An independent Chl b-less mutant line M4437_2 carried a nonsynonymous substitution(F263 L) in the C domain of HvCAO. | 14 | Adsorption of humic acid from aqueous solution on bilayer hexadecyltrimethyl ammonium bromide-modified zeolite显示文摘Surfactant-modified natural zeolites(SMNZ)with different coverage types were prepared by loading hexadecyltrimethyl ammonium bromide(HTAB)onto the surface of a natural zeolite.The adsorption behavior of humic acid(HA)on SMNZ was investigated.Results indicate that the adsorbent SMNZ exhibited a higher affinity toward HA than the natural zeolite.HA removal efficiency by SMNZ increased with HTAB loading.Coexisting Ca2+in solution favored HA adsorption onto SMNZ.Adsorption capacity decreased with an increasing solution pH.For typical SMNZ with bilayer HTAB coverage,HA adsorption process is well described by a pseudo-second-order kinetic model.The experimental isotherm data fitted well with the Langmuir model.Calculated maximum HA adsorption capacities for SMNZ with bilayer HTAB coverage at pH 5.5 and 7.5 were 63 and 41 mg·g^(-1),respectively.E2/E3(absorbance at 250 nm to that at 365 nm)and E4/E6(absorbance at 465 nm to that at 665 nm)ratios of the residual HA in solution were lower than that of the original HA solution.This indicates that the HA fractions with high polar functional groups,low molecular weight(MW),and aromaticity had a stronger tendency for adsorption onto SMNZ with bilayer HTAB coverage.Results show that HTABmodified natural zeolite is a promising adsorbent for removal of HA from aqueous solution. | Yanhui ZHAN Jianwei LIN Yanling QIU Naiyun GAO Zhiliang ZHU | 2011 | Frontiers of Environmental Science & Engineering2011,5,1: | 2 |
| 15 | Phenotypic analysis and molecular characterization of an allelic mutant of the D61 gene in rice显示文摘Brassinosteroids(BRs) are a class of plant-specific steroidal hormones that play important roles in multiple biological processes. In this paper, a classic rice mutant gsor300084,showing erect leaves and semi-dwarf stature, was characterized. Morphological analysis in darkness showed that the mesocotyl of the gsor300084 mutant did not elongate when grown in darkness. Coleoptile elongation and root growth were less affected by the exogenous application of brassinolide(BL), the most active form of BR, in gsor300084 than in the wild-type rice variety Matsumae. Lamina joint bending analysis also showed that gsor300084 was less sensitive to exogenous BL than Matsumae. These results suggested that the gsor300084 mutant is defective in BR sensitivity. Map-based cloning indicated that gsor300084 is a novel allelic mutant of the DWARF61(D61) gene, which encodes the putative BR receptor OsBRI1. A single-base mutation appears in the LRR domain of OsBRI1, changing the 444 th amino acid from tryptophan(W) to arginine(R). Subcellular localization analysis suggested that both the wild-type and mutant OsBRI1 protein are localized at the cytoplasmic membrane. | 18 | Efficacy and safety of single-bolus tenecteplase compared with front-loaded alteplase in Chinese patients with acute myocardial infarction显示文摘Background and Objective Previous study showed tenecteplase and alteplaxe were equovalent for 30-day mortality in the treatment of acute myocardial infarction. The purpose of this open-label,randomized,multi-center,angiographic trial was to assess the efficacy and safety of tenecteplase compared with alteplase in Chinese patients with acute myocardial infarction. Methods We recruited patients with acute ST-elevation myocardial infarction presenting within 6 hours of symptom onset from October,2002 to March,2004,in 5 hospitals in Beijing. After giving informed consent,patients were randomly assigned a single-bolus injection of tenecteplase(30-50 mg according to body weight) or front loaded alteplase(100 mg) ,and underwent coronary angiography at 90 min after starting the study drug. All patients received aspirin and heparin(target activated partial thromboplastin time 50-70 s) . The primary efficacy end point was the rate of TIMI grade 3 flow at 90 minutes. Other efficacy end points included TIMI grade 2/3 flow at 90 minutes. Safety end points included all stroke,intracranial hemorrhage(ICH) ,moderate/severe hemorrhage(except for ICH) ,all-cause mortality at 30-days,and major non-fatal cardiac events at 30 days. Results Overall 110 patients were eligible for statistical analysis,with 58 patients assigned to receive tenecteplase and 52 patients to alteplase. Tenecteplase produced a rate of TIMI grade 3 flow at 90 minutes after the start of thrombolysis(68.4%) similar to that of alteplase(66.7%,P=1.0) ;the rates of TIMI grade 2 or 3 were similar for patients treated with tenecteplase versus alteplase(89.5% versus 80.4%,respectively,P=0.278) . At 30 days,rates for all strokes were similar for the two groups(5.17% for tenecteplase and 1.92% for alteplase,P=0.62) ;rates of ICH were 3.45% and 1.92%(tenecteplase and rt-PA,P=1.00) respectively. The rate of moderate/severe hemorrhage was 8.62% with tenecteplase and 5.77% with alteplase(P=0.72) ;total mortality was almost identical in the two groups(13.8% versus 9.6%,respectively,P=0.565) while the rates of non-fatal cardiac complications were 10.35% and 11.54%(tenecteplase and alteplase,P=1.0) . Conclusions The efficacy of a single-bolus,weight-adjusted tenecteplase fibrinolytic regimen is equivalent to front-loaded alteplase in terms of the rates of TIMI grade 3 flow,and TIMI 2 or 3 flow,but the 30-day mortality and ICH in both groups was so high that the use of tenecteplase is not permitted in China.
