| 1 | Experimental Study on the Protection of Agrimony Extracts from Different Extracting Methods against Cerebral Ischemia-Reperfusion Injury显示文摘Objective To study the protective effect of agrimony extracts from different extracting methods on cerebral ischemia-reperfusion injury in rats, in order to optimize the extraction scheme of agrimony.Methods Male rats were randomly assigned into seven groups: 1. Sham-operated group, 2. Untreated MCAO group (MCAO), 3. Petroleum ether extract of Agrimonia pilosa treated MCAO group (PEA), 4. Ethyl acetate extract of Agrimonia pilosa treated MCAO group (EAEA), 5. Ethanol extract of Agrimonia pilosa treated MCAO group (EEA), 6. Water extract of Agrimonia pilosa treated MCAO group (WEA), 7. Nimodipine treated MCAO group (NP). Intragastrical drug administration (i.g) was performed at 0 and 6 hours after MCAO.Neurological function tests were performed after reperfusion for 24 hours, then the brain was removed for the evaluations of the cerebral infarction volume (percentage of total brain volume) by immunohistochemistry,histological changes (hematoxylin-eosin staining), Na+/K+-ATPase, Ca2+-ATPase (modified method of Svoboda and Mosinger), mRNA expression of Tumor suppressor gene (P53) and hot shock protein (HSP70)(quantitative real-time PCR).Results The neurological function of MCAO group had significantly higher scores than the sham group (P<0.01). The WEA group showed a significantly lower neurological score than the MCAO group (P<0.05),indicating the protective effect of WEA on neurological deficits. The mean infarction volumes of WEA (13.5±6.6%, F=4.75, P<0.01), EEA (19.90±6.90%, F=5.23, P<0.01), PEA (20.40±5.30%, F=4.68,P<0.01) and EAEA (22.50±10.50%, F=6.25, P<0.05) group were all significantly smaller than that of MCAO group (29.40±6.50%). HE staining demonstrated that, compared to the treated groups, the infarcted cerebral tissue of MCAO group had more swelling neural cells, lighter stained nucleus, fewer and irregularly distributed neurons. The activity of Na+/K+-ATPase and Ca2+-ATPase reduced in the MCAO group (3.67±0.48 U/mg,1.28±0.26 U/mg, respectively), and were significantly higher in WEA group (7.56±0.85 U/mg, F=12.65,P=0.010; 3.59±0.22 U/mg, F=8.32, P=0.041, respectively). The MCAO group showed significantly elevated P53 and HSP70 mRNA expressions compared to the sham group (P<0.01, P<0.05). P53 mRNA expressions in Agrimony extracts treated groups were significantly lower than that of the MCAO group (all P<0.01), with the WEA group showing the greatest difference from MCAO group. | 5 | Tailoring the mercaptan ligands for high performance inverted perovskite solar cells with efficiency exceeding 21%显示文摘Interface passivation engineering has been recognized as an effective way to simultaneously contribute to the optoelectronic characteristic and stability of perovskite solar cells(PSCs). Herein, a p-conjugated dual-ligand 1,4-phenylmercaptan(PHMT) is explored to rationally tailor the surface of perovskite film.The experimental and theoretical results show that the PHMT presents planar structure and obvious electron delocalization characteristics, which allow it to anchor on the surface of perovskite with a certain orientation, thereby promoting the transport of interface charge. Moreover, the two sulfhydryl groups in PHMT reduce the trap density of the perovskite film by passivating under-coordinated lead ions.Consequently, the PHMT-modified inverted device based on MAPbI_(3)(MA: methylammonium) achieves enhanced efficiency from 18.11%(control) to 21.11%, along with the ambient stability up to 3500 h.After being placed at 85 °C for 500 h or illuminated for 600 h, the modified device remains over 89%or 86% of initial efficiency. | 7 | High-precision potassium isotope analysis using the Nu Sapphire collision cell(CC)-MC-ICP-MS显示文摘This study presents high-precision analyses of stable potassium(K)isotope ratio using the recently-developed,collision-cell multi-collector inductively coupled plasma mass spectrometry(CC-MC-ICP-MS,Nu Sapphire).The accuracy of our analyses is confirmed by measuring well-characterized geostandards(including rocks and seawater).Our results are consistent with literature values and a precision of 0.04‰(2SD)has been achieved based on multiple measurements of BCR-2 geostandard over a six-month period.We also evaluate factors that may lead to artificial isotope fractionations,including the mismatches in K concentration and acid molarity between samples and bracketing standards,as well as potential matrices.As the K adsorption capacity of AGW50-X8(200-400 mesh)is reduced with an increasing amount of matrix elements,less than 150µg K was loaded during the column chemistry.To evaluate the potential use of K isotopes as an archive of paleo seawater composition,δ^(41)K values of an international seawater standard(IAPSO),a Mn-nodule(NOD-P-1),and two iron formation standards(FeR-2 and FeR-4)are reported.The δ^(41)K value of IAPSO is consistent with other seawater samples reported previously,further substantiating a homogeneous K isotopic distribution in modern global oceans.The K isotopes in Mn-nodule(NOD-P-1:−0.121±0.013‰)and iron formation samples(FeR-2:−0.538±0.009‰;FeR-4:−0.401±0.008‰)seem to be an effective tracer of their formation genesis and compositional changes of ancient seawater.Our results suggest that high-precision measurements of stable K isotopes can be routinely obtained and open up a large variety of geological applications,such as continental weathering,hydrothermal circulation and alteration of oceanic crust. | Wenjun LI Mengmeng CUI Qiqi PAN Jing WANG Bingyu GAO Shanke LIU Meng YUAN Benxun SU Ye ZHAO Fang-Zhen TENG Guilin HAN | 2022 | Science China Earth Sciences2022,65,8: | 1 |
| 8 | MAVS-loaded unanchored Lys63-linked polyubiquitin chains activate the RIG-I-MAVS signaling cascade显示文摘The adaptor molecule MAVS forms prion-like aggregates to govern the RIG-I-like receptor(RLR)signaling cascade.Lys63(K63)-linked polyubiquitination is critical for MAVS aggregation,yet the underlying mechanism and the corresponding E3 ligases and deubiquitinating enzymes(DUBs)remain elusive.Here,we found that the K63-linked polyubiquitin chains loaded on MAVS can be directly recognized by RIG-I to initiate RIG-I-mediated MAVS aggregation with the prerequisite of the CARDRIG-I-CARDMAVS interaction.Interestingly,many K63-linked polyubiquitin chains attach to MAVS via an unanchored linkage.We identified Ube2N as a major ubiquitin-conjugating enzyme for MAVS and revealed that Ube2N cooperates with the E3 ligase Riplet and TRIM31 to promote the unanchored K63-linked polyubiquitination of MAVS.In addition,we identified USP10 as a direct DUB that removes unanchored K63-linked polyubiquitin chains from MAVS.Consistently,USP10 attenuates RIG-I-mediated MAVS aggregation and the production of type I interferon.Mice with a deficiency in USP10 show more potent resistance to RNA virus infection.Our work proposes a previously unknown mechanism for the activation of the RLR signaling cascade triggered by MAVS-attached unanchored K63-linked polyubiquitin chains and establishes the DUB USP10 and the E2:E3 pair Ube2N-Riplet/TRIM31 as a specific regulatory system for the unanchored K63-linked ubiquitination and aggregation of MAVS upon viral infection. | Feng Liu Wanxin Zhuang Bin Song Yuan Yang Junqi Liu Yi Zheng Bingyu Liu Jie Zheng Wei Zhao Chengjiang Gao | 2023 | Cellular & Molecular Immunology2023,20,10: | 0 |
| 9 | Analysis of the Correlation between Middle-season Rice Yield and Meteorological Factors in Jingdong County from 2009 to 2016显示文摘Middle-season rice is an important food crop in southern rice areas of China,especially in Yunnan,the main rice-producing region.However,due to the impact of low temperature at the seedling stage and high or low temperature at the booting and heading stage of middle-season rice,the yield is not stable.Based on the data of yield factors of different middle-season rice varieties planted in the same ecological site in Jingdong County from 2009 to 2016,average development period was calculated using the data of development period measured in field during 2009-2016,and the average of meteorological factors(daily average temperature,daily maximum temperature,daily minimum temperature,and sunshine hours)and total precipitation were calculated.The correlation between meteorological factors in different development periods of each year and corresponding per unit area yield was analyzed.The results show that temperature is the most important factor affecting rice yield.Sufficient light is beneficial to the increase in the number of grains per spike and thousand seed weight at the sowing-seedling emergence stage and milk maturity-maturity stage.Excessive precipitation will reduce the number of grains per spike at the booting-heading stage.Excessive precipitation decreases the number of filled grains per spike at the jointing-booting stage,and proper drainage helps increase the formation rate of ears.This study provides scientific reference for rice production and management in this county in future. | Bingyu YANG Zhengyou WANG Min GAO Baofeng HUANG | 2020 | Asian Agricultural Research2020,12,8: | 0 |
| 10 | No consistent daily variation in DNA methylation detected in Populus nigra leaves by methylation-sensitive amplification polymorphism analysis显示文摘DNA methylation, an epigenetic mechanism used by cells to control gene expression, has an important biological role in plant development and environmental fitness. Since plant DNA methylation is closely related to environmental conditions, variation during the day is expected. Here, in genetically identical plants of Populus nigra clone N46, DNA methylation changes in leaves over a 24 h period were detected using the methylation-sensitive amplification polymorphism method. The results showed different DNA methylation patterns in mature poplar leaves: not only in individuals at the same time, but also in samples at each of the six time during the day. In addition,night samples had a higher percentage of methylation than in morning samples. However, no statistically significant differences were found among the samples gathered at different times. Similar results were obtained for three other P. nigra clones with different genetic backgrounds.Real time q PCR showed that the DNA methyltransferase genes Pt-MET1 and Pt-SOM1 involved in CG DNA methylation in poplar were stable over a 24 h period in leaves of P. nigra N46 compared with circadian-controlled genes. That could be part of the reason that methylation of CCGG sites is stable in those leaves. That DNA methylation differed even in genetically identical plants indicates the specificity of DNA methylation changes in their genomes. | 11 | The action mechanism by which C1q/tumor necrosis factor-related protein-6 alleviates cerebral ischemia/reperfusion injury in diabetic mice显示文摘Studies have shown that C1q/tumor necrosis factor-related protein-6 (CTRP6) can alleviate renal ischemia/reperfusion injury in mice. However, its role in the brain remains poorly understood. To investigate the role of CTRP6 in cerebral ischemia/reperfusion injury associated with diabetes mellitus, a diabetes mellitus mouse model of cerebral ischemia/reperfusion injury was established by occlusion of the middle cerebral artery. To overexpress CTRP6 in the brain, an adeno-associated virus carrying CTRP6 was injected into the lateral ventricle. The result was that oxygen injury and inflammation in brain tissue were clearly attenuated, and the number of neurons was greatly reduced. In vitro experiments showed that CTRP6 knockout exacerbated oxidative damage, inflammatory reaction, and apoptosis in cerebral cortical neurons in high glucose hypoxia-simulated diabetic cerebral ischemia/reperfusion injury. CTRP6 overexpression enhanced the sirtuin-1 signaling pathway in diabetic brains after ischemia/reperfusion injury. To investigate the mechanism underlying these effects, we examined mice with depletion of brain tissue-specific sirtuin-1. CTRP6-like protection was achieved by activating the sirtuin-1 signaling pathway. | 13 | Microstructure, mechanical properties and magnetic properties of FeCoNiCuTiSi_x high-entropy alloys显示文摘The microstructure, mechanical properties and magnetic properties of FeCoNiCuTiSi_x(x=0, 0.05, 0.1, 0.2, 0.4 and 0.5) highentropy alloys were studied in detail in the present paper. The crystal structure changes from FCC+Laves to FCC+BCC+Laves phases, as the Si content increases. Cu is more segregated with the increase in Si content, which can be explained from a thermodynamic point of view. The FeCoNiCuTi high-entropy alloy has good compression mechanical properties, and the fracture strength, fracture strain, and Vickers hardness are 1584.2 MPa, 3.3% and 483.2 HV, respectively. However, the plasticity of the alloy decreases slightly as the Si content increases. | 14 | Production evolution patterns and development stage division of waterflooding oilfields显示文摘The continuous growth of recoverable reserves in a waterflooding oilfield has a significant impact on the patterns of production evolution. A new production evolution model is established by improving the Weng Cycle model. With the new model, the statistical correspondence between the production decline stage and the reserve-production imbalance is clarified,and the correlation of water cut with the recovery percent of recoverable reserves is discussed, providing quantitative basis of reservoir engineering for dividing development stages of oilfield and defining mature oilfields. According to the statistics of oilfields in eastern China, the time point corresponding to the reserve-production balance coefficient dropping to less than 1dramatically is well correlated the beginning point of production decline, thus the time when the reserve-production balance coefficient drops dramatically can be taken as the initiation point of production decline stage. The research results show that the water cut and the recovery percent of recoverable reserves have a good statistical match in the high water cut stage, and it is more rational to take both the start point of production decline stage and the water cut of 90%(or the recovery percent of recoverable reserves of 80%) as the critical criteria for defining a mature oilfield.
