1Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli,but are also actively engaged in inflammation and immunity,whereas the intrinsic regulatory mechanism is poorly understood.Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans,which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation,which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain,mitigated the imbalance in downstream SHP-1-pAKT signaling,and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance.Thus,our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects.Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang 2022Cell Research2022,32,5:3
2Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients.We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment.We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients.The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone.The cure rate,changes of oxygen saturation and interference,and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group,and 32 patients to the control group.The cure rate in the tocilizumab group was higher than that in the control group,but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions,the hypoxia ameliorated earlier after tocilizumab treatment,and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative.For patients with bilateral pulmonary lesions and elevated IL-6 levels,tocilizumab could be recommended to improve outcome.Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu 2021Frontiers of Medicine2021,15,3:2
3Mechanical properties and deformation mechanisms of a novel fine-grained Mg-Gd-Y-Ag-Zr-Ce alloy with high strength-ductility synergy显示文摘Grain boundary precipitation and segregation play an important role in determining mechanical properties of Mg alloys. In the present work, we studied work focuses on the strengthening and deformation mechanism of coarse-grained(CG) and fine-grained(FG) Mg-Gd-Y-Ag-Zr-Ce alloy. The CG alloy is strengthened by means of age-strengthening with the formation of both basal plate γ' and prismatic plate β’ precipitates in the grain interior. While the strengthening of FC alloy is completed by intergranular alloying segregation and intragranular precipitates γ' and β’. The segregation of alloying elements at the grain boundary and formation of sub-micron particles can stabilize the grain boundary and suppress the intergranular deformation. Consequently, dislocations could be trapped near γ' and β’ precipitates in the grain interior. Unlike CG alloys, the FG alloys exhibit a heterogeneous transition from elastic to plastic deformation via the Lüders plateau. The rapid gliding dislocation multiplications and fine-grained size are necessary and sufficient conditions for the Lüders strains. 5Distribution of the indigenous microorganisms and mechanisms of their orientational activation in Daqing Oilfield显示文摘The distribution of indigenous microorganisms was surveyed in Block 1 of Daqing Oilfield. Based on this survey,the indigenous microorganisms in the formation water were activated with different activator systems at the simulated stratum ecological environment. The changes of the number of bacteria of various physiological groups were determined during the process of activation. Also changes of pH value and composition of gas productions were analyzed at the end of culturing. The results showed that the selected block formation water contained a great number of saprophytic bacteria,hydrocarbon-oxidizing bacteria,fermentative bacteria,methane-producing bacteria and sulfate-reducing bacteria. Under the conditions that the growth of sulfate-reducing bacteria was controlled the block had the potential to enhance oil recovery by activating beneficial bacteria. The growth of sulfate-reducing bacteria can be inhibited through the activation of nitrate-reducing bacteria. The number of nitratereducing bacteria reached 106―107 cells/mL,but sulfate-reducing bacteria reached only 0―45 cells/mL in A system. Methane-producing bacteria can be activated by C,D activators. The relative content of biological methane in the light hydrocarbon gas reached 80% in C,D systems. B activator was conducive to the propagation of acid-producing bacteria,so that the pH value of the culture medium decreased from 7.5 to around 5.0. Hydrocarbon-oxidizing bacteria can be activated by various activator systems. There was low molecular light hydrocarbon in gas production according to the analysis of gas chromatograph. According to the content of methane and the number of methane-producing bacteria,methane only can be generated through activating methane-producing bacteria. 6Heterogeneous fiberous structured Mg-Zn-Zr alloy with superior strength-ductility synergy显示文摘Here we reported a heterogeneous fiberous structured Mg-5.6Zn-0.6Zr(wt%)alloy obtained by conventional extrusion method,which exhibited high yield strength of∼345 MPa,ultimate tensile strength of∼370 MPa,and high tensile strain of∼20.5%,superior to most of the Mg-Zn based alloys reported so far.The extraordinarily high mechanical properties were mainly attributed to the heterogeneous fiberous structure consisting of alternating coarse-and fine-grain layers.Grains in the different layers grew into the neighboring layers,ensuring a good layer bonding.A high Schmid factor and geometric compatibility factor for pyramidal slip led to full slip transfer between the neighboring coarse grains and fine grains,which could help to release the stress concentration and avoid early fracture.The profuse acti-vatedglide dislocations could render the unprecedented high tensile strain.The constraint by the hard fine-grain domains made the soft coarse-grain domains strong like the hard fine-grain domains,as well as the nanoscale precipitates pinning dislocations,contributed to the high strength.The hetero-geneous microstructure design was shown to have synergistic improvement in strength-ductility balance,which could be an inspiring strategy to improve mechanical properties of hexagonal close-packed(hcp)metals.Wei Fu Pengfei Dang Shengwu Guo Zijun Ren Daqing Fang Xiangdong Ding Jun Sun 2023Journal of Materials Science & Technology2023,,3:1
7Enhancing corrosion resistance of ZK60 magnesium alloys via Ca microalloying: The impact of nanoscale precipitates显示文摘Enhancing corrosion resistance of Mg-Zn alloys with high strength and low cost was critical for broadening their large-scale practical applications. Here we prepared solutionized, peak-and over-aged ZK60 alloys with and without microalloying Ca(0.26 wt.%) to explore the effects of nanoscale precipitates on their corrosion behavior in detail via experimental analyses and theoretical calculations. The results suggested the peak-aged ZK60 alloy with Ca addition showed improved corrosion resistance in comparison with the alloys without Ca,owing to the contribution of Ca on the refinement of precipitates and increase in their number density. Although the precipitates and Mg matrix formed micro-galvanic couples leading to dissolution, the fine and dense precipitates could generate “in-situ pinning” effect on the corrosion products, forming a spider-web-like structure and improving the corrosion inhibition ability accordingly. The pinning effect was closely related to the size and number density of precipitates. 10Knowledge-based System Prototype in Structural Component Design Based on FM显示文摘A knowledge-based system in structural component design based on fracture mechanics is developed in this paper. The system consists of several functional parts: a general inference engine, a set of knowledge bases and data-bases, an interpretation engine, a bases administration system and the interface. It can simulate a human expert to make analysis and design scheme mainly for four kinds of typical structural components widely used in shipbuilding industry: pressure vessels, huge rotation constructions, pump-rod and welded structures. It is an open system which may be broadened and perfected to cover a wider range of engineering application through the modification and enlargement of knowledge bases and data-bases. 11Comparing mineral weathering and elemental transport between earth's critical zone with different parent rocks in Yanshan Mountain,Hebei province,China显示文摘Bedrock weathering performs a significant influence on the evolution of Earth’s critical zone.Carbonate rock(dolostone),metamorphic rock(gneiss),and sedimentary rock(sandstone)geological formations in Yanshan Mountain,Hebei Province,are taken as objects to probe the controlling of geological formations on weathering characteristics,migration,and enrichment of elements as well as structure of Earth’s critical zone under the identical climate conditions through geological field survey,analysis on minerals component,element distribution in the weathering profile.The dolostone geological formation(DGF)is lithologically dominated by dolostone,characterized by the strongest and predominant chemical weathering.During bedrock weathering and pedogenesis,DGF is marked by significant depletion of CaO,Mg O,S,Mn,Mo and enrichment of N,K,Fe_(2)O_(3),and Zn with concentrations of P,Cu,and B keeping stable.Shortage of soil-forming materials and significant loss are driven by soil erosion,which results in thin regolith and soil.The soil thickness is less than 10 cm,and the regolith thickness is less than 30 cm.The vegetation community is predominantly rock arbor or brush,which is calcivorous and tolerant of barrenness.Plagiogneiss is a dominant rock type of gneiss geological formation(GGF),characterized by the weakest weathering and fast chemical and physical weathering rate.GGF is masked by significant depletion of P,K,CaO,MgO,Fe_(2)O_(3),Mn,Cu and enrichment of N,S,Mo,and B,with contents of Zn keeping stable.Both soil and regolith developed in GGF are relatively thick for one of the reasons that biotite expands during weathering.The soil thickness is more than 50 cm,and the regolith thickness ranges from 100 to 200 cm.The vegetation community is predominantly high-quality economic forests and various arbors because of the enrichment of nutrients in GGF.Sandstone is primarily a rock type of sandstone geological formation(SGF),characterized by moderate weathering degree and slow chemical weathering rate.SGF is marked by significant depletion of P,K,CaO,MgO,Fe_(2)O_(3),and enrichment of N,S,Mn,Cu,Zn,and Mo,with fluctuant changes of Zn and B.The thickness of soil developed in SGF varies between that of DGF and GGF.The soil thickness ranges from 30 to 50 cm,and the regolith thickness ranges from 50 to 100 cm.Chinese pines are widely spread on the shady slopes of SGF.Research provides theoretical support for screening dominant ecological resource areas,ecological industry development and ecological protection and restoration for Yanshan Mountain,Hebei Province.Zijian Sun Zhen Liao Wei Shen Daqing Fu Xiaofeng Wei Huiqiong Zhang Ziran Chen Lianghui Xiong Tianhao An Hao Wei 2022Acta Geochimica2022,41,6:0
12Reliable assessment approach of landslide susceptibility in broad areas based on optimal slope units and negative samples involving priori knowledge显示文摘Reliable assessment of landslide susceptibility in broad areas of terrain remains challenging due to complex topography and poor representation of randomly selected negative samples.Assessment in broad areas is now primarily based on grid units,which do not have a clear physical meaning like slope units,and their accuracy is not ideal.Nevertheless,the large amount of manual editing,due to the incorrectly generated horizontal and vertical lines during slope unit partitioning,limits using slope units for rapid assessment over large areas.Hence,this paper proposes a reliable susceptibility assessment approach to solve this problem based on optimal slope units and negative samples involving prior knowledge.Precisely,an algorithm to automatically extract slope units is designed to eliminate fragmented and erroneous units.Second,a samples labeling index(SLI)is defined based on the certainty factors model to select negative samples reasonably.Sichuan Province,China is selected for experimental analysis,with the results demonstrate that the optimized slope unit and the negative samples selection strategy consider prior knowledge achieve better results in the random forest model,support vector machine model,and artificial neural network model.In particular,the composite performance index AUC of artificial neural network model improved from 0.81 to 0.90.Xiao Fu Yuefan Liu Qing Zhu Daqing Ge Yun Li Haowei Zeng 2022International Journal of Digital Earth2022,15,1:0
13Microstructure and yield phenomenon of an extruded Mg-Y-Cu alloy with LPSO phase显示文摘A yield phenomenon was firstly reported in an extruded Mg-6.8Y-2.5Cu alloy and the corresponding microstructure was also investigated in this work,The cast alloy is mainly composed ofα-Mg,18R long period stacking order(LPSO)phase,eutectic phase(Mg_(20)Cu_(4)Y_(1)),and Mg_(2)Cu phase.The 18R LPSO phase at the dendritic grain boundary transforms into the 14H LPSO phase in the grain interior during homogenization.After extrusion,the grain size of the homogenized alloy is remarkably refined to-3.69μm and the second phase is significantly broken and distributed in the extrusion direction.Tensile testing curves of the extrude alloy at room temperature indicate that the yield strength and ultimate tensile strength increase while the elongation of the alloy decreases with increasing strain rate.Interestingly,a yield plateau fo rms and gradually decreases with increasing strain rate.The yield phenomenon is related to the dislocation multiplication and the interaction between the movable dislocations and solute atoms.Guangli Bi Niuming Zhang Jing Jiang Yuandong Li Tijun Chen Wei Fu Xiaoru Zhang Daqing Fang Xiangdong Ding 2023Journal of Rare Earths2023,41,3:0

15Analysis of the Value of Elojumab in Patients with Acute Coronary Syndrome after Percutaneous Coronary Intervention显示文摘Objective:To analyze the application value of Elouzmab in patients with acute coronary syndrome(AcS)after percutaneous coronary intervention(PCI),so as to lay a foundation for the follow-up treatment.Merthods:84 ACS patients who underwent PCI inour hospital from December 1,2018 to December 1,2019 were selected and divided into control group(n=42)and study group(n=42)according to the random number table.The control group was treated with statins,and the study group was treated with alloxan combined therapy.The changes of blood lipid index,quality of life score,adverse cardiovascular and cerebrovascular events and adverse reactions were compared before and after treatment.Results:There was no significant difference in TCHO,TG,HDL-C and LDL-Cbetween the two groups before treatment(P>0.05);After treatment,the levels of TCHO,TG,HDL-Cand LDL-C in the study group were significantly lower than those in the control group(P<0.05);There was no significant difference im the scores of WHOQOL BREF before treatment(P>0.05);After treatment,the WHOQOL-BREF scores of the two groups were improved,and the study group was significantly higher than the control group(P<0.05);The incidence of adverse cardiovascular and cerebrovascular events and adverse reactions in the study group was lower than that in the control group,but the difference was not statistically sigmificant(P>0.05).Conclusion:After percutaneous coronary intervention in patients with acute coronary syndrome,the use of Elojumab can effectively reduice the blood lipid index,improve the quality df patients and reduce the incidence of adverse cardiovascular and cerebrovascular events and adverse reactions,which can be effectively promoted in clinical practice.Baojun Fu Daqing Song Shuyin Sun 2020Journal of Clinical and Nursing Research2020,4,4:0
