1Effect of Arctium lappa L.in the dextran sulfate sodium colitis mouse model显示文摘AIM:To analyze the possible protective role of Arctium lappa L.(AL)in a murine model of ulcerative colitis(UC).METHODS:BALB/c mice were administered 100 mg/kg AL powder orally each day.After 7 d,colitis was induced by administration of dextran sulfate sodium(DSS)(5% W/V)in drinking water for a further 8 consecutive days.Diarrhea and bloody stools as well as colonic histology were observed.The level of interleukin-6(IL-6)and tu-mor necrosis factor-α(TNF-α)in colonic sections were detected by immunohistochemistry.RESULTS:There were significant differences in mean body weight values and disease activity indices between controls and AL-treated animals.Moreover,the histological findings showed that AL treatment can prevent mucosal edema,submucosal erosions,ulceration,inflammatory cell infiltration and colon damage.In addition,immunohistochemistry analysis showed that the levels of the inflammatory cytokines,IL-6 and TNF-α were also decreased in AL-treated groups.CONCLUSION:We suggest that AL can prevent intestinal damage and decrease inflammatory cytokines in mice with DSS-induced colitis.Thus,AL could prove to be a useful food for UC.Tzou-Chi Huang Shinn-Shyong Tsai Li-Fang Liu Yu Lin Liu Hung-Jen Liu Kuo Pin Chuang 2010World Journal of Gastroenterology2010,16,33:10
2High degree of pharmacokinetic compatibility exists between the five-herb medicine XueBiJing and antibiotics comedicated in sepsis care显示文摘Managing the dysregulated host response to infection remains a major challenge in sepsis care. Chinese treatment guideline recommends adding Xue Bi Jing, a five-herb medicine, to antibioticbased sepsis care. Although adding Xue Bi Jing further reduced 28-day mortality via modulating the host response, pharmacokinetic herbedrug interaction is a widely recognized issue that needs to be studied.Building on our earlier systematic chemical and human pharmacokinetic investigations of Xue Bi Jing, we evaluated the degree of pharmacokinetic compatibility for Xue Bi Jing/antibiotic combination based on mechanistic evidence of interaction risk. Considering both Xue Bi Jing-antibiotic and antibiotic-Xue Bi Jing interaction potential, we integrated informatics-based approach with experimental approach and developed a compound pair-based method for data processing. To reflect clinical reality, we selected for study Xue Bi Jing compounds bioavailable for drug interactions and 45 antibiotics commonly used in sepsis care in China. Based on the data of interacting with drug metabolizing enzymes and transporters, no Xue Bi Jing compound could pair, as perpetrator, with the antibiotics. Although some antibiotics could,due to their inhibition of uridine 50-diphosphoglucuronosyltransferase 2 B15, organic anion transporters1/2 and/or organic anion-transporting polypeptide 1 B3, pair with senkyunolide I, tanshinol and salvianolic acid B, the potential interactions(resulting in increased exposure) are likely desirable due to these Xue Bi Jing compounds’ low baseline exposure levels. Inhibition of aldehyde dehydrogenase by 7 antibiotics probably results in undesirable reduction of exposure to protocatechuic acid from Xue Bi Jing.Collectively, Xue Bi Jing/antibiotic combination exhibited a high degree of pharmacokinetic compatibility at clinically relevant doses. 5Effect of Post-weld Heat Treatment on Properties of Friction Welded Joint Between TC4 Titanium Alloy and 40Cr Steel Rods显示文摘Dissimilar metal joining of Ti–6Al–4V(TC4) titanium alloy to as-rolled 40 Cr steel rods was conducted with friction welding, and the effect of post-weld heat treatment(PWHT) on the microstructure and mechanical properties of the resultant joints was investigated. The average tensile strength of the as-welded joints reached 766 MPa and failure occurred in 40 Cr steel base metal. However, after PWHT at 600 °C for 0.5, 1, 2 and 3 h, the tensile strength of the joints decreased and fracture happened through the interface with quasi-cleavage features. The bending angle of specimens was improved from 9.6° in as-welded state to 32.5° after PWHT for 2 h. The tensile strength of the joint was enhanced by martensitic transformation near the interface in as-welded state. Sorbite formed near the interface in PWHT state and improved the bending ductility of the joint. Ti C brittle phase formed at the interface after PWHT for 0.5 h and deteriorated the tensile strength and bending ductility of the joint. After PWHT for 2 h, no Ti C phase was detected at the interface. 6Effect of long-term weightlessness on retina and optic nerve in tail-suspension rats显示文摘AIM: To evaluate the effect of long-term weightlessness on retina and optic nerve in tail-suspension(TS) rats.METHODS: A stimulated weightlessness model was established by suspending rats' tail. After 12 wk, the ultrastructure and the number of optic nerve axons were observed by transmission electron microscope. The number of survival retinal ganglion cells(RGCs) was calculated by fluorescent gold retrograde labeling. Retina cells apoptosis was detected by TUNEL staining. The function of optic nerve and retina was evaluated by the visual evoked potential(VEP) and oscillatory potentials(Ops).RESULTS: The optic nerve axons were swollen and sparsely aligned, and the lamellar separation and myelin disintegration occurred after 12 wk in TS rats. 7Bioinspired hierarchical helical nanocomposite macrofibers based on bacterial cellulose nanofibers显示文摘Bio-sourced nanocellulosic materials are promising candidates for spinning high-performance sustainable macrofibers for advanced applications.Various strategies have been pursued to gain nanocellulose-based macrofibers with improved strength.However,nearly all of them have been achieved at the expense of their elongation and toughness.Inspired by the widely existed hierarchical helical and nanocomposite structural features in biosynthesized fibers exhibiting exceptional combinations of strength and toughness,we report a design strategy to make nanocellulose-based macrofibers with similar characteristics.By combining a facile wet-spinning process with a subsequent multiple wet-twisting procedure,we successfully obtain biomimetic hierarchical helical nanocomposite macrofibers based on bacterial cellulose nanofibers,realizing impressive improvement in their tensile strength,elongation and toughness simultaneously.The achievement certifies the validity of the bioinspired hierarchical helical and nanocomposite structural design proposed here.This bioinspired design strategy provides a potential platform for further optimizing or creating many more strong and tough nanocomposite fiber materials for diverse applications.Huai-Ling Gao Ran Zhao Chen Cui Yin-Bo Zhu Si-Ming Chen Zhao Pan Yu-Feng Meng Shao-Meng Wen Chuang Liu Heng-An Wu Shu-Hong Yu 2020National Science Review2020,7,1:4
8Co-infection of SENV-D among chronic hepatitis C patients treated with combination therapy with high-dose interferon-alfa and ribavirin显示文摘AIM: The clinical significance of co-infection of SENV-D among patients with chronic hepatitis C (CHC) and response of both viruses to combination therapy with high-dose interferon-alfa (IFN) plus ribavirin remain uncertain and are being investigated.METHODS: Total 164 (97 males and 67 females, the mean age 48.1±11.4 years, range: 20-73 years, 128histologically proved) naive CHC patients were enrolled in this study. SENV-D DNA was tested by PCR method.Detection of serum HCV RNA was performed using a standardized automated qualitative RT-PCR assay (COBAS AMPLICOR HCV Test, version 2.0). HCV genotypes 1a,1b, 2a, 2b, and 3a were determined by using genotypespecific primers. Pretreatment HCV RNA levels were determined by using the branched DNA assay (Quantiplex HCV RNA 3.0). There are 156 patients receiving combination therapy with IFN 6 MU plus ribavirin for 24 wk and the response to therapy is determined.RESULTS: Sixty-one (37.2%) patients were positive for SENV-D DNA and had higher mean age than those who were negative (50.7±10.6 years vs46.6±11.6 years,P = 0.026). The rate of sustained viral response (SVR)for HCV and SENV-D were 67.3% (105/156) and 56.3%(27/48), respectively. By univariate analysis, the higher rate of SVR was significantly related to HCV genotype non-1b (P<0.001), younger ages (P = 0.014), lower pretreatment levels of HCV RNA (P = 0.019) and higher histological activity index (HAI) score for intralobular regeneration and focal necrosis (P = 0.037). By multivariate analyses, HCV genotype non-1b, younger age and lower pretreatment HCV RNA levels were significantly associated with HCV SVR (odds ratio (OR)/95% confidence interval (CI): 12.098/0.02-0.19, 0.936/0.890-0.998, and 3.131/1.080-9.077, respectively). The SVR of SENV-D was higher among patients clearing SENV-D than those who had viremia at the end of therapy (P = 0.04).CONCLUSION: Coexistent SENV-D infection, apparently associated with higher ages, is found in more than onethird Taiwan Residents CHC patients. Both HCV and SENV-D are highly susceptible to combination therapy with high-dose IFN and ribavirin and SENV-D co-infection does not affect the HCV response. 9Differences in pathological changes between two rat models of severe traumatic brain injury显示文摘The rat high-impact free weight drop model mimics the diffuse axonal injury caused by severe traumatic brain injury in humans,while severe controlled cortical impact can produce a severe traumatic brain injury model using precise strike parameters.In this study,we compare the pathological mechanisms and pathological changes between two rat severe brain injury models to identify the similarities and differences.The severe controlled cortical impact model was produced by an electronic controlled cortical impact device,while the severe free weight drop model was produced by dropping a 500 g free weight from a height of 1.8 m through a plastic tube.Body temperature and mortality were recorded,and neurological deficits were assessed with the modified neurological severity score.Brain edema and bloodbrain barrier damage were evaluated by assessing brain water content and Evans blue extravasation.In addition,a cytokine array kit was used to detect inflammatory cytokines.Neuronal apoptosis in the brain and brainstem was quantified by immunofluorescence staining.Both the severe controlled cortical impact and severe free weight drop models exhibited significant neurological impairments and body temperature fluctuations.More severe motor dysfunction was observed in the severe controlled cortical impact model,while more severe cognitive dysfunction was observed in the severe free weight drop model.Brain edema,inflammatory cytokine changes and cortical neuronal apoptosis were more substantial and blood-brain barrier damage was more focal in the severe controlled cortical impact group compared with the severe free weight drop group.The severe free weight drop model presented with more significant apoptosis in the brainstem and diffused blood-brain barrier damage,with higher mortality and lower repeatability compared with the severe controlled cortical impact group.Severe brainstem damage was not found in the severe controlled cortical impact model.These results indicate that the severe controlled cortical impact model is relatively more stable,more reproducible,and shows obvious cerebral pathological changes at an earlier stage.Therefore,the severe controlled cortical impact model is likely more suitable for studies on severe focal traumatic brain injury,while the severe free weight drop model may be more apt for studies on diffuse axonal injury.All experimental procedures were approved by the Ethics Committee of Animal Experiments of Tianjin Medical University,China(approval No.IRB2012-028-02)in Febru ary 2012.Yi-Ming Song Yu Qian Wan-Qiang Su Xuan-Hui Liu Jin-Hao Huang Zhi-Tao Gong Hong-Liang Luo Chuang Gao Rong-Cai Jiang 2019Neural Regeneration Research2019,14,10:4

15Dynamic Analysis of Semi-Submersible Production Platform Under the Failure of Mooring Lines显示文摘This paper quantitatively studies the transient dynamic response of a semi-submersible production platform with the loss of one or several positioning mooring lines.A semi-submersible platform,production risers,and positioning mooring lines are all included in the numerical simulation.Increased motion of the semi-submersible platform,tension variation of the remaining mooring lines/risers and the risk of mooring line or riser clashing are all investigated through fully coupled time-domain analysis.Combined environmental loads are selected from irregular waves and the steady current varying from very rough to extreme sea conditions.Three dimension radiation/diffraction theories and Morison’s equation are applied to calculate first-order wave force and second-order mean drift force of floating semi-submersible platform.Nonlinear time-domain finite element methods are employed to analyze the behavior of mooring lines and risers.Results show that the failure of mooring lines seriously reduce the platform’s stability performance.The tension of the rest lines is also increased accordingly.Remaining lines which are closer to the failed lines will have larger tension increase to compensate.Line-Line distance provides practical information for the risk of clashing investigation.CHUANG Zhen-ju LIU She-wen LI Chun-zheng LU Yu CHANG Xin 2021China Ocean Engineering2021,35,1:3
16Improvement of stability and solid-state battery performances of annealed 70Li_(2)S–30P_(2)S_(5) electrolytes by additives显示文摘The replacement of liquid electrolyte with solid electrolyte can significantly improve the safety and power/energy density of lithium batteries.70Li_(2)S–30P_(2)S_(5) is one of the most promising solid electrolytes with high conductivity for solid–state batteries.In this work,the ionic conductivity and stability toward moisture and lithium metal of 70Li_(2)S–30P_(2)S_(5) were enhanced by introducing the different amounts of Li_(2)O additives.65Li_(2)S–30P_(2)S_(5)–1%Li_(2)O delivered the highest conductivity,while 65Li_(2)S–30P_(2)S_(5)–5%Li_(2)O showed the best moisture stability and improved lithium compatibility.Solid-state batteries using 65Li_(2)S–30P_(2)S_(5)–5%Li_(2)O electrolyte and high-voltage LiNi_(0.6)Mn_(0.2)Co_(0.2)O_(2) cathode exhibited low initial discharge capacity(100 mAh·g^(-1))and Coulombic efficiency(69%).Li_(3)InCl_(6) electrolytes were introduced both in the cathode mixture to replace sulfide electrolyte and in the interface layer to improve the cathode compatibility for the solid-state batteries,showing enhanced discharge capacity(175 mAh·g^(-1))and improved initial Coulombic efficiency(86%).Moreover,it also exhibited good performance at-20℃.Hao-Tian Ren Zi-Qi Zhang Jun-Zhao Zhang Lin-Feng Peng Zhen-Yuan He Ming Yu Chuang Yu Long Zhang Jia Xie Shi-Jie Cheng 2022Rare Metals2022,41,1:3
17Simultaneous evaluation of two branches of a multifunctional integrated optic chip with an ultra-simple dual-channel configuration显示文摘We propose an ultra-simple dual-channel configuration for simultaneously evaluating two branches of a multifunctional integrated optic chip(MFIOC). In the configuration, the MFIOC is employed as a beam splitter to construct the demodulation interferometer together with a 2 × 2 fiber coupler. Interference happens between polarization modes traveling through different channels of the MFIOC. The cross-couplings of each channel are respectively characterized by the interference peaks which distribute on opposite sides of the central interference peak. Temperature responses of the MFIOC are experimentally measured from-40°C to 80°C. Results show that the proposed configuration can achieve simultaneous dual-channel transient measurements with resolution of-90 d B and dynamic range of 90 d B. 18Investigation of Li-ion transport in Li7P3S11 and solid-state lithium batteries显示文摘The high Li-ion conductivity of the Li7P3S11 sulfide-based solid electrolyte makes it a promising candidate for all-solid-state lithium batteries. The Li-ion transport over electrode-electrolyte and electrolyteelectrolyte interfaces, vital for the performance of solid-state batteries, is investigated by impedance spectroscopy and solid-state NMR experiments. An all-solid-state Li-ion battery is assembled with the Li7P3S11 electrolyte, nano-Li2S cathode and Li-In foil anode, showing a relatively large initial discharge capacity of 1139.5 m Ah/g at a current density of 0.064 m A/cm^ 2 retaining 850.0 m Ah/g after 30 cycles. Electrochemical impedance spectroscopy suggests that the decrease in capacity over cycling is due to the increased interfacial resistance between the electrode and the electrolyte. 1D exchange ^7Li NMR quantifies the interfacial Li-ion transport between the uncycled electrode and the electrolyte, resulting in a diffusion coefficient of 1.70(3) ×10^-14cm^2/s at 333 K and an energy barrier of 0.132 e V for the Li-ion transport between Li2S cathode and Li7P3S11 electrolyte. This indicates that the barrier for Li-ion transport over the electrode-electrolyte interface is small. 20Nuciferine protects against high-fat diet-induced hepatic steatosis and insulin resistance via activating TFEB-mediated autophagy——lysosomal pathway显示文摘Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its treatment.Hyperactivation of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of NAFLD.Accordingly,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against NAFLD.The objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of action.Here we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent manner.Mechanistic investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin resistance.Our present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.Xiliang Du Chiara Di Malta Zhiyuan Fang Taiyu Shen Xiaodi Niu Meng Chen Bo Jin Hao Yu Lin Lei Wenwen Gao Yuxiang Song Zhe Wang Chuang Xu Zhijun Cao Guowen Liu Xinwei Li 2022Acta Pharmaceutica Sinica B2022,12,6:2
