1Biological effect of velvet antler polypeptides on neural stem cells from embryonic rat brain显示文摘Background Velvet antler polypeptides (VAPs), which are derived from the antler velvets, have been reported to maintain survival and promote growth and differentiation of neural cells and, especially the development of neural tissues This study was designed to explore the influence of VAPs on neural stem cells in vitro derived from embryonic rat brain Methods Neural stem cells derived from E12 14 rat brain were isolated, cultured, and expanded for 7 days until neural stem cell aggregations and neurospheres were generated The neurospheres were cultured under the condition of different concentration of VAPs followed by immunocytochemistry to detect the differentiation of neural stem cells Results VAPs could remarkablely promote differentiation of neural stem cells and most neural stem cells were induced to differentiate towards the direction of neurons under certain concentration of VAPs Conclusion Neural stem cells can be successfully induced into neurons by VAPs in vitro , which could provide a basis for regeneration of the nervousLULai-jin CHENLei MENGXiao-ting YANGFan ZHANGZhi-xin CHENDong 2005Chinese Medical Journal2005,,1:14
2VdPKS1 is required for melanin formation and virulence in a cotton wilt pathogen Verticillium dahliae显示文摘Verticillium dahliae is a soil-borne phytopathogenic fungus that causes vascular wilt disease in a broad range of hosts.This pathogen survives for many years in soil in the form of melanized microsclerotia.To investigate the melanin synthesis in V.dahliae,we identified a polyketide synthase gene in V.dahliae,namely VdPKS1.PKS1 is known to involve in the dihydroxynaphthalene melanin synthesis pathway in many fungi.We found that VdPKS1 was required for melanin formation but not for microsclerotial production in V dahliae.The VdPKS1 gene-disruption mutant (vdpksl) formed melanin-deficient albino microsclerotia,which did not affect the fungal colonization in host tissues but significantly reduced the disease severity.Gene transcription analysis in the wild-type and the vdpksl strains suggested that VdPKS1 gene-disruption influenced the expression of a series of genes involved in ethylene biosynthesis,microsclerotial formation and pathogenesis.Our results suggest that the VdPKS1-mediated melanin synthesis is important for virulence and developmental traits of V.dahliae.Ting Zhang Bosen Zhang Chenlei Hua Pei Meng Sheng Wang Zhirong Chen Yejuan Du Feng Gao Jiafeng Huang 2017Science China(Life Sciences)2017,60,8:14

5Expression of pathogenesis-related genes in cotton roots in response to Verticillium dahliae PAMP molecules显示文摘VerticiUium wilt disease becomes a major threat to many economically important crops.It is unclear whether and how plant immunity takes place during cotton-Verticillium interaction due to the lack of marker genes.Taking advantage of cotton(Gossypium hirsutum) genome,we discovered pathogenesis-related(PR) gene families,which have been widely used as markers of immune responses in plants.To profile the expression of G.hirsutum PR genes in the process of plant immunity,we treated cotton roots with two immunogenic peptides,flg22 and nlp20 known as pathogen-associated molecular patterns,as well as three VerticiUium dahliae-derived peptides,nlp20^(Vd2),nlp23^(Vd3),and nlp23^(Vd4) which are highly identical to nlp20.Quantitative real-time PCR results revealed that 14 G hirsutum PR gene(GhPR) families were induced or suppressed independently in response to flg22,nlp20,nlp20^(Vd2),nlp23^(Vd3),and nlp23^(Vd4).Most GhPR genes are expressed highest at 3 h post incubation of immunogenic peptides.Compared to flg22 and nlp20,nlp20^(Vd2) is more effective to trigger up-regulated expression of GhPR genes.Notably,both nlp23^(Vd3) and nlp23^(Vd4) are able to induce GhPR gene up-regulation,although they do not induce necrosis on cotton leaves.Thus,our results provide marker genes and new immunogenic peptides for further investigation of cotton-V.dahliae interaction.Xuan Du Sheng Wang Feng Gao Lisha Zhang Jian-Hua Zhao Hui-Shan Guo Chenlei Hua 2017Science China(Life Sciences)2017,60,8:6
6Nonuniform Three-dimensional Configuration Distributed SAR Signal Reconstruction Clutter Suppression显示文摘This paper deals with the problem of clutter suppression in spaceborne distributed synthetic aperture radar (D-SAR) with nonuniform three-dimensional (3D) configuration geometry. In order to make a breakthrough of the configuration limitation of the traditional space time adaptive processing (STAP) based on uniform array and improve the inhomogeneous clutter suppression performance, this paper considers signal reconstruction technique using array interpolation to process the D-SAR signal. An array interpolation signal reconstruction method based on pitching-partition is derived then a signal reconstruction 3D-STAP clutter suppression method applied to nonuniform 3D configuration is proposed. In particular, the proposed method is compared with conventional methods and the performance analysis is carried out based on simulations. 73D microgroove electrical impedance sensing to examine 3D cell cultures for antineoplastic drug assessment显示文摘In recent decades,three-dimensional(3D)cancer cell models have attracted increasing interest in the field of drug screening due to their significant advantages in more accurate simulations of heterogeneous tumor behavior in vivo compared to two-dimensional models.Furthermore,drug sensitivity testing based on 3D cancer cell models can provide more reliable in vivo efficacy prediction.The gold standard fluorescence staining is hard to achieve real-time and label-free viability monitoring in 3D cancer cell models.In this study,a microgroove impedance sensor(MGIS)was specially developed for the dynamic and noninvasive monitoring of 3D cell viability.3D cancer cells were trapped in microgrooves with gold electrodes on opposite walls for in situ impedance measurement.The change in the number of live cells caused inversely proportional changes to the impedance magnitude of the entire cell/Matrigel construct and reflected the proliferation and apoptosis of the 3D cells.It was confirmed that the 3D cell viability detected by the MGIS was highly consistent with the standard live/dead staining by confocal microscope characterization.Furthermore,the accuracy of the MGIS was validated quantitatively using a 3D lung cancer model and sophisticated drug sensitivity testing.In addition,the parameters of the MGIS in the measurement experiments were optimized in detail using simulations and experimental validation.The results demonstrated that the MGIS coupled with 3D cell culture would be a promising platform to improve the efficiency and accuracy of cell-based anticancer drug screening in vitro.Yuxiang Pan Deming Jiang Chenlei Gu Yong Qiu Hao Wan Ping Wang 2020Microsystems & Nanoengineering2020,6,1:2

11Silicon-based on-chip hybrid(de)multiplexers显示文摘A review is given on the recent progress of silicon-based on-chip hybrid multiplexers, which are the key elements to enable more than one(de)multiplexing techniques simultaneously, including wavelengthdivision-multiplexing(WDM), polarization-division-multiplexing(PDM), and mode-division-multiplexing(MDM). This helps enhance the link capacity of optical interconnects multiplexed with many channels.The first part gives a review on the recent developed silicon-based hybrid WDM-PDM(de)multiplexers enabling WDM and PDM simultaneously, which helps achieve 2 N channels by introducing N wavelengths and dual polarizations. The recent progress of silicon-based hybrid WDM-MDM(de)multiplexers developed is reviewed in the second part. With the hybrid WDM-MDM(de)multiplexers, one can achieve N × M channels by using N wavelengths and M guided-modes. 15Research on Leak Location Method of Water Supply Pipeline Based on MVMD显示文摘At present,the leakage rate of the water distribution network in China is still high,and the waste of water resources caused by water distribution network leakage is quite serious every year.Therefore,the location of pipeline leakage is of great significance for saving water resources and reducing economic losses.Acoustic emission technology is the most widely used pipeline leak location technology.The traditional non-stationary random signal de-noising method mainly relies on the estimation of noise parameters,ignoring periodic noise and components unrelated to pipeline leakage.Aiming at the above problems,this paper proposes a leak location method for water supply pipelines based on a multivariate variational mode decomposition algorithm.This method combines the two parameters of the energy loss coefficient and the correlation coefficient between adjacent modes,and adaptively determines the decomposition mode number K according to the characteristics of the signal itself.According to the correlation coefficient,the effective component is selected to reconstruct the signal and the cross-correlation time delay is estimated to determine the location of the pipeline leakage point.The experimental results show that this method has higher accuracy than the cross-correlation method based on VMD and the cross-correlation method based on EMD,and the average relative positioning error is less than 2.2%.Qiansheng Fang Haojie Wang Chenlei Xie Jie Chen 2023Computer Modeling in Engineering & Sciences2023,,2:1
