2The combined genotypes effect of ESR and FSHβ genes on litter size traits in five different pig breeds显示文摘Estrogen receptor (ESR) and Follicular-stimula-ting hormone beta subunit (FSHβ) genes were chosen as candidates to determine whether they control litter size and some other reproductive traits in swine. 269 sows from five different pig breeds were genotyped by an established PCR-RFLPs protocol at both ESR and FSHβ loci. The effects of both ESR and FSHβ on pig reproductive traits, including total number born (TNB) and number born alive (NBA), are analyzed by SAS software (version 6.12). These computation results demonstrated that both ESR locus and FSHβ locus are the major genes influencing litter size in pigs. The sows of BBBB combined genotype of ESR and FSHβ loci generally produce 1.85-3.01 TNB and 2.0-3.0 NBA more than those of ABAA combined genotype. The notable effect of ESR locus and FSHβ locus on litter size of pigs have made it possible to improve the pig reproduction by Marker-assisted selection (MAS). 3Cloned pigs derived from somatic cell nuclear transferembryos cultured in vitro atlow oxygen tension显示文摘Pig cloning has great potential to human xenotransplantation. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. 5Online Unsupervised Learning Classification of Pedestrian and Vehicle for Video Surveillance显示文摘This paper presents an online unsupervised learning classification of pedestrians and vehicles for video surveillance. Different from traditional methods depending on offline training, our method adopts the online label strategy based on temporal and morphological features,which saves time and labor to a large extent. It extract the moving objects with their features from the original video.An online filtering procedure is adopted to label the moving objects according to certain threshold of speed and area feature. The labeled objects are sent into a SVM classifier to generate the pedestrian & vehicle classifier. 6A transceptor-channel complex couples nitrate sensing to calcium signaling in Ambidopsis显示文摘Nitrate-induced Ca^(2+) signaling is crucial for the primary nitrate response in plants.However,the molecular mechanism underlying the generation of the nitrate-specific calcium signature remains unknown.We report here that a cyclic nucleotide-gated channel(CNGC)protein,CNGC15,and the nitrate transceptor(NRT1.1)constitute a molecular switch that controls calcium influx depending on nitrate levels.The expression of CNGC15 is induced by nitrate,and its protein is localized at the plasma membrane after establishment of young seedlings.We found that disruption of CNGC15 results in the loss of the nitrate-induced Ca^(2+) signature(primary nitrate response)and retards root growth,reminiscent of the phenotype observed in the nrt1.1 mutant.We further showed that CNGC15 is an active Ca^(2+)-permeable channel that physically interacts with the NRT1.1 protein in the plasma membrane.Importantly,we discovered that CNGC15-NRT1.1 interaction silences the channel activity of the heterocomplex,which dissociates upon a rise in nitrate levels,leading to reactivation of the CNGC15 channel.The dynamic interactions between CNGC15 and NRT1.1 therefore control the channel activity and Ca^(2+) influx in a nitrate-dependent manner.Our study reveals a new nutrient-sensing mechanism that utilizes a nutrient transceptor-channel complex assembly to couple nutrient status to a specific Ca^(2+) signature.Xiaohan Wang Changxin Feng LiLi Tian Congcong Hou Wang Tian Bin Hu Qian Zhang Zhijie Ren Qi Niu Jiali Song Dongdong Kong Liangyu Liu Yikun He Ligeng Ma Chengcai Chu Sheng Luan Legong Li 2021Molecular Plant2021,14,5:5

9Trade-offs and Synergies of Ecosystem Services in the Taihu Lake Ba- sin of China显示文摘Understanding the spatial interactions among multiple ecosystem services is crucial for ecosystem services management. Ecosystem services, including crop production, freshwater supply, aquatic production, net primary production, soil conservation, water conservation, flood regulation, forest recreation, were measured at 1-km grid scale covering the Taihu Lake Basin(TLB) of China. Our objective is to get a comprehensive understanding of the spatial distributions, trade-offs, synergies of multiple ecosystem services across the TLB. Our results found that: 1) majority of ecosystem services were clustered in space and had a similar spatial distribution pattern with the geographical resource endowment. Most of the landscape contributed a high supply of no services, one or two, and a low supply of three to seven services. 2) There were high correlation between forest recreation and freshwater supply and regulating services. Aquatic production had low correlation with other services. 3) The changes of provisioning services led to trade-offs between regulating services and cultural services in the TLB, while synergies mainly occurred among the provisioning service. 4) The spatial relationships of multiple services are consistent at 1-km spatial scale, counties and provinces. 10CNN-Based Intelligent Safety Surveillance in Green IoT Applications显示文摘Safety surveillance is considered one of the most important factors in many constructing industries for green internet of things(IoT)applications.However,traditional safety monitoring methods require a lot of labor source.In this paper,we propose intelligent safety surveillance(ISS)method using a convolutional neural network(CNN),which is an autosupervised method to detect workers whether or not wearing helmets.First,to train the CNN-based ISS model,the labeled datasets mainly come from two aspects:1)our labeled datasets with the full labeled on both helmet and pedestrian;2)public labeled datasets with the parts labeled either on the helmet or pedestrian.To fully take advantage of all datasets,we redesign CNN structure of network and loss functions based on YOLOv3.Then,we test our proposed ISS method based on the specific detection evaluation metrics.Finally,experimental results are given to show that our proposed ISS method enables the model to fully learn the labeled information from all datasets.When the threshold of intersection over union(IoU)between the predicted box and ground truth is set to 0.5,the average precision of pedestrians and helmets can reach 0.864 and 0.891,respectively.Wengang Cao Jianing Zhang Changxin Cai Quan Chen Yu Zhao Yimo Lou Wei Jiang Guan Gui 2021China Communications2021,18,1:4
11Integrative analysis of miRNA and mRNA profiles reveals that gga-miR-106-5p inhibits adipogenesis by targeting the KLF15 gene in chickens显示文摘Background:Excessive abdominal fat deposition in commercial broilers presents an obstacle to profitable meat quality,feed utilization,and reproduction.Abdominal fat deposition depends on the proliferation of preadipocytes and their maturation into adipocytes,which involves a cascade of regulatory molecules.Accumulating evidence has shown that microRNAs(miRNAs)serve as post-transcriptional regulators of adipogenic differentiation in mammals.However,the miRNA-mediated molecular mechanisms underlying abdominal fat deposition in chickens are still poorly understood.This study aimed to investigate the biological functions and regulatory mechanism of miRNAs in chicken abdominal adipogenesis.Results:We established a chicken model of abdominal adipocyte differentiation and analyzed miRNA and mRNA expression in abdominal adipocytes at different stages of differentiation(0,12,48,72,and 120 h).A total of 217 differentially expressed miRNAs(DE-miRNAs)and 3520 differentially expressed genes were identified.Target prediction of DE-miRNAs and functional enrichment analysis revealed that the differentially expressed targets were significantly enriched in lipid metabolism-related signaling pathways,including the PPAR signaling and MAPK signaling pathways.A candidate miRNA,gga-miR-106-5p,exhibited decreased expression during the proliferation and differentiation of abdominal preadipocytes and was downregulated in the abdominal adipose tissues of fat chickens compared to that of lean chickens.gga-miR-106-5p was found to inhibit the proliferation and adipogenic differentiation of chicken abdominal preadipocytes.A dual-luciferase reporter assay suggested that the KLF15 gene,which encodes a transcriptional factor,is a direct target of gga-miR-106-5p.gga-miR-106-5p suppressed the posttranscriptional activity of KLF15,which is an activator of abdominal preadipocyte proliferation and differentiation,as determined with gain-and loss-of-function experiments.Conclusions:gga-miR-106-5p functions as an inhibitor of abdominal adipogenesis by targeting the KLF15 gene in chickens.These findings not only improve our understanding of the specific functions of miRNAs in avian adipogenesis but also provide potential targets for the genetic improvement of excessive abdominal fat deposition in poultry.Weihua Tian Xin Hao Ruixue Nie Yao Ling Bo Zhang Hao Zhang Changxin Wu 2022Journal of Animal Science and Biotechnology2022,13,6:3
12An Active De-anonymizing Attack Against Tor Web Traffic显示文摘Tor is pervasively used to conceal target websites that users are visiting. A de-anonymization technique against Tor, referred to as website fingerprinting attack, aims to infer the websites accessed by Tor clients by passively analyzing the patterns of encrypted traffic at the Tor client side. However, HTTP pipeline and Tor circuit multiplexing techniques can affect the accuracy of the attack by mixing the traffic that carries web objects in a single TCP connection. In this paper, we propose a novel active website fingerprinting attack by identifying and delaying the HTTP requests at the first hop Tor node. Then, we can separate the traffic that carries distinct web objects to derive a more distinguishable traffic pattern. To fulfill this goal, two algorithms based on statistical analysis and objective function optimization are proposed to construct a general packet delay scheme. We evaluate our active attack against Tor in empirical experiments and obtain the highest accuracy of 98.64%, compared with 85.95% of passive attack. We also perform experiments in the open-world scenario. 13Single nucleotide polymorphism analysis of the encoding region of EX-FABP gene and its association with fattiness trait in chicken显示文摘Fattiness is an important parameter to estimate meat quality, which has high heritability. In this experiment, F2 chickens derived from Broilers crossing to Silky were used to study the effect of extracellular fatty acid inding protein (EX-FABP) gene on abdominal fat accumulation. Five exons and partial introns of the gene were amplified by six pairs of primers, and then single nucleotide polymorphisms (SNPs) were detected by single strand conformation polymorphism (SSCP) and subsequently confirmed by sequencing. There were four nucleotides variations found, T-C at 659, C-T at 924, CA-TG at 968 and C-T at 1985 respectively. The result of least square analysis suggests that the chickens with HH genotype defined by the third pair of primers have a lower abdominal fat weight and abdominal fat percentage than the chickens with other genotypes (GG, GI, GH and II). 14Crustal thickness and composition in the South China Block:Constraints from earthquake receiver function显示文摘Crustal thickness and composition are closely related to geology and tectonic evolution of the region.Studying the differences in the crustal thickness and composition of the South China Block(SCB)is important to gain a comprehensive understanding of multi-phase amalgamation,breakup,reworking,and regional geodynamic processes.In this study,teleseismic data from 135 high-density portable broadband stations from Sino Probe were processed using Common Conversion Point(CCP)stacking and H-κstacking methods.The Moho depth and P-wave and S-wave velocity ratio(V_(p)/V_(s))were studied.Our results revealed several insights about the tectonic processes in the SCB.First,the crustal structure and V_(p)/V_(s)ratios of the Cathaysia Block,Jiangnan Orogenic Belt,and Yangtze Block were significantly different.The average depth of the Moho in the Cathaysia Block was approximately 31 km,and the V_(p)/V_(s)ratios increased from the inland area(1.66)to the coastal area(1.78),indicating the oceanward increase of mafic proportion in the lower crust,which is related to the influence of the Paleo-Pacific Plate westward subduction.Second,the crustal thickness of Jiangnan Orogenic Belt deepens from east ca.31 to the west ca.42 km and the V_(p)/V_(s)ratios varied from 1.75 to 1.64,illustrating a relatively felsic crust,which could have been related to the Mesozoic upper crustal thickening under compression followed by the lower crust removal under the extensional background.Third,the average crustal thickness of the Yangtze Block was 42 km,and the V_(p)/V_(s)ratios ranged from 1.64 to 1.84,presenting a positive correlation between the Moho depth and the V_(p)/V_(s)ratio,which is explained by the relatively thick mafic lower crust.Based on the variations in the crustal structure and V_(p)/V_(s)ratios of the profile,we inferred that the central part of the Jiangnan Orogenic Belt was bounded by the Jiangshan-Shaoxing-Hengyang in the east and the Jiujiang-Shitai-Jishou in the west.The small-scale Moho depth undulations at the intersection of the Cathaysia Block and the Jiangnan Orogenic Belt could be related to the Mesozoic compression-extension geodynamic transformation.Changxin CHEN Qingtian LÜ Ling CHEN Danian SHI Jiayong YAN Yinshuang AI 2022Science China Earth Sciences2022,65,4:3
15Experimental Investigation of Nozzle Effects on Thrust and Inlet Pressure of an Air-breathing Pulse Detonation Engine显示文摘Nozzle effects on thrust and inlet pressure of a multi-cycle air-breathing pulse detonation engine (APDE) are investigated experimentally. An APDE with 68 mm in diameter and 2 050 mm in length is operated using gasoline/air mixture. Straight nozzle, converging nozzle, converging-diverging nozzle and diverging nozzle are tested. The results show that thrust augmentation of converging-diverging nozzle, diverging nozzle or straight nozzle is better than that of converging nozzle on the whole. Thrust augmentation of straight nozzle is worse than those of converging-diverging nozzle and diverging nozzle. Thrust augmentations of diverging nozzle with larger expansion ratio and converging-diverging nozzle with larger throat area range from 20% to 40% on tested frequencies and are better than those of congeneric other nozzles respectively. Nozzle effects on inlet pressure are also researched. At each frequency it is indicated that filling pressures and average peak pressures of inlet with diverging nozzle and converging-diverging nozzle with large throat cross section area are higher than those with straight nozzle and converging nozzle. 16A small scale expression screen identifies tissue specific markers in the Dugesia japonica strain Pek-1显示文摘Freshwater planaria has tremendous capacity to reform the missing part of the body and therefore is considered as one of the most important model organism for regeneration study. At present, Schmidtea mediterranea and Dugesia japonica are the two major species utilized for laboratory manipulations. Dugesia japonica flatworms are widely distributed in the Far East including Cherry Valley region in the northwest area of Beijing, China. We reported here the establishment of an asexual Dugesia japonica strain Pek1, as a suitable system for regeneration study. Using morphological, karyotypical as well as phylogenetic analyses, we confirmed that these flatworms indeed belonged to Dugesia japonica. We went on to show that the commonly used in situ probes and immunohistochemistry reagents and protocols were applicable to the Pek1 strain. Using this strain, we carried out small scale analysis on EST, RNAi and gene expression. We identified 193 unique EST sequences and 65 of them had not been reported in planarian. By RNAi analysis, we showed that 48 genes, when downregulated individually, had no effect on regeneration. Furthermore, we identified 3 groups of tissue specific expressing genes that were useful for cell lineage analysis. 17Effects of Dielectric Barrier Discharge Plasma on the Combustion Performances of Reverse-Flow Combustor in an Aero-Engine显示文摘In order to apply plasma assisted combustion(PAC) into a reverse-flow aero-engine and verify the improvement of combustion performance, a feasible approach was proposed in this work. In this approach, based on the structure characteristics of the reverse-flow combustor, a parallel plate double dielectric barrier discharge(DBD) PAC actuator was designed to generate plasma. It was installed at the front of combustor. When the actuator is driven, the original air flow is not disturbed for the device’s structure and installation. Using aviation kerosene as fuel, the effects of plasma on ignition boundary and outlet temperature of the combustor were experimentally investigated at atmosphere pressures. Through the dual high voltage differential power supply, the large gap, large area and uniform plasma discharge was achieved. The results of PAC actuator discharge indicate that inlet air temperature has a small increase of 4–9 K. After PAC is applied, the combustion performances of reverse-flow combustor in an aero-engine are remarkably improved. Experimental results indicate that ignition boundary is widened by 3.7%–12.5% because of the impact of plasma. Outlet highest temperature of combustor is raised by 19–75 K;outlet temperature distribution coefficient is reduced by 11.1%–26.6%. 20Chronological Video Synopsis via Events Rearrangement Optimization显示文摘Video synopsis aims at retrieving interested events and reducing human labor on browsing long surveillance video. Traditional video synopsis methods based on energy function minimization is high computational and time consuming. Besides, the unchronological tubes shifting strategy may cause chaotic temporality and uncomfortable collisions. We propose a spatiotemporal events rearrangement optimization algorithm,which formulates the events rearrangement problem as iteration judgment on trajectory correlation and events compactability to generate the output synopsis video. An events subsection modification strategy has been presented to solve the spatial incompleteness of certain single event caused by imperfect object tracking.
