1Side Effects during Treatment of Advanced Gastric Carcinoma by Chemotherapy Combined with CIK-cell Transfusion in Elderly People显示文摘OBJECTIVE To study the side effects and therapeutic results of autologous cytokine-induced killer(CIK)cell treatment in elderly patients with advanced gastric cancer. METHODS CIK cells were induced and cultured using biotechnics in vitro,and then the cells were infused back into the patients.Sixty elderly gastric cancer patients treated by chemotherapy(FOLFOX4 protocol)were followed-up.Among them,29 patients were treated with CIK cells during application of chemotherapy.Short-term curative effects and adverse events from the CIK transfusion and chemotherapy were observed. RESULTS Eight cases developed partial remission(PR),9 cases moderate remission(MR),7 cases stable disease(SD)and 5 cases progressive disease(PD).Out of a total of 29 patients who received chemotherapy combined with autologous CIK therapy, the total remission rate(PR+MR)was 58.6%.The total remission rate following chemotherapy alone was 45.2%,including 5 PR cases,9 MR cases,7 SD cases,and 10 PD cases.There was a relatively lower rate of severe chemotherapic toxicities in the CIK- cell transfusion group.Side e?ects of autologous CIK transfusion included chills(13 cases),fever(9 cases),nausea and vomiting (1 case)and general malaise(3 cases).Side effects were treated with conventional therapy resulting in their amelioration.No patients developed shock,blood capillary leakage syndrome,or abnormalities in routine blood,urine,liver and renal function tests. CONCLUSION Adoptive immunotherapy with autologous CIK cells may decrease the clinical signs and symptoms of elderly patients who suffer from advanced gastric cancer.Adverse reactions of patients can be alleviated by conventional therapy. 2Pertuzumab in combination with trastuzumab and chemotherapy for Chinese patients with HER2-positive metastatic gastric or gastroesophageal junction cancer:a subpopulation analysis of the JACOB trial显示文摘Background:The JACOB trial(NCT01774786)was a double-blinded,placebo-controlled,randomized,multicenter,international,phase III trial evaluating the efficacy and safety of adding pertuzumab to trastuzumab and chemo-therapy in first-line treatment of human epidermal growth factor receptor 2(HER2)-positive metastatic gastric cancer/gastroesophageal junction cancer(GEJC).The aim of this analysis was to investigate efficacy and safety outcomes in the Chinese subpopulation from the JACOB trial.Methods:This post hoc subpopulation analysis included all patients recruited in China's Mainland(n=163;20.9%)between June 2013 and January 2016.The patients were randomly assigned in a 1:1 ratio to receive pertuzumab plus trastuzumab and chemotherapy(pertuzumab group;n=82)or placebo plus trastuzumab and chemotherapy(con-trol group;n=81).Intravenous pertuzumab(840 mg)and trastuzumab(8 mg/kg loading and 6 mg/kg maintenance doses)were given every 3 weeks until disease progression or unacceptable toxicity.Chemotherapy was given as per standard regimens/doses of capecitabine or 5-fluorouracil plus cisplatin.The primary endpoint was overall survival(OS);secondary efficacy endpoints included progression-free survival(PFS),and overall objective response rate(ORR).Results:The median OS was 18.7 months in the pertuzumab group and 16.1 months in the control group(hazard ratio[HR]0.75;95%confidence interval[CI]0.49 to 1.14).The median PFS was 10.5 and 8.6 months in the pertuzumab and control groups,respectively(HR 0.85;95%CI 0.60 to 1.21),and the median ORRs were 68.9%and 55.7%,respectively.The treatment effect in this Chinese subpopulation showed consistency with that in the global ITT population with numerically lower HR for OS and PFS compared with the control group.The safety profiles of the pertuzumab and control groups in this Chinese subpopulation analysis were generally comparable.The most common grade 3-5adverse events were neutropenia,anemia,and leukopenia.However,due to the nature of being a post hoc subgroup analysis,the results presented here are descriptive only and need to be interpreted with caution.Conclusions:OS and PFS were numerically improved by adding pertuzumab to trastuzumab and chemotherapy as first-line treatment in Chinese HER2-positive gastric cancer/GEJC patients,and this regimen demonstrated an acceptable safety profile.Tianshu Liu Yanru Qin Jin Li Ruihua Xu Jianming Xu Shujun Yang Shukui Qin Yuxian Bai Changping Wu Yixiang Mao Haiyan Wu Yilin Ge Lin Shen 2019Cancer Communications2019,39,1:6
3N-acetyltransferase 10 promotes colon cancer progression by inhibiting ferroptosis through N4-acetylation and stabilization of ferroptosis suppressor protein 1 (FSP1) mRNA显示文摘Background:N-acetyltransferase 10(NAT10)is the only enzyme known tomediate the N4-acetylcytidine(ac4C)modification of mRNA and is crucial formRNA stability and translation efficiency.However,its role in cancer development and prognosis has not yet been explored.This study aimed to examine the possible role of NAT10 in colon cancer.Methods:The expression levels ofNAT10were evaluated by immunohistochemical analyses with a colon cancer tissue microarray,and its prognostic value in patients was further analyzed.Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blotting were performed to analyze NAT10 expression in harvested colon cancer tissues and cell lines.Stable NAT10-knockdown and NAT10-overexpressing colon cancer cell lines were constructed using lentivirus.The biological functions of NAT10 in colon cancer cell lines were analyzed in vitro by Cell Counting Kit-8(CCK-8),wound healing,Transwell,cell cycle,and ferroptosis assays.Xenograft models were used to analyze the effect of NAT10 on the tumorigenesis and metastasis of colon cancer cells in vivo.Dot blotting,acetylated RNA immunoprecipitation-qPCR,and RNA stability analyses were performed to explore the mechanism by which NAT10 functions in colon cancer progression.Results:NAT10 was upregulated in colon cancer tissues and various colon cancer cell lines.This increased NAT10 expression was associated with shorter patient survival.Knockdown of NAT10 in two colon cancer cell lines(HT-29 and LoVo)impaired the proliferation,migration,invasion,tumor formation and metastasis of these cells,whereas overexpression of NAT10 promoted these abilities.Further analysis revealed that NAT10 exerted a strong effect on the mRNA stability and expression of ferroptosis suppressor protein 1(FSP1)in HT-29 and LoVo cells.In these cells,FSP1 mRNA was found to be modified by ac4C acetylation,and this epigenetic modification was associated with the inhibition of ferroptosis.Conclusions:Our study revealed that NAT10 plays a critical role in colon cancer development by affecting FSP1 mRNA stability and ferroptosis,suggesting that NAT10 could be a novel prognostic and therapeutic target in colon cancer.Xiao Zheng Qi Wang You Zhou Dachuan Zhang Yiting Geng Wenwei Hu Changping Wu Yufang Shi Jingting Jiang 2022Cancer Communications2022,42,12:3
4Unveiling the mechanical response and accommodation mechanism of pre-rolled AZ31 magnesium alloy under high-speed impact loading显示文摘Split Hopkinson pressure bar(SHPB)tests were conducted on pre-rolled AZ31 magnesium alloy at 150–350℃ with strain rates of 2150s-1,3430s^(-1) and 4160s-1.The mechanical response,microstructural evolution and accommodation mechanism of the pre-rolled AZ31 magnesium alloy under high-speed impact loading were investigated.The twin and shear band are prevailing at low temperature,and the coexistence of twins and recrystallized grains is the dominant microstructure at medium temperature,while at high temperature,dynamic recrystallization(DRX)is almost complete.The increment of temperature reduces the critical condition difference between twinning and DRX,and the recrystallized temperature decreases with increasing strain rate.The mechanical response is related to the competition among the shear band strengthen,the twin strengthen and the fine grain strengthen and determined by the prevailing grain structure.The fine grain strengthen could compensate soften caused by the temperature increase and the reduction of twin and shear band.During high-speed deformation,different twin variants,introduced by pre-rolling,induce different deformation mechanism to accommodate plastic deformation and are in favor for non-basal slip.At low temperature,the high-speed deformation is achieved by twinning,dislocation slip and the following deformation shear band at different deformation stages.At high temperature,the high-speed deformation is realized by twinning and dislocation slip of early deformation stage,transition shear band of medium deformation stage and DRX of final deformation stage.Xiao Liu Hui Yang Biwu Zhu Yuanzhi Wu Wenhui Liu Changping Tang 2022Journal of Magnesium and Alloys2022,10,4:2

6Polyploidy events shaped the expansion of transcription factors in Cucurbitaceae and exploitation of genes for tendril development显示文摘Cucurbitaceae is one of the most important plant families distributed worldwide.Transcription factors(TFs)regulate plant growth at the transcription level.Here,we performed a systematic analysis of 42641 TFs from 63 families in 14 Cucurbitaceae and 10 non-cucurbit species.Whole-genome duplication(WGD)was the dominant event type in almost all Cucurbitaceae plants.The TF families were divided into 1210 orthogroups(OGs),of which,112 were unique to Cucurbitaceae.Although the loss of several gene families was detected in Cucurbitaceae,the gene families expanded in five species that experienced a WGD event comparing with grape.Our findings revealed that the recent WGD events that had occurred in Cucurbitaceae played important roles in the expansion of most TF families.The functional enrichment analysis of the genes that significantly expanded or contracted uncovered five gene families,AUX/IAA,NAC,NBS,HB,and NF-YB.Finally,we conducted a comprehensive analysis of the TCP gene family and identified 16 tendril-related(TEN)genes in 11 Cucurbitaceae species.Interestingly,the characteristic sequence changed from CNNFYFP to CNNFYLP in the TEN gene(Bhi06M000087)of Benincasa hispida.Furthermore,we identified a new characteristic sequence,YNN,which could be used for TEN gene exploitation in Cucurbitaceae.In conclusion,this study will serve as a reference for studying the relationship between gene family evolution and genome duplication.Moreover,it will provide rich genetic resources for functional Cucurbitaceae studies in the future.Yu Zhang Yingchao Zhang Bing Li Xiao Tan Changping Zhu Tong Wu Shuyan Feng Qihang Yang Shaoqin Shen Tong Yu Zhuo Liu Xiaoming Song 2022Horticultural Plant Journal2022,8,5:2

19Inhibitory Activities on α-Glucosidase and Composition Analysis of Different Fractions of Ethanol Extracts from Actinidia chinensis Radix显示文摘Tlie inhibitory actions of ethyl acetate,n-butanol and water fractions from Actinidia chinensis Radix (A. chinensis Radix) ethanol extracts on glucosidase were investigated. We found that the inhibitory activity of different ethanol extract fractions on α-glucosidase is concentration-dependent. At the same concentration, the ethyl acetate and n-butaiiol fractions showed higher inliibitory activities on gglucosidase in comparison witli acarbose. We then determined the chemical constituents of tlie ethyl acetate and n-butanol fractions through ultra performance liquid cliromatography-triple-time of flight/mass spectrometry(UPLC-Triple-TOF/MS). Based on the mass spectrometry analysis and the databases Scitinder and Reaxys, 21 compoiinds(A1-A21) were identified from the ethyl acetate fraction. In addition, 11 of those compounds(A2, A3, A5, A6, A8, A10, A11, A13, A15, A19, A20) were identified for the first time from A. chinensis Radix. We also identified 14 compounds(A22-A32 and A4-A6) from tlie w-butanol fraction and compound A29 is a new natural product. 20Morphology and distribution of the marine diatom Azpeitia africana(Janisch ex A. Schmidt) G. Fryxell & T. P. Watkins in the South China Sea显示文摘The purpose of the present paper is to study the morphological structure and variability of Azpeitia africana and to determine its geographical distribution in the surface sediments of the South China Sea(SCS). Sediment samples were collected with grabs or box corers in one cruise in 2001 and two cruises in 2007. The sampling stations were located between 3°56.61′–20°59.37′N and 108°30.68′–116°46.70′E,where the water depth ranged from 72 m to 4 238 m. The diatom was observed by phase contrast microscopy and scanning electron microscopy. Microscopical observation showed that A. africana had circular valves with the areolar lines radiating from the eccentric ring. The central rimoportula had an external tube recessed on the edge of a central ring. The marginal rimoportulae were not evenly spaced, and they were positioned more closely together in one quadrant than in the others. Azpeitia africana is the most abundant diatom species in the southern region of the SCS, and accounted for 0.9%–5.6% of all diatom species in the Xisha Islands area. Average cell density of A. africana was 1.1×10~5 valves/g. The percentage abundance of A.africana was low(0%–2.5%) in the northern regions of the SCS and the Sunda Shelf, and it was not detected in the northwestern continental shelf(shallow water area) and northern Kalimantan Island shelf.
