1Chlorinated paraffins in the technosphere:A review of available information and data gaps demonstrating the need to support the Stockholm Convention implementation显示文摘Chlorinated paraffins(CPs)have been produced for a wide range of applications,mostly in open uses,such as metalworking fluids,lubricants,coolants or additives in consumer goods.The production volume is more than one million tonnes requiring control of the lifecycle of these persistent and bioaccumulative chemicals.In May 2017,the Stockholm Convention amended its Annex A to list short chain chlorinated paraffins(SCCPs)as a Persistent Organic Pollutant(POP).Additionally,a limit for the presence of SCCPs in other CP mixtures was set at 1%by weight.CPs can be released to the environment throughout their lifecycle.Therefore,the major objective of this review was to assess and compile information on SCCPs and other CPs regarding their lifecycle in the technosphere to support the Stockholm Convention implementation.A few studies have assessed CP production plants and contamination in the surrounding environments.However,there was no systematic investigation of release routes from production and no assessment of associated landfills,even though these are known major pollution sources at other organochlorine production sites.Some studies have reported that industrial areas,where CPs have likely been used,have elevated levels of CPs in sediments and soil.However,although CPs are largely released when used as metalworking fluids or lubricants,no systematic assessment of releases has been conducted at the thousands of sites where CPs are used in high volumes.Many CPs,mainly medium chain chlorinated paraffins(MCCPs)and SCCPs,are used as additives in the production of consumer goods,resulting in exposure risks.Levels above the European Union regulation for SCCPs of 1500 mg kg^-1 and up to approx.20%are frequently found.The end-of-life management of CP-containing products is difficult since no labelling requirement exists even for products containing SCCPs.The Stockholm Convention prohibits the recycling of SCCP-containing products,which will result in challenges recycling the impacted waste categories.The activities under the Stockholm Convention related to SCCPs,such as the inventory,phase-out,and management of impacted products,provide opportunities to address existing data gaps and challenges.Special attention needs to be given to developing countries with lacking analytical capacity as well as waste management and destruction capacity.Yago Guida Raquel Capella Roland Weber 2020Emerging Contaminants2020,6,1:4
2DMP1 prevents osteocyte alterations,FGF23 elevation and left ventricular hypertrophy in mice with chronic kidney disease显示文摘During chronic kidney disease (CKD),alterations in bone and mineral metabolism include increased production of the hormone fibroblast growth factor 23 (FGF23) that may contribute to cardiovascular mortality.The osteocyte protein dentin matrix protein 1 (DMP1) reduces FGF23 and enhances bone mineralization,but its effects in CKD are unknown.We tested the hypothesis that DMP1 supplementation in CKD would improve bone health,prevent FGF23 elevations and minimize consequent adverse cardiovascular outcomes.We investigated DMP1 regulation and effects in wild-type (WT) mice and the Col4a3^-/- mouse model of CKD.Col4a3^-/- mice demonstrated impaired kidney function,reduced bone DMP1 expression,reduced bone mass,altered osteocyte morphology and connectivity,increased osteocyte apoptosis,increased serum FGF23,hyperphosphatemia,left ventricular hypertrophy (LVH),and reduced survival.Genetic or pharmacological supplementation of DMP1 in Col4a3^-/- mice prevented osteocyte apoptosis,preserved osteocyte networks,corrected bone mass,partially lowered FGF23 levels by attenuating NFAT-induced FGF23 transcription,and further increased serum phosphate.Despite impaired kidney function and worsened hyperphosphatemia,DMP1 prevented development of LVH and improved Col4a3^-/- survival.Our data suggest that CKD reduces DMP1 expression,whereas its restoration represents a potential therapeutic approach to lower FGF23 and improve bone and cardiac health in CKD.Corey Dussold Claire Gerber Samantha White Xueyan Wang Lixin Qi Connor Francis Maralee Capella Guillaume Courbon Jingya Wang Chaoyuan Li Jian Q. 3Histotype-based prognostic classification of gastric cancer显示文摘AIM:To test the efficiency of a recently proposed histotype-based grading system in a consecutive series of gastric cancers.METHODS:Two hundred advanced gastric cancers operated upon in 1980-1987 and followed for a median 159 mo were investigated on hematoxylin-eosinstained sections to identify low-grade [muconodular,well differentiated tubular,diffuse desmoplastic and high lymphoid response (HLR)],high-grade (anaplastic and mucinous invasive) and intermediate-grade (ordinary cohesive,diffuse and mucinous) cancers,in parallel with a previously investigated series of 292 cases.In addition,immunohistochemical analyses for CD8,CD11 and HLA-DR antigens,pancytokeratin and podoplanin,as well as immunohistochemical and molecular tests for microsatellite DNA instability and in situ hybridization for the Epstein-Barr virus (EBV) EBER1 gene were performed.Patient survival was assessed with death rates per 100 person-years and with Kaplan-Meier or Cox model estimates.RESULTS:Collectively,the four low-grade histotypes accounted for 22% and the two high-grade histotypes for 7% of the consecutive cancers investigated,while the remaining 71% of cases were intermediate-grade cancers,with highly significant,stage-independent,survival differences among the three tumor grades (P=0.004 for grade 1 vs 2 and P=0.0019 for grade 2 vs grade 3),thus confirming the results in the original series.A combined analysis of 492 cases showed an improved prognostic value of histotype-based grading compared with the Lauren classification.In addition,it allowed better characterization of rare histotypes,particularly the three subsets of prognostically different mucinous neoplasms,of which 10 ordinary mucinous cancers showed stage-inclusive survival worse than that of 20 muconodular (P=0.037) and better than that of 21 high-grade (P<0.001) cases.Tumors with high-level microsatellite DNA instability (MSI-H) or EBV infection,together with a third subset negative for both conditions,formed the T8 cell-rich HLR group,the largest group among low-grade histotypes.Coexisting HLR proved to be a factor in improved prognosis in tumors with microsatellite instability (P=0.0015 vs HLR-/MSI-H tumors) or DR type human leukocyte antigen expression (P=0.033 vs HLR/HLA-DR+ tumors).CONCLUSION:Identification of lowand high-grade histotypes can improve the prognostic assessment of a substantial proportion of gastric cancers in routine diagnostic practice.Anna Maria Chiaravalli Catherine Klersy Alessandro Vanoli Andrea Ferretti Carlo Capella Enrico Solcia 2012World Journal of Gastroenterology2012,18,9:3
