2Cholesterol transport through the peroxisome-ER membrane contacts tethered by PI(4,5)P2 and extended synaptotagmins显示文摘Most mammalian cells take up cholesterol from low-density lipoproteins(LDLs) via receptor-mediated endocytosis.After reaching lysosomes,LDL-derived cholesterol continues to transport to downstream organelles including the ER for specific structural and functional needs.Peroxisomes are recently found to receive cholesterol from lysosomes through lysosomeperoxisome membrane contacts.However,whether and how cholesterol is conveyed from peroxisomes to the ER remain unknown.Here,by combining high-resolution microscopic analyses and in vitro reconstitution of highly purified organelles or artificial liposomes,we demonstrate that peroxisomes form membrane contacts with the ER through the interaction between peroxisomal PI(4,5)P2 and ER-resident extended synaptotagmin-1,2 and 3(E-Syts).Depletion of peroxisomal PI(4,5)P2 or ESyts markedly decreases peroxisome-ER membrane contacts and induces cholesterol accumulation in lysosomes.Furthermore,we show that cholesterol is delivered from 3H-labeled peroxisomes or PI(4,5)P2-containing liposomes to the ER in vitro,and that the presence of peroxisomes augments cholesterol transfer from lysosomes to the ER.Together,our study reveals a new cholesterol transport pathway along the lysosome-peroxisome-ER membrane contacts in the cell.Jian Xiao Jie Luo Ao Hu Ting Xiao Meixin Li Zekai Kong Luyi Jiang Zimu Zhou Yacheng Liao Chang Xie Beibei Chu Honghua Miao Boliang Li Xiongjie Shi Bao-Liang Song 2019Science China(Life Sciences)2019,62,9:8

7Factors Influencing Diurnal Variations of Cloud and Precipitation in the Yushu Area of the Tibetan Plateau显示文摘Using the cloud radar,ground observations,and ECMWF Reanalysis v5(ERA5)data,we investigate the factors influencing nighttime precipitation during summer in the Yushu area of the Tibetan Plateau(TP).The cloud top height(CTH),cloud base height(CBH),and liquid water content(LWC)are compared between non-precipitation and precipitation days.The results show that the average CTH on precipitation days in Yushu is below 10 km above ground level(AGL)in the daytime,whereas it exceeds 10 km AGL at night,with the maximum at 2300 BT(Beijing Time).The CBH is in-phase with the dewpoint spread.The precipitation intensity and CTH are in-phase with the LWC.The hourly averaged precipitation intensity and convective available potential energy in ERA5 reach their maximums at2100 BT,which is 3 h ahead of their observed counterparts.There is descending motion in the mid day on non-precipitation days,whereas there is ascending motion at night on precipitation days.In addition,the horizontal wind direction in the lower level(below 5000 m)shows clockwise rotation from morning to night.Wind shear occurs in the mid level of the atmosphere,accompanied by a subtropical westerly jet in the upper level.The difference in horizontal wind speed between 200 and 500 hPa is positively related to the LWC,thereby contributing to the formation of upper-level cloud.Bangjun CAO Shihua LYU Yu ZHANG Xianyu YANG Boliang LI Liang YUAN Meilin LI 2022Journal of Meteorological Research2022,36,2:2

12In situ printing of liquid superlenses for subdiffraction-limited color imaging of nanobiostructures in nature显示文摘The nanostructures and patterns that exist in nature have inspired researchers to develop revolutionary components for use in modern technologies and our daily lives.The nanoscale imaging of biological samples with sophisticated analytical tools,such as scanning electron microscopy(SEM)and transmission electron microscopy(TEM),has afforded a precise understanding of structures and has helped reveal the mechanisms contributing to the behaviors of the samples but has done so with the loss of photonic properties.Here,we present a new method for printing biocompatible“superlenses”directly on biological objects to observe subdiffraction-limited features under an optical microscope in color.We demonstrate the nanoscale imaging of butterfly wing scales with a super-resolution and larger field-of-view(FOV)than those of previous dielectric microsphere techniques.Our approach creates a fast and flexible path for the direct color observation of nanoscale biological features in the visible range and enables potential optical measurements at the subdiffraction-limited scale.Boliang Jia Feifei Wang Hoyin Chan Guanglie Zhang Wen Jung Li 2019Microsystems & Nanoengineering2019,5,1:1
13An estimation model of time-varying origin-destination flows in expressway corridors based on unscented Kalman filter显示文摘On the basis of measurable time series of mainline and ramp flows from traffic counts and the assumption of travel time distributions, this research presents a dynamic system model and its on-line estimation algorithm for recursive estimation of time-varying origin-destination (OD) matrices in expressway corridors. The proposed model employs a macro-traffic flow model to estimate travel times of OD flows and uses parameters of the traffic model as state variables, which are added to the constrained function of the system. To improve the model efficiency, we revise the travel time distribution based on the feature of normal distribution. The research employs a newly developed filtering technique, called unscented Kalman filter. The proposed model is evaluated with simulation experiments. 14Plasma exchange therapy for acute necrotizing encephalopathy of childhood显示文摘Importance Acute necrotizing encephalopathy(ANE)is a rare disease with high mortality.Plasma exchange(PLEX)has recently been reported to treat ANE of childhood(ANEC),but its efficacy is uncertain.Objective This study aimed to investigate the effectiveness of PLEX on ANEC.Methods A retrospective study was conducted in four pediatric intensive care units from December 2014 to December 2020.All patients who were diagnosed with ANEC were included;however,these patients were excluded if their length of stay was less than 24 h.Participants were classified into PLEX and non-PLEX groups.Results Twenty-nine patients with ANEC were identified,10 in the PLEX group and 19 in the non-PLEX group.In the PLEX group,C-reactive protein,procalcitonin,alanine aminotransferase,and aspartate aminotransaminase levels were significantly lower after 3 days of treatment than before treatment(13.1 vs.8.0,P=0.043;9.8 vs.1.5,P=0.028;133.4 vs.31.9,P=0.028;282.4 vs.50.5,P=0.046,respectively).Nine patients(31.0%,9/29)died at discharge,and a significantly difference was found between the PLEX group and non-PLEX group[0 vs.47.4%(9/19),P=0.011].The median follow-up period was 27 months,and three patients were lost to follow-up.Thirteen patients(50.0%,13/26)died at the last follow-up,comprising three(33.3%,3/9)in the PLEX group and ten(58.8%,10/17)in the non-PLEX group,but there was no significant difference between the two groups(P=0.411).Three patients(10.3%,3/29)fully recovered.Interpretation PLEX may reduce serum C-reactive protein and procalcitonin levels and improve liver function in the short term.PLEX may improve the prognosis of ANEC,and further studies are needed.Kechun Li Tao Zhang Gang Liu Ping Jin Yeqing Wang Lijie Wang Meixian Xu Chunyi Liu Yingchao Liu Tao Zhou Yan Xu Ying Yang Boliang Fang Xin Yang Chunfeng Liu Suyun Qian 2021Pediatric Investigation2021,5,2:0
15Phylogenetically diverse,acetaldehyde-degrading bacterial community in the deep sea water of the West Pacific Ocean显示文摘As a major aldehyde pollutant widely existing in industry and our daily life, acetaldehyde is more and more harmful to human health. As characteristic habitat niche, bacteria from deep sea environments are abundant and distinctive in heredity, physiology and ecological functions. Thus, the development of acetaldehyde-degrading bacteria from deep sea provides a new method to harness acetaldehyde pollutant. Firstly, in this study,acetaldehyde-degrading bacteria in the deep sea water of the West Pacific Ocean were enriched in situ and in the laboratory respectively, and then the diversity of uncultured bacteria was studied by using 16 S r RNA genes. Then acetaldehyde-degrading strains were isolated from two samples, including enrichment in situ and enrichment in laboratory samples of deep sea water from the West Pacific Ocean using acetaldehyde as the sole carbon source,and then the ability of acetaldehyde degradation was detected. Our results showed that the main uncultured bacteria of two samples with different enrichment approaches were similar, including Proteobacteria,Actinobacteria, Firmicutes, Cyanobacteria, but the structure of bacterial community were significant different.Four subgroups, α, γ, δ and ε, were found in Proteobacteria group. The γ-Proteobacteria was dominant(63.5%clones in laboratory enriched sample, 75% clones in situ enriched sample). The species belonged to γ-Proteobacteria and their proportion was nearly identical between the two enrichment samples, and Vibrio was the predominant genus(45% in laboratory enriched sample, 48.5% in situ enriched sample), followed by Halomonas(9% in situ enriched sample) and Streptococcus(6% in laboratory enriched sample). A total of 12 acetaldehyde-degrading strains were isolated from the two samples, which belonged to Vibrio, Halomonas,Pseudoalteromonas, Pseudomonas and Bacillus of γ-Proteobacteria. Strains ACH-L-5, ACH-L-8 and ACH-S-12,belonging to Vibrio and Halomonas, have strong ability of acetaldehyde degradation, which could tolerate 1.5 g/L acetaldehyde and degrade 350 mg/L acetaldehyde within 24 hours.
