1Qinghu zircon:A working reference for microbeam analysis of U-Pb age and Hf and O isotopes显示文摘Zircon is the most useful mineral for studies in U-Pb geochronology and Hf and O isotope geochemistry.Matrix effect is a major problem of the microbeam techniques such as SIMS and LA-(MC)-ICPMS.Therefore,external standardization using well-characterized natural zircon standards is fundamental for accurate microbeam measurements.While the isotopic geochronology and geochemistry laboratories equipped with microbeam analytical facilities have been increasingly established in China during the past decade,applications of the isotopic microanalysis are still limited due to shortage of available standards.We report here the Qinghu zircon as a potential new working reference for microbeam analysis of zircon U-Pb age and O-Hf isotopes.This zircon was separated from the Qinghu quartz monzonite from the western Nanling Range,Southeast China.It is fairly homogeneous in U-Pb age and Hf and O isotopes in terms of large amounts of mircobeam measurements by LA-MC-ICPMS and SIMS at the scales of 20–60 m.SIMS measurements yield consistent206Pb/238U age within analytical uncertainties with that obtained by ID-TIMS.Precise determinations of O isotopes by IRMS and Hf isotopes by solution MC-ICPMS are in good agreement with the statistical mean of microbeam measurements.We recommend U-Pb age of=159.5±0.2 Ma(2SE),18O=5.4‰±0.2‰(2SD)and176Hf/177Hf=0.283002±0.000004(2SD)as the best reference values for the Qinghu zircon.LI XianHua TANG GuoQiang GONG Bing YANG YueHeng HOU KeJun HU ZhaoChu LI QiuLi LIU Yu LI WuXian 2013Chinese Science Bulletin2013,58,36:60

4Hydraulic fracture initiation theory for a horizontal well in a coal seam显示文摘A series of experiments were performed to determine rock mechanical parameters related to hydraulic fracturing of coal.The effect of confining pressure and pore pressure on the strength of coal was studied.Experimental results show that the coal seam in the study areas has a relatively low elastic modulus,high Poisson's ratio,high fragility and is easily broken and compressed.The coal seam is considered as a transversely isotropic medium,since the physical properties in the direction of bedding plane and orthogonal to the bedding plane vary markedly.Based on the generalized plane strain model,stress distribution for an arbitrarily orientated wellbore in the coal seam was determined.In a horizontal well,hydraulic fracturing was found to initiate in the coal seam mass due to tensile failure,or from cleats due to shear or tensile failure.For those coal seams with abundant natural cleats,hydraulic fracture initiation can be induced by any of these mechanisms.In this study,hydraulic fracture initiation criteria for a horizontal well in a coal seam were established.Hou Bing Chen Mian Wang Zheng Yuan Jianbo Liu Ming 2013Petroleum Science2013,10,2:18
5One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients显示文摘The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. 6Near-inertial waves in the wake of 2011 Typhoon Nesat in the northern South China Sea显示文摘In September 2011, Typhoon Nesat passed over a moored array of instruments recording current and temperature in the northern South China Sea(SCS). A wake of baroclinic near-inertial waves(NIWs) commenced after Nesat passed the array. The associated near-inertial currents are surface-intensified and clockwise-polarized. The vertical range of NIWs reached 300 m, where the vertical range is defined as the maximum depth of the horizontal near-inertial velocity 5 cm/s. The current oscillations have a frequency of 0.709 9 cycles per day(cpd), which is 0.025 f higher than the local inertial frequency. The NIWs have an e-folding time-scale of 10 d based on the evolution of the near-inertial kinetic energy. The depth-leading phase of near-inertial currents indicates downward group velocity and energy flux. The estimated vertical phase velocity and group velocity are 0.27 and 0.08 cm/s respectively, corresponding to a vertical wavelength of 329 m. A spectral analysis reveals that NIWs act as a crucial process to redistribute the energy injected by Typhoon Nesat. A normal mode and an empirical orthogonal function analysis indicate that the second mode has a dominant variance contribution of 81%, and the corresponding horizontal phase velocity and wavelength are 3.50 m/s and 420 km respectively. The remarkable large horizontal phase velocity is relevant to the rotation of the earth, and a quantitative analysis suggests that the phase velocity of the NIWs with a blue-shift of 0.025 f overwhelms that of internal gravity waves by a factor of 4.6.YANG Bing HOU Yijun 2014Acta Oceanologica Sinica2014,33,11:14
7Understanding hydraulic fracture propagation behavior in tight sandstone–coal interbedded formations: an experimentalinvestigation显示文摘Whether hydraulic fractures could connect multiple gas zones in the vertical plane is the key to fracturing treatment to jointly exploit coalbed methane and tight sandstone gas through integrative hydraulic fracturing in tight sandstone–coal interbedded formations. Laboratory true triaxial hydraulic fracturing experiments were conducted on layered specimens with di erent combination types of natural sandstone and coal to simulate the propagation behavior of hydraulic fractures. The effects of the fracture initiation position, fracturing fluid viscosity and injection rate were discussed. The results showed that di erent fracture morphologies could be found. When initiating from coal seams, three patterns of fracture initiation and propagation were obtained:(1) The main hydraulic fracture initiated and propagated along the natural fractures and then diverged due to the effects of in situ stress and formed secondary fractures.(2) The hydraulic fracture initiated and propagated in the direction of the maximum horizontal stress.(3) Multiple fractures initiated and propagated at the same time. With the same fracturing fluid viscosity and injection rate, the hydraulic fractures initiating in sandstones had greater chances than those in coal seams to penetrate interfaces and enter neighboring layers. Excessively small or large fracturing fluid viscosity and injection rate would do harm to the vertical extension height of the induced fracture and improvement of the stimulated reservoir volume. Compared with operation parameters(fracturing fluid viscosity and injection rate), the natural weak planes in coals were considered to be the key factor that a ected the fracture propagation path. 9Phase-field simulation for the evolution of solid/liquid interface front in directional solidification process显示文摘In this study, the phase field method was used to study the multi-controlling factors of dendrite growth in directional solidification. The effects of temperature gradient, propelling velocity, thermal disturbance and growth orientation angle on the growth morphology of the dendritic growth in the solid/liquid interface were discussed. It is found that the redistribution of solute leads to multilevel cavity and multilevel fusion to form multistage solute segregation, and the increase of temperature gradient and propelling velocity can accelerate the dendrite growth of directional solidification, and also make the second dendrites more developed, which reduces the primary distance and the solute segregation. When the temperature gradient is large, the solid-liquid interface will move forward in a flat interface mode,and the thermal disturbance does not affect the steady state behavior of the directionally solidified dendrite tip. It only promotes the generation and growth of the second dendrites and forms the asymmetric dendrite. Meanwhile, it is found that the inclined dendrite is at a disadvantage in the competitive growth compared to the normal dendrite, and generally it will disappear. 10The long rather than the short allele of 5-HTTLPR predisposes Han Chinese to anxiety and reduced connectivity between prefrontal cortex and amygdala显示文摘The short allele of the serotonin-transporter gene is associated with higher risk for anxiety and depression in Caucasians, but this association is still unclear in Asians. Here, we addressed this issue using behavioral and multi-modal MRI approaches in a large group of healthy Han Chinese participants (n = 233). In contrast to findings in Caucasians, we found that long-allele (L) carriers had higher anxiety scores. In another group (n = 64) experiencing significant levels of depression or anxiety, the L-allele frequency was also significantly higher. In healthy participants, L-carriers had reduced functional and anatomical connectivity between the amygdala and prefrontal cortex (PFC), which was correlated with anxiety or depression scores. Our findings demonstrated that in Chinese Han participants, in contrast to Caucasians, the L-allele confers vulnerability to anxiety or depression and weakens top-down emotional control between the PFC and amygdala. 11Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro.Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan 2020Cell Research2020,30,5:8

13Separate Neural Networks for Gains and Losses in Intertemporal Choice显示文摘An important and unresolved question is how human brain regions process information and interact with each other in intertemporal choice related to gains and losses. Using psychophysiological interaction and dynamic causal modeling analyses, we investigated the functional interactions between regions involved in the decisionmaking process while participants performed temporal discounting tasks in both the gains and losses domains. We found two distinct intrinsic valuation systems underlying temporal discounting in the gains and losses domains:gains were specifically evaluated in the medial regions,including the medial prefrontal and orbitofrontal cortices,and losses were evaluated in the lateral dorsolateral prefrontal cortex. In addition, immediate reward or punishment was found to modulate the functional interactions between the dorsolateral prefrontal cortex and distinct regions in both the gains and losses domains: in the gains domain, the mesolimbic regions; in the losses domain, the medial prefrontal cortex, anterior cingulate cortex, and insula. 14Design Bistatic Interferometric DEM Generation Algorithm and Its Theoretical Accuracy Analysis for LuTan-1 Satellites显示文摘LuTan-1(LT-1)is a constellation with two full-polarization L-band radar satellites designed by China,and the first satellite was scheduled to be launched in December 2021 and the second one in January 2022.The LT-1 will be operated for deformation monitoring in repeat-pass mode,and for DEM generation in bistatic mode,improving self-sufficiency of SAR data for the field of geology,earthquake,disaster reduction,geomatics,forestry and so on.In this paper,we focused on designing an algorithm for interferometric DEM generation using LT-1 bistatic satellites.The basic principle,main error sources and errors control of the DEM generation algorithm of LT-1 were systematically analyzed.The experiment results demonstrated that:①The implemented algorithm had rigorous resolution with a theoretic accuracy better than 0.03 m for DEM generation.②The errors in satellite velocity and Doppler centroid had no obvious effect on DEM accuracy and they could be neglected.While the errors in position,baseline,slant range and interferometric phase had a significant effect on DEM accuracy.And the DEM error caused by baseline error was dominated,followed by the slant range error,interferometric phase error and satellite position error.③To obtain an expected DEM accuracy of 2 m,the baseline error must be strictly controlled and its accuracy shall be 1.0 mm or better for Cross-Track and Normal-Direction component,respectively.And the slant range error and interferometric phase error shall be reasonably controlled.The research results were of great significance for accurately grasping the accuracy of LT-1 data products and their errors control,and could provide a scientific auxiliary basis for LT-1 in promoting global SAR technology progress and the generation of high-precision basic geographic data.Bing XU Liqun LIU Zhiwei LI Yan ZHU Jingxin HOU Wenxiang MAO 2022Journal of Geodesy and Geoinformation Science2022,5,1:6
15Polymorphisms of chemokine receptors and its ligand alleles influencing genetic suscepti-bity to HIV-1 infection in eight ethnic groups in Chinese mainland显示文摘Limited genetic information is available concerning the polymorphisms of HIV-1 resistant genes in indigenous Chinese populations. The aim of this study is to identify the allelic frequencies of the chemokine and chemokine receptor genes in the Chinese mainland. Genomic DNA samples extracted from whole blood of 2318 subjects were analyzed by using PCR or PCR/restriction fragment length polymorphism (RFLP) assays, and further confirmed by direct DNA sequencing. Higher frequencies of mutant CCR2-64I (19.15%-28.79%) and SDF1-3’A (19.10% -29.86%) alleles were found in subjects of 8 ethnic groups in the Chinese mainland. In contrast, the △32 mutation in CCRS gene occurs at a very low frequency (0.0016, n=1287) in Han population. A relatively high frequency of CCR5-wt/△32 heterozygotes was observed in Uygurian and Mongolian populations. No A32 mutation allele was detected in Tibetan and other 4 ethnic groups in Yunnan Province. 16Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5
17Improvement in affinity and thermostability of a fully human antibody against interleukin-17A by yeast-display technology and CDR grafting显示文摘Monoclonal antibodies(m Abs) are widely used in many fields due to their high specificity and ability to recognize a broad range of antigens. IL-17 A can induce a rapid inflammatory response both alone and synergistically with other proinflammatory cytokines. Accumulating evidence suggests that therapeutic intervention of IL-17 A signaling offers an attractive treatment option for autoimmune diseases and cancer. Here, we present a combinatorial approach for optimizing the affinity and thermostability of a novel anti-h IL-17 A antibody. From a large na?ve phage-displayed library, we isolated the anti-IL-17 A m Ab 7 H9 that can neutralize the effects of recombinant human IL-17 A. However, the modest neutralization potency and poor thermostability limit its therapeutic applications. In vitro affinity optimization was then used to generate 8 D3 by using yeast-displayed random mutagenesis libraries.This resulted in four key amino acid changes and provided an approximately 15-fold potency increase in a cell-based neutralization assay. Complementarity-determining regions(CDRs) of 8 D3 were further grafted onto the stable framework of the hu Fv 4 D5 to improve thermostability. The resulting hybrid antibody9 NT/S has superior stabilization and affinities beyond its original antibody. Human fibrosarcoma cellbased assays and in vivo analyses in mice indicated that the anti-IL-17 A antibody 9 NT/S efficiently inhibited the secretion of IL-17 A-induced proinflammatory cytokines. 20Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research.
