| 1 | Immunohistochemical analysis of p53,cyclinD1,RB1,c-fos and N-ras gene expression in hepatocellular carcinoma in Iran显示文摘AIM: To study the effect of some genes especially those involved in cell cycle regulation on hepatocellular carcinoma. METHODS: Paraffin-embedded tissue samples of 25 patients (18 males and 7 females) with hepatocellular carcinoma were collected from 22 pathology centers in Tehran during 2000-2001, and stained using immunohistochemistry method (avidin-biotin-peroxidase) for detection of p53, cyclinD1, RB1, c-fos and N-ras proteins. RESULTS: Six (24%), 5 (20%), 12 (48%) and 2 samples (8%) were positive for p53, cyclinD1, C-fos and N-ras expression, respectively. Twenty-two (88%) samples had alterations in the G1 cell-cycle checkpoint protein expression (RB1 or cyclinD1). P53 positive samples showed a higher (9 times) risk of being positive for RB1 protein than p53 negative samples. Loss of expression of RB1 in association with p53 over-expression was observed in 4 (66.7%) of 6 samples. Loss of expression of RB1 was seen in all cyclinD1 positive, 20 (90.9%) N-ras negative, and 11 (50%) C-fos positive samples, respectively. CyclinD1 positive samples showed a higher (2.85 and 4.75 times) risk of being positive for c-fos and N-ras expression than cyclinD1 negative samples. CONCLUSION: The expression of p53, RB1 and c-fos genes appears to have a key role in the pathogenesis of hepatocellular carcinoma in Iran. | 3 | WONCA研究论文摘要汇编——慢性阻塞性肺疾病对亚太地区的影响:基于亚洲人群的EPIC调查显示文摘背景慢性阻塞性肺疾病(COPD)是一种慢性、进行性呼吸减弱的临床病症,主要临床表现为不完全可逆性气流受限。在亚太地区,关于COPD患病率的研究较多,但关于COPD影响的研究较少,本研究旨在对COPD的现患病率及其对亚太地区的影响进行综合阐述。方法 2012年2月1日—5月16日,在亚太地区的9个区域进行人群调查。共有112 300个家庭接受调查,从中抽取符合本研究纳入标准(≥40岁;确诊为COPD、慢性支气管炎或肺气肿,或伴有慢性支气管炎症状)的居民69 279例,其中确诊为COPD的居民为4 289例。采用结构式调查问卷,通过面对面采访或电话随访的形式收集居民的相关资料,最终纳入患者1 841例。结果估算COPD总患病率为6.2%,其中19.1%为重症。调查前的12个月内,近一半患者(46.0%)的病情加重,其中19.0%的患者因病情加重而入院治疗。关于COPD对工作的影响,23.0%的患者表示无法工作,42.0%的患者认为该病限制了工作或活动能力。20.0%服用处方药的患者不知晓自身服用药物的名称;44.0%的患者在调查前12个月内服用过常见口服皮质类固醇处方,而吸入器的使用率(25.0%)较低。仅有37.0%的患者进行了肺功能检查,但89.0%接受检测的患者不知晓检查结果。结论亚太地区的COPD患病率较高,因患病带来的社会经济负担较大。COPD管理能力提升空间较大,患者和医生的疾病相关教育有待加强。 | Lim1 S Chi-Leung Lam D M AR | 2015 | 中国全科医学2015,18,28: | 20 |
| 4 | 桃品种特异性荧光SSR分子标记数据库构建显示文摘【目的】选择通用的荧光SSR分子标记建立桃种质资源基因型数据库便于进行种质特异性鉴定、资源交换、管理和利用。【方法】根据48个SSR标记对国内外669份桃品系的遗传多样性测试结果,从中选择出一部分多态信息含量较高、适合品种鉴别的标记,采用不同荧光标识组合在ABI3130测序仪测试,为了使不同批次的测试结果准确可靠,6—8个参照品种纳入每次的测试中。结果用16个SSR标记基本可以判断品种的特异性,它们可以用4种荧光标识进行标记,便于分为4组在ABI遗传分析仪上测试。目前数据库包含641份材料.基因型一致的品系多为同一品种突变体和自交系演化的地方品种。文中列出了100份中国代表性种质资源分子标记数据库,并建议8个中国品系为参照。对17个桃品种进行系谱验证发现,我国资源圃所育成的7个品种系谱信息正确,但日本桃品种系谱信息与文献出现偏差。【结论】应用16个多态性高的SSR荧光标记可以构建精确的桃品种分子指纹数据库,测试方法及结果对品种特异性和系谱鉴定提供重要的参考.推动资源分子指纹数据库标准化建设工作。 | 李雄伟 孟宪桥 贾惠娟 王力荣 王志强 马瑞娟 吴大军 董冰 Maria Jose Aranzana Pere Arús 高中山 | 2013 | 果树学报2013,30,6: | 17 |
| 5 | ApoB-100, ApoE and CYP7A1 gene polymorphisms in Mexican patients with cholesterol gallstone disease显示文摘AIM: To determine the possible association of the ApoB100 (Xba Ⅰ ), ApoE (Hha Ⅰ ) and CYP7A1 (Bsa Ⅰ ) gene polymorphisms, with the development of cholesterol gallstone disease (GD) in a Mexican population. METHODS: The polymorphisms were analyzed by polymerase chain reaction followed by restriction fragment length polymorphism, in two groups matched by ethnicity, age and sex: patients with GD (n = 101) and stone-free control subjects (n = 101). RESULTS: Allelic frequencies in patients and controls were: 34.16% vs 41.58% (P = 0.124) for X+of ApoB-100; 4.46% vs 5.94% (P = 0.501) for E2, 85.64% vs 78.22% (P = 0.052) for E3, 9.90% vs 15.84% (P = 0.075) for E4 of ApoE; and 25.74% vs 27.72% (P = 0.653) for C of CYP7A1. Differences in genotypic frequencies between the studied groups were not significant (P < 0.05). | 6 | Helicobacter pylori infection as a cause of iron deficiency anaemia of unknown origin显示文摘AIM: To assess the aetiological role of Helicobacter pylori (H. pylori) infection in adult patients with ironrefractory or iron-dependent anaemia of previously unknown origin. METHODS: Consecutive patients with chronic irondeficient anaemia (IDA) with H. pylori infection and a negative standard work-up were prospectively evaluated. All of them had either iron refractoriness or iron dependency. Response to H. pylori eradication was assessed at 6 and 12 mo from follow-up. H. pylori infection was considered to be the cause of the anaemia when a complete anaemia resolution without iron supplements was observed after eradication. RESULTS: H. pylori was eradicated in 88 of the 89 patients. In the non-eradicated patient the four eradicating regimens failed. There were violations of protocol in 4 patients, for whom it was not possible to ascertain the cause of the anaemia. Thus, 84 H. pylori eradicated patients (10 men; 74 women) were available to assess the effect of eradication on IDA. H. pylori infection was considered to be the aetiology of IDA in 32 patients (38.1%; 95%CI: 28.4%-48.8%). This was more frequent in men/postmenopausal women than in premenopausal women (75% vs 23.3%; P < 0.0001) with an OR of 9.8 (95%CI: 3.3-29.6). In these patients, anaemia resolution occurred in the first follow-up visit at 6 mo, and no anaemia or iron deficiency relapse was observed after a mean follow-up of 21 ± 2 mo. | 8 | 急性脊髓损伤患者治疗的临床操作指南:关于MRI基线在临床治疗决策与结果预测中作用的几点建议(英文)显示文摘The objective of this guideline is to outline the role of magnetic resonance imaging(MRI) in clinical decision making and outcome prediction in patients with traumatic spinal cord injury(SCI).Methods A systematic review of the literature was conducted to address key questions related to the use of MRI in patients with traumatic SCI.This review focused on longitudinal studies that controlled for baseline neurologic status.A multidisciplinary Guideline Development Group(GDG) used this information,their clinical expertise,and patient input to develop recommendations on the use of MRI for SCI patients.Based on GRADE(Grading of Recommendation,Assessment,Development and Evaluation),a strong recommendation is worded as ' we recommend,' whereas a weaker recommendation is indicated by 'we suggest.' | 9 | Growth factor-and cytokine-driven pathways governing liver stemness and differentiation显示文摘Liver is unique in its capacity to regenerate in response to injury or tissue loss. Hepatocytes and other liver cells are able to proliferate and repopulate the liver. However, when this response is impaired, the contribution of hepatic progenitors becomes very relevant. Here, we present an update of recent studies on growth factors and cytokine-driven intracellular pathways that govern liver stem/pro-genitor cell expansion and differentiation, and the rel-evance of these signals in liver development, regeneration and carcinogenesis. Tyrosine kinase receptor signaling, in particular, c-Met, epidermal growth factor receptors or fibroblast growth factor receptors, contribute to prolifera-tion, survival and differentiation of liver stem/progenitor cells. Different evidence suggests a dual role for the trans-forming growth factor (TGF)-β signaling pathway in liver stemness and differentiation. On the one hand, TGF-βmediates progression of differentiation from a progenitor stage, but on the other hand, it contributes to the expan-sion of liver stem cells. Hedgehog family ligands are nec-essary to promote hepatoblast proliferation but need to be shut off to permit subsequent hepatoblast differentiation. In the same line, the Wnt family and β-catenin/T-cell fac-tor pathway is clearly involved in the maintenance of liver stemness phenotype, and its repression is necessary for liver differentiation during development. Collectively, data indicate that liver stem/progenitor cells follow their own rules and regulations. The same signals that are essential for their activation, expansion and differentiation are good candidates to contribute, under adequate conditions, to the paradigm of transformation from a pro-regenerative to a pro-tumorigenic role.
