1Biogenic production of DMSP and its degradation to DMS—their roles in the global sulfur cycle显示文摘Dimethyl sulfide(DMS) is the most abundant form of volatile sulfur in Earth’s oceans, and is mainly produced by the enzymatic clevage of dimethylsulfoniopropionate(DMSP). DMS and DMSP play important roles in driving the global sulfur cycle and may affect climate. DMSP is proposed to serve as an osmolyte, a grazing deterrent, a signaling molecule, an antioxidant, a cryoprotectant and/or as a sink for excess sulfur. It was long believed that only marine eukaryotes such as phytoplankton produce DMSP. However, we recently discovered that marine heterotrophic bacteria can also produce DMSP, making them a potentially important source of DMSP. At present, one prokaryotic and two eukaryotic DMSP synthesis enzymes have been identified.Marine heterotrophic bacteria are likely the major degraders of DMSP, using two known pathways: demethylation and cleavage.Many phytoplankton and some fungi can also cleave DMSP. So far seven different prokaryotic and one eukaryotic DMSP lyases have been identified. 2White Matter Abnormalities in Major Depression Biotypes Identified by Diffusion Tensor Imaging显示文摘Identifying data-driven biotypes of major depressive disorder(MDD) has promise for the clarification of diagnostic heterogeneity. However, few studies have focused on white-matter abnormalities for MDD subtyping. This study included 116 patients with MDD and118 demographically-matched healthy controls assessed by diffusion tensor imaging and neurocognitive evaluation.Hierarchical clustering was applied to the major fiber tracts, in conjunction with tract-based spatial statistics, to reveal white-matter alterations associated with MDD.Clinical and neurocognitive differences were compared between identified subgroups and healthy controls. With fractional anisotropy extracted from 20 fiber tracts, cluster analysis revealed 3 subgroups based on the patterns of abnormalities. Patients in each subgroup versus healthy controls showed a stepwise pattern of white-matter alterations as follows: subgroup 1(25.9% of patient sample),widespread white-matter disruption;subgroup 2(43.1% of patient sample), intermediate and more localized abnormalities in aspects of the corpus callosum and left cingulate;and subgroup 3(31.0% of patient sample),possible mild alterations, but no statistically significant tract disruption after controlling for family-wise error. The neurocognitive impairment in each subgroup accompanied the white-matter alterations: subgroup 1, deficits in sustained attention and delayed memory;subgroup 2, dysfunction in delayed memory;and subgroup 3, no significant deficits. 3Transcriptome analysis reveals long noncoding RNAs involved in fiber development in cotton (Gossypium arboreum)显示文摘Long noncoding RNAs(lnc RNAs)play important roles in various biological regulatory processes in yeast,mammals,and plants.However,no systematic identification of lnc RNAs has been reported in Gossypium arboreum.In this study,the strand-specific RNA sequencing(ss RNA-seq)of samples from cotton fibers and leaves was performed,and lnc RNAs involved in fiber initiation and elongation processes were systematically identified and analyzed.We identified 5,996 lnc RNAs,of which 3,510 and 2,486 can be classified as long intergenic noncoding RNAs(linc RNAs)and natural antisense transcripts(lnc NAT),respectively.Linc RNAs and lnc NATs are similar in many aspects,but have some differences in exon number,exon length,and transcript length.Expression analysis revealed that 51.9%of linc RNAs and 54.5%of lnc NATs transcripts were preferentially expressed at one stage of fiber development,and were significantly highly expressed than protein-coding transcripts(21.7%).During the fiber and rapid elongation stages,rapid and dynamic changes in lnc RNAs may contribute to fiber development in cotton.This work describes a set of lnc RNAs that are involved in fiber development.The characterization and expression analysis of lnc RNAs will facilitate future studies on their roles in fiber development in cotton.Changsong Zou Qiaolian Wang Cairui Lu Wencui Yang Youping Zhang Hailiang Cheng Xiaoxu Feng Mtawa Andrew Prosper Guoli Song 2016Science China(Life Sciences)2016,59,2:9

5Application of a collaborative modelling and strategic fuzzy decision support system for selecting appropriate resilience strategies for seaport operations显示文摘The selection of an appropriate resilience investment strategy to optimize the operational efficiency of a seaport is a challenging task given that many criteria need to be considered and modelled under an uncertain environment.The design of such a complex decision system consists of many subjective and imprecise parameters contained in different quantitative and qualitative forms.This paper proposes a fuzzy multi-attribute decision making methodology for the selection of an appropriate resilience investment strategy in a succinct and straightforward manner.The decision support model allows for a collaborative modelling of the system by multiple analysts in a group decision making process.Fuzzy analytical hierarchy process(FAHP) was utilized to analyse the complex structure of the system to obtain the weights of all the criteria while fuzzy technique for order of preference by similarity to ideal solution(TOPSIS) was employed to facilitate the ranking process of the resilience strategies.Given that it is often financially difficult to invest in all the resilience strategies,it is envisaged that the proposed approach could provide decision makers with a flexible and transparent tool for selecting appropriate resilience strategies aimed at increasing the resilience of seaport operations.Andrew John Zaili Yang Ramin Riahi Jin Wang 2014Journal of Traffic and Transportation Engineering(English Edition)2014,1,3:6
6Mammalian ALKBH1 serves as an N^6-mA demethylase of unpairing DNA显示文摘N^6-methyladenine(N^6-mA)of DNA is an emerging epigenetic mark in mammalian genome.Levels of N^6-mA undergo drastic fluctuation during early embryogenesis,indicative of active regulation.Here we show that the 2-oxoglutarate-dependent oxygenase ALKBH1 functions as a nuclear eraser of N^6-mA in unpairing regions(e.g.,SIDD,Stress-Induced DNA Double Helix Destabilization regions)of mammalian genomes.Enzymatic profiling studies revealed that ALKBH1 prefers bubbled or bulged DNAs as substrate,instead of single-stranded(ss-)or double-stranded(ds-)DNAs.Structural studies of ALKBH1 revealed an unexpected“stretch-out”conformation of its“Flip1”motif,a conserved element that usually bends over catalytic center to facilitate substrate base flipping in other DNA demethylases.Thus,lack of a bending“Flip1”explains the observed preference of ALKBH1 for unpairing substrates,in which the flipped N^6-mA is primed for catalysis.Co-crystal structural studies of ALKBH1 bound to a 21-mer bulged DNA explained the need of both flanking duplexes and a flipped base for recognition and catalysis.Key elements(e.g.,an ALKBH1-specificα1 helix)as well as residues contributing to structural integrity and catalytic activity were validated by structurebased mutagenesis studies.Furthermore,ssDNA-seq and DIP-seq analyses revealed significant co-occurrence of base unpairing regions with N^6-mA in mouse genome.Collectively,our biochemical,structural and genomic studies suggest that ALKBH1 is an important DNA demethylase that regulates genome N^6-mA turnover of unpairing regions associated with dynamic chromosome regulation.Min Zhang Shumin Yang Raman Nelakanti Wentao Zhao Gaochao Liu Zheng Li Xiaohui Liu Tao Wu Andrew Xiao Haitao Li 2020Cell Research2020,30,3:6
7Herbal hepatoxicity from Chinese skullcap: A case report显示文摘The use of herbal supplements has increased considerably over the last decade. We report a case of an elderly woman who began taking Move Free Advanced for arthritis, which in addition to glucosamine and chondroitin, contained two herbal ingredients, Chinese skullcap and Black Catechu. Our patient presented with significant cholestasis and hepatitis which significantly improved after discontinuation of the supplement. Since neither the patient nor the treating physician recognized this supplement as a potential hepatotoxin, she resumed taking the supplement and again suffered from considerable hepatotoxicity. Liver biopsy at that time was consistent with acute drug induced liver injury. She, once again, recovered after discontinuation of the supplement. 8Inferring the Tree of Life of the order Cypriniformes, the earth's most diverse clade of freshwater fishes: Implications of varied taxon and character sampling显示文摘The phylogenetic relationships of species are fundamental to any biological investigation, including all evolutionary studies. Accurate inferences of sister group relationships provide the researcher with an historical framework within which the attributes or geographic origin of species (or supraspecific groups) evolved. Taken out of this phylogenetic context, interpretations of evolutionary processes or origins, geographic distributions, or speciation rates and mechanisms, are subject to nothing less than a biological experiment without controls. Cypriniformes is the most diverse clade of freshwater fishes with estimates of diversity of nearly 3,500 species. These fishes display an amazing array of morphological, ecological, behavioral, and geographic diversity and offer a tremendous opportunity to enhance our understanding of the biotic and abiotic factors associated with diversification and adaptation to environments. Given the nearly global distribution of these fishes, they serve as an important model group for a plethora of biological investigations, including indicator species for future cli-matic changes. The occurrence of the zebrafish, Danio rerio, in this order makes this clade a critical component in understanding and predicting the relationship between mutagenesis and phenotypic expressions in vertebrates, including humans. With the tremendous diversity in Cypriniformes, our understanding of their phylogenetic relationships has not proceeded at an acceptable rate, despite a plethora of morphological and more recent mo-lecular studies. Most studies are pre-Hennigian in origin or include relatively small numbers of taxa. Given that analyses of small numbers of taxa for molecular characters can be compromised by peculiarities of long-branch attraction and nodal-density effect, it is critical that significant progress in our understanding of the relationships of these important fishes occurs with increasing sampling of species to mitigate these potential problems. The recent Cypriniformes Tree of Life initiative is an effort to achieve this goal with morphological and molecular (mitochondrial and nuclear) data. In this early synthesis of our understanding of the phylogenetic relationships of these fishes, all types of data have contributed historically to improving our understanding, but not all analyses are complementary in taxon sampling, thus precluding direct understanding of the impact of taxon sampling on achieving accurate phylogenetic inferences. However, recent molecular studies do provide some insight and in some instances taxon sampling can be implicated as a variable that can influence sister group relationships. Other instances may also exist but without inclusion of more taxa for both mitochondrial and nuclear genes, one cannot distinguish between inferences being dictated by taxon sampling or the origins of the molecular data.Richard L. 9SuperDARN CUTLASS Finland radar observations of high-latitude magnetic reconnections under northward interplanetary magnetic field (IMF) conditions显示文摘A number of backscatter power enhancement events with 'equatorward-moving radar auroral forms' in the high-latitude ionosphere were observed by SuperDARN CUTLASS Finland radar when the IMF was northward during 09:00 -10:00 UT on 26 March 2004. These events were also associated with sunward flow enhancements at each location in the Northern Hemisphere which were shown in ionospheric convections measured by the SuperDARN radars. These are typical features of high-latitude (lobe) magnetic reconnections. The durations of the velocity enhancements imply that the evolution time of the lobe reconnections is about 8-16 min from their origin at the reconnection site to their addition to the magnetotail lobe again. In additional, the Double Star TC-1 spacecraft was moving from magnetosheath into magnetosphere, and crossing the magnetopause near the subsolar region during this interval, and observed typical low-latitude magnetic reconnection signatures. 10Practical high-performance lead-free piezoelectrics:structural flexibility beyond utilizing multiphase coexistence显示文摘Due to growing concern for the environment and human health, searching for high-performance lead-free piezoceramics has been a hot topic of scientific and industrial research. Despite the significant progress achieved toward enhancing piezoelectricity, further efforts should be devoted to the synergistic improvement of piezoelectricity and its thermal stability. This study provides new insight into these topics.A new KNN-based lead-free ceramic material is presented, which features a large piezoelectric coefficient(d33) exceeding 500 pC/N and a high Curie temperature(Tc) of ~200℃. The superior piezoelectric response strongly relies on the increased composition-induced structural flexibility due to lattice softening and decreased unit cell distortion. In contrast to piezoelectricity anomalies induced via polymorphic transition, this piezoelectricity enhancement is effective within a broad temperature range rather than a specific small range. 15Humus-Rich Compost Increases Lettuce Growth, Nutrient Uptake, Mycorrhizal Colonisation, and Soil Fertility显示文摘Sandy soils, typical of Australia's west, either have little or no habitat protection for microbes including arbuscular mycorrhizal(AM) fungi, which are essential for nutrient cycling. To minimize this problem, the application of organic matter, such as humus-rich composts, is necessary during vegetable crop production. This study aimed at determining the effects of humus-rich composts on either indigenous or inoculated AM fungal colonisation in roots, lettuce(Lactuca sativa L. var. Quechua) growth, and soil fertility improvement. Four different humus-rich composts with varying humus contents were applied at the same standard rate to lettuce grown under glasshouse conditions for 10 weeks after sowing and compared with two low-humus composts and non-amended soil(control). Humus-rich composts significantly increased lettuce shoot growth, root growth, and AM fungal colonisation in roots. Humus contents in the composts were also correlated with lettuce shoot and root growth. Soil dissolved organic carbon, microbial biomass carbon, and fertility were increased with the application of humus-rich composts. These humus-rich composts, especially the compost of higher humic acid with and without AM inoculation, might have a significant role in sustainable vegetable production, for example lettuce growth. 17Involvement of 90-kuD ribosomal S6 kinase in collagen type Ⅰ expression in rat hepatic fibrosis显示文摘AIM: To investigate the relationship between 90-kuD ribosomal S6 kinase (p90RSK) and collagen type □expression during the development of hepatic fibrosis in vivo and in vitro.METHODS: Rat hepatic fibrosis was induced by intraperitoneal injection of dimethylnitrosamine. The protein expression and cell location of p90RSK and their relationship with collagen type □were determined by co-immunofluoresence and confocal microscopy. Subsequently, RNAi strategy was employed to silence p90RSK mRNA expression in HSC-T6, an activated hepatic stellate cell (HSC) line. The expression of collagen type □in HSC-T6 cells was assessed by Western blotting and real-time polymerase chain reaction. Furthermore, HSCs were transfected with expression vectors or RNAi constructs of p90RSK to increase or decrease the p90RSK expression, then collagen type □promoter activity in the transfected HSCs was examined by reporter assay. Lastly HSC-T6 cells transfected with p90RSK siRNA was treated with or without platelet-derived growth factor (PDGF)-BB at a final concentration of 20 μg/L and the cell growth was determined by MTS conversion.RESULTS: In fibrotic liver tissues, p90RSK was overexpressed in activated HSCs and had a significant positive correlation with collagen type □levels.In HSC-T6 cells transfected with RNAi targeted to p90RSK, the expression of collagen type □was down-regulated (61.8% in mRNA, P < 0.01, 89.1% in protein, P < 0.01). However, collagen type □promoter activity was not increased with over-expression of p90RSK and not decreased with low expression either, compared with controls in the same cell line (P = 0.076).
