1Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 2Reduced graphene oxide-nano zero value iron(rGO-nZVI) micro-electrolysis accelerating Cr(Ⅵ) removal in aquifer显示文摘Nanoscale zero-valent iron(nZVI) assembled on graphene oxide(GO)(rGO-nZVI) composites were synthesized by reduction of GO and ferrous ions with potassium borohydride,for use in Cr(VI) removal from aqueous solution.The results showed that the two-dimensional structure of GO could provide a skeleton support for Fe^0,thus overcoming the bottleneck of aggregation for nZVI.Also,rGO-nZVI would form a ferric-carbon micro-electrolysis system in Cr(VI)-contaminated aquifers,enhancing and accelerating electron transfer,exhibiting high rate and capacity for Cr(VI) removal.The optimum dosage of the applied r GO-nZVI was linearly correlated with the initial Cr(VI) concentration.Characterization of rGO-nZVI before and after reaction with Cr(VI) revealed the process of Cr(VI) removal:r GO-nZVI firstly transferred electrons from Fe^0 cores via their Fe(II)/Fe(III) shells to the GO sheet;there,negatively charged Cr(VI) received electrons and changed into positively charged Cr(III),which was adsorbed by the negatively charged GO sheet,avoiding the capping and passivating of nZVI.rGO-nZVI formed a good electrically conductive network,and thus had long-term electron releasing properties,which was important for groundwater remediation.Liming Ren Jun Dong Zifang Chi Huazheng Huang 2018Journal of Environmental Sciences2018,30,11:12
3An innovative wood-chip-framework substrate used as slow-release carbon source to treat high-strength nitrogen wastewater显示文摘Removal of nitrogen in wastewater before discharge into receiving water courses is an important consideration in treatment systems.However,nitrogen removal efficiency is usually limited due to the low carbon/nitrogen(C/N) ratio.A common solution is to add external carbon sources,but amount of liquid is difficult to determine.Therefore,a combined wood-chip-framework substrate(with wood,slag and gravel) as a slow-release carbon source was constructed in baffled subsurface-flow constructed wetlands to overcome the problem.Results show that the removal rate of ammonia nitrogen(NH_4^+-N),total nitrogen(TN) and chemical oxygen demand(COD) could reach 37.5%-85%,57.4%-86%,32.4%-78%,respectively,indicating the combined substrate could diffuse sufficient oxygen for the nitrification process(slag and gravel zone) and provide carbon source for denitrification process(wood-chip zone).The nitrification and denitrification were determined according to the location of slag/gravel and wood-chip,respectively.Nitrogen removal was efficient at the steady phase before a shock loading using slag-wood-gravel combined substrate because of nitrification-denitrification process,while nitrogen removal was efficient under a shock loading with wood-slag-gravel combined substrate because of ANAMMOX process.This study provides a new idea for wetland treatment of high-strength nitrogen wastewater.Huai Li Zifang Chi Baixing Yan Long Cheng Jianzheng Li 2017Journal of Environmental Sciences2017,29,1:10
4Geochemical Characteristics and Origin of Natural Gases in the Qaidam Basin,China显示文摘Sixty-five natural gas samples were collected from 19 oil-gasfields in the Qaidam basin, China. The chemical composition and carbon isotope values of the samples were measured, and the geochemical characteristics and origin of the natural gases were studied. The gases can be divided into biogenic gases, sapropelic oil-type gases, mixed type oil-type gases, coal-type gases and mixed gas. The δ13C1 values of the biogenic gases are very small and the C2+ contents of them are very low, ranging from -68.2‰ to -61.8‰ and 0.06% to 0.20% respectively. They have heavy δD and δ13CCO2, showing a CO2 reduction pathway. They are distributed in the East depression region and derived from the Quaternary source rocks. The sapropelic oil-type gases have small δ13C2 values and high C2+ ranging from -36.6‰ to -28.6‰ and from 33.01% to 47.15% respectively. The mixed type oil-type gases have δ13C2 values and C2+ contents varying from -28.6‰ to -24.8‰ and from 4.81% to 26.06% respectively. Both sapropelic oil-type gases and mixed type oil-type gases are associated with oil-type oils, distributed in the West depression region and derived from the Tertiary saltwater lacustrine sapropelic source rocks and humic source rocks respectively. The δ13C2 values of the coal-type gases are extremely high and the C2+ contents are very low, changing from -23.3‰ to -12.5‰ and from 0.06% to 18.07% respectively. The coal-type gases in the Nanbaxian gasfield and the Lenghu oil-gasfields in the North fault block belt are derived from the Middle Jurassic coal-measures source rocks, whereas those in the West depression region are derived from the Tertiary saltwater lacustrine humic source rocks. Compared with some other basins in China, the natural gases there have obviously heavier δ13C due to the heavier δ13C of different types of kerogens of the Tertiary saltwater lacustrine source rocks in the West depression region of the basin. The mixing of natural gases is common in the West depression region, but the mixed gases are formed by sapropelic oil-type gases, mixed type oil-type gases or coal-type gases, respectively, of different levels of maturity. 5Long-term impacts of graphene oxide and Ag nanoparticles on anammox process: Performance, microbial community and toxic mechanism显示文摘The increasing application of engineered nanoparticles(NPs) has posed an emerging challenge to constructed wetland wastewater treatment. The performance, microbial community and toxic mechanism of anammox-based unplanted subsurface-flow constructed wetlands(USFCWs) were investigated under the long-term exposure of different graphene oxides(GOs) and Ag NP concentrations. Results showed that the addition of GO could promote TN removal, manifesting as function anammox bacteria C. Anammoxoglobus having a relative high abundance, for GO did not cause significant damage to the cell integrity though there was an increase in ROS concentrations. TN removal would not be obviously affected under exposure of 1 mg/L Ag NPs, for the function gene related to cell biogenesis and repair was up-regulated; while the addition of 10 mg/L Ag NPs would have an inhibiting effect on TN removal in the USFCWs, for the disappearance of some species having anammox ability. 6Partially hydroxylated ultrathin iridium nanosheets as efficient electrocatalysts for water splitting显示文摘Ultrathin two-dimensional(2D)materials have attracted considerable attention for their unique physicochemical properties and promising applications;however,preparation of freestanding ultrathin 2D noble metal remains a significant challenge.Here,for the first time,we report use of a wet-chemical method to synthesize partially hydroxylated ultrathin Ir nanosheets(ir-NSs)of only five to six atomic layers*thickness.Detailed analysis indicates that the growth confinement effect of carbon monoxide and the partially hydroxylated surface play a critical role in formation of the ultrathin structure.The ultrathin Ir-NSs exhib让excellent performance for both the hydrogen evolution reaction and oxygen evolution reaction in a wide pH range,outperforming the state-of the-art Pt/C and IrO2,respectively.Density-functional theory calculations reveal that the partial hydroxylation not only enhances the surface electron transfer between Ir-sites and intermediate O-species,but also guarantees efficient irdtial activation ofbond cleavage of H-O-H for first-step H2O splitting.This;ultimately,breaks through barriers to full water splitting,with efficient electron transfer essentially maintained.Zifang Cheng Bolong Huang Yecan Pi Leigang Li Qi Shao Xiaoqing Huang 2020National Science Review2020,7,8:4
7Maternal supply of cysteamine alleviates oxidative stress and enhances angiogenesis in porcine placenta显示文摘Background:Oxidative stress in placenta is associated with the occurrence of adverse pregnancy outcomes in sow,but there are few satisfactory treatment strategies for these conditions.This study investigated the potential of cysteamine(CS)as an antioxidant protectant for regulating the reproductive performance,redox status,and placental angiogenesis of sows.Methods:The placental oxidative stress status and vascular density of piglets with different birth weights:<1.0 kg(low birth weight,LBW)and 1.4–1.6 kg(normal birth weight,NBW)were evaluated,followed by allotting 84 sows to four treatments(n=21)and feeding them with a basal diet supplemented with 0,100,300,or 500 mg/kg of CS from d 85 of gestation to d 21 of lactation,respectively.Placenta,serum,and colostrum samples of sows or piglets were collected,and the characteristics of sows and piglets were recorded.Furthermore,the in vivo results were validated using porcine vascular endothelial cells(PVECs).Results:Compared with the NBW placentae,the LBW placentae showed increased oxidative damage and were vulnerable to angiogenesis impairment.Particularly,H2O2-induced oxidative stress prompted intracellular reactive oxygen species generation and inhibited the tube formation and migration of PVECs as well as the expression of vascular endothelial growth factor-A(VEGF-A)in vitro.However,dietary CS supplementation can alleviate oxidative stress and improve the reproductive performance of sows.Specifically,compared with the control group,dietary 100 mg/kg CS could(1)decrease the stillbirth and invalid rates,and increase both the piglet birth weight in the low yield sows and the placental efficiency;(2)increase glutathione and reduce malondialdehyde in both the serum and the colostrum of sows;(3)increase the levels of total antioxidant capacity and glutathione in LBW placentae;(4)increase the vascular density,the mRNA level of VEGF-A,and the immune-staining intensity of platelet endothelial cell adhesion molecule-1 in the LBW placentae.Furthermore,the in vitro experiment indicated that CS pre-treatment could significantly reverse the NADPH oxidase 2-ROS-mediated inactivation of signal transducer and activator of transcription-3(Stat3)signaling pathway induced by H2O2 inhibition of the proliferation,tube formation,and migration of PVECs.Meanwhile,inhibition of Stat3 significantly decreased the cell viability,tube formation and the VEGF-A protein level in CS pretreated with H_(2)O_(2)-cultured PVECs.Conclusions:The results indicated that oxidative stress and impaired angiogenesis might contribute to the occurrence of LBW piglets during pregnancy,but CS supplementation at 100 mg/kg during late gestation and lactation of sows could alleviate oxidative stress and enhance angiogenesis in placenta,thereby increasing birth weight in low yield sows and reducing stillbirth rate.The in vitro data showed that the underlying mechanism for the positive effects of CS might be related to the activation of Stat3 in PVECs.Shuangbo Huang Zifang Wu Zihao Huang Xiangyu Hao Longmiao Zhang Chengjun Hu Jianfu Wei Jinping Deng Chengquan Tan 2022Journal of Animal Science and Biotechnology2022,13,1:4
8Kinetics of nitrobenzene degradation coupled to indigenous microorganism dissimilatory iron reduction stimulated by emulsified vegetable oil显示文摘Widespread contamination by nitrobenzene(NB) in sediments and groundwater requires better understanding of the biogeochemical removal process of the pollutant. NB degradation, coupled with dissimilatory iron reduction, is one of the most efficient pollutant removal methods. However, research on NB degradation coupled to indigenous microorganism dissimilatory iron reduction stimulated by electron donors is still experimental. A model for remediation in an actual polluted site does not currently exist.Therefore, in this study, the dynamics was derived from the Michaelis–Menten model(when the mass ratio of emulsified vegetable oil and NB reached the critical value 91:1). The effect of SO_4^(2-), NO_3^-, Ca^(2+)/Mg^(2+), and the grain size of aquifer media on the dynamics were studied, and the NB degradation dynamic model was then modified based on the most significant factors. 9Activated carbon felts with exfoliated graphene nanosheets for flexible all-solid-state supercapacitors显示文摘The recent development of portable electronics promotes the growing demand for flexible energy storage devices. Supercapacitors are promising candidates due to their high power density. Therefore, flexible supercapacitors are desired. Here, the porous activated carbon felts(ACFs) with exfoliated graphene nanosheets and rich oxygen-containing groups were fabricated by a facile thermal treatment strategy.Such ACFs can act as the flexible electrodes of all-solid-state supercapacitors directly without the use of binder and conductive materials. They exhibit excellent electrochemical properties, such as high specific areal capacitance, superior rate ability and long-term cycling stability. 10肝细胞肝癌自发破裂出血:87例回顾性分析(英文)显示文摘Objective: The aim of the study was to review the management of ruptured hepatocellular carcinoma (HCC) in a single teaching hospital over 13-year period; to determine the prognostic factor of in-hospital mortality and evaluate the safety and efficacy of liver resection. Methods: A retrospective collection of medical records of 87 patients with spontaneous ruptured HCC was carried out. The 28 patients underwent emergency intervention including transarterial chemoembolization (TACE) and laparotomy with/without liver resection. Conservative treatment was performed in 59 patients and 16 of which underwent delayed hepatectomy or TACE. Results: The overall in-hospital mortality and median survival time was 54% and 22 days respectively. Albumin level (OR = 0.874, 95% CI: 0.778-0.973, P = 0.024), number of tumors (OR = 5.011, 95% CI: 1.015-24.750, P = 0.048) and laparotomy (OR = 0.069, 95% CI: 0.012-0.406, P = 0.003) were all independent factors affecting overall in-hospital mortality, but for patients undergone laparotomy, only total bilirubin level (OR = 1.138, 95% CI: 1.024-1.264, P = 0.016) was independent factor affecting overall in-hospital mortality. Age, total bilirubin level, maximum tumor size, number of tumors, portal vein tumor thrombosis and extra-hepatic metastasis were all significantly different between groups with laparotomy and without. There were no significant differences between emergency and delayed liver resection groups in in-hospital mortality (0 vs. 0), median survival time (788 vs. 750 days respectively) as well as 1-year and 3-year survival rates (66.7%, 44.4% vs. 70%, 30%, respectively) (P = 0.763, log-rank test). Conclusion: Both underlying chronic liver disease and tumor stage can affect the in-hospital mortality, but for patients undergone laparotomy, only total bilirubin level is independent factor. Surgeons are more prone to choose patients with younger age, better liver function and earlier tumor stage to do surgery. 11The Strategies of Pathogen-Oriented Therapy on Circumventing Antimicrobial Resistance显示文摘The emerging antimicrobial resistance(AMR)poses serious threats to the global public health.Conventional antibiotics have been eclipsed in combating with drug-resistant bacteria.Moreover,the developing and deploying of novel antimicrobial drugs have trudged,as few new antibiotics are being developed over time and even fewer of them can hit the market.Alternative therapeutic strategies to resolve the AMR crisis are urgently required.Pathogen-oriented therapy(POT)springs up as a promising approach in circumventing antibiotic resistance.The tactic underling POT is applying antibacterial compounds or materials directly to infected regions to treat specific bacteria species or strains with goals of improving the drug efficacy and reducing nontargeting and the development of drug resistance.This review exemplifies recent trends in the development of POTs for circumventing AMR,including the adoption of antibiotic-antibiotic conjugates,antimicrobial peptides,therapeutic monoclonal antibodies,nanotechnologies,CRISPR-Cas systems,and microbiota modulations.Employing these alternative approaches alone or in combination shows promising advantages for addressing the growing clinical embarrassment of antibiotics in fighting drug-resistant bacteria.Zifang Shang Siew Yin Chan Qing Song Peng Li Wei Huang 2020Research2020,,1:2
12Improved adaptive fuzzy control for MIMO nonlinear time-delay systems显示文摘This paper presents an improved observer-based indirect adaptive fuzzy control scheme for multiinput-multioutput (MIMO) nonlinear time-delay systems.The control scheme synthesizes adaptive fuzzy control with adaptive fuzzy identification.An observer is designed to observe the system state,and an identifier is developed to identify the unknown parts of the system.The update laws for parameters utilize two types of errors in the adaptive time-delay fuzzy logic systems,the observation error and the identification error.Performance analysis proves the superiority of the update laws in terms of faster and improved tracking and parameter convergence.Simulation results of two-link manipulator demonstrate the effectiveness of the improved control scheme.Du, Zhenbin Qu, Zifang 2011控制理论与应用(英文版)2011,9,2:2
13Differences in the primary culture, purification and biological characteristics between endothelial cells and smooth muscle cells from rat aorta显示文摘Objective: To investigate the differences of primary culture, purification and biological characteristics between endothelial cells and smooth muscle cells from rat aorta. Methods: Endothelial cells were obtained using the vascular ring adherence, collagenase digestion method and an improved vascular ring adherence method, while smooth muscle cells were separated from tissue sections of rat aorta. Clones of endothelial cells were selected by limiting dilution assay. Both cell types were identified using specific cell immunofluorescent markers, and phase contrast microscopy was used to observe the morphological disparity between endothelial cells and smooth muscle cells at the single cell and colony level. Cell proliferation was determined by the cell counting kit-8. Differences between endothelial cells and smooth muscle cells were evaluated in trypsin digestion time, attachment time and recovery after cryopreservation. Results: Endothelial cells were obtained by all three methods. The improved vascular ring method provided the most reproducible results. Cells were in good condition, and of high purity. Smooth muscle cells were cultured successfully by the tissue fragment culture method. Clonal expansion of single endothelial cells was attained. The two cell types expressed their respective specific markers, and the rate of proliferation of smooth muscle cells exceeded that of endothelial cells. Endothelial cells were more sensitive to trypsin digestion than smooth muscle cells. In addition, they had a shorter adherence time and better recovery following cryopreservation than smooth muscle cells. Conclusion: The improved vascular ring method was optimal for yielding endothelial cells. Limiting dilution is a novel and valid method for purifying primary endothelial cells from rat aorta. Primary rat endothelial cell and vascular smooth muscle cell cultures exhibited different morphological characteristics, proliferation rate, adherence time, susceptibility to trypsin digestion and recovery after cryopreservation. 16One-dimensional iridium-based nanowires for efficient water electrooxidation and beyond显示文摘The sluggish reaction kinetics of oxygen evolution reaction(OER)has largely lowered the efficiency of electrochemical water splitting.Ir represents one of the state-of-the-art electrocatalysts for promoting OER especially in acidic electrolytes.However,it remains a formidable challenge to synthesize high-quality one-dimensional(1D)Ir-based nanostructures for improved electrocatalytic performance.Herein,a template-assisted synthesis method is reported wherein 1D porous Ir-Te nanowires(Ir-Te NWs)are synthesized with Te NWs serving as the template.The Ir-Te NWs exhibit highly enhanced OER performance compared to commercial IrO_(2) and Ir/C.In detail,the overpotentials to reach 10 mA cm^(-2) are 248 and 284 mV in 1 M KOH and 0.5 M H2S04,respectively,much lower than those of commercial catalysts.The Ir-Te NWs also show smaller Tafel slopes than commercial IrO_(2) and Ir/C,signifying faster reaction kinetics.Besides,much more durable OER activity can be maintained for Ir-Te NWs with negligible decay during 25 and 20 h stability tests in 1 M KOH and 0.5 M H_(2)SO_(4),respectively.Further analysis indicates that the significantly improved OER performance of Ir-Te NWs could be ascribed to the larger electrochemical surface area and smaller electrical resistance.More significantly,the templated synthesis of Ir-Te NWs can be facilely extended to the fabrication of other metal-Te NWs including Ru-Te,Rh-Te and Pt-Te NWs.The design and synthesis of 1D metal-based NWs in this work provide important inspiration for the synthesis of diversified 1D metallic nanostructures with distinctly enhanced catalytic performance and beyond.Leigang Li Pengtang Wang Zifang Cheng Qi Shao Xiaoqing Huang 2022Nano Research2022,15,2:1

18肝细胞肝癌经肝动脉化疗栓塞后破裂:2例报告及系统回顾(英文)显示文摘Objective: Rupture of hepatocellular carcinoma (HCC) following transarterial embolization/chemoembolization (TAE/TACE) is a rare but life-threatening complication. The aim of the study was to explore the incidence, risk factors, clinical characteristics, treatment, and outcomes of this complication. Methods: We described two cases and reviewed all cases of ruptured HCC after TAE/TACE reported in the literature. Results: Our search yielded 32 cases of ruptured HCC after TAE/TACE. The overall incidences were 0.45% per patient and 0.21% per session. The mean age of the patients was 57.4 years (range 28-90 years, n=26, No. of cases with available information). Males accounted for 81% of cases (21/26). The 50% of the cases had histories of primary hypertension, diabetes or peripheral artery disease (6/12). Mean diameter of the tumors was 11.4 cm (range 3-20 cm, n=27). The 100% of cases had superficial or exophytic tumors (23/23). Portal vein thrombosis was presented in 61.5% of patients (8/13). The median interval between TAE/TACE and rupture was 2 days (range 0 hour-30 days, n=31). Management choices included emergency TAE, surgery, and conservative treatment. The overall median survival time was 7 days (n=19). Conclusion: Rupture of HCC following TAE/TACE is relatively rare but potentially life-threatening. The management is difficult and prognosis is poor.
