| 3 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |
| 4 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 6 | Synergetic Improvement of Interlaminar Fracture Toughness in Carbon Fiber/Epoxy Composites Interleaved with PES/PEK‑C Hybrid Nanofiber Veils显示文摘In this study,two types of soluble thermoplastic resins were added to epoxy resin at a fixed weight ratio to prepare a three-phase cast body.The cast was then manufactured into hybrid nanofiber as interleaves for interlaminar toughening of carbon fiber/epoxy resin(CF/EP)composites using a co-solvent method.The results revealed that when the hybrid components reached 15 wt%,Polyethersulfone(PES)and polyaryletherketone cardo(PEK-C)exhibited the best synergistic toughening effect,and the fracture toughness increased by 99.8%and 39.8%,respectively,compared with the reference or the same proportion of the single PES toughened sample.We used PES/PEK-C hybrid nanofibers with an areal density of 19.2 g per square meter(gsm)as composite toughening layers.Apart from the lack of significant influence of PES nanofiber on CF/EP composites,the interlaminar fracture toughness of mode I and mode II layers increased by 88.3%and 46.9%,respectively,compared to the reference sample.Scanning Electron Microscopy of the fracture surface and cross-section micromorphology of the laminate displayed that the thermoplastic microspheres of different sizes contribute differently to crack resistance:PEK-C consumes more energy due to the debonding and extraction of microspheres and resin,whereas the presence of the PES phase can induce more plastic deformation and crack deflection. | Jinli Zhou Chenyu Zhang Chao Cheng Ming Wang Zhihui Yang Yanfei Yang Hongying Yang Muhuo Yu | 2022 | Advanced Fiber Materials2022,4,5: | 3 |
| 7 | SSR markers development and their application in genetic diversity evaluation of garlic(Allium sativum)germplasm显示文摘Garlic(Allium sativum),an asexually propagated vegetable and medicinal crop,has abundant genetic variation.Genetic diversity evaluation based on molecular markers has apparent advantages since their genomic abundance,environment insensitivity,and non-tissue specific features.However,the limited number of available DNA markers,especially SSR markers,are insufficient to conduct related genetic diversity assessment studies in garlic.In this study,4372 EST-SSR markers were newly developed,and 12 polymorphic markers together with other 17 garlic SSR markers were used to assess the genetic diversity and population structure of 127 garlic accessions.The averaged polymorphism information content(PIC)of these 29 SSR markers was 0.36,ranging from 0.22 to 0.49.Seventy-nine polymorphic loci were detected among these accessions,with an average of 3.48 polymorphic loci per SSR.Both the clustering analyses based on either the genotype data of SSR markers or the phenotypic data of morphological traits obtained genetic distance divided the 127 garlic accessions into three clusters.Moreover,the Mantel test showed that genetic distance had no significant correlations with geographic distance,and weak correlations were found between genetic distance and the phenotypic traits.AMOVA analysis showed that the main genetic variation of this garlic germplasm collection existed in the within-population or cluster.Results of this study will be of great value for the genetic/breeding studies in garlic and enhance the utilization of these garlic germplasms. | Xiaxia Li Lijun Qiao Birong Chen Yujie Zheng Chengchen Zhi Siyu Zhang Yupeng Pan Zhihui Cheng | 2022 | Plant Diversity2022,44,5: | 2 |

| 16 | Sapphirine-Bearing Pelitic Granulite from Ailaoshan Orogen,West Yunnan,China: Metamorphic Conditions and Tectonic Setting显示文摘To reveal the petrological characteristics, metamorphic evolution histon and tectonic setting of the pelitic granulites from Ailaoshan Orogen, Uest Yunnan, China, a comprehensive study in mineral chemistry, petrogeochemistry and geochronology studies is presented in this paper. Two metamorphic stages of the granulites can be established:(1) the peak metamorphism recorded by the mineral assemblage of garnet, kyanite, K-feidspar and rutile, and the initial retrograde metamorphism shown by the mineral assemblage of garnet, sillimanite, sapphirine, spinel, K-feldspar, plagioclase and biotite;(2) the superim-posed metamorphism recorded by the mineral assemblage of biotite, muscovite, plagioclase, quartz and ilmenite. Zircon LA-ICP-MS U-Pb dating indicates that the protolith of the granulite was deposited after 337 Ma. The initial retrograde metamorphism occurred at P-T conditions of 8.6-12 kbar at 850-920℃ estimated by mineral assemblages, the low pressure limit of kyanite stability and GBPQ geothermobarometer in Indosinian (about 235 Ma), and the late superimposed metamorphism occurred at the P-T condition of 3.5-3.9 kbar at 572-576℃ estimated by GBPQ geothermobarometer since 33Ma. The first stage was related to the amalgamation of the South China and Indochina blocks during the Triassic, and the second stage was possibly related with the large scale sinistral slip-shearing since the Oligocene.
