1Improvement of wheat drought and salt tolerance by expression of a stress- inducible transcription factorGmDREB of soybean (Glycinemax)显示文摘Under stress conditions such as droughthigh-salinity and low-temperature, the transcription factorof DREB (dehydration responsive element binding proteins)improved efficiently stress resistance by regulating the ex-pression of its downstream genes with various environmentastress resistance in plants. GmDREB gene (GenBank Acces-sion No. AF514908) encoding a stress-inducible transcriptionfactor was cloned by screening a cDNA library of Glycinemax cv. Jinong 27 with yeast one-hybrid method. GmDREBgene was 910 bp in length and encoded 174 amino acids con-taining a conserved AP2/EREBP DNA-binding domain of 58amino acids. Two conserved functional amino acids, valineand glutamic acid, were located on the 14th and the 19thamino acid residues in the conserved structural domain. Analkaline amino acid region (KKR) related to a nuclear local-ization signal was at the N-terminal, while an acidic aminoacid region (DDD) related to trans-activation was at theC-terminal. Plant expression vectors were constructed andtransformed into wheat by bombardment. In total, 13 trans-genic plants with Ubi::GmDREB and 11 transgenic plantswith rd29A::GmDREB were identified from 103 regenerationplants by molecular analysis. The drought and salt tolerancesof T1 transgenic lines with Ubi::GmDREB orrd29A::GmDREB were demonstrated to be improved ascompared to wild type. 2Intratumor heterogeneity,microenvironment,and mechanisms of drug resistance in glioma recurrence and evolution显示文摘Glioma is the most common lethal tumor of the human brain.The median survival of patients with primary World Health Organization grade IV glioma is only 14.6 months.The World Health Organization classification of tumors of the central nervous system categorized gliomas into lower-grade gliomas and glioblastomas.Unlike primary glioblastoma that usually develop de novo in the elderly,secondary glioblastoma enriched with an isocitrate dehydrogenase mutant typically progresses from lower-grade glioma within 5-10 years from the time of diagnosis.Based on various evolutional trajectories brought on by clonal and subclonal alterations,the evolution patterns of glioma vary according to different theories.Some important features distinguish the normal brain from other tissues,e.g.,the composition of the microenvironment around the tumor cells,the presence of the blood-brain barrier,and others.The underlying mechanism of glioma recurrence and evolution patterns of glioma are different from those of other types of cancer.Several studies correlated tumor recurrence with tumor heterogeneity and the immune microenvironment.However,the detailed reasons for the progression and recurrence of glioma remain controversial.In this review,we introduce the different mechanisms involved in glioma progression,including tumor heterogeneity,the tumor microenvironment and drug resistance,and their pre-clinical implements in clinical trials.This review aimed to provide new insights into further clinical strategies for the treatment of patients with recurrent and secondary glioma.Zhaoshi Bao Yongzhi Wang Qiangwei Wang Shengyu Fang Xia Shan Jiguang Wang Tao Jiang 2021Frontiers of Medicine2021,15,4:10
3Expression of an(E)-β-farnesene synthase gene from Asian peppermint in tobacco affected aphid infestation显示文摘Aphids are major agricultural pests that cause significant yield losses in crop plants each year.(E)-β-farnesene(EβF) is the main or only component of an alarm pheromone involved in chemical communication within aphid species and particularly in the avoidance of predation. EβF also occurs in the essential oil of some plant species, and is catalyzed by EβF synthase. By using oligonucleotide primers designed from the known sequence of an EβF synthase gene from black peppermint(Mentha × piperita), two cDNA sequences, MaβFS1 and MaβFS2, were isolated from Asian peppermint(Mentha asiatica). Expression pattern analysis showed that the MaβFS1 gene exhibited higher expression in flowers than in roots, stems and leaves at the transcriptional level. Overexpression of MaβFS1 in tobacco plants resulted in emission of pure EβF ranging from 2.62 to 4.85 ng d-1g-1of fresh tissue. Tritrophic interactions involving peach aphids(Myzus persicae), and predatory lacewing(Chrysopa septempunctata) larvae demonstrated that transgenic tobacco expressing MaβFS1 had lower aphid infestation. 4Autophagy and cancer treatment: four functional forms of autophagy and their therapeutic applications显示文摘Cancer is the leading cause of death worldwide. Drugs play a pivotal role in cancer treatment, but the complex biological processes of cancer cells seriously limit the efficacy of various anticancer drugs. Autophagy, a self-degradative system that maintains cellular homeostasis, universally operates under normal and stress conditions in cancer cells. The roles of autophagy in cancer treatment are still controversial because both stimulation and inhibition of autophagy have been reported to enhance the effects of anticancer drugs. Thus, the important question arises as to whether we should try to strengthen or suppress autophagy during cancer therapy. Currently, autophagy can be divided into four main forms according to its different functions during cancer treatment: cytoprotective(cell survival), cytotoxic(cell death), cytostatic(growth arrest), and nonprotective(no contribution to cell death or survival). In addition, various cell death modes, such as apoptosis, necrosis, ferroptosis, senescence, and mitotic catastrophe, all contribute to the anticancer effects of drugs. The interaction between autophagy and these cell death modes is complex and can lead to anticancer drugs having different or even completely opposite effects on treatment. Therefore, it is important to understand the underlying contexts in which autophagy inhibition or activation will be beneficial or detrimental.That is, appropriate therapeutic strategies should be adopted in light of the different functions of autophagy. 20Event-Triggered Consensus for Discrete-Time Multi-agent Systems with Parameter Uncertainties Based on a Predictive Control Scheme显示文摘In this paper,the event-triggered consensus for linear discrete-time multi-agent systems with parameter uncertainties is investigated.The parameter uncertainty is assumed to be normbounded.An event-triggered consensus protocol based on the predictive control method is proposed to make the multi-agent system achieve consensus.And for the design of the consensus protocol,the problem of estimating the control input is transformed into the problem of estimating state differences between agents.Furthermore,the event-triggered consensus protocol proposed in this paper only demands each agent to mornitor its state to determine its event-triggered instants.A sufficient existence condition for the consensus protocol is proposed based on the linear matrix inequality.And a sufficient condition for the nonexistence of the Zeno-like behaviour is also derived.Finally,a numerical example is given to illustrate that the event-triggered consensus protocol proposed in this paper can make the multi-agent system with parameter uncertainties achieve consensus effectively.LIU Xiaoyu XUAN Yongbo ZHANG Zhongyu DIAO Zhaoshi MU Zhenxing LI Zhitao 2020Journal of Systems Science & Complexity2020,33,3:1
