2miR-155 aggravates impaired autophagy of pancreatic acinar cells through targeting Rictor显示文摘The aim of this study was to investigate the role and mechanism of miR-155 in regulating autophagy in a caerulein-induced acute pancreatitis(AP)cellular model.GFP-LC3 immunofluo-rescence assay was performed to detect autophagy vesicle formation in pancreatic acinar cell line AR42J.AR42J cells were transfected with miR-155 mimic,inhibitor,and corresponding controls to explore the effect of miR-155 on autophagy.The protein levels of LC3-I,LC3-11,Beclin-1,and p62 were analyzed by western blot analysis.Dual-luciferase reporter assay was performed to verify the interaction between miR-155 and Rictor(RPTOR independent companion of MTOR complex 2).The results showed that caerulein treatment induced impaired autophagy as evi-denced by an increase in the accumulation of p62 together with LC3-11 in AR42J cells,accompanied by miR-155 upregulation.Furthermore,miR-155 overexpression aggravated,whereas miR-155 silencing reduced the caerulein-induced impairment of autophagy.Mechanistically,Rictor was confirmed to be a direct target of miR-155,which could rescue the miR-155 overexpression-mediated aggravation of im paired autophagy.Collectively,these findings indicate that miR-155 aggravates impaired autophagy in caerulein-treated pancreatic acinar cells by targeting Rictor.Xueming Zhang Jiangtao Chu Haijun Sun Dali Zhao Biao Ma Dongbo Xue Weihui Zhang Zhituo Li 2020Acta Biochimica et Biophysica Sinica2020,52,2:4
3Optimization of a synthetic medium for ethanol production by xylose-fermenting Zymomonas mobilis using response surface methodology显示文摘A synthetic medium for ethanol production by recombinant xylose-fermenting Zymomonas mobilis was optimized using Plackett-Burman(PB) design and response surface methodology(RSM).The effects of 19 medium components were investigated by PB design.Eight of these components were determined to have significant effects on ethanol production.The statistical model was constructed via central composite design(CCD) using five selected variables,including xylose,trisodium citrate,choline chloride,pyridoxine,and thiamine.The validity of the developed model was verified.The ethanol concentration in the optimized medium was 6.7% higher than in the RM medium.The optimized medium was then simplified to give the final synthetic medium(S2),in which the ethanol concentration was 20.7% higher than in the RM medium.Xylose and trisodium citrate were the key medium components influencing ethanol production,while calcium pantothenate was the only growth factor required by recombinant Z.mobilis.Ethanol production and growth response were directly proportional to the logarithm of the concentration of trisodium citrate in the range of 0.1-1.6 g/L.DONG HuiNa ZHAO XueMing MA YuanYuan ZHANG MinHua 2012Chinese Science Bulletin2012,57,28:3
4A high-efficient triboelectric-electromagnetic hybrid nanogenerator for vibration energy harvesting and wireless monitoring显示文摘Scavenging mechanical vibration energy by combining triboelectric nanogenerators(TENGs) and electromagnetic generators(EMGs) is a renewable and low-carbon way to generate electric power. In this paper, we fabricate a cylindrical structure hybrid nanogenerator based on spring structure to effectively scavenge vibration energy. The introduction of spring structure not only enhances the space utilization, but also supports the moving magnet. Owing to its innovative structure, the instantaneous output power of TENG and EMG are 0.88 mW at a load of 10 MΩ and 216.7 mW at a load of 60 Ω, respectively. With the aids of a power management circuit, the hybrid nanogenerator can provide continuous power for two hygrothermographs connected in series. In addition, a self-powered wireless acceleration sensing monitoring system(SWAM) is developed to measure the acceleration signals of vibration sources. 5Spatio-temporal Differentiation of Residential Land for Coastal Town: A Case Study of Dalian Jinshitan显示文摘Based on SPOT5 satellite remote sensing images and land use data in five time phases covering the period 1988–2012, the distribution characteristics, spatial differentiation processes, and driving mechanisms affecting the residential land of the Jinshitan Tourist Resort were studied. The work included a combination of GIS technology, adoption of the differentiation index D, and the multi-group differentiation-measure model D(m). The results provided a basis for the following conclusions: 1) From 1988 to 2012, the degree of differentiation of the rural residential land first decreased, and then increased. The general residential, commercial residential and garden-house land-types all showed declining trends, of which the garden-house type presented the most intense volatility. 2) The overall background of the traditional residential land type was gradually replaced by a new residential land type, and the total area of residential land increased year by year, significantly improving the overall residential conditions. The areas of the new residential land type showed growth, and were increasingly widely distributed in the coastal tourist resorts, transportation corridors, and scenic areas. 3) Government policy, social differentiation, market mechanisms, and individual choices(along with other factors) were mutually promoted, although still restrained. 6The 2012 Flash Drought Threatened US Midwest Agroecosystems显示文摘In the summer of 2012, the US Midwest, the most productive agricultural region in the world, experienced the most intense and widespread drought on record for the past hundred years. The 2012 drought, characterized as ‘flash drought’, developed in May with a rapid intensification afterwards, and peaked in mid-July. ~76% of crop region and 60% of grassland and pasture regions have been under moderate to severe dry conditions. This study used multiple lines of evidences, i.e., in-situ AmeriFlux measurements, spatial satellite observations, and scaled ecosystem modeling, to provide independent and complementary analysis on the impact of 2012 flash drought on the US Midwest vegetation greenness and photosynthesis carbon uptake. Three datasets consistently showed that 1) phenological activities of all biomes advanced 1–2 weeks earlier in 2012 compared to the other years of 2010–2014;2) the drought had a more severe impact on agroecosystems(crop and grassland) than on forests;3) the growth of crop and grassland was suppressed from June with significant reduction of vegetation index, sun-induced fluorescence(SIF) and gross primary production(GPP), and did not recover until the end of growing season. The modeling results showed that regional total GPP in 2012 was the lowest(1.76 Pg C/yr) during 2010–2014, and decreased by 63 Tg C compared with the other-year mean. Agroecosystems, accounting for 84% of regional GPP assimilation, were the most impacted by 2012 drought with total GPP reduction of 9%, 7%, 6%, and 29% for maize, soybean, cropland, and grassland, respectively. The frequency and severity of droughts have been predicted to increase in future.
