Serum thymosin β4 levels in patients with hepatitis B virus-related liver failure显示文摘AIM:To investigate whether serum thymosinβ4 can provide diagnostic or prognostic information in liver failure patients caused by chronic hepatitis B virus(HBV) infection. METHODS:Serum thymosinβ4 levels were measured in 30 patients with acute-on-chronic liver failure(ACLF), 31 patients with chronic liver failure(CLF),30 patients with compensated liver cirrhosis(CR)and 32 patients with chronic hepatitis B and 30 healthy controls.Serum thymosinβ4 levels were measured by enzyme-linked immunosorbent assay and Child-Pugh and model for end-stage liver disease(MELD)scores were calculated for each patient on admission.RESULTS:Compared with healthy controls, serum thymosinβ4 levels in ACLF,CLF,CR and chronic hepatitis B patients were significantly lower,6.5047 (4.7879-10.5314)μg/mL vs 0.4632(0.2759-0.8768) μg/mL,0.6981(0.5209-1.2008)μg/mL,1.8053 (0.8110-2.3397)μg/mL,3.7803(1.8570-6.4722)μg/mL, respectively(P<0.001).The levels of thymosinβ4 in liver failure(ACLF or CLF)patients were markedly lower than that in CR(P<0.001),and a difference was also found between CLF and ACLF patients(P=0.038).In patients with chronic liver disease, there was a positive relationship between thymosinβ4 levels and albumin, choline esterase, and platelet(P<0.001),and negative relationship with alanine aminotransferase(P=0.020), aspartate aminotransferase, total bilirubin, international normalized ratio of prothrombin time, and Child-Pugh and MELD scores(P<0.001).Of the 61 liver failure patients, the thymosinβ4 levels of non-survivors were significantly lower than that of survivors(P=0.007). Receiver operating characteristics analysis identified a thymosinβ4 cutoff level of 0.5708μg/mL for predicting poor prognosis in all liver failure patients. The serial thymosinβ4 values were observed in 13 liver failure inpatients.Lower initial values were observed in the death. While greater improvement in thymosinβ4 value was found in those who recovered from the disease. CONCLUSION:Serum thymosinβ4 can be used as an important potential predictor for liver failure caused by chronic HBV infection. | Tao Han,Ying Liu,Huan Liu,Zheng-Yan Zhu,Yan Li,Shi-Xiang Xiao,Zhen Guo,Zhen-Gang Zhao,Department of Hepatology,Tianjin Institute of Hepatobiliary Disease,Tianjin Key Laboratory of Artificial Cells,Tianjin Third Central Hospital,Tianjin Medical University,83 Jintang Road,Tianjin 300170, China | 2010 | World Journal of Gastroenterology2010,16,5: | 20
B-type natriuretic peptide in predicting the severity of community-acquired pneumonia显示文摘BACKGROUND: Although pneumonia severity index(PSI) is widely used to evaluate the severity of community-acquired pneumonia(CAP), the calculation of PSI is very complicated. The present study aimed to evaluate the role of B-type natriuretic peptide(BNP) in predicting the severity of CAP.METHODS: For 202 patients with CAP admitted to the emergency department, BNP levels, cardiac load indexes, inf lammatory indexes including C-reactive protein(CRP), white blood cell count(WBC), and PSI were detected. The correlation between the indexes and PSI was investigated. BNP levels for survivor and non-survivor groups were compared, and a receiver operating characteristic(ROC) curve analysis was performed on the BNP levels versus PSI.RESULTS: The BNP levels increased with CAP severity(r=0.782, P<0.001). The BNP levels of the high-risk group(PSI classes IV and V) were signifi cantly higher than those of the low-risk group(PSI classes I–III)(P<0.001). The BNP levels were signifi cantly higher in the non-survivor group than in the survivor group(P<0.001). In addition, there were positive correlations between BNP levels and PSI scores(r=0.782, P<0.001). The BNP level was highly accurate in predicting the severity of CAP(AUC=0.952). The optimal cut-off point of BNP level for distinguishing high-risk patients from low-risk ones was 125.0 pg/m L, with a sensitivity of 0.891 and a specifi city of 0.946. Moreover, BNP level was accurate in predicting mortality(AUC=0.823). Its optimal cut-off point for predicting death was 299.0 pg/m L, with a sensitivity of 0.675 and a specifi city of 0.816. Its negative predictive cut-off value was 0.926, and the positive predictive cut-off value was 0.426.CONCLUSION: BNP level is positively correlated with the severity of CAP, and may be used as a biomarker for evaluating the severity of CAP. | Jing Li Huan Ye Li Zhao | 2015 | World Journal of Emergency Medicine2015,6,2: | 18
A general end-to-end diagnosis framework for manufacturing systems显示文摘The manufacturing sector is envisioned to be heavily influenced by artificial-intelligence-based technologies with the extraordinary increases in computational power and data volumes. A central challenge in the manufacturing sector lies in the requirement of a general framework to ensure satisfied diagnosis and monitoring performances in different manufacturing applications. Here, we propose a general data-driven,end-to-end framework for the monitoring of manufacturing systems. This framework, derived from deep-learning techniques, evaluates fused sensory measurements to detect and even predict faults and wearing conditions. This work exploits the predictive power of deep learning to automatically extract hidden degradation features from noisy, time-course data. We have experimented the proposed framework on 10 representative data sets drawn from a wide variety of manufacturing applications. Results reveal that the framework performs well in examined benchmark applications and can be applied in diverse contexts, indicating its potential use as a critical cornerstone in smart manufacturing. | Ye Yuan Guijun Ma Cheng Cheng Beitong Zhou Huan Zhao Hai-Tao Zhang Han Ding | 2020 | National Science Review2020,7,2: | 18

Coinfection with influenza A virus enhances SARS-CoV-2 infectivity显示文摘The upcoming flu season in the Northern Hemisphere merging with the current COVID-19 pandemic raises a potentially severe threat to public health.Through experimental coinfection with influenza A virus(IAV)and either pseudotyped or live SARS-CoV-2 virus,we found that IAV preinfection significantly promoted the infectivity of SARS-CoV-2 in a broad range of cell types.Remarkably,in vivo, increased SARS-CoV-2 viral load and more severe lung damage were observed in mice coinfected with IAV.Moreover, such enhancement of SARS-CoV-2 infectivity was not observed with several other respiratory viruses, likely due to a unique feature of IAV to elevate ACE2 expression. This study illustrates that IAV has a unique ability to aggravate SARS-CoV-2 infection, and thus, prevention of IAV infection is of great significance during the COVID-19 pandemic. | Lei Bai Yongliang Zhao Jiazhen Dong Simeng Liang Ming Guo Xinjin Liu Xin Wang Zhixiang Huang Xiaoyi Sun Zhen Zhang Lianghui Dong Qianyun Liu Yucheng Zheng Danping Niu Min Xiang Kun Song Jiajie Ye Wenchao Zheng Zhidong Tang Mingliang Tang Yu Zhou Chao Shen Li Zhou Yu Chen Huan Yan Ke Lan Ke Xu | 2021 | Cell Research2021,31,4: | 16
Electroacupuncture activates enteric glial cells and protects the gut barrier in hemorrhaged rats显示文摘AIM:To investigate whether electroacupuncture ST36 activates enteric glial cells, and alleviates gut inflammation and barrier dysfunction following hemorrhagic shock.METHODS:Sprague-Dawley rats were subjected to approximately 45% total blood loss and randomly divided into seven groups:(1) sham:cannulation, but no hemorrhage;(2) subjected to hemorrhagic shock(HS);(3) electroacupuncture(EA) ST36 after hemorrhage;(4) vagotomy(VGX)/EA:VGX before hemorrhage, then EA ST36;(5) VGX:VGX before hemorrhage;(6) a-bungarotoxin(BGT)/EA:intraperitoneal injection of a-BGT before hemorrhage, then EA ST36; and(7) a-BGT group:a-BGT injection before hemorrhage.Morphological changes in enteric glial cells(EGCs) were observed by immunofluorescence, and glial fibrillary acidic protein(GFAP; a protein marker of enteric glial activation) was evaluated using reverse transcriptase polymerase chain reaction and western blot analysis.Intestinal cytokine levels, gut permeability to 4-k Da fluorescein isothiocyanate(FITC)-dextran,and the expression and distribution of tight junction protein zona occludens(ZO)-1 were also determined.RESULTS:EGCs were distorted following hemorrhage and showed morphological abnormalities.EA ST36 attenuated the morphological changes in EGCs at 6 h,as compared with the VGX,a-BGT and HS groups.EA ST36 increased GFAP expression to a greater degree than in the other groups.EA ST36 decreased intestinal permeability to FITC-dextran(760.5 ± 96.43 ng/m L vs 2466.7 ± 131.60 ng/m L,P < 0.05) and preserved ZO-1 protein expression and localization at 6 h afterhemorrhage compared with the HS group. However, abdominal VGX and a-BGT treatment weakened or eliminated the effects of EA ST36.EA ST36 reduced tumor necrosis factor-a levels in intestinal homogenates after blood loss, while vagotomy or intraperitoneal injection of a-BGT before EA ST36 abolished its antiinflammatory effects.CONCLUSION:EA ST36 attenuates hemorrhageinduced intestinal inflammatory insult, and protects the intestinal barrier integrity, partly via activation of EGCs. | Sen Hu Zeng-Kai Zhao Rui Liu Hai-Bin Wang Chun-Yu Gu Hong-Min Luo Huan Wang Ming-Hua Du Yi Lv Xian Shi | 2015 | World Journal of Gastroenterology2015,21,5: | 15
Factors Affecting the Performance of Single-Chamber Soil Microbial Fuel Cells for Power Generation显示文摘There is limited information about the factors that affect the power generation of single-chamber microbial fuel cells(MFCs) using soil organic matter as a fuel source. We examined the effect of soil and water depths, and temperature on the performance of soil MFCs with anode being embedded in the flooded soil and cathode in the overlaying water. Results showed that the MFC with 5 cm deep soil and 3 cm overlaying water exhibited the highest open circuit voltage of 562 mV and a power density of 0.72 mW m-2. The ohmic resistance increased with more soil and water. The polarization resistance of cathode increased with more soil while that of anode increased with more water. During the 30 d operation, the cell voltage positively correlated with temperature and reached a maximum of 162 mV with a 500 ? external load. After the operation, the bacterial 16S rRNA gene from the soil and anode was sequenced. The bacteria in the soil were more diverse than those adhere to the anode where the bacteria were mainly affiliated to Escherichia coli and Deltaproteobacteria. In summary, the two bacterial groups may generate electricity and the electrical properties were affected by temperature and the depth of soil and water. | DENG Huan WU Yi-Cheng ZHANG Fan HUANG Zong-Chuan CHEN Zheng XU Hui-Juan ZHAO Feng | 2014 | Pedosphere2014,24,3: | 15
Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. Therefore, LOCS+hPMSCs implantation holds a great promise for bridging the nerve defect and may be clinically useful in the near future. | Sufang Han Zhifeng Xiao Xing Li Huan Zhao Bin Wang Zhixue Qiu Zhi Li Xin Mei Bai Xu Caixia Fan Bing Chen Jin Han Yanzheng Gu Huilin Yang Qin Shi Jianwu Dai | 2018 | Science China(Life Sciences)2018,61,1: | 15
Evolution of boundary layer ozone in Shijiazhuang,a suburban site on the North China Plain显示文摘The structure of the boundary layer affects the evolution of ozone(O3), and research into this structure will provide important insights for understanding photochemical pollution. In this study, we conducted a one-month observation(from June 15 to July 14, 2016) of the boundary layer meteorological factors as well as O3 and its precursors in Luancheng County,Shijiazhuang(37°53′N, 114°38′E). Our research showed that photochemical pollution in Shijiazhuang is serious, and the mean hourly maximum and mean 8-hr maximum O3 concentrations are 97.9 ± 26.1 and 84.4 ± 22.4 ppbV, respectively. Meteorological factors play a significant role in the formation of O3. High temperatures and southeasterly winds lead to elevated O3 values, and at moderate relative humidity(40%–50%) and medium boundary layer heights(1200–1500 m), O3 production sensitivity occurred in the transitional region between volatile organic compounds(VOC) and nitrogen oxides(NOx) limitations, and the O3 concentration was the highest. The vertical profiles of O3 were also measured by a tethered balloon. The results showed that a large amount of O3 was stored in the residual layer, and the concentration was positively correlated with the O3 concentration measured the previous day. During the daytime of the following day, the contribution of O3 stored in the residual layer to the boundary layer reached 27%± 7% on average. | Wei Zhao Guiqian Tang Huan Yu Yang Yang Yinghong Wang Lili Wang Junlin An Wenkang Gao Bo Hu Mengtian Cheng Xingqin An Xin Li Yuesi Wang | 2019 | Journal of Environmental Sciences2019,31,9: | 15

Cloning and characterization of squalene synthase and cycloartenol synthase from Siraitia grosvenorii显示文摘Mogrosides and steroid saponins are tetracyclic triterpenoids found in Siraitia grosvenorii.Squalene synthase(SQS) and cycloartenol synthase(CAS) are key enzymes in triterpenoid and steroid biosynthesis. In this study,full-length cDNAs of SgSQS and SgCAS were cloned by a rapid amplification of cDNA-ends with polymerase chain reaction(RACE-PCR) approach. The SgSQS cDNA has a 1254 bp open reading frame(ORF) encoding 417 amino acids, and the SgCAS cDNA contains a 2298 bp ORF encoding 765 amino acids.Bioinformatic analysis showed that the deduced SgSQS protein has two transmembrane regions in the C-terminal.Both SgSQS and SgCAS have significantly higher levels in fruits than in other tissues, suggesting that steroids and mogrosides are competitors for the same precursors in fruits.Combined in silico prediction and subcellular localization, experiments in tobacco indicated that SgSQS was probably in the cytoplasm or on the cytoskeleton, and SgCAS was likely located in the nucleus or cytosol. These results will provide a foundation for further study of SgSQS and SgCAS gene functions in S.grosvenorii, and may facilitate improvements in mogroside content in fruit by regulating gene expression. | Huan Zhao Qi Tang Changming Mo Longhua Bai Dongping Tu Xiaojun Ma | 2017 | Acta Pharmaceutica Sinica B2017,7,2: | 10
Dendrobium officinale Kimura et Migo and American ginseng mixture: A Chinese herbal formulation for gut microbiota modulation显示文摘Dendrobium officinale Kimura et Migo(D. officinale) is a famous traditional Chinese medicine(TCM). A mixture of D. officinale and American ginseng has been shown to enhance cell-mediated immunity, humoral immunity, and monocyte/macrophage functions in mice. Here, the effects of a D. officinale and American ginseng mixture on the structure of gut microbial community in dogs were examined using high-throughput 16S rRNA gene amplicon sequencing. The data revealed that while the mixture did not change the diversity of gut microbial community significantly, differences among individuals were significantly reduced. Furthermore, the mixture-responsive operational taxonomic units(OTUs) exhibited a phase-dependent expression pattern. Fifty-five OTUs were found to exhibit a mixture-induced expression pattern, among which one third were short-chain fatty acid(SCFA)-producing genera and the others were probiotic genera included Lactobacillus spp., Sutterella, Alistipes, Anaerovorax, Bilophila, Coprococcus, Gordonibacter, Oscillibacter, among others. By contrast, 36% of the OTUs exhibiting a mixture-repressed expression pattern were disease-associated microorganisms, and six genera, namely Actinomyces, Escherichia/Shigella, Fusobacterium, Slackia, Streptococcus and Solobacterium, were associated with cancer. In addition, five genera were closely associated with diabetes, namely Collinsella,Rothia, Howardella, Slackia and Intestinibacter. Our results indicate that this D. officinale and American ginseng mixture may be used as a prebiotic agent to enhance SCFA-producing genera and prevent gut dysbiosis. | LIU Cheng-Zhi CHEN Wei WANG Mei-Xia WANG Ying CHEN Li-Qing ZHAO Feng SHI Ya LIU Hui-Jun DOU Xiao-Bing LIU Chao CHEN Huan | 2020 | Chinese Journal of Natural Medicines2020,18,6: | 10
A bHLH transcription activator regulates defense signaling by nucleo-cytosolic trafficking in rice显示文摘Crosstalk between plant hormone signaling pathways is vital for controlling the immune response during pathogen invasion.Salicylic acid(SA)and jas-monic acid(JA)often play important but antagonistic roles in the immune responses of higher plants. Here,we identify a basic helix-loop-helix transcription acti-vator,OsbHLH6,which confers disease resistance in rice by regulating SA and JA signaling via nucleo-cytosolic trafficking in rice(Oryza sativa).OsbHLH6 expression was upregulated during Magnaporthe oryzae infection.Transgenic rice plants overexpressing OsbHLH6 display increased JA responsive gene expression and enhanced disease susceptibility to the pathogen.Nucleus-localized OsbHLH6 activates JA signaling and suppresses SA signaling;however, the SA regulator OsNPR1(Nonexpressor of PR genes 1)sequesters OsbHLH6 in the cytosol to alleviate its effect. Our data suggest that OsbHLH6 controls disease re-sistance by dynamically regulating SA and JA signaling. | Fanwei Meng Chao Yang Jidong Cao Huan Chen Jinhuan Pang Qiqi Zhao Zongyi Wang Zheng Qing Fu Jun Liu | 2020 | Journal of Integrative Plant Biology2020,62,10: | 10
