1Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 2Three interpenetrated copper(Ⅱ) coordination polymers based on a V-shaped ligand:Synthesis,structures,sorption and magnetic properties显示文摘Three new CuⅡ coordination polymers,[Cu2(oba)2(H2O)(DMF)].3H2O(1),[Cu4(oba)4(DMF)4].4H2O.DMF(2) and [Cu2(oba)(hmp)2](3)(H2oba = 4,4'-oxybis(benzoic acid),Hhmp = 2-pyridine methanol,DMF = N,N'-dimethylformamide) were synthesized by solvothermal reactions and characterized by single-crystal X-ray diffraction.Both complexes 1 and 2 take three-dimensional(3D) three-fold interpenetrating PtS topology with 1D channels.In complex 3,Cu4 clusters were formed by the chelating hmp ligands,and then the clusters were further linked by oba ligands to form a 3D three-fold interpenetrating dia topology network.For 2,N2 sorption shows type I isotherm with BET surface areas of 562 m2 g-1 and Langmuir surface area of 747 m2 g-1,respectively.At 77 and 87 K,the hydrogen uptakes of complex 2 are 0.89 wt% and 0.57 wt%,respectively.Magnetic investigation showed the ferromagnetic coupling between CuII ions and very weak antiferromagnetic interaction between Cu4 clusters in complex 3.ZHANG XiaoFeng YANG Qian ZHAO JiongPeng HU Tong-Liang CHANG Ze BU Xian-He 2011Science China Chemistry2011,54,9:7
3High salt diet accelerates the progression of murine lupus through dendritic cells via the p38 MAPK and STAT1 signaling pathways显示文摘The increased incidence of systemic lupus erythematosus(SLE)in recent decades might be related to changes in modern dietary habits.Since sodium chloride(NaCl)promotes pathogenic T cell responses,we hypothesize that excessive salt intake contributes to the increased incidence of autoimmune diseases,including SLE.Given the importance of dendritic cells(DCs)in the pathogenesis of SLE,we explored the influence of an excessive sodium chloride diet on DCs in a murine SLE model.We used an induced lupus model in which bone marrow-derived dendritic cells(BMDCs)were incubated with activated lymphocyte-derived DNA(ALD-DNA)and transferred into C57BL/6 recipient mice.We observed that a high-salt diet(HSD)markedly exacerbated lupus progression,which was accompanied by increased DC activation.NaCl treatment also stimulated the maturation,activation and antigenpresenting ability of DCs in vitro.Pretreatment of BMDCs with NaCl also exacerbated BMDC-ALD-DNA-induced lupus.These mice had increased production of autoantibodies and proinflammatory cytokines,more pronounced splenomegaly and lymphadenopathy,and enhanced pathological renal lesions.The p38 MAPK–STAT1 pathway played an important role in NaClinduced DC immune activities.Taken together,our results demonstrate that HSD intake promotes immune activation of DCs through the p38 MAPK–STAT1 signaling pathway and exacerbates the features of SLE.Thus,changes in diet may provide a novel strategy for the prevention or amelioration of lupus or other autoimmune diseases.Ze Xiu Xiao Xiaojiang Hu Ximei Zhang Zhigang Chen Julie Wang Ke Jin Feng Lin Cao Baoqing Sun Joseph A.Bellanti Nancy Olsen Song Guo Zheng 2020Signal Transduction and Targeted Therapy2020,5,1:6
4Syntheses, structures and magnetic properties of three Co(Ⅱ)coordination architectures based on a flexible multidentate carboxylate ligand and different N-donor ligands显示文摘Three new cobalt complexes,{[Co5(tci)2(bimb)3(3-O)2(H2O)2]·3DMF·4H2O}n(1),{[Co3(tci)2(bib)]·2DMF·2H2O}n(2)and{[Co(Htci)(bpea)0.5]·H2O}n(3)(H3tci=tris(2-carboxyethyl)isocyanurate,bimb=4,4′-bis(imidazol-1-yl)biphenyl,bib=1,4-bis(imidazol-1-yl)benzene,bpea=1,2-bis(4-pyridyl)ethane,DMF=N,N′-dimethylformamide),have been successfully synthesized through the assembly of Co(II)ions,H3tci and different N-donor ligands,respectively.All complexes were structurally characterized by single crystal X-ray diffraction,elemental analyses,IR spectra,thermogravimetric(TG)analyses and X-ray powder diffraction(XRPD).Complex 1 exhibits a 3D three-fold parallel interpenetrated 3D→3D structure with(65·8)CdSO4topology.Complex 2 is built from[Co3(2-Ocarboxyl)2(CO2)4]clusters and linear bib ligands,displaying a two-fold parallel interpenetrated(3,8)-connected(43)2(46·618·84)topology,while complex 3 is a 3D pillar-layered structure involving an infinite–Co–(2-Ocarboxyl)(CO2)–Co–chain.The diverse structures of the three complexes indicate that the skeletons of different N-donor ligands play an important role in the assembly of such different frameworks.In addition,magnetic investigation indicates that besides spin-orbit coupling of Co(II)ions,there exist antiferromagnetic exchange interactions in Co5and Co3clusters of 1 and 2,respectively.TIAN Dan LIU SuiJun ZHANG DaShuai CHANG Ze HU TongLiang BU XianHe 2013Science China Chemistry2013,56,12:5
5APOE and APOC1 gene polymorphisms are associated with cognitive impairment progression in Chinese patients with late-onset Alzheimer's disease显示文摘Current evidence shows that apolipoprotein E(APOE), apolipoprotein CI(APOC1) and low density lipoprotein receptor-related protein(LRP) variations are related to late-onset Alzheimer's disease. However, it remains unclear if genetic polymorphisms in these genes are associated with cognitive decline in late-onset Alzheimer's disease patients. We performed a 30-month longitudinal cohort study to investigate the relationship between Alzheimer's disease and APOE, APOC1, and LRP. In this study, 78 Chinese Han patients with late-onset Alzheimer's disease were recruited form Guangxi Zhuang Autonomous Region in China. APOE, APOC1, and LRP genotyping was performed using polymerase chain reaction-restriction fragment length polymorphisms. The Mini-Mental State Examination and Clinical Dementia Rating Scale were used to assess patients' cognitive function. After a 30-month follow-up period, we found a significant reduction in Mini-Mental State Examination total score, a higher proportion of patients fulfilling cognitive impairment progression criteria, and a higher proportion of APOC1 H2 carriers in APOE ε4 carriers compared with non-carriers. In addition, the APOE ε4 allele frequency was significantly higher in the cognitive impairment progression group compared with the non-cognitive impairment progression group. 8Thermodynamically Revealing the Essence of Order and Disorder Structures in Layered Cathode Materials显示文摘Layered transition metal(TM) oxides are one of the most widely used cathode materials in lithium-ion batteries. The atomic configuration in TM layer of these materials is often known to be random when multiple TM elements co-exist in the layer(e.g. Ni, Co and Mn). By contrast, the configuration tends to be ordered if the elements are Li and Mn. Here, by using special quasi-random structures(SQS) algorithm, the essential reasons of the ordering in a promising Li-rich Mn-based cathode material Li2MnO3 are investigated. The difference of internal energy and entropy between ordered and disordered materials is calculated. As a result, based on the Gibbs free energy, it is found that Li2MnO3 should have an ordered structure in TM layer. 9Protective effects of medical ozone combined with traditional Chinese medicine against chemically-induced hepatic injury in dogs显示文摘AIM: To investigate the protective effect of medical ozone (O3) combined with Traditional Chinese Medicine (TCM) Yigan Fuzheng Paidu Capsules (YC) against carbon tetrachloride (CCl4)-induced hepatic injury in dogs. METHODS: Thirty healthy dogs were divided randomly into fi ve groups (n = 6 in each group), namely control, oleanolic acid tablet (OAT), O3, YC and O3 + YC, given either no particular pre-treatment, oral OAT, medical ozone rectal insulfflation every other day, oral YC, or oral YC plus medical ozone rectal insulfflation every other day, respectively, for 30 consecutive days. After pre-treatment, acute hepatic injury was induced in all dogs with a single-dose intraperitoneal injection of CCl4. General condition and survival time were recorded. The biochemical and hematological indexes of alanine aminotransferase (ALT), aspartate aminotransferase/ alanine aminotransferase (AST/ALT), serum total bilirubin (TBIL), prothrombin time (PT), blood ammonia (AMMO), and blood urea nitrogen (BUN) were measured after CCl4 injection. Hepatic pathological changes were also observed.RESULTS: Compared to the other four groups, the changes of group O3 + YC dogs’ general conditions (motoricity, mental state, eating, urination and defecation) could be better controlled. In group O3 + YC the survival rates were higher (P < 0.05 vs group control). AST/ALT values were kept within a normal level in group O3 + YC. Hepatic histopathology showed that hepatic injury in group O3 + YC was less serious than those in the other four groups. 11Exosomal PD-L1 induces osteogenic differentiation and promotes fracture healing by acting as an immunosuppressant显示文摘A moderate inflammatory response at the early stages of fracture healing is necessary for callus formation.Over-active and continuous inflammation,however,impairs fracture healing and leads to excessive tissue damage.Adequate fracture healing could be promoted through suppression of local over-active immune cells in the fracture site.In the present study,we achieved an enriched concentration of PD-L1 from exosomes(Exos)of a genetically engineered Human Umbilical Vein Endothelial Cell(HUVECs),and demonstrated that exosomes overexpressing PD-L1 specifically bind to PD-1 on the T cell surface,suppressing the activation of T cells.Furthermore,exosomal PD-L1 induced Mesenchymal Stem Cells(MSCs)towards osteogenic differentiation when pre-cultured with T cells.Moreover,embedding of Exos into an injectable hydrogel allowed Exos delivery to the surrounding microenvironment in a time-released manner.Additionally,exosomal PD-L1,embedded in a hydrogel,markedly promoted callus formation and fracture healing in a murine model at the early over-active inflammation phase.Importantly,our results suggested that activation of T cells in the peripheral lymphatic tissues was inhibited after local administration of PD-L1-enriched Exos to the fracture sites,while T cells in distant immune organs such as the spleen were not affected.In summary,this study provides the first example of using PD-L1-enriched Exos for bone fracture repair,and highlights the potential of Hydrogel@Exos systems for bone fracture therapy through immune inhibitory effects.Ze Lin Yuan Xiong Weilin Meng Yiqiang Hu Lili Chen Lang Chen Hang Xue Adriana CPanayi Wu Zhou Yun Sun Faqi Cao Guodong Liu Liangcong Hu Chenchen Yan Xudong Xie Chuanchuan Lin Kaiyong Cai Qian Feng Bobin Mi Guohui Liu 2022Bioactive Materials2022,7,7:4
12Dynamic Airy imaging through high_efficiency broadband phase microelements by femtosecond laser direct writing显示文摘Manipulating Airy beams to symmetric Airy beams(SABs)with abruptly autofocusing and self accelerating properties has attracted much attention.With such a particular propagation dynamic,SABs may provide great potential in dynamic signal imaging.On the other hand,the generation of SABs by spatial light modulators suffers from the limitations of phase gradient accuracy,low optical efficiency(<40%),and a bulky footprint.Therefore,exploring imaging applications and optimal generation methods of these Airy-type beams deserves further research.Here,based on the coordinate transformation of SAB,an asymmetric Airy beam(AAB)is realized.Symmetric/asymmetric cubic phase microplates(SCPPs/ACPPs)are designed and fabricated for generating SAB/AAB.The SCPP/ACPP demonstrates superior performance:compact construction(60μm × 60μm × 1.1μm),continuous variation of phase,high efficiency(~81%at 532 nm),and broadband operation from 405 to 780 nm.Dynamic imaging under monochromatic and polychromatic lights is realized by the SAB/AAB,indicating various results at different propagation distances with a certain initial signal.Further investigation reveals that the SCPP on a soft substrate maintains its physical dimensions and optical properties unchanged during stretching.Our work enables wide potential applications in integrated optics,beam manipulation,and imaging.ZE CAI XINBO QI DENG PAN SHENGYUN JI JINCHENG NI ZHAOXIN LAO CHEN XIN JIAWEN LI YANLEI HU DONG Wu JIARU CHI 2020Photonics Research2020,8,6:3
13First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 14Specially shaped Bessel-like self-accelerating beams along predesigned trajectories显示文摘Over the past several years, spatially shaped self-accelerating beams along different trajectories have been studied extensively. Due to their useful properties such as resistance to diffraction, self-healing, and selfbending even in free space, these beams have attracted great attention with many proposed applications. Interestingly, some of these beams could be designed with controllable spatial profiles and thus propagate along various desired trajectories such as parabolic, snake-like, hyperbolic, hyperbolic secant, three-dimensional spiraling, and even self-propelling trajectories. Experimentally, suchbeams are realized typically by using a spatial light modulator so as to imprint a desired phase distribution on a Gaussian-like input wave front propagating under paraxial or nonparaxial conditions. In this paper, we provide a brief overview of our recent work on specially shaped self-accelerating beams, including Bessel-like, breathing Bessellike, and vortex Bessel-like beams. In addition, we propose and demonstrate a new type of dynamical Bessel-like beams that can exhibit not only self-accelerating but also self-propelling during propagation. 15Optimal Waist Circumference Cut-off values for Identifying Metabolic Risk Factors in Middle-aged and Elderly Subjects in Shandong Province of China显示文摘Objective To study the optimal waist circumference(WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China. Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome(MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men. 17Association of CD14/-260 polymorphism with gastric cancer risk in Highland Tibetans显示文摘AIM:To investigate the relationship between CD14-260and-651 polymorphisms and the risk of developing gastric cancer.METHODS:DNA was extracted from peripheral blood samples obtained from 225 Tibetans with gastric cancer and 237 healthy Tibetans,and analyzed using the polymerase chain reaction/ligase detection(PCR/LDR)method to determine the genotypes at-260 and-651loci of the CD14 promoter.The allele frequencies,genotype frequencies,and haplotypes were analyzed for their association with gastric cancer risk using online SHEsis software.The luciferase reporter assay and point mutation analysis were used to construct in vitro plasmids expressing a C/T homozygote at the-260 lo-cus of the CD14 promoter.RESULTS:The frequencies of CC,CT and TT genotypes in the CD14-260 C/T locus in gastric cancer patients were 19.1%,38.7%and 42.2%,respectively,whereas they were 33.3%,32.5%and 34.2%,respectively,in healthy control subjects.CT genotype carriers were more frequently found among gastric cancer patients than healthy controls(OR=2.076;95%CI:1.282-3.360).Also,TT genotype carriers were more frequently found among gastric cancer patients(OR=2.155;95%CI:1.340-3.466).Compared to the C allele of CD14/-260,the T allele was associated with an increased risk for gastric cancer(OR=1.574;95%CI:1.121-2.045).Furthermore,the frequencies of CC,CT and TT in the CD14-651 C/T locus in gastric cancer patients were 64.4%,29.3%and 6.2%,respectively,while they were 56.5%,35.0%and 8.4%,respectively,in the healthy control subjects(P>0.05).Data obtained using the luciferase reporter assay showed that the p260T homozygote was associated with greater CD14 promoter activity(P<0.01).CONCLUSION:CD14/-260 polymorphism is associated with gastric cancer risk in Highland Tibetans and affects CD14 promoter activity,thereby regulating CD14expression.Kang Li Zeng Dan Xue-Jun Hu Luo-Bu Gesang Yong-Ge Ze Zha-Xi Bianba Cuo-Mu Ciren Yu-Qiang Nie 2014World Journal of Gastroenterology2014,20,10:3
18Insulin-like growth factor 1-induced enolase 2 deacetylation by HDAC3 promotes metastasis of pancreatic cancer显示文摘Enolase 2(ENO2)is a key glycolytic enzyme in the metabolic process of glycolysis,but its potential function in pancreatic ductal adenocarcinoma(PDAC)is unclear.In this study,we observed a significant overexpression of ENO2 in PDAC tissues,and its expression was correlated with metastasis and poor prognosis in PDAC patients.K394 was identified as a major acetylation site in ENO2 that regulates its enzymatic activity,cell metabolism and PDAC progression.Knockdown of ENO2 suppressed tumor growth and liver metastasis in PDAC.Re-expression of wild-type(WT)ENO2,but not the K394 acetylation mimetic mutant,could reverse the decreased tumor malignancy.We further characterized histone deacetylase 3(HDAC3)and P300/CBP-associated factor(PCAF)as the potential deacetylase and acetyltransferase for ENO2,respectively.HDAC3-mediated deacetylation was shown to lead to ENO2 activation and enhancement of glycolysis.Importantly,insulin-like growth factor-1(IGF-1)was found to decrease K394 acetylation and stimulate ENO2 activity in a dose-and time-dependent manner.The PI3K/AKT/mTOR pathway facilitated the phosphorylation of HDAC3 on S424,which promoted K394 deacetylation and activation of ENO2.Linsitinib,an oral small-molecule inhibitor of IGF-1R,could inhibit IGF-1-induced ENO2 deacetylation by HDAC3 and the PI3K/AKT/mTOR pathway.Furthermore,linsitinib showed a different effect on the growth and metastasis of PDAC depending on the overexpression of WT versus K394-mutant ENO2.Our results reveal a novel mechanism by which acetylation negatively regulates ENO2 activity in the metastasis of PDAC by modulating glycolysis.Blockade of IGF-1-induced ENO2 deacetylation represents a promising strategy to prevent the development of PDAC.Yan Zheng Chao Wu Jimeng Yang Yue Zhao Huliang Jia Min Xue Da Xu Feng Yang Deliang Fu Chaoqun Wang Beiyuan Hu Ze Zhang Tianen Li Shican Yan Xuan Wang Peter J.Nelson Christiane Bruns Lunxiu Qin Qiongzhu Dong 2020Signal Transduction and Targeted Therapy2020,5,1:3

20Analysis of Phenolic Compounds in Coke Plant Wastewater by Capillary Zone Electrophoresis with Inhibited Chemiluminescence Detection显示文摘A capillary electrophoresis(CE) with on-line inhibited chemiluminescence (CL) detection was firstly used for the simultaneous analysis of benzenediol isomers and phenol. It is based on the quenching effect of benzenediol isomers and phenol on the chemiluminescence reaction of luminol with potassium ferricyanide in sodium hydroxide medium. Under the optimum conditions,the four phenols were baseline separated and detected in less than 10 min. The detection limits (S/N=3) for hydroquinone,resorcinol,catechol and phenol were 2.9×10-8 mol/L,3.7×10-7 mol/L,8.4×10-8 mol/L and 4.4×10-6 mol/L,respectively.
