| 1 | Identification of ferroptosis as a novel mechanism for antitumor activity of natural product derivative a2 in gastric cancer显示文摘Ferroptosis is a type of cell death accompanied by iron-dependent lipid peroxidation,thus stimulating ferroptosis may be a potential strategy for treating gastric cancer,therapeutic agents against which are urgently required.Jiyuan oridonin A(JDA) is a natural compound isolated from Jiyuan Rabdosia rubescens with anti-tumor activity,unclear anti-tumor mechanisms and limited water solubility hamper its clinical application.Here,we showed a2,a new JDA derivative,inhibited the growth of gastric cancer cells.Subsequently,we discovered for the first time that a2 induced ferroptosis.Importantly,compound a2 decreased GPX4 expression and overexpressing GPX4 antagonized the anti-proliferative activity of a2.Furthermore,we demonstrated that a2 caused ferrous iron accumulation through the autophagy pathway,prevention of which rescued a2 induced ferrous iron elevation and cell growth inhibition.Moreover,a2 exhibited more potent anti-cancer activity than 5-fluorouracil in gastric canc er cell line-derived xenograft mice models.Patient-derived tumor xenograft models from different patients displayed varied sensitivity to a2,and GPX4 downregulation indicated the sensitivity of tumors to a2.Finally,a2 exhibited well pharmacokinetic characteristic s.Overall,our data suggest that inducing ferroptosis is the major mechanism mediating anti-tumor activity of a2,and a2 will hopefully serve as a promising compound for gastric cancer treatment. | Ying Liu Zan Song Yajie Liu Xubin Ma Wang Wang Yu Ke Yichao Xu Dequan Yu Hongmin Liu | 2021 | Acta Pharmaceutica Sinica B2021,11,6: | 10 |
| 2 | Magnetic properties of surface soils across the southern Tarim Basin and their relationship with climate and source materials显示文摘The magnetic susceptibility (χ) of surface soils on the Chinese Loess Plateau (CLP) has strong positive correlation with mean annual precipitation.It is widely accepted that ultrafine magnetite/maghemite grains produced during pedogenesis are responsible for the enhancement of χ,and loess χ on the CLP has been widely used as a proxy for the intensity of the East Asian summer monsoon.However,few works have investigated mechanisms for the enhancement of χ in the case of surface aeolian sediments in westerly-dominated inland China,north of the Tibetan Plateau.Here detailed rock magnetism and grain size studies of 49 surface samples taken across the southern Tarim Basin for different rainfall/temperature/altitude conditions are presented.The results show that samples taken from desert have the lowest χ values and that χ of loessic sand and loess samples decreases with increasing altitude.In addition,the rock magnetism studies suggest that the magnetic properties of surface soils across the southern Tarim Basin are mainly controlled by the concentration of coarse aeolian ferrimagnetic minerals.The contribution of ultrafine pedogenically produced magnetic grains to χ is very limited. | ZAN JinBo FANG XiaoMin NIE JunSheng YANG ShengLi SONG ChunHui DAI Shuang | 2011 | Chinese Science Bulletin2011,56,3: | 5 |
| 3 | Controlled release of hydrogen by implantation of magnesium induces P53-mediated tumor cells apoptosis显示文摘Hydrogen has been used to suppress tumor growth with considerable efficacy.Inhalation of hydrogen gas and oral ingestion of hydrogen-rich saline are two common systemic routes of hydrogen administration.We have developed a topical delivery method of hydrogen at targeted sites through the degradation of magnesium-based biomaterials.However,the underlying mechanism of hydrogen’s role in cancer treatment remains ambiguous.Here,we investigate the mechanism of tumor cell apoptosis triggered by the hydrogen released from magnesium-based biomaterials.We find that the localized release of hydrogen increases the expression level of P53 tumor suppressor proteins,as demonstrated by the in vitro RNA sequencing and protein expression analysis.Then,the P53 proteins disrupt the membrane potential of mitochondria,activate autophagy,suppress the reactive oxygen species in cancer cells,and finally result in tumor suppression.The anti-tumor efficacy of magnesium-based biomaterials is further validated in vivo by inserting magnesium wire into the subcutaneous tumor in a mouse.We also discovered that the minimal hydrogen concentration from magnesium wires to trigger substantial tumor apoptosis is 91.2μL/mm^(3)per day,which is much lower than that required for hydrogen inhalation.Taken together,these findings reveal the release of H2 from magnesium-based biomaterial exerts its anti-tumoral activity by activating the P53-mediated lysosome-mitochondria apoptosis signaling pathway,which strengthens the therapeutic potential of this biomaterial as localized anti-tumor treatment. | Rui Zan Hao Wang Weijie Cai Jiahua Ni Berengere J.C.Luthringer-Feyerabend Wenhui Wang Hongzhou Peng Weiping Ji Jun Yan Jiazeng Xia Yang Song Xiaonong Zhang | 2022 | Bioactive Materials2022,7,3: | 3 |
| 4 | miRNA-dysregulation associated with tenderness variation induced by acute stress in Angus cattle显示文摘miRNAs are a class of small, single-stranded, non-coding RNAs that perform post-transcriptional repression of target genes by binding to 3' untranslated regions. Research has found that miRNAs involved in the regulation of many metabolic processes. Here we uncovered that the beef quality of Angus cattle sharply diversified after acute stress. By performing miRNA microarray analysis, 13 miRNAs were significantly differentially expressed in stressed group compared to control group. Using a bioinformatics method, 135 protein-coding genes were predicted as the targets of significant differentially expressed miRNAs. | 7 | Compound Gaoziban tablet(复方高滋斑片) alleviates depression via toll-like receptor 4/myeloid differentiation factor 88/nuclear factor-kappa B pathway显示文摘OBJECTIVE: To evaluate the effect of compound Gaoziban tablet(复方高滋斑片, CGZBT) on depression, and to investigate the underlying mechanism. METHODS: The components of CGZBT were analysed by high-performance liquid chromatography. Then, we assessed the effects of varying doses of CGZBT on an established chronic unpredictable mild stress(CUMS) model in rats. Whether animals were depressed was evaluated by sucrose preference test, open field test and forced swimming test. Neurotransmitters of hippocampus were detected by liquid chromatography-mass spectrometry. Serum levels of tumor necrosis factor-alpha(TNF-α), interleukin(IL)-1β, IL-6, IL-4, and IL-10 were measured by enzyme-linked immunosorbent assay. Expressions of toll-like receptor 4(TLR4), myeloid differentiation factor 88(My D88), phospho-nuclear factorkappa B(p-NF-κB), cyclooxygenase-2(COX-2), ionized calcium binding adapter molecule-1(IBA-1) were assessed by immunohistochemical staining and western blotting. RESULTS: Eight compounds were identified from CGZBT, moreover, our results showed that CGZBT effectively reversed the CUMS-induced decrease in sucrose preference, shortened the movement distance and prolonged immobility time. CGZBT significantly increased levels of 5-hydroxytryptamine, dopamine, norepinephrine, 5-hydroxyindoleacetic acid levels, and reduced the expression of TNF-α, IL-1β, IL-6, yet increased IL-4 and IL-10. Furthermore, the expressions of TLR4, My D88, COX-2, p-NF-κB and IBA-1 in hippocampus were effectively reversed after treatment with CGZBT. | 9 | Aeolian dust dynamics in the Fergana Valley, Central Asia, since -30 ka inferred from loess deposits显示文摘Knowledge of the interactions among atmospheric dynamics,dust emissions and climate system is essential to understand the physical mechanisms for the dust lifecycle,their role in loess formation as well as the predictions of future dust concentration.However,these issues still remain relatively poorly known in Central Asia(CA).The extensive loess deposits on the CA pediments provide a promising archive to explore atmospheric dust dynamics and climatic conditions in the past and their association with loess formation.This study investigates the granulometric and magnetic properties of a loess section(named Osh section)in the Fergana Valley,which provides a sensitive record of atmospheric dust dynamics since 30 ka based on radiometric(AMS ^(14)C)dating.The frequency-dependent magnetic susceptibility(χfd)and the mean grain size are used to reconstruct the broadscale effective moisture and summer atmospheric dynamics pattern in CA,respectively.The results show that the precession forcing exerts a huge influence on the wind-regime variabilities in CA,but with different physical processes under the impact of the Northern Hemisphere ice sheet(NHIS)before and after 15 ka.The origin of the sedimentation rate variations in the Osh loess is also linked to the NHIS-modulated changes of the atmospheric circulation patterns.Either the strengthened westerlies or the increased surface roughness from higher vegetation cover in loess-deposition areas have significantly accelerated the loess accumulation.As a result,these complicated influence factors of sedimentation rate change in the Osh loess section,especially during the Holocene epoch,may hamper accessibility of the authentic dust emission flux and atmospheric dust concentration in CA. | Yue Li Yougui Song Dimitris G.Kaskaoutis Jinbo Zan Rustam Orozbaev Liangcheng Tan Xiuling Chen | 2021 | Geoscience Frontiers2021,12,5: | 1 |

| 15 | New insights on the age of the Mengyejing Formation in the Simao Basin,SE Tethyan domain and its geological implications显示文摘Knowledge on the origin and mechanism of the large potash formation in the southeast Tethyan tectonic domain are still under debate,one of which mainly due to the controversial ages of the potash-bearing strata.Here we present a detailed detrital zircon U-Pb geochronologic-magnetostratigraphic study of the potash-bearing Mengyejing(MYJ)Formation(Fm)from the Simao Basin.948 paleomagnetic cores and 4 detrital zircon U-Pb samples were collected from a 932-m thick Jiangcheng section.Thermal demagnetization isolated 857 primary characteristic remnant magnetization(ChRM)directions,indicated by positive reversals and fold tests and petrologic examinations.Seven normal and seven reversed polarity zones were identified and correlated well with chrons C27r to C34n of the geomagnetic polarity timescale(GPTS2012),yielding the first known age sequence of>112 to~63 Ma for the MYJ Fm.The ages for the potash bearing strata in the Simao commenced~20 Ma earlier than those in the adjacent Khorat basins,opposite to the traditional viewpoint that the former would rather correspond to the middle and lower parts of the latter.Given the paleogeographic reconstruction of likely a pan-Simao-Khorat basin during the Cretaceous,~85 Ma would be another important potential period for future potash exploration in the Simao Basin. | Maodu YAN Dawen ZHANG Xiaomin FANG Weilin ZHANG Chunhui SONG Chenglin LIU Jinbo ZAN Miaomiao SHEN | 2021 | Science China Earth Sciences2021,64,2: | 1 |
| 16 | Modified graphite filled natural rubber composites with good thermal conductivity显示文摘The rubber composites with good thermal conductivity contribute to heat dissipation of tires. Graphite filled natural rubber composites were developed in this study to provide good thermal conductivity. Graphite was coated with polyacrylate polymerized by monomers including methyl methacrylate, n-butyl acrylate and acrylic acid. The ratios between a filler and acrylate polymerization emulsion and those between monomers were varied. Eight types of surface modification formulas were experimentally investigated. Modification formula can affect coating results and composite properties greatly. The best coating type was achieved by a ratio of1:1 between methyl methacrylate and n-butyl acrylate. The coating of graphite was thermally stable in a running tire. Filled with modified graphite, the tire thermal conductivity reached up to 0.517–0.569 W·m-1·K-1. | 17 | Integrating transcriptomic and proteomic analyses of photoperiod-sensitive in near isogenic maize line under long-day conditions显示文摘As a short-day plant species, maize requires an optimal photoperiod for inducing reproductive growth. However, there is a lack of information regarding photoperiod-induced changes in maize mRNA and protein levels. In this study, a photoperiod-insensitive maize inbred line and its near isogenic photoperiod-sensitive line were used. By integrating RNAbased transcriptomic and iTRAQ LC-MS/MS-based proteomic approaches, we generated a comprehensive inventory of the transcripts and proteins with altered abundances in response to a long photoperiod(LP) during growth stage transitions. We detected 22 000 transcripts in RNA-sequence runs and 5 259 proteins from an iTRAQ-based analysis. A weak correlation between mRNA-and protein-level changes was observed, suggesting the LP-induced transition between maize growth stages is largely regulated post-transcriptionally. Differentially expressed genes influenced by LP conditions were associated with several regulatory processes in both maize inbred lines, especially phosphate ion transport and the circadian rhythm. Additionally, 31 transcripts and six proteins related to photoperiodic flowering in maize were identified by comparing transcriptomic and proteomic data.
