1Netrin-1 works with UNC5B to regulate angiogenesis in diabetic kidney disease显示文摘Netrin-1,an axon guidance factor,and its receptor UNC5B play important roles in axonal development and angiogenesis.This study examined netrin-1 and UNC5B expression in kidneys with diabetic kidney disease (DKD) and investigated their roles in angiogenesis.Netrin-1 and UNC5B were upregulated in streptozotocininduced DKD Wistar rats,and their expression was compared with that in healthy controls.However,exogenous netrin-1 in UNC5B-depleted human renal glomerular endothelial cells (HRGECs) inhibited cell migration and tubulogenesis.This effect was likely associated with SRC pathway deactivation.Netrin-1 treatment also eliminated the pro-angiogenic effects of exogenous VEGF-165 on UNC5B-silenced HRGECs.These results indicate that UNC5B antagonizes netrin-1 and that UNC5B upregulation contributes partly to enhancing angiogenesis in DKD.Therefore,introducing exogenous netrin-1 and depleting endogenous UNC5B are potential strategies for reducing the incidence of early angiogenesis and mitigating kidney injury in DKD.Xiaojing Jiao Dong Zhang Quan Hong Lei Yan Qiuxia Han Fengmin Shao Guangyan Cai Xiangmei Chen Hanyu Zhu 2020Frontiers of Medicine2020,14,3:7
2Hydrologic balance, net primary productivity and water use efficiency of the introduced exotic Eucalyptus grandis × Eucalyptus urophylla plantation in south-western China显示文摘Aims Land cover changes can disrupt water balance and alter the partitioning of precipitation into surface runoff,evapotranspiration and groundwater recharge.The widely planted Eucalyptus trees in south-western China have the potential to bring about hydrologic impacts.Our research aims to elucidate the hydrologic balance characteristics of the introduced exotic Eucalyptus grandis×Eucalyptus urophylla plantation and to assess whether its high productivity results from high water use efficiency(WUE)or large water consumption.Methods A 400-m^(2) experimental plot was established in an E.grandis×E.urophylla plantation in south-western China.Water balance components,including stand transpiration(T_(r)),evapotranspiration(E_(t))and runoff(R)were obtained as follows:T_(r) was estimated based on sap flow measurements,E_(t) was estimated as the average of surface transpiration and evaporation weighted by the fractional green vegetation cover using a modeling approach,and R was collected using the installed metal frame.Net primary productivity(NPP)was obtained from allometric equation and annual diameter at breast height(DBH)increment determination.Important Findings Annual E_(t) and T_(r) were 430±31 and 239±17 mm,respectively.Annual T_(r) accounts for 56±8%of total evapotranspiration on average.WUE(NPP/T_(r))of the E.grandis×E.urophylla was estimated to be 3.3–3.9 mmol·mol^(−1).Based on the comparative analysis of T_(r) and WUE,E.grandis×E.urophylla had a high productivity due to its high WUE without exhibiting prodigal water use.Meteorological factors including vapor pressure deficit and global solar radiation(R_(s))were key factors regulating E_(t) and T_(r) in our research site.Annual surface runoff,E_(t) and canopy interception occupied 7%,27–30%and 16%of total precipitation,while the remaining 46–50%of precipitation was used for sustaining groundwater recharge and altering soil water storage.The higher runoff coefficient(7.1%)indicated the weaker capability of E.grandis×E.urophylla to reserve water resource than natural forests and less disturbed plantations.The planting and protection of understory vegetation may decrease the surface runoff and exert beneficial effects on water conservation capacity of Eucalyptus plantation.Yanting Hu Ping Zhao Yuqing Huang Liwei Zhu Guangyan Ni Xiuhua Zhao Zhihong Huang 2019Journal of Plant Ecology2019,12,6:2
3Comparative analysis of membranous and other nephropathy subtypes and establishment of a diagnostic model显示文摘This study aimed to compare clinical features between membranous nephropathy (MN) and nonmembranous nephropathy (non-MN), to explore the clinically difTerential diagnosis of these two types, and to establish a diagnostic model of MN. After renal biopsy was obtained, 798 patients were divided into two groups based on their examination results: primary MN group (n = 248) and non-MN group (n = 550). Their data were statistically analyzed. Logistic regression analysis indicated that anti-PLA2R antibodies, IgG, and Cr were independently correlated with MN, and these three parameters were then used to establish the MN diagnostic model. A receiver operating characteristic (ROC) curve confirmed that our diagnostic model could distinguish between patients with and without MN, and their corresponding sensitivity, specificity, and AUC were 79.9%, 89.4%, and 0.917, respectively. The cutoff value for this combination in MN diagnosis was 0.34. 6Cold-induced Yes-associated-protein expression through miR-429 mediates the browning of white adipose tissue显示文摘Targeting the white-to-brown fat conversion is important for developing potential strategies to counteract metabolic diseases;yet the mechanisms are not fully understood.Yes-associated-protein(YAP),a transcription co-activator,was demonstrated to regulate adipose tissue functions;however,its effects on browning of subcutaneous white adipose tissue(sWAT)are unclear.We demonstrated that YAP was highly expressed in cold-induced beige fat.Mechanistically,YAP was found as a target gene of miR-429,which downregulated YAP expression in vivo and in vitro.In addition,miR-429 level was decreased in cold-induced beige fat.Additionally,pharmacological inhibition of the interaction between YAP and transcriptional enhanced associate domains by verteporfin dampened the browning of sWAT.Although adipose tissue-specific YAP overexpression increased energy expenditure with increased basal uncoupling protein 1 expression,it had no additional effects on the browning of sWAT in young mice.However,we found age-related impairment of sWAT browning along with decreased YAP expression.Under these circumstances,YAP overexpression significantly improved the impaired WAT browning in middle-aged mice.In conclusion,YAP as a regulator of sWAT browning,was upregulated by lowering miR-429 level in cold-induced beige fat.Targeting the miR-429-YAP pathway could be exploited for therapeutic strategies for age-related impairment of sWAT browning.Chenji Ye Jinjie Duan Xuejiao Zhang Liu Yao Yayue Song Guangyan Wang Qi Li Biqing Wang Ding Ai Chunjiong Wang Yi Zhu 2021Science China(Life Sciences)2021,64,3:1

13Interleukin-13 promotes cellular senescence through inducing mitochondrial dysfunction in IgG4-related sialadenitis显示文摘Immunoglobulin G4-related sialadenitis(IgG4-RS)is an immune-mediated fibro-inflammatory disease and the pathogenesis is still not fully understood.The aim of this study was to explore the role and mechanism of interleukin-13(IL-13)in the cellular senescence during the progress of IgG4-RS.We found that the expression of IL-13 and IL-13 receptorα1(IL-13Rα1)as well as the number of senescent cells were significantly higher in the submandibular glands(SMGs)of IgG4-RS patients.IL-13 directly induced senescence as shown by the elevated activity of senescence-associatedβ-galactosidase(SA-β-gal),the decreased cell proliferation,and the upregulation of senescence markers(p53 and p16)and senescence-associated secretory phenotype(SASP)factors(IL-1βand IL-6)in SMG-C6 cells.Mechanistically,IL-13 increased the level of phosphorylated signal transducer and activator of transcription 6(p-STAT6)and mitochondrial-reactive oxygen species(mt ROS),while decreased the mitochondrial membrane potential,ATP level,and the expression and activity of superoxide dismutase 2(SOD2).Notably,the IL-13-induced cellular senescence and mitochondrial dysfunction could be inhibited by pretreatment with either STAT6 inhibitor AS1517499 or mitochondria-targeted ROS scavenger Mito TEMPO.Moreover,IL-13 increased the interaction between p-STAT6 and c AMP-response element binding protein(CREB)-binding protein(CBP)and decreased the transcriptional activity of CREB on SOD2.Taken together,our findings revealed a critical role of IL-13 in the induction of salivary gland epithelial cell senescence through the elevated mitochondrial oxidative stress in a STAT6–CREB–SOD2-dependent pathway in IgG4-RS.Mengqi Zhu Sainan Min Xiangdi Mao Yuan Zhou Yan Zhang Wei Li Li Li Liling Wu Xin Cong Guangyan Yu 2022International Journal of Oral Science2022,14,3:1

17PLX8394,a RAF inhibitor,inhibits enterovirus 71 replication by blocking RAF/MEK/ERK signaling显示文摘Enterovirus 71(EV71)poses a serious threat to human health,with scattered outbreaks worldwide.There are several vaccines against a few EV71 strains but no efficient drug for the treatment of EV71 infection.Therefore,it is urgent and of significance to develop anti-EV71 drugs.Here,we found that PLX8394,a RAF inhibitor,possesses high antiviral activity against EV71 in vitro,being superior to the traditional clinical drug ribavirin.Moreover,PLX8394 exhibits broad-spectrum antiviral activity against enteroviruses.Notably,in a suckling mouse model,PLX8394 provided a 70%protection rate for EV71-infected mice,reduced the viral load in liver and heart tissues,and relieved the inflammatory response.A mechanistic study showed that PLX8394 inhibited EV71 by suppressing the RAF/MEK/ERK signaling pathway.Thus,PLX8394 lays a foundation for the development of new drugs against EV71.Chengyuan Wu Guangyan Zhu Fang Qiu Fuli Ren Binbin Lin Dingyu Zhang Qingyu Yang Chaolin Huang 2023Virologica Sinica2023,38,2:0
18The microbuckling failure of Dyneema~? composites under compression显示文摘Two grades of Dyneema~?composite laminates with the commercial designations of HB26 and HB50 were cut into blocks with or without an edge crack and compressed in the longitudinal fiber direction. The cracked and uncracked specimens show similar compressive responses including failure pattern and failure load. The two grades of Dyneema~? composites exhibits different failure modes: a diffuse, sinusoidal buckling pattern for Dyneema~? HB50 due to its weak matrix constituent and a kink band for Dyneema~? HB26 due to its relatively stronger matrix constituent. An effective finite element model is used to simulate the collapse of Dyneema~? composites, and the sensitivity of laminate compressive responses to the overall effective shear modulus, interlaminar shear strength, thickness and imperfection angle are investigated. 19Water transport of native and exotic tree species in relation to xylem anatomical characteristics in low subtropical China显示文摘Aims Exotic fast-growing tree species have been commonly planted as pioneer species to facilitate ecological restoration in South China.Their growth and resource utilization behavior related to intrinsic physiology and structural properties have profound influences on forest ecosystem.However,the contrastive research focusing on water utilization features along with xylem anatomical properties between native and exotic species is scarce in South China.The objective of this study is to investigate the sapwood anatomical characteristics and water utilization conditions of native and exotic fast-growing species,and to elucidate the relationship between sap-flux density and conduit features.Methods We measured sap-flux density,conduit length,diameter and density of four native species(Schima superba,Michelia mac-clurei,Castanopsis hystrix and Castanopsis fissa)and four exotic species(Eucalyptus citriodora,Eucalyptus urophylla×grandis,Acacia auriculaeformis and Acacia mangium).Sap flux density was measured based on the Granier’s thermal dissipation probe method.The whole-tree water transport was quantified by mul-tiplying sap-flux density by sapwood area.The measurements of conduit characteristics were conducted by using segregation and slice method.Important Findings Sapwood area increased with the growing diameter at breast height(DBH)as a power function.Native species had a larger water-conducting tissue area than exotic species at the same DBH value when trees grew to a size with a certain value of DBH.The con-duit diameter of exotic species was significantly larger than that of native species.Conversely,native species,such as S.superba and M.macclurei,had longer conduit length and higher conduit den-sity than other tree species.Based on a physiological interpretation of the measured conduit characteristics,native tree species devel-oped a safe water transport system while exotic fast-growing tree species come into being an efficient system instead.Water trans-port increased with the growing DBH as a power function,and the exponent for native species(1.60)was higher than that for exotic species(1.22).Under the combined impact of sap-flux density and sapwood area,native species presented a larger water transport at a larger DBH value,indicating that growth advantage of exotic fast-growing species might weaken as DBH increased.Yanting Hu Jianguo Gao Ping Zhao Weijun Shen Peiqiang Zhao Liwei Zhu Guangyan Ni Junfeng Niu Lei Ouyang 2018Journal of Plant Ecology2018,11,3:0
20miR-378 and its host gene Ppargc1β exhibit independent expression in mouse skeletal muscle显示文摘MicroRNAs (miRNAs) are implicated in multiple biological processes in physiological and pathological settings. Nearly half of the known miRNAs are classified as ‘intronic’ miRNAs because they are embedded within the introns of protein-coding or noncoding genes. Such miRNAs were thought to be processed from primary host gene transcripts and share the promoter of their host. Recent analyses predicted that some intronic miRNAs might be transcribed and regulated as independent units, but there is little direct evidence for this in a specific biological context. Here, we focused on miR-378, which is located within the first intron of the peroxisome proliferator-activated receptor γ coactivator 1-beta (Ppargc1β) gene and critically regulates skeletal muscle cell differentiation and muscle regeneration. We demonstrate that miR-378 and Ppargc1β exhibit distinct expression patterns during skeletal muscle cell differentiation. In terminally differentiated adult skeletal muscle tissues of mice, miR-378 is predominantly expressed in glycolytic muscle, whereas Ppargc1β is mainly expressed in oxidative soleus muscle. Mechanistically, miR-378, but not Ppargc1β, is regulated by the transcription factor, MyoD, in muscle cells.
