1pORF5 plasmid protein of Chlamydia trachomatis induces MAPK-mediated pro-inflammatory cytokines via TLR2 activation in THP-1 cells显示文摘Infection with Chlamydia trachomatis induces inflammatory pathologies in the urogenital tract that can lead to infertility and ectopic pregnancy.Pathogenesis of infection has been mostly attributed to excessive cytokine production.However,precise mechanisms on how C.trachomatis triggers this production,and which protein(s) stimulate inflammatory cytokines remains unknown.In the present study,the C.trachomatis pORF5 protein induced tumor necrosis factor alpha(TNF-α),interleukin-1 beta(IL-1β) and interleukin-8(IL-8) in dose-and time-dependent manners in the THP-1 human monocyte cell line.We found that intracellular p38/mitogen-activated protein kinase(MAPK) and extracellular signal-regulated kinase(ERK)/MAPK signaling pathways were required for the induction of TNF-α,IL-1β and IL-8.Blockade of toll-like receptor 2(TLR2) signaling reduced induction levels of TNF-α,IL-8 and IL-1β.We concluded that the C.trachomatis pORF5 protein might contribute to the inflammatory processes associated with chlamydial infections.ZHOU Hui HUANG QiuLin LI ZhongYu WU YiMou XIE XiaoBing MA KangKang CAO WenJuan ZHOU Zhou LU ChunXue ZHONG GuangMing 2013Science China(Life Sciences)2013,56,5:26
2Effects of surfactants on enzyme-containing reversed micellar system显示文摘With the development of colloid interface and enzyme technologies,enzyme-containing reversed micellar system has been receiving much attention in bioseparation and bioconversion. Because of its high efficiency,it has brought new opportunities for the development of molecular biotechnology. Reversed micelles represent nano-sized aqueous droplets stabilized by surfactant amphiphiles inside the bulk organic solvents. The entrapped enzymes have enhanced activities under those conditions as suited in the lipid bilayers of biological membranes. The fundamentals of enzyme-containing reversed micellar system are described in this paper,with special emphasis on the effects of surfactants varying in concentrations and structures. The latest study progress on the surfactants application in enzyme-containing reversed micelles is reviewed. 3Localization and characterization of the hypothetical protein CT440 in Chlamydia trachomatis-infected cells显示文摘The inclusion membrane proteins play potentially important roles in chlamydial biology and pathogenesis.Here we localized and characterized the hypothetical protein CT440 in Chlamydia trachomatis-infected cells.The open reading frame(ORF) encoding the CT440 protein from the C.trachomatis serovar D genome was cloned into the prokaryotic expression vector pGEX-6p and expressed as a glutathione-S-transferase(GST) fusion protein in E.coli XL1-Blue.The CT440 fusion protein was used to immunize mice to raise antigen-specific antibody.After verification by Western blot and immunofluorescence assay(IFA),the specific antibody was used to localize the endogenous CT440 protein and to detect its expression pattern in Chlamydia-infected cells.Cytosolic expression of CT440 in HeLa cells was also carried out to evaluate the effect of the CT440 protein on the subsequent chlamydial infection.The results showed that the hypothetical protein CT440 was localized in the C.trachomatis inclusion membrane,and was detectable 12 h after chlamydial infection.Expression of CT440 in the cytoplasm did not inhibit the subsequent chlamydial infection.In summary,we have identified a new inclusion membrane protein that may be an important candidate for understanding C.trachomatis pathogenesis.LI ZhongYu HUANG QiuLin SU ShengMei ZHOU Zhou CHEN ChaoQun ZHONG GuangMing WU YiMou 2011Science China(Life Sciences)2011,54,11:5
4Aberrant Resting-State Functional Connectivity in the Default Mode Network in Pediatric Bipolar Disorder Patients with and without Psychotic Symptoms显示文摘Mood disorders/psychosis have been associated with dysfunctions in the default mode network(DMN).However,the relative contributions of DMN regions to state and trait disturbances in pediatric bipolar disorder(PBD)remain unclear.The aim of this study was to investigate the possible mechanisms of PBD through brain imaging and explore the influence of psychotic symptoms on functional alterations in PBD patients.Twenty-nine psychotic and 26 non-psychotic PBD patients,as well as 19 age-and sex-matched healthy controls underwent a restingstate functional MRI scan and the data were analyzed by independent component analysis.The DMN component from the fMRI data was extracted for each participant.Spearman's rank correlation analysis was performed between aberrant connectivity and clinical measurements.The results demonstrated that psychotic PBD was characterized by aberrant DMN connectivity in the anterior cingulate cortex/medial prefrontal cortex,bilateral caudate nucleus,bilateral angular gyri,and left middle temporal gyrus,while non-psychotic PBD was not,suggesting further impairment with the development of psychosis.In summary,we demonstrated unique impairment in DMN functional connectivity in the psychotic PBD group.These specific neuroanatomical abnormalities may shed light on the underlying pathophysiology and presentation of PBD.Yuan Zhong Chun Wang Weijia Gao Qian Xiao Dali Lu Qing Jiao Linyan Su Guangming Lu 2019Neuroscience Bulletin2019,35,4:5
5A mathematical modeling study of the HIV epidemics at two rural townships in the Liangshan Prefecture of the Sichuan Province of China显示文摘Background:As a response to a severe HIV epidemic in the Liangshan Prefecture,one of the worst in China,population based HIV interventions,including two population-wide HIV screening,have been carried out since 2005 at two townships in a remote mountainous region of Liangshan.The objective of our mathematical modeling study is to assess the temporal dynamics of the HIV epidemic in the two townships based on the data collected in the study area during the period 2005e2010.Methods:A mathematical model was set up to describe the population dynamics of HIV transmission in study area.The model was calibrated by fitting it to the HIV testing and treatment data from 2005 to 2008.Validation of the model was done by comparing its predicted value of HIV prevalence in 2010 to the prevalence data obtained in the 2010 population wide HIV testing.The validated model was used to produce estimation of HIV incidence,prevalence and death.Results:Our model estimations show that population-based HIV interventions have significantly slowed down the rise of the HIV epidemic in the two townships.Over the five-year period from 2005 to 2010,the year-over-year rate of increase in HIV incidence,prevalence,and death has declined by 91.5%,28.7%,and 52.3%,respectively.Conclusion:Mathematical models,when integrated with epidemiological and surveillance data,can be an effective tool for predicting the temporal dynamics of HIV and assessing the impacts of HIV interventions.Zhimin Su Caiting Dong Ping Li Hongxia Deng Yuhan Gong Shiyong Zhong Min Wu Yuhua Ruan Guangming Qin Wen Yang Yiming Shao Michael Li 2016Infectious Disease Modelling2016,1,1:2

12Analysis of PR and near-PR cosine-modulated filter banks with odd and even numbers of channels显示文摘It is well known that M-channel filter banks, satisfying either the perfect-reconstruction (PR) or near-PR property, can be obtained by cosine modulation of a linear-phase prototype filter. In this work, we investigated the effects of choosing odd or even number of channels on the performance of both PR and near-PR cosine-modulated filter banks (CMFBs). Given the same m, a performance degradation happens in the PR case with odd numbered M. Contrarily, the choice of even or odd M has little effects on the whole system quality for near-PR one. In addition, we made some comparisons between the constrained optimization and the Parks-McClellan algorithm with cosine roll-off characteristic in designing near-PR CMFBs. Detailed analyses and numerical comparisons show that the PR and near-PR systems have their individual characteristics in terms of M and m selection. 14Method of determining grid sizes of finite element method of water quality models for geometrically complex river course显示文摘The method of determining grid sizes of finite element method of water quality models is presented on the basis of characteristics of pollutant distributed in the just downstream grid of discharge. A given length of the grid is applied to calculate the minimum distance for the pollutant to complete mixing in the lateral direction in the just downstream grid of the discharge, and the minimum distance is regarded as the maximum width of the grid. Application of the maximum width of the grid and flow rate per unit width of the river results in the number of flow zones in the lateral direction of the river. Consequently division of grids and numerical calculation of water quality with finite element method can be carried out on the basis of available method and topographic map of the river course. Analysis of sensitivity shows that the effect of the width of the river and the effect of the length of the grids on the minimum number of flow zones in the lateral direction is relatively small, and that the effect of the given lateral mixing degree of the pollutants on the minimum number of flow zones is acceptable, which indicates that the method presented is relatively stable. The method can avoid the blindness and randomness in determining the number of flow zones, and can decrease the amount of computation under the condition that the accuracy requirement is met. 18Successive digestion of tilapia collagen by serine proteinase and proline specific endopeptidase to produce novel angiotensin l-converting enzyme inhibitory peptides显示文摘Serine proteinase,purified from the hepatopancreas of Pacific white shrimp(Litopenaeus vannamei), was used to hydrolyze acid solubilized collagen(ASC)isolated from Nile tilapia(Oreochromis sp.)skin to produce angiotensin I-converting enzyme(ACE)inhibitory peptides(ACEIPs).A series of column chromatography assays were used to separate the ACEIPs.A peptide,NPARTCR,was isolated as it exhibited high ACE inhibition potential.Further digestion of this peptide by a proline specific endopeptidase(PSEP),produced a pentapeptide ARTCR with ACE inhibitory activity(IC_(50))of 77.0 pmol/L.Both NPARTCR and ARTCR inhibited ACE in a non-competitive manner.An in vivo study in rats demonstrated that ARTCR has ACE inhibitory activity via lowering systolic blood pressure in spontaneously hypertensive rats(SHRs).These results suggest that processing by-products from shrimp and tilapia are ideal raw materials for the production of serine proteinase and collagen,respectively.Serine proteinase and collagen are both ideal raw materials that can be used to derive ACE inhibitory active peptides against hypertension.Xin Hua Lechang Sun Chan Zhong Qiang Wu Panpan Xiao Asami Yoshida Guangming Liu Minjie Cao 2020Marine Life Science & Technology2020,2,3:0
19Deep-Learning-Assisted Underwater 3D Tactile Tensegrity显示文摘The growth of underwater robotic applications in ocean exploration and research has created an urgent need for effective tactile sensing.Here,we propose an underwater 3-dimensional tactile tensegrity(U3DTT)based on soft self-powered triboelectric nanogenerators and deep-learning-assisted data analytics.This device can measure and distinguish the magnitude,location,and orientation of perturbations in real time from both flow field and interaction with obstacles and provide collision protection for underwater vehicles operation.It is enabled by the structure that mimics terrestrial animals’musculoskeletal systems composed of both stiff bones and stretchable muscles.Moreover,when successfully integrated with underwater vehicles,the U3DTT shows advantages of multiple degrees of freedom in its shape modes,an ultrahigh sensitivity,and fast response times with a low cost and conformability.The real-time 3-dimensional pose of the U3DTT has been predicted with an average root-mean-square error of 0.76 in a water pool,indicating that this developed U3DTT is a promising technology in vehicles with tactile feedback.Peng Xu Jiaxi Zheng Jianhua Liu Xiangyu Liu Xinyu Wang Siyuan Wang Tangzhen Guan Xianping Fu Minyi Xu Guangming Xie Zhong Lin Wang 2023Research2023,,3:0
