| 1 | Thermal regimes and degradation modes of permafrost along the Qinghai-Tibet Highway显示文摘Permafrost on the Qinghai-Tibet Plateau (QTP) is widespread, thin, and thermally unstable. Under a warming climate during the past few decades, it has been degrading extensively with generally rising ground temperatures, the deepening of the maximum summer thaw, and with lessening of the winter frost penetration. The permafrost has degraded downward, upward and laterally. Permafrost has thinned or, in some areas, has totally disappeared. The modes of permafrost degradation have great significance in geocryology, in cold regions engineering and in cold regions environmental management. Permafrost in the interior of the QTP is well represented along the Qing-hai-Tibet Highway (QTH), which crosses the Plateau through north to south and traverses 560 km of permafrost-impacted ground. Horizontally, the degradation of permafrost occurs more visibly in the sporadic permafrost zone in the vicinity of the lower limit of permafrost (LLP), along the margins of taliks, and around permafrost islands. Downward degradation develops when the maximum depth of seasonal thaw exceeds the maximum depth of seasonal frost, and it generally results in the formation of a layered talik disconnecting the permafrost from the seasonal frost layer. The downward degrada- tion is divided into four stages: 1) initial degradation, 2) accelerated degradation, 3) layered talik and 4) finally the conversion of permafrost to seasonally frozen ground (SFG). The upward degradation occurs when the geothermal gradient in permafrost drops to less than the geothermal gradients in the underlying thawed soil layers. Three types of permafrost temperature curves (stable, degrading, and phase-changing transitory permafrost) illustrate these modes. Although strong differentiations in local conditions and permafrost types exist, the various combinations of the three degradation modes will ultimately transform permafrost into SFG. Along the QTH, the downward degradation has been proceeding at annual rates of 6 to 25 cm, upward degradation at 12 to 30 cm, and lateral degradation in the sporadic permafrost zone at 62 to 94 cm during the last quarter century. | 3 | Cardiac autonomic nerve fiber regeneration in chronic heart failure Do Akt gene-transduced mesenchymal stem cells promote repair?显示文摘BACKGROUND: Transplantation of Akt-over-expressing mesenchymal stem cells (Akt-MSCs) has been shown to repair infarcted myocardium and improve cardiac function. However, little is known about the therapeutic effects of Akt-MSCs on cardiac autonomic neuropathy in chronic heart failure (CHF). OBJECTIVE: The present study used adriamycin-induced CHF rat models to observe the effect of Akt-MSCs on cardiac autonomic nervous regeneration and the factors mediating this effect. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Central Laboratory of Basic Medical College, China Medical University, between September 2008 and April 2009. MATERIALS: Rabbit anti-choline acetyltransferase (ChAT), growth associated protein-43 (GAP-43), synaptophysin (SYN) polyclonal antibodies and the secondary antibody (goat anti-rabbit IgG) were purchased from Boster, China. Cat-A-Kit assay system was provided by Amersham, USA. METHODS: (1) Adult rat MSCs were isolated and cultured for the preparation of Akt-MSCs. (2) Forty male Wistar rats were intramyocardially administered adriamycin at 2 mg/kg over 3 days for a total of five times and once a week for additional five times thereafter to establish CHF models. At 2 weeks after final adriamycin treatment, 34 successful CHF rat models were randomized to three groups: Akt-MSCs (n = 11), simple MSCs (s-MSCs, n =11), and control (n = 12). Each group was intravenously administered Akt-MSCs (2×106 cells in 100 μL PBS), s-MSCs (2×106 cells in 100 μL PBS) or equal volume of phosphate buffered saline, once a day for a total of three times. MAIN OUTCOME MEASURES: At 4 weeks after final adriamycin treatment, myocardial norepi- nephrine (NE) content was detected using a Cat-A-Kit assay system. Myocardial ChAT, SYN and GAP-43 were performed by immunohistochemistry and Western blot analysis. Prior to, 2 and 4 weeks after adriamycin treatment, echocardiographic examination was performed and left ventricular ejection fraction (LVEF) was determined. RESULTS: Myocardial NE content, as well as SYN-positive and GAP-43-positive nerve fiber density and expression, and LVEF, was the greatest in the Akt-MSCs group, followed by the s-MSCs group, and lastly the control group (P < 0.05 or P < 0.01). ChAT expression was similar between Akt-MSCs and s-MSCs groups, but it was higher compared with the control group (P < 0.05). NE contents were negatively correlated to LVEF (r = -0.64, P = 0.015). | 5 | Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers. | 7 | Meaningful comparison of photocatalytic properties of {001} and {101} faceted anatase TiO2 nanocrystals显示文摘The facet-dependent photocatalytic performance of TiO_2 nanocrystals has been extensively investigated due to their promising applications in renewable energy and environmental fields. However, the intrinsic distinction in the photocatalytic oxidation activities between the {001}and {101} facets of anatase TiO_2 nanocrystals is still unclear and under debate. In this work, a simple photoelectrochemical method was employed to meaningfully quantify the intrinsic photocatalytic activities of {001} and{101} faceted TiO_2 nanocrystal photoanodes. The effective surface areas of photoanodes with different facets were measured based on the monolayer adsorption of phthalic acid on TiO_2 photoanode surface by an ex situ photoelectrochemical method, which were used to normalize the photocurrents obtained from different faceted photoanodes for meaningful comparison of their photocatalytic activities. The results demonstrated that the {001} facets of anatase TiO_2 nanocrystals exhibited much better photocatalytic activity than that of {101} facets of anatase TiO_2 nanocrystals toward photocatalytic oxidation of water and organic compounds with different functional groups(e.g.,–OH, –CHO, –COOH). In order to optimize the relative volume fraction of secondary phases, the as-HIPed ODS steel was annealed at 800°C, 1000°C, 1200°C for 5 h, respectively. The microstructures and different secondary phases of the as-HIPed and annealed ODS samples were identified by scanning electron microscopy(SEM), transmission electron microscopy(TEM), and X-ray diffraction(XRD). The tensile properties of all the ODS steels at room temperature were also investigated. The results indicate that annealing is an effective way to control the microstructure and the integral secondary phases. The annealing process promotes the dissolution of M23C6 particles, thus promoting the precipitation of TiC.No obvious coarsening of Y2Ti2O7 nanoparticles can be observed during annealing. This functionalized nickel catalyst demonstrated an enhanced catalytic performance for selective hydrogenation of FAL, in which almost 100% FOL(<80℃) and THFOL(>100℃) selectivity with complete conversion was obtained, respectively. More importantly, the conversion of transfer hydrogenation of FAL to FOL also can reach almost 100% under optimal conditions(140℃, 4.0 h). In DAFPN, the attention mechanism is introduced by calculating the topdown pathway and lateral pathway, where the spatial attention, as well as channel attention, would participate, respectively, such that the pyramidal feature maps can be generated with enhanced spatial and channel interdependencies, which bring more semantical information for the feature pyramid. Herein, we review the recent progress in synthesizing {001} facets dominated anatase TiO2 crystals with different morphologies by various synthetic methods. Furthermore, our review is mainly focused on the formation/etching mechanisms of {001} facets dominated anatase TiO2 crystals based on our and other studies. The extensive application potentials of the anatase TiO2 crystals with exposed {001} facets have been summarized in this review such as photocatalysis, photoelectrocatalysis, solar energy conversion, lithium ion battery, and hydrogen generation. | 18 | Layered Fe(Ⅲ) doped TiO_2 thin-film electrodes for the photoelectrocatalytic oxidation of glucose and potassium hydrogen phthalate显示文摘Four types of TiO 2 thin-film electrodes were fabricated from TiO 2 and Fe(III) doped TiO 2 sols using a layer-by-layer dip-coating technique. Electrodes fabricated were TF (pure TiO 2 surface, Fe(III)-TiO 2 bottom layer), FT (Fe(III)-TiO 2 surface, pure TiO 2 bottom layer), TT (both layers pure TiO 2 ) and FF (both layers Fe(III)-TiO 2 ). The photoelectrochemical behavior of these electrodes was characterized using linear sweep voltammetry (LSV), electrochemical impedance spectroscopy (EIS) and steady-state photocurrent measurements in aqueous 0.1 mol L –1 NaNO 3 containing varying concentrations of glucose or potassium hydrogen phthalate (KHP). EIS and LSV results revealed that exciton separation efficiency followed the sequence of TF﹥TT﹥FT > FF. Under a constant potential of +0.3 V, steady-state photocurrent profiles were recorded with varying organic compound concentrations. The TF electrode possessed the greatest photocatalytic capacity for oxidizing glucose and KHP, and possessed a KHP anti-poisoning effect.
