| 2 | Engineering a folic acid-decorated ultrasmall gemcitabine nanocarrier for breast cancer therapy: Dual targeting of tumor cells and tumor-associated macrophages显示文摘Combination of passive targeting with active targeting is a promising approach to improve the therapeutic efficacy of nanotherapy.However,most reported polymeric systems have sizes above100 nm,which limits effective extravasation into tumors that are poorly vascularized and have dense stroma.This will,in turn,limit the overall effectiveness of the subsequent uptake by tumor cells via active targeting.In this study,we combined the passive targeting via ultra-small-sized gemcitabine(GEM)-based nanoparticles(NPs)with the active targeting provided by folic acid(FA)conjugation for enhanced dual targeted delivery to tumor cells and tumor-associated macrophages(TAMs).We developed an FAmodified prodrug carrier based on GEM(PGEM)to load doxorubicin(DOX),for co-delivery of GEM and DOX to tumors.The co-delivery system showed small particle size of~10 nm in diameter.The ligand-free and FA-targeted micelles showed comparable drug loading efficiency and a sustained DOX release profile.The FA-conjugated micelles effectively increased DOX uptake in cultured KB cancer cells that express a high level of folate receptor(FR),but no obvious increase was observed in 4T1.2 breast cancer cells that have a low-level expression of FR.Interestingly,in vivo,systemic delivery of FAPGEM/DOX led to enhanced accumulation of the NPs in tumor and drastic reduction of tumor growth in a murine 4T1.2 breast cancer model.Mechanistic study showed that 4T1.2 tumor grown in mice expressed a significantly higher level of FOLR2,which was selectively expressed on TAMs.Thus,targeting of TAM may also contribute to the improved in vivo targeted delivery and therapeutic efficacy. | Pearl Moharil Zhuoya Wan Apurva Pardeshi Jiang Li Haozhe Huang Zhangyi Luo Sanjay Rathod Ziqian Zhang Yuang Chen Bei Zhang Christian A.Fernandez Jingjing Sun Song Li | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 1 |
| 3 | New paleomagnetic evidences of paleogene tectonic rotation of the Qaidam Basin and adjacent region: mechanism and its tectonic implication显示文摘In order to better understand the tectonic evolution of the Qaidam Basin. The authors carried out a paleomagnetic study on 39 samples of 7 sites from Hongsanhan section, northwest Qaidam Basin. Stepwise thermo-demagnetization isolated a high-temperature component (HTC), which passes the fold and reversal tests at 95% confidence level. It suggests that the HTC should be primary. Unblocking temperature (about 685℃) and the experiment of isothermal remanence indicate that hematite is the mainly carrier of the remanence. A tilt-corrected mean direction is: Ds=7.1°, Is=38.5°, α95=7.4°. Corresponding to a palaeopole at, φ=250.1°E, λ=72.0°N, A95=6.8°, Compared with the reference APWP of Eurasia, the sampling area occurred a non-significant rotation (3.4°±5.5°) relative to Eurasia since Eocene due to the affect of left-lateral Altyn fault, Based on the paleomagnetic results of early Cretaceous and Teritary within the Qaidam block, we can obtained the mean clockwise rotations from the samples deposited from 160 Ma to 45 Ma is 24.5°±9.0°, and from 38 Ma to present is -0.5°±7.5° relative to present geomagnetism respectively. | 5 | Adaptive step-size forward advection method for aerosol process simulation显示文摘Outdoor aerosol processes are often associated with disasters and diseases,which threaten human life and health.Outdoor aerosols are afluid system affected by meteorological conditions and three-dimensional complex terrain.Their variable wind speed and direction and complex terrain boundary conditions make simulating advection processes difficult.Based on incompressibleflow conditions,we designed an adaptive time step algorithm for forward advection for the rapid simulation of aerosol processes.The method is based on thefirst-order forward semi-Lagrangian advection method with unconditional mass conservation.Thefirst-order truncated error coefficient function theory generates an adaptive time step to control the accuracy of forward advection.Smoke aerosol simulation experiments in two small outdoor scenes were designed,and the effects of the traditional backward advection and forwardfixed step methods were compared with the algorithm in this study.The proposed simulation method showed improved accuracy compared with the other two methods in experimental scenarios;moreover,compared with those of the traditional backward method,the computation time was significantly reduced and the conservation of mass was significantly improved.Thus,the proposed method is a fast simulation method for outdoor aerosol numerical prediction.KEY POLICY HIGHLIGHTS.The first-order forward semi-Lagrangian method,which requires no iteration and less computation and offers unconditional conservation,was used..The law of truncation error coefficient of thefirst-order forward method was studied and an adaptive step algorithm was designed..Full-size real aerosol experiments in small-scale complex outdoor scenes were conducted for verification and comparison of simulation effects. | Yuang Wu Shuo Liu Bowen Shu Weichao Sun Sheng Wang Hongyang Zhang Chenchen Chen | 2023 | International Journal of Digital Earth2023,16,1: | 0 |
| 6 | Predicting the capacity of perfobond rib shear connector using an ANN model and GSA method显示文摘Due to recent advances in the field of artificial neural networks(ANN)and the global sensitivity analysis(GSA)method,the application of these techniques in structural analysis has become feasible.A connector is an important part of a composite beam,and its shear strength can have a significant impact on structural design.In this paper,the shear performance of perfobond rib shear connectors(PRSCs)is predicted based on the back propagation(BP)ANN model,the Genetic Algorithm(GA)method and GSA method.A database was created using push-out test test and related references,where the input variables were based on different empirical formulas and the output variables were the corresponding shear strengths.The results predicted by the ANN models and empirical equations were compared,and the factors affecting shear strength were examined by the GSA method.The results show that the use of ANN model optimization by GA method has fewer errors compared to the empirical equations.Furthermore,penetrating reinforcement has the greatest sensitivity to shear performance,while the bonding force between steel plate and concrete has the least sensitivity to shear strength. | Guorui SUN Jun SHI Yuang DENG | 2022 | Frontiers of Structural and Civil Engineering2022,16,10: | 0 |
| 7 | Enhancing photodynamic inactivation via tunning spatial constraint on photosensitizer显示文摘The critical factor of spatial constraint,provided by the external confinement(e.g.,matrix),is often overlooked during photodynamic inactivation,despite playing a crucial role in determining the molecular photophysical process and subsequent antipathogen performance.Here,as a proof-of-concept model,we employed two types of polymers with varying interaction energies with dopants to investigate the intrinsic relationship between spatial constraint and the essential excited-state behaviors of doped photosensitizer(4-(2-(5-(4-(diphenylamino)phenyl)thiophen-2-yl)ethyl)-1-methylquinolin-1-ium iodine,TPP).Through experimental investigation and theoretical calculations,we found that TPP tends to remain in the excited state for a shorter dwell time under weaker spatial constraints due to less restricted molecular motion in polyurethane(PU) nanofibers.Consequently,the singlet oxygen(^(1)O_(2)) generated from doped-TPP shows a 9.23-fold enhancement in PU than in the polyvinylchloride(PVC) matrix.Under light irradiation,the PU@TPP nanofiber can efficiently eliminate the coronavirus MHV-A59(≥99.9997%) at a 220,000-fold higher concentration than the infected space.Its antibacterial efficacy has also been demonstrated,with a killing rate of ≥99%. | Chong Li Minghao Han Ji Gao Shuxian Wang Song-Bo Lu Yaxi Li Zhao Liu Chen Zhang Zhaojin Wang Fangliang Wang Yuang Sun Chun Loong Ho Kai Wang Yun Shen Kai Li | 2024 | Science China Chemistry2024,67,2: | 0 |
| 8 | piRT-IFC:Physics-informed real-time impedance flow cytometry for the characterization of cellular intrinsic electrical properties显示文摘Real-time transformation was important for the practical implementation of impedance flow cytometry.The major obstacle was the time-consuming step of translating raw data to cellular intrinsic electrical properties(e.g.,specific membrane capacitance C_(sm) and cytoplasm conductivityσ_(cyto)).Although optimization strategies such as neural network-aided strategies were recently reported to provide an impressive boost to the translation process,simultaneously achieving high speed,accuracy,and generalization capability is still challenging.To this end,we proposed a fast parallel physical fitting solver that could characterize single cells’C_(sm)andσ_(cyto)within 0.62 ms/cell without any data preacquisition or pretraining requirements.We achieved the 27000-fold acceleration without loss of accuracy compared with the traditional solver.Based on the solver,we implemented physics-informed real-time impedance flow cytometry(piRT-IFC),which was able to characterize up to 100,902 cells’C_(sm) andσ_(cyto)within 50 min in a real-time manner.Compared to the fully connected neural network(FCNN)predictor,the proposed real-time solver showed comparable processing speed but higher accuracy.Furthermore,we used a neutrophil degranulation cell model to represent tasks to test unfamiliar samples without data for pretraining.After being treated with cytochalasin B and N-Formyl-Met-Leu-Phe,HL-60 cells underwent dynamic degranulation processes,and we characterized cell’s C_(sm)andσ_(cyto)using piRT-IFC.Compared to the results from our solver,accuracy loss was observed in the results predicted by the FCNN,revealing the advantages of high speed,accuracy,and generalizability of the proposed piRT-IFC. | Xiaofeng Luan Pengbin Liu Di Huang Haiping Zhao Yuang Li Sheng Sun Wenchang Zhang Lingqian Zhang Mingxiao Li Tian Zhi Yang Zhao Chengjun Huang | 2023 | Microsystems & Nanoengineering2023,9,3: | 0 |
