| 1 | A New Species of the Genus Protobothrops(Squamata:Viperidae:Crotalinae) from the Dabie Mountains,Anhui,China显示文摘During a biological survey in July 2011, a pit viper was collected from the Yaoluoping Nature Reserve in the Dabie Mountains, Anhui, China. The pit viper's the total length measured 836 mm with a dorsal ground color of yellowbrown, decorated by 56 dark-brown transverse bands. The 4thsupralabial squama of the new species is separated from subocular by two squamae, and the tip of tail is orange; this combination of characters distinguish it from other species of the genus Protobothrops. | 2 | Prepartum body conditions affect insulin signaling pathways in postpartum adipose tissues in transition dairy cows显示文摘Background: Overconditioned dairy cows are susceptible to excessive lipolysis and increased insulin resistance during the transition period.The associations among body fat reserve,insulin resistance,and lipolysis in adipose tissues(AT) remain to be elucidated.Therefore,this study aimed to investigate whether excessive fat reserves influence the insulin signaling pathway in AT postpartum.Results: Twenty multiparous dairy cows were selected and assigned to one of two groups,according to prepartum body condition score(BCS): Control group(BCS = 3.0–3.5;n = 10) and Overconditioned group(BCS ≥ 4.0;n = 10).Blood samples were collected on days-14,-7,-4,-2,-1,0,1,2,4,7,and 14 relative to parturition.Subcutaneous AT were collected on day 2 following parturition for quantitative real-time polymerase chain reaction and western blot analyses.No differences were observed between the two groups in serum glucose,non-esterified fatty acids,β-hydroxybutyric acid,tumor necrosis factor(TNF)α,insulin,or leptin concentrations during the experimental period.Compared with the control cows,the overconditioned cows had lower serum triglyceride levels and higher adiponectin concentrations.In the AT postpartum,insulin receptor mRNA and protein levels were lower in the overconditioned cows than in the control cows,and no differences were found in glucose transporter 4 mRNA.Compared with the control cows,the overconditioned cows had lower mRNA levels of TNFα and higher mRNA levels of peroxisome proliferator-activated receptor gamma(PPARγ) in AT postpartum.The phosphorylated protein kinase B(AKT) content and phosphorylation rate of AKT were increased in the overconditioned cows compared with the control cows,which suggested that the downstream insulin signaling in AT was affected.Conclusions: In the present study,transition dairy cows with higher BCS did not show more fat mobilization.The changes of insulin signaling pathway in AT postpartum of overconditioned cows may be partly related to the expression of PPARγ and TNFα,and the secretion of adiponectin. | Fanjian Zhang Dan Li Qiong Wu Jian Sun Wenyi Guan Yinxu Hou Yaohong Zhu Jiufeng Wang | 2019 | Journal of Animal Science and Biotechnology2019,10,3: | 2 |
| 3 | Effect of Different Thawing Methods on Post-thaw Motility of Cow Semen in Cryopreservation Straws显示文摘The study was to investigate the effects of different thawing temperatures(5,15,40,75,90 ℃) and times(1- 120 s) on properties of post-thaw cow semen by detecting frozen-thawed semen motility,acrosome integrity and tail membrane integrity,further obtaining the optimal thawing method of straw frozen semen from dairy cow. The results showed that(1) Thawing of the straw frozen semen of dairy cow at 75 ℃ for 3 s yielded the highest semen motility,followed by 40 ℃for 20 s,and the least by low temperature 5 ℃ and room temperature 15 ℃ with a semen motility of 0. 3,moreover thawing at high temperature 90 ℃ was not suitable for large scale production due to the difficult control of the temperature;(2) The acrosome intact rate and plasma membrane integrity rate of semens thawed at90 ℃ were remarkably lower than that at 40 ℃ and 75 ℃ significantly(P < 0. 05),while its semen malformation rate was significantly higher than that at 40 ℃and 75 ℃(P < 0. 05);(3) The Survival time of semens at 37 ℃ varied largely among different thawing temperature,in detail by 40 ℃ > 75 ℃ > 90 ℃. In practice,the thawing method of straw frozen semen of dairy cow should be selected according to the specific circumstance and inseminated immediately,with the recommended condition of thawing at 75 ℃ for 3 s. | 4 | Genetic structure of the oriental white stork (Ciconia boyciana): implications for a breeding colony in a non-breeding area显示文摘The oriental white stork (Ciconia boyciana) is a threatened species, and their numbers are still in decline due tohabitat loss and poaching. China is a breeding and main wintering area for this animal and in recent years someindividuals have been found breeding in wintering areas and at some stopover sites. These new breeding coloniesare an exciting sign, however, little is understood of the genetic structure of this species. Based on the analysis ofa 463-bp mitochondrial DNA (mtDNA) control region, we investigated the genetic structure and genetic diversity of66 wild oriental white storks from a Chinese population. We analyzed the sequences of 66 storks obtained in thisstudy and the data of 17 storks from a Japanese population. Thirty-seven different haplotypes were detected amongthe 83 samples. An analysis of molecular variance showed a significant population subdivision between the twopopulations (FST = 0.316, P < 0.05). However, the phylogenetic analysis revealed that the samples from the differentpopulations did not form separate clusters and that there were genetic exchanges between the two populations.Compared with the Japanese population, the Chinese population had a relatively higher genetic diversity with ahaplotype diversity (h ± SD) of 0.953 ± 0.013 and a nucleotide diversity (π ± SD) of 0.013 ± 0.007. The high haplotypediversity and low nucleotide diversity indicate that this population might be in a rapidly increasing period from asmall effective population. A neighbor-joining tree analysis indicated that genetic exchange had occurred betweenthe newly arisen southern breeding colony and the northern breeding colony wintering in the middle and lowerYangtze River floodplain. | 5 | Effects of Transport,Storage Temperature and Time on Parthenogenetically Developmental Competence of Sheep Oocyte显示文摘This study was to investigate the effects of transport,storage temperature and time on parthenogenetical developmental competence of sheep oocyte. The ovaries were collected and randomly immersed in physiological saline at( 10- 15) ℃,( 20- 25) ℃ and( 30- 35) ℃ and transported to the laboratory for culture.They were divided into three groups randomly in an optimized transport temperature,kept in physiological saline at 4 ℃,( 14- 18) ℃ and( 25- 30) ℃ respectively; 15- 17 h later,the ovaries were cutured for the maturation and parthenogenetic activation of oocytes. The results showed that the optimal transport temperature of ovaries was 20- 25 ℃,when the maturation rate and blastocysts rate reached 67. 44% and 35. 93% respectively. Incubating sheep ovaries at( 14- 18) ℃for( 15- 17) h did not reduce the maturation rate( 61. 81%) and blastocysts rate( 29. 03%) of oocyte significantly. However,incubating sheep ovaries at 4 ℃or( 25- 30) ℃ significantly reduced the maturation rate( 41. 90%,18. 40%) and cleavage rate( 9. 09%,13. 04%),and the ovaries could not develop into blastocysts at these two temperature ranges. | 6 | Effects of Storage Temperature on the Effective Activity of Straw Frozen Semen after Thawing显示文摘The aim was to discuss the optimal storage environment and proper insemination time after thawing of 0. 25 m L straw frozen semen. Straw frozen semen was thawed at 40 ℃ for 20 s,and then stored at 0- 4 ℃,14- 16 ℃,25- 27 ℃ for 2,4,6,8 and 10 h,respectively. The sperm motility was detected. After thawing,semen was stored at 0- 4 ℃ and 14- 16 ℃ for 10 h. Their sperm motilities( 0. 434 ± 0. 016 7 and 0. 423 ± 0. 019 6) had no significant differences( P> 0. 05) with initial thawing motility( 0. 441 ± 0. 030). Sperm motility reduced as the storage time prolonged at 25- 27 ℃. Sperm motility after 6 h had significant differences with that of initial thawing motility( P < 0. 05),and sperm motilities after 8 and 10 h showed extremely significant differences( P < 0. 01).Thus,sperm motility after thawing was still very high after stored at 0- 4 ℃ and 14- 16 ℃ within 10 h,which met the requirements for insemination.
