| 4 | Transcription factors AS1 and AS2 interact with LHP1 to repress KNOX genes in Arabidopsis显示文摘Polycomb group proteins are important repressors of numerous genes in higher eukaryotes. However, the mechanism by which Polycomb group proteins are recruited to specific genes is poorly understood. In Arabidopsis, LIKE HETEROCHROMATIN PROTEIN 1(LHP1), also known as TERMINAL FLOWER 2, was originally proposed as a subunit of polycomb repressive complex 1(PRC1) that could bind the tri-methylated lysine 27 of histone H3(H3K27me3) established by the PRC2. In this work, we show that LHP1 mainly functions with PRC2 to establish H3K27me3, but not with PRC1 to catalyze monoubiquitination at lysine 119 of histone H2 A. Our results show that complexes of the transcription factors ASYMMETRIC LEAVES 1(AS1) and AS2 could help to establish the H3K27me3 modification at the chromatin regions of Class-I KNOTTED1-like homeobox(KNOX) genes BREVIPEDICELLUS andKNAT2 via direct interactions with LHP1. | 5 | MicroRNA-132 in the Adult Dentate Gyrus is Involved in Opioid Addiction Via Modifying the Differentiation of Neural Stem Cells显示文摘MicroRNA-132(miR-132), a small RNA that regulates gene expression, is known to promote neurogenesis in the embryonic nervous system and adult brain.Although exposure to psychoactive substances can increase miR-132 expression in cultured neural stem cells(NSCs)and the adult brain of rodents, little is known about its role in opioid addiction. So, we set out to determine the effect of miR-132 on differentiation of the NSCs and whether this effect is involved in opioid addiction using the rat morphine self-administration(MSA) model. We found that miR-132 overexpression enhanced the differentiation of NSCs in vivo and in vitro. Similarly, speci?c overexpression of miR-132 in NSCs of the adult hippocampal dentate gyrus(DG) during the acquisition stage of MSA potentiated morphine-seeking behavior. | 6 | Biofilm-inspired adhesive and antibacterial hydrogel with tough tissue integration performance for sealing hemostasis and wound healing显示文摘Uncontrolled bleeding and infection can cause significant increases in mortalities.Hydrogel sealants have attracted extensive attention for their ability to control bleeding.However,because interfacial water is a formidable barrier to strong surface bonding,a challenge remains in finding a product that offers robust tissue adhesion combined with anti-infection properties.Inspired by the strong adhesive mechanism of biofilm and mussels,we report a novel dual bionic adhesive hydrogel(DBAH)based on chitosan grafted with methacrylate(CS-MA),dopamine(DA),and N-hydroxymethyl acrylamide(NMA)via a facile radical polymerization process.CS-MA and DA were simultaneously included in the adhesive polymer for imitating the two key adhesive components:polysaccharide intercellular adhesin(PIA)of staphylococci biofilm and 3,4-dihydroxy-L-phenylalanine(Dopa)of mussel foot protein,respectively.DBAH presented strong adhesion at 34 kPa even upon three cycles of full immersion in water and was able to withstand up to 168 mm Hg blood pressure,which is significantly higher than the 60–160 mm Hg measured in most clinical settings.Most importantly,these hydrogels presented outstanding hemostatic capability under wet and dynamic in vivo movements while displaying excellent antibacterial properties and biocompatibility.Therefore,DBAH represents a promising class of biomaterials for high-efficiency hemostasis and wound healing. | Wei Han Bo Zhou Kai Yang Xin Xiong Shifang Luan Yu Wang Zheng Xu Peng Lei Zhengshan Luo Jian Gao Yijing Zhan Guopu Chen Lei Liang Rui Wang Sha Li Hong Xu | 2020 | Bioactive Materials2020,5,4: | 6 |

| 8 | Ethylenediurea(EDU) pretreatment alleviated the adverse effects of elevated O3 on Populus alba 'Berolinensis' in an urban area显示文摘Ethylenediurea(EDU)has been used as a chemical protectant against ozone(O3).However,its protective effect and physiological mechanisms are still uncertain.The present study aimed to investigate the changes of foliar visible injury,physiological characteristics and emission rates of volatile organic compounds(VOCs)in one-year-old Populus alba'Berolinensis'saplings pretreated with EDU and exposed to elevated O3(EO,120μg/m3).The results showed that foliar visible injury symptoms under EO were significantly alleviated in plants with EDU application(p<0.05).Under EO,net photosynthetic rate,the maximum photochemical efficiency of PSII and the photochemical efficiency of PSII of plants pretreated with 300 and600 mg/L EDU were similar to unexposed controls and significantly higher compared to EOstressed plants without EDU pretreatment,respectively.Malondialdehyde content was highest in EO without EDU and decreased significantly by 14.9%and 21.3%with 300 and600 mg/L EDU pretreatment,respectively.EDU pretreatment alone increased superoxide dismutase activity by 10-fold in unexposed plants with further increases of 88.4%and 37.5%in EO plants pretreated with 300 and 600 mg/L EDU pretreatment,respectively(p<0.05).Abscisic acid content declined under EO relative to unexposed controls with the effect partially reversed by EDU pretreatments.Similarly,VOCs emission rate declined under EO relative to unexposed plants with a recovery of emission rate observed with 300 and 600 mg/L EDU pretreatment.These findings provided significant evidence that EDU exerted a beneficial effect and protection on the tested plants against O3 stress. | Sheng Xu Xingyuan He Kent Burkey Wei Chen Pin Li Yan Li Bo Li Yijing Wang | 2019 | Journal of Environmental Sciences2019,31,10: | 4 |
| 9 | Biochemical Composition and Nutritional Value of Different Shell Color Strains of Pacific Oyster Crassostrea gigas显示文摘The shell color of Pacific oyster(Crassostrea gigas) is a desirable trait; but the nutritional studies on C. gigas with different shell colors have not been conducted. Through successive selective breeding, five shell color strains of black(B), purple(P), orange(O), golden(G) and white(W) C. gigas have been developed. The aim of this study was to evaluate the chemical composition and nutritional value of five shell color strains and one commercial population with a common color. The biochemical composition including moisture, total protein, glycogen, ash, total fat, fatty acids(FA), amino acids and minerals was detected. The results indicated that the protein(50.76%–56.57%) was the major component. The content of glycogen showed a significant difference between orange shell and golden shell strains, as well as between commercial population and golden shell strain. In addition, all shell color strains contained a large amount of essential amino acids(12.20–14.15 g(100 g)^(-1)), of them leucine(2.81–3.29 g(100 g)^(-1)) and lysine(2.79–3.28 g(100 g)^(-1)) were predominant. The oysters were rich in polyunsaturated fatty acids(42.26%–45.24% of total fatty acid) with high levels of DHA(18.53%–21.16% of total fatty acid) and EPA(17.23%–18.68% of total fatty acid). Significant differences of mineral contents(Mg, Zn, Fe and Cu) were identified among the six populations. These results indicated that C. gigas with different shell colors presented rich nutritional value with high protein, glycogen, essential amino acids and polyunsaturated fatty acids. A pair of primers was designed with Oligo6.0 and used to amplify PLF N gene. It was in accordance with the characters of translational fusions from gene and expression vector plasmid. A 1 077 bp fragment of the gene from PLF was cloned from mammary gland tissue of the lactating sow on the third day by RT-PCR;the gene was connected with the vector plasmid pPG612.1 and transformed into the host strain JM109. The recombinant expression vector plasmid pPG612-PLF N was created and identified by using plasmid extraction,PCR,restriction enzyme digestion and sequence analysis. The recombinant plasmid was transformed into Lactobacillus casei ATCC393,Lactobacillus plantarum KLDS 1.0344,Lactobacillus paracasei KLDS 1.0652 and Lactobacillus pentosus KLDS1.0413 by electroporation,and produced the recombinant strains of pPG612-PLF N /L. casei,pPG612-PLF N /L. plantarum,pPG612-PLF N /L. paracasei and pPG612-PLF N /L. pentosus,respectively. The results indicated that PLF N gene had inserted into the expression vectors and achieved multiple Lactobacillus expression systems. | 15 | Ecological selection of bacterial taxa with larger genome sizes in response to polycyclic aromatic hydrocarbons stress显示文摘Polycyclic aromatic hydrocarbons(PAHs)are ubiquitous priority pollutants that cause great damage to the natural environment and health.Average genome size in a community is critical for shedding light on microbiome's functional response to pollution stress within an environment.Here,microcosms under different concentrations were performed to evaluate the selection of PAHs stress on the average genome size in a community.We found the distinct communities of significantly larger genome size with the increase of PAHs concentration gradients in soils,and consistent trends were discovered in soils at different latitudes.The abundance of Proteobacteria and Deinococcus-Thermus with relatively larger genomes increased along with PAHs stress and well adapted to polluted environments.In contrast,the abundance of Patescibacteria with a highly streamlined and smaller genome decreased,implying complex interactions between environmental selection and functional fitness resulted in bacteria with larger genomes becoming more abundant.Moreover,we confirmed the increased capacity for horizontal transfer of degrading genes between communities by showing an increased connection number per node positively related to the nid A gene along the concentration gradients in the co-occurrence network.Our findings suggest PAHs tend to select bacterial taxa with larger genome sizes,with significant consequences for community stability and potential biodegradation strategies. | Yuzhu Dong Shanghua Wu Haonan Fan Xianglong Li Yijing Li Shengjun Xu Zhihui Bai Xuliang Zhuang | 2022 | Journal of Environmental Sciences2022,34,2: | 2 |
| 16 | Distinct Roles of Dopamine Receptors in the Lateral Thalamus in a Rat Model of Decisional Impulsivity显示文摘The thalamus and central dopamine signaling have been shown to play important roles in high-level cognitive processes including impulsivity. However, little is known about the role of dopamine receptors in the thalamus in decisional impulsivity. In the present study,rats were tested using a delay discounting task and divided into three groups: high impulsivity(HI), medium impulsivity(MI), and low impulsivity(LI). Subsequent in vivo voxel-based magnetic resonance imaging revealed that the HI rats displayed a markedly reduced density of gray matter in the lateral thalamus compared with the LI rats. In the MI rats, the dopamine D1 receptor antagonist SCH23390 or the D2 receptor antagonist eticlopride was microinjected into the lateral thalamus. SCH23390 significantly decreased their choice of a large, delayed reward and increased their omission of lever presses. In contrast,eticlopride increased the choice of a large, delayed reward but had no effect on the omissions. Together, our results indicate that the lateral thalamus is involved in decisional impulsivity, and dopamine D1 and D2 receptors in the lateral thalamus have distinct effects on decisional impulsive behaviors in rats.
